    Parasitic Energy Loss in the LEP Superconducting Cavities

    The energy loss of bunches in the LEP superconducting (SC) cavities has been determined by measuring the closed orbit as a function of current with the beam position monitors located at finite dispersion. This method has already been used in earlier experiments to determine the distribution of the longitudinal impedance of different parts of LEP. In the present experiment the energy loss in two straight sections, containing only SC cavities, was compared with that in sections having both copper cavities and SC cavities. The results confirm the impedance calculations for the two types of cavities. The accuracy of the measurements was considerably improved by determining simultaneously the orbits of bunches with different currents. At the same time with these beam-based impedance measurements, the power dissipation was observed directly by local temperature monitors in different elements: the inter-cavity bellows inside the cryostat, the warm intermodule bellows, and Ferrite absorbers which were installed in two places to reduce the energy leaking out of cavities. These observations were correlated with the change of cryogenics power consumption, and showed an unexpected dependence of energy loss on beam energy

    Dispersion of carbon nanotubes in polyamide 6 for microinjection moulding

    The focus of this study was to investigate the dispersion state of pure and functionalized carbon nanotubes in polyamide 6, on composites prepared by twin-screw extrusion and then processed by microinjection moulding. Nanocomposites were prepared with different carbonvnanotube compositions, with and without functionalization. The nanotubes were functionalized by the 1,3-dipolar cycloaddition reaction. The dispersion of the carbon nanotube agglomerates was quantified using optical microscopy and image analysis. The effect of functionalization on the polyamide 6/carbon nanotube interface, the nanocomposite morphology and the mechanical and electrical properties were studied. It was observed that the microinjected composites with functionalized carbon nanotubes presented improved dispersion, with smaller carbon nanotube agglomerate area ratio compared to the composites with pure nanotubes. The functionalized nanotubes showed better adhesion to polyamide 6 compared to pure nanotubes, as observed by scanning electron microscopy. The incorporation of carbon nanotubes considerably improved the mechanical properties. The effect of high polymer shear rate on carbon nanotube alignment during microinjection moulding was assessed by comparing the electrical resistivity of the composite after extrusion and after microinjection moulding, through the thickness and along the flow direction.     Near-infrared molecular imaging of tumors via chemokine receptors CXCR4 and CXCR7

    The chemokine CXCL12/SDF-1 and its receptors CXCR4 and CXCR7 play a major role in tumor invasion, proliferation and metastasis. Since both receptors are overexpressed on distinct tumor cells and on the tumor vasculature, we evaluated their potential as targets for detection of cancers by molecular imaging. We synthesized conjugates of CXCL12 and the near-infrared (NIR) fluorescent dye IRDye®800CW, tested their selectivity, sensitivity and biological activity in vitro and their feasibility to visualize tumors in vivo. Purified CXCL12-conjugates detected in vitro as low as 500 A764 human glioma cells or MCF-7 breast cancer cells that express CXCR7 alone or together with CXCR4. Binding was time- and concentration-dependent, and the label could be competitively displaced by the native peptide. Control conjugates with bovine serum albumin or lactalbumin failed to label the cells. In mice, the conjugate distributed rapidly. After 1–92 h, subcutaneous tumors of human MCF-7 and A764 cells in immunodeficient mice were detected with high sensitivity. Background was observed in particular in liver within the first 24 h, but also skull and hind limbs yielded some background. Overall, fluorescent CXCL12-conjugates are sensitive and selective probes to detect solid and metastatic tumors by targeting tumor cells and tumor vasculature

    Cultivation of a novel cold-adapted nitrite oxidizing betaproteobacterium from the Siberian Arctic

    Permafrost-affected soils of the Siberian Arctic were investigated with regard to identification of nitrite oxidizing bacteria active at low temperature. Analysis of the fatty acid profiles of enrichment cultures grown at 4°C, 10°C and 17°C revealed a pattern that was different from that of known nitrite oxidizers but was similar to fatty acid profiles of Betaproteobacteria. Electron microscopy of two enrichment cultures grown at 10°C showed prevalent cells with a conspicuous ultrastructure. Sequence analysis of the 16S rRNA genes allocated the organisms to a so far uncultivated cluster of the Betaproteobacteria, with Gallionella ferruginea as next related taxonomically described organism. The results demonstrate that a novel genus of chemolithoautotrophic nitrite oxidizing bacteria is present in polygonal tundra soils and can be enriched at low temperatures up to 17°C. Cloned sequences with high sequence similarities were previously reported from mesophilic habitats like activated sludge and therefore an involvement of this taxon in nitrite oxidation in nonarctic habitats is suggested. The presented culture will provide an opportunity to correlate nitrification with nonidentified environmental clones in moderate habitats and give insights into mechanisms of cold adaptation. We propose provisional classification of the novel nitrite oxidizing bacterium as 'Candidatus Nitrotoga arctica'

    Barotropic tides in the Northeast Atlantic inferred from moored current meter data

    Current data obtained from 7 moorings in the Northeast Atlantic in the course of many years are analysed with respect to semi-diurnal barotropic and baroclinic tides and diurnal barotropic tides. For semi-diurnal tides M2 and S2 the energy distribution is usually dominated by the barotropic mode; only in a few cases does the first-order baroclinic mode contain higher energy. Barotropic tidal ellipse orientations are found to be consistent with results from earlier tide gauge observations in this area. Significant deviations occur, however, in amplitudes. Results for the diurnal component K1 are also presented. With few exceptions, tides are found to be progressive waves in this area. The current ellipse pattern is similar to results obtained indirectly by Cartwright, Edden, Spencer et al. [1980] from tide gauge observations

    Transcription Factor SP4 Is a Susceptibility Gene for Bipolar Disorder

    The Sp4 transcription factor plays a critical role for both development and function of mouse hippocampus. Reduced expression of the mouse Sp4 gene results in a variety of behavioral abnormalities relevant to human psychiatric disorders. The human SP4 gene is therefore examined for its association with both bipolar disorder and schizophrenia in European Caucasian and Chinese populations respectively. Out of ten SNPs selected from human SP4 genomic locus, four displayed significant association with bipolar disorder in European Caucasian families (rs12668354, p = 0.022; rs12673091, p = 0.0005; rs3735440, p = 0.019; rs11974306, p = 0.018). To replicate the genetic association, the same set of SNPs was examined in a Chinese bipolar case control sample. Four SNPs displayed significant association (rs40245, p = 0.009; rs12673091, p = 0.002; rs1018954, p = 0.001; rs3735440, p = 0.029), and two of them (rs12673091, rs3735440) were shared with positive SNPs from European Caucasian families. Considering the genetic overlap between bipolar disorder and schizophrenia, we extended our studies in Chinese trios families for schizophrenia. The SNP7 (rs12673091, p = 0.012) also displayed a significant association. The SNP7 (rs12673091) was therefore significantly associated in all three samples, and shared the same susceptibility allele (A) across all three samples. On the other hand, we found a gene dosage effect for mouse Sp4 gene in the modulation of sensorimotor gating, a putative endophenotype for both schizophrenia and bipolar disorder. The deficient sensorimotor gating in Sp4 hypomorphic mice was partially reversed by the administration of dopamine D2 antagonist or mood stabilizers. Both human genetic and mouse pharmacogenetic studies support Sp4 gene as a susceptibility gene for bipolar disorder or schizophrenia. The studies on the role of Sp4 gene in hippocampal development may provide novel insights for the contribution of hippocampal abnormalities in these psychiatric disorders

    “Microbiota, symbiosis and individuality summer school” meeting report

    How does microbiota research impact our understanding of biological individuality? We summarize the interdisciplinary summer school on “Microbiota, symbiosis and individuality: conceptual and philosophical issues” (July 2019), which was supported by a European Research Council starting grant project “Immunity, DEvelopment, and the Microbiota” (IDEM). The summer school centered around interdisciplinary group work on four facets of microbiota research: holobionts, individuality, causation, and human health. The conceptual discussion of cutting-edge empirical research provided new insights into microbiota and highlights the value of incorporating into meetings experts from other disciplines, such as philosophy and history of science
