    A Research on Mathematical Thinking Skills: Mathematical Thinking Skills of Athletes in Individual and Team Sports

    The aim of this research is to examine the mathematical thinking skills of licensed athletes engaged in individual and team sports. The research is designed as a survey model. The sample of the research is composed of 59 female and 170 male licensed athletes (n = 229) and (aged 14 to 52) licensed who do the sports of shooting, billiards, archery, tennis, basketball, football, volleyball in various clubs in Turkey. The "Mathematical Thinking Scale" developed by Ersoy (2012) has been employed in the research. Individual sports athletes are more likely to have higher mathematical thinking scores than team athletes. In sports types; those who play billiards and archery have higher scores of mathematical thinking skills compared to other sports types. According to the type of sports the lowest scores of thinking skills were obtained by basketball players. These differences are valid for higher-level thinking tendencies, reasoning, mathematical thinking skills and problem-solving skills, which are sub-dimensions of the mathematical thinking scale

    Optimisation of bacillus amyloliquefaciens fe-k1 extracellular peptidase production by response surface methodology

    In this study, it was aimed to optimise the extracellular peptidase production of Bacillus amyloliquefaciens FE-K1, previously isolated from ropy wholemeal bread, by using response surface methodology (RSM) based on central composite design (CCD). The temperature (20-45°C), initial pH of the enzyme production medium (pH 5-9) and inoculation level (1-5%, v/v) were used as the factors for RSM, and the fermentation time was determined for each trial separately. Results showed that the optimum peptidase production occurred at 33.4°C, pH 6.62 and 2.3% inoculation. It was determined that the fermentation time was only 7h, the crude enzyme had a peptidase activity of 49.17U/mL and a specific activity of 504.77U/mg under the optimised conditions.Bu çalışmada, daha önce sünmüş ekmekten izole edilmiş olan Bacillus amyloliquefaciens FE-K1’in ekstraselüler peptidaz üretiminin, merkezi kompozit tasarıma (MKT) dayalı yanıt yüzey yöntemi (YYY) ile optimize edilmesi amaçlanmıştır. Yanıt yüzey yönteminde sıcaklık (20-45°C), enzim üretim ortamının başlangıç pH değeri (pH 5-9) ve inokülasyon seviyesi (%1-5, v/v) faktör olarak kullanılmış, fermentasyon süresi her deneme için ayrı ayrı belirlenmiştir. Sonuçlar optimum peptidaz üretiminin 33,4°C, pH 6,62 ve %2,3 inokülasyon seviyesinde elde edildiğini göstermiştir. Optimum koşullar altında fermentasyon süresinin sadece 7 saat, ham enzimin aktivitesinin 49,17U/mL, spesifik aktivitesinin ise 504,77U/mg olduğu tespit edilmiştir

    NSAIDs and butyrate sensitize a human colorectal cancer cell line to TNF-α and Fas ligation: the role of reactive oxygen species

    AbstractThe nonsteroidal antiinflammatory drugs (NSAIDs) indomethacin and salicylic acid and the short chain fatty acid butyrate are effective colon cancer chemopreventive agents that increase reactive oxygen species (ROS) generation in colon cancer cells. Here we demonstrate that these agents sensitize the normally resistant human HT-29 colon cancer cell line to apoptosis induced by TNF-α or a Fas ligating antibody. The role of ROS in this sensitization is supported by the finding that direct exposure of the cells to H2O2 is sufficient for sensitization. Neither TNF-α nor Fas ligation alter basal or chemopreventive agent-activated ROS generation, suggesting that the death ligands and chemopreventive agents act in a complementary fashion. The dual chemopreventive agent/death ligand treatments do not increase Fas, TNF receptor 1, Bak or c-myc expression (although salicylic acid moderately induces of Fas expression). Cell death does correlate with alterations in NF-κB activity: the NSAIDs, butyrate and H2O2 enhance c-Rel complex formation by TNF-α and provide an overall enhancement of NF-κB activation by Fas. The antioxidant N-acetylcysteine (NAC) blocks cell death and NF-κB activation induced by Fas ligation, suggesting a potential role for NF-κB in Fas-induced apoptosis in these cells. The effects of NAC on TNF-α-induced cell death are more complex, with NAC being marginally protective and itself enhancing the formation of c-Rel containing complexes at higher concentrations (25 mM). The influence of NSAIDs and butyrate on ROS generation and death ligand sensitivity may be relevant to their ability to suppress colon carcinogenesis

    Examination of Socialization Level of University Students Engaged in Sports Activities According to Their Locus of Control

    In this study, the university students who participate in sports have been examined in terms of their socialization relative to the participation in sport activities and the locus of control. Students are thought to be engaged in many activities in addition to their lessons during their student tenure at higher education institutions. Their participation in such activities is examined to find out whether the students take part in the mentioned activities on their own accord, or through the manipulation of others. The sample of this research consisted of students who studied at the Marmara, Istanbul Technical (ITU), and Haliç universities (n=174). Data collection was based on Şahan’s (2007) scale of “Role of the Sportive activities in Socialization Process of the University Students” (r= 0.77, r= 0.84) and Dağ’s (2002) “Scale on Locus of Control” (α= .92). The obtained results demonstrate significant differences between the items in the socialization scales and the characteristics of locus of control of the university students. Significant differences have been detected between the items of socialization scales and university students’ interest (i.e., amateur, spectator, no interest) in sports. The only significant difference in terms of gender was determined in sport and socialization skills

    Partial Purification and Characterization of An Extracellular Metallopeptidase Produced by Bacillus Amyloliquefaciens Fe-K1

    DergiPark: 647525trkjnatBu çalışmanın amacı, sünmüş ekmeklerden izole edilen Bacillus amyloliquefaciens (Fukumoto) (suş FE-K1) ile elde edilen peptidazı saflaştırmak ve karakterize etmektir. Peptidazlar ham enzim çözeltisinden afinite kromatografisi ile % 25 verim ve 1,53 saflaştırma katsayısı ile saflaştırılmıştır. Kısmi olarak saflaştırılmış peptidaz (PPPaz) çözeltisinin optimum pH değeri 7,5 olarak tespit edilmiş olup, pH 7,5-8,0 aralığında peptidaz, 37°C’de 2 saat inkübasyonun ardından başlangıç aktivitesini yaklaşık % 90 oranında korumuştur. PPPaz’ın optimum sıcaklığı 60°C’dir. PPPaz’ın yaklaşık molekül ağırlığı 36 kDa olarak belirlenmiştir. PPPaz’ın O-FEN ve EDTA varlığında inaktive olması, enzimin metallopeptidaz olduğunu göstermiştir. Ayrıca 5 mM K+1 ve 5 mM Mn+2, enzimin aktivitesini sırasıyla % 4 ve % 6,15 oranında artırmıştır. Hg+2, Fe+3 ve SDS (% 0,1-1,0 w/v) varlığı enzimin inaktivasyonuna neden olurken, % 0,1-1,0 (v/v) Triton X-100, Tween 20 ve Tween 80; ve % 1-20 (v/v) ksilen, etanol, aseton ve asetonitril varlığında enzim aktivitesini büyük ölçüde korumuştur. PPPaz’ın karakterizasyonu, enzimin bazı organik çözücü ve yüzey aktif maddelerle ile uyumlu nötr bir serin metalopeptidaz olduğunu ortaya çıkarmıştır.The aim of this study was to purify and characterize the peptidase of Bacillus amyloliquefaciens (Fukumoto) (strain FE-K1) isolated from ropey bread. Peptidases were purified from crude enzyme solution by affinity chromatography with an efficiency of 25 % and a purification coefficient of 1.53. The optimum pH of partially purified peptidase (PPPase) solution was determined as 7.5 and the peptidases retained approximately 90 % of their initial activity in the pH range 7.0-8.5 following incubation at 37°C for 2 h. The optimum temperature for the PPPase was 60°C. The approximate molecular weight of the PPPase was determined as 36 kDa. Inactivation of the PPPase in the presence of O-FEN and EDTA showed them to be metallopeptidases and 5 mM of K+1 and 5 mM of Mn+2 ions increased the enzyme activity by 4 % and 6.15 %, respectively. The presence of Hg+2, Fe+3 and SDS (0.1-1.0 % w/v) caused inactivation whereas the enzyme retained most of its activity in the presence of 0.1-1.0 % (v/v) Triton X-100, Tween 20 and Tween 80 and 1-20 % (v/v) xylene, ethanol, acetone and acetonitrile. Characterization of the PPPase revealed the enzyme as a neutral serine metallopeptidase compatible with some organic solvents and surfactants

    COMPARISON OF KNOWLEDGE LEVELS OF SWIMMING COACHES ABOUT DOPING AND ERGOGENIC AIDS

    Most athletes consider their coaches as the most important source of information on nutrition and doping. The aim of this study was to determine the level of knowledge of swimming coaches about doping and ergogenic aids. Method: This study consisted of 148 randomly selected swimming coaches educated in the development Seminar of Swimming Coaches of Rize Ministry of Youth and Sports between April 23-27, 2019. The survey prepared by taking expert opinion. It totally consists of 19 items, 9 of which aim to determine demographic information and 10 of them to determine doping knowledge level. Frequency (f), percentage (%), chi-square test, independent sample t-test and one-way analysis of variance (ANOVA) used for data analysis. Results and discussion: According to the results of the study, when the participants' coaching experience and degree of coaching certificate were compared with the other variables, a statistically significant difference was found (p <0.05) but there was no significant difference between the other variables (p> 0.05). According to the results of the study, it is unclear to what extent the participants who have more coaching experience transfer information to their athletes despite the high level of knowledge about doping and ergogenic aids. In addition, it is thought that the education given in doping and ergogenic aid in universities is insufficient.  Article visualizations
