Detailed diagnostics of an X-ray flare in the single giant HR 9024
We analyze a 96 ks Chandra/HETGS observation of the single G-type giant HR
9024. The high flux allows us to examine spectral line and continuum
diagnostics at high temporal resolution, to derive plasma parameters. A
time-dependent 1D hydrodynamic model of a loop with half-length cm (), cross-section radius
cm, with a heat pulse of 15 ks and ~erg cm s
deposited at the loop footpoints, satisfactorily reproduces the observed
evolution of temperature and emission measure, derived from the analysis of the
strong continuum emission. For the first time we can compare predictions from
the hydrodynamic model with single spectral features, other than with global
spectral properties. We find that the model closely matches the observed line
emission, especially for the hot ( K) plasma emission of the FeXXV
complex at \AA. The model loop has and aspect
ratio as typically derived for flares observed in active stellar
coronae, suggesting that the underlying physics is the same for these very
dynamic and extreme phenomena in stellar coronae independently on stellar
parameters and evolutionary stage.Comment: 26 pages. Accepted for publication on the Astrophysical Journa
Spectroscopic Study of IRAS 19285+0517(PDS 100): A Rapidly Rotating Li-Rich K Giant
We report on photometry and high-resolution spectroscopy for IRAS 19285+0517.
The spectral energy distribution based on visible and near-IR photometry and
far-IR fluxes shows that the star is surrounded by dust at a temperature of
250 K. Spectral line analysis shows that the star is a K
giant with a projected rotational velocity = 9 2 km s.
We determined the atmospheric parameters: = 4500 K, log =
2.5, = 1.5 km s, and [Fe/H] = 0.14 dex. The LTE abundance
analysis shows that the star is Li-rich (log (Li) = 2.50.15),
but with essentially normal C, N, and O, and metal abundances. Spectral
synthesis of molecular CN lines yields the carbon isotopic ratio
C/C = 9 3, a signature of post-main sequence evolution and
dredge-up on the RGB. Analysis of the Li resonance line at 6707 \AA for
different ratios Li/Li shows that the Li profile can be fitted best
with a predicted profile for pure Li. Far-IR excess, large Li abundance,
and rapid rotation suggest that a planet has been swallowed or, perhaps, that
an instability in the RGB outer layers triggered a sudden enrichment of Li and
caused mass-loss.Comment: To appear in AJ; 40 pages, 9 figure
Sucrose breakdown within guard cells provides substrates for glycolysis and glutamine biosynthesis during light- induced stomatal opening
Sucrose has long been thought to play an osmolytic role in stomatal opening. However, recent evidence supports the idea that the role of sucrose in this process is primarily energetic. Here we used a combination of stomatal aperture assays and kinetic [U- 13 C]-sucrose isotope labelling experiments to confirm that sucrose is degraded during light-induced stomatal opening and to define the fate of the C released from sucrose breakdown. We additionally show that addition of sucrose to the medium did not enhance light- induced stomatal opening. The isotope experiment showed a consistent 13 C enrichment in fructose and glu- cose, indicating that during light-induced stomatal opening sucrose is indeed degraded. We also observed a clear 13 C enrichment in glutamate and glutamine (Gln), suggesting a concerted activation of sucrose degra- dation, glycolysis and the tricarboxylic acid cycle. This is in contrast to the situation for Gln biosynthesis in leaves under light, which has been demonstrated to rely on previously stored C. Our results thus collectively allow us to redraw current models concerning the influence of sucrose during light-induced stomatal open- ing, in which, instead of being accumulated, sucrose is degraded providing C skeletons for Gln biosynthesis
Medulloblastoma has a global impact on health related quality of life: Findings from an international cohort.
BackgroundUnderstanding the global impact of medulloblastoma on health related quality of life (HRQL) is critical to characterizing the broad impact of this disease and realizing the benefits of modern treatments. We evaluated HRQL in an international cohort of pediatric medulloblastoma patients.MethodsSeventy-six patients were selected from 10 sites across North America, Europe, and Asia, who participated in the Medulloblastoma Advanced Genomics International Consortium (MAGIC). The Health Utilities Index (HUI) was administered to patients and/or parents at each site. Responses were used to determine overall HRQL and attributes (ie specific subdomains). The impact of various demographic and medical variables on HRQL was considered-including molecular subgroup.ResultsThe majority of patients reported having moderate or severe overall burden of morbidity for both the HUI2 and HUI3 (HUI2 = 60%; HUI3 = 72.1%) when proxy-assessed. Self-care in the HUI2 was rated as higher (ie better outcome) for patients from Western versus Eastern sites, P = .02. Patients with nonmetastatic status had higher values (ie better outcomes) for the HUI3 hearing, HUI3 pain, and HUI2 pain, all P < .05. Patients treated with a gross total resection also had better outcomes for the HUI3 hearing (P = .04). However, those who underwent a gross total resection reported having worse outcomes on the HUI3 vision (P = .02). No differences in HRQL were evident as a function of subgroup.ConclusionsBy examining an international sample of survivors, we characterized the worldwide impact of medulloblastoma. This is a critical first step in developing global standards for evaluating long-term outcomes
Modifications in Organic Acid Profiles During Fruit Development and Ripening: Correlation or Causation?
The pivotal role of phytohormones during fruit development and ripening is considered established knowledge in plant biology. Perhaps less well-known is the growing body of evidence suggesting that organic acids play a key function in plant development and, in particular, in fruit development, maturation and ripening. Here, we critically review the connection between organic acids and the development of both climacteric and non-climacteric fruits. By analyzing the metabolic content of different fruits during their ontogenetic trajectory, we noticed that the content of organic acids in the early stages of fruit development is directly related to the supply of substrates for respiratory processes. Although different organic acid species can be found during fruit development in general, it appears that citrate and malate play major roles in this process, as they accumulate on a broad range of climacteric and non-climacteric fruits. We further highlight the functional significance of changes in organic acid profile in fruits due to either the manipulation of fruit-specific genes or the use of fruit-specific promoters. Despite the complexity behind the fluctuation in organic acid content during fruit development and ripening, we extend our understanding on the importance of organic acids on fruit metabolism and the need to further boost future research. We suggest that engineering organic acid metabolism could improve both qualitative and quantitative traits of crop fruits
Statistical Mechanics of Vacancy and Interstitial Strings in Hexagonal Columnar Crystals
Columnar crystals contain defects in the form of vacancy/interstitial loops
or strings of vacancies and interstitials bounded by column ``heads'' and
``tails''. These defect strings are oriented by the columnar lattice and can
change size and shape by movement of the ends and forming kinks along the
length. Hence an analysis in terms of directed living polymers is appropriate
to study their size and shape distribution, volume fraction, etc. If the
entropy of transverse fluctuations overcomes the string line tension in the
crystalline phase, a string proliferation transition occurs, leading to a
supersolid phase. We estimate the wandering entropy and examine the behaviour
in the transition regime. We also calculate numerically the line tension of
various species of vacancies and interstitials in a triangular lattice for
power-law potentials as well as for a modified Bessel function interaction
between columns as occurs in the case of flux lines in type-II superconductors
or long polyelectrolytes in an ionic solution. We find that the centered
interstitial is the lowest energy defect for a very wide range of interactions;
the symmetric vacancy is preferred only for extremely short interaction ranges.Comment: 22 pages (revtex), 15 figures (encapsulated postscript

The E3 ubiquitin ligase SCF(Fbxo7) mediates proteasomal degradation of UXT isoform 2 (UXT-V2) to inhibit the NF-κB signaling pathway.
BACKGROUND: Ubiquitously eXpressed Transcript isoform 2 (UXTV2) is a prefoldin-like protein involved in NF-κB signaling, apoptosis, and the androgen and estrogen response. UXT-V2 is a cofactor in the NF-κB transcriptional enhanceosome, and its knockdown inhibits TNF-α -induced NF-κB activation. Fbxo7 is an F-box protein that interacts with SKP1, Cullin1 and RBX1 proteins to form an SCF(Fbxo7) E3 ubiquitin ligase complex. Fbxo7 negatively regulates NF-κB signaling through TRAF2 and cIAP1 ubiquitination. METHODS: We combine co-immunoprecipitation, ubiquitination in vitro and in vivo, cycloheximide chase assay, ubiquitin chain restriction analysis and microscopy to investigate interaction between Fbxo7 and overexpressed UXT-V2-HA. RESULTS: The Ubl domain of Fbxo7 contributes to interaction with UXTV2. This substrate is polyubiquitinated by SCF(Fbxo7) with K48 and K63 ubiquitin chain linkages in vitro and in vivo. This post-translational modification decreases UXT-V2 stability and promotes its proteasomal degradation. We further show that UXTV1, an alternatively spliced isoform of UXT, containing 12 additional amino acids at the N-terminus as compared to UXTV2, also interacts with and is ubiquitinated by Fbxo7. Moreover, FBXO7 knockdown promotes UXT-V2 accumulation, and the overexpression of Fbxo7-ΔF-box protects UXT-V2 from proteasomal degradation and enhances the responsiveness of NF-κB reporter. We find that UXT-V2 colocalizes with Fbxo7 in the cell nucleus. CONCLUSIONS: Together, our study reveals that SCF(Fbxo7) mediates the proteasomal degradation of UXT-V2 causing the inhibition of the NF-κB signaling pathway. GENERAL SIGNIFICANCE: Discovering new substrates of E3 ubiquitin-ligase SCF(Fbxo7) contributes to understand its function in different diseases such as cancer and Parkinson.Biotechnology and Biological Science Research Council [BB/J007846/1]
Modulation of auxin signalling through DIAGETROPICA and ENTIRE differentially affects tomato plant growth via changes in photosynthetic and mitochondrial metabolism
Auxin modulates a range of plant developmental processes including embryogenesis, organogenesis, and shoot and root development. Recent studies have shown that plant hormones also strongly influence metabolic networks, which results in altered growth phenotypes. Modulating auxin signalling pathways may therefore provide an opportunity to alter crop performance. Here, we performed a detailed physiological and metabolic characterization of tomato (Solanum lycopersicum) mutants with either increased (entire) or reduced (diageotropica—dgt) auxin signalling to investigate the consequences of altered auxin signalling on photosynthesis, water use, and primary metabolism. We show that reduced auxin sensitivity in dgt led to anatomical and physiological modifications, including altered stomatal distribution along the leaf blade and reduced stomatal conductance, resulting in clear reductions in both photosynthesis and water loss in detached leaves. By contrast, plants with higher auxin sensitivity (entire) increased the photosynthetic capacity, as deduced by higher Vcmax and Jmax coupled with reduced stomatal limitation. Remarkably, our results demonstrate that auxin‐sensitive mutants (dgt) are characterized by impairments in the usage of starch that led to lower growth, most likely associated with decreased respiration. Collectively, our findings suggest that mutations in different components of the auxin signalling pathway specifically modulate photosynthetic and respiratory processes
Pediatricians and nutritionists knowledge about treatment of cow milk allergy in infants
OBJECTIVE: Evaluate the knowledge of pediatricians and nutritionists regarding the exclusion diet of cow milk and derivates, with emphasis on questions related to the nutrition of children submitted to such diet. METHODS: Cross-sectional study that enrolled pediatricians (n=53) and nutritionists (n=29) from public hospitals in São Paulo, Brazil, during 2005. Data was collected through self-administered questionnaires. RESULTS: The age of the professionals varied from 21 to 50 years old. Regarding professional experience, 41.2% were graduated for less than five years and 91.6% had a specialization course, masters and/or PhD degree. The vast majority of professionals (97.5%) confirmed that they regularly evaluated the diet of children that needed exclusion of cow milk. However, only 48% of the professionals conducted a more detailed evaluation of the diet, including calculations of food ingestion. Only 38.7% of the professionals compared child s food ingestion with some recommended pattern. Recommendations for daily ingestion of calcium by children up to the age of 36 months were properly mentioned by 22% of the pediatricians and 60.7% of the nutritionists (p=0.001). Inadequate cow milk substitute products were recommended by 66% of the pediatricians and by 48.3% of the nutritionists. Regarding labels of industrialized products, 81.6% of the pediatricians and 96.4% of the nutritionists advised the parents to look for all terms that could indicate the presence of cow milk protein. CONCLUSIONS: Pediatricians and nutritionists made conceptual errors in their main recommendations regarding the treatment of cow milk protein allergy.OBJETIVO: Avaliar o conhecimento de pediatras e nutricionistas sobre a dieta de exclusão do leite de vaca e seus derivados, com ênfase em questões relacionadas à nutrição da criança. MÉTODOS: Estudo transversal descritivo, do qual participaram pediatras (n=53) e nutricionistas (n=29), vinculados a hospitais públicos do Município de São Paulo, no ano de 2005.
