    Ceratoscopelus maderensis : pecular sound-scattering layer identified with this myctophid fish

Originally issued as Reference No. 68-58, series later renamed WHOI-.A sound- scattering layer, composed of discrete hyperbolic echo-sequences and apparently restricted to the Slope Water region of the western North Atlantic, has been identified from the Deep Submergence Research Vehicle ALVIN with schools of the myctophid fish Ceratoscopelus maderensis. By diving into the layer and using ALVIN's echo-ranging sonar, we approached and visually identified the sound scatterers. The number of echo sequences observed with the surface echo-sounder (1 /23. 76 x 105 cubic meters of water) checked roughly with the number of sonar targets observed from the submarine (1/7. 45 x 105 cubic meters) . The fish schools appeared to be 5 to 10 meters thick, 10 to 100 meters in diameter, and on centers 100 to 200 meters apart.     Events in a Non-Commutative Space-Time

    We treat the events determined by a quantum physical state in a noncommutative space-time, generalizing the analogous treatment in the usual Minkowski space-time based on positive-operator-valued measures (POVMs). We consider in detail the model proposed by Snyder in 1947 and calculate the POVMs defined on the real line that describe the measurement of a single coordinate. The approximate joint measurement of all the four space-time coordinates is described in terms of a generalized Wigner function (GWF). We derive lower bounds for the dispersion of the coordinate observables and discuss the covariance of the model under the Poincare' group. The unusual transformation law of the coordinates under space-time translations is interpreted as a failure of the absolute character of the concept of space-time coincidence.     Localization of Events in Space-Time

    The present paper deals with the quantum coordinates of an event in space-time, individuated by a quantum object. It is known that these observables cannot be described by self-adjoint operators or by the corresponding spectral projection-valued measure. We describe them by means of a positive-operator-valued (POV) measure in the Minkowski space-time, satisfying a suitable covariance condition with respect to the Poincare' group. This POV measure determines the probability that a measurement of the coordinates of the event gives results belonging to a given set in space-time. We show that this measure must vanish on the vacuum and the one-particle states, which cannot define any event. We give a general expression for the Poincare' covariant POV measures. We define the baricentric events, which lie on the world-line of the centre-of-mass, and we find a simple expression for the average values of their coordinates. Finally, we discuss the conditions which permit the determination of the coordinates with an arbitrary accuracy.Comment: 31 pages, latex, no figure

    Morphological predictors of BRCA1 germline mutations in young women with breast cancer

    BACKGROUND: Knowing a young woman with newly diagnosed breast cancer has a germline BRCA1 mutation informs her clinical management and that of her relatives. We sought an optimal strategy for identifying carriers using family history, breast cancer morphology and hormone receptor status data.METHODS: We studied a population-based sample of 452 Australian women with invasive breast cancer diagnosed before age 40 years for whom we conducted extensive germline mutation testing (29 carried a BRCA1 mutation) and a systematic pathology review, and collected three-generational family history and tumour ER and PR status. Predictors of mutation status were identified using multiple logistic regression. Areas under receiver operator characteristic (ROC) curves were estimated using five-fold stratified cross-validation.RESULTS: The probability of being a BRCA1 mutation carrier increased with number of selected histology features even after adjusting for family history and ER and PR status (Po0.0001). From the most parsimonious multivariate model, the odds ratio for being a carrier were: 9.7 (95% confidence interval: 2.6-47.0) for trabecular growth pattern (P=0.001); 7.8 (2.7-25.7) for mitotic index over 50 mitoses per 10 high-powered field (P 0.0003); and 2.7 (1.3-5.9) for each first-degree relative with breast cancer diagnosed before age 60 years (P 0.01). The area under the ROC curve was 0.87 (0.83-0.90).CONCLUSION: Pathology review, with attention to a few specific morphological features of invasive breast cancers, can identify almost all BRCA1 germline mutation carriers among women with early-onset breast cancer without taking into account family history.     Fatigue in primary Sjögren's syndrome (pSS) is associated with lower levels of proinflammatory cytokines: a validation study

    Primary Sjögren’s syndrome (pSS) is a chronic autoimmune rheumatic disease with symptoms including dryness, fatigue, and pain. The previous work by our group has suggested that certain proinflammatory cytokines are inversely related to patient-reported levels of fatigue. To date, these findings have not been validated. This study aims to validate this observation. Blood levels of seven cytokines were measured in 120 patients with pSS from the United Kingdom Primary Sjögren’s Syndrome Registry and 30 age-matched healthy non-fatigued controls. Patient-reported scores for fatigue were classified according to severity and compared to cytokine levels using analysis of variance. The differences between cytokines in cases and controls were evaluated using Wilcoxon test. A logistic regression model was used to determine the most important identifiers of fatigue. Five cytokines, interferon-γ-induced protein-10 (IP-10), tumour necrosis factor-α (TNFα), interferon-α (IFNα), interferon-γ (IFN-γ), and lymphotoxin-α (LT-α) were significantly higher in patients with pSS (n = 120) compared to non-fatigued controls (n = 30). Levels of two proinflammatory cytokines, TNF-α (p = 0.021) and LT-α (p = 0.043), were inversely related to patient-reported levels of fatigue. Cytokine levels, disease-specific and clinical parameters as well as pain, anxiety, and depression were used as predictors in our validation model. The model correctly identifies fatigue levels with 85% accuracy. Consistent with the original study, pain, depression, and proinflammatory cytokines appear to be the most powerful predictors of fatigue in pSS. TNF-α and LT-α have an inverse relationship with fatigue severity in pSS challenging the notion that proinflammatory cytokines directly mediate fatigue in chronic immunological conditions

    Symptom-based stratification of patients with primary Sjögren's syndrome: multi-dimensional characterisation of international observational cohorts and reanalyses of randomised clinical trials

    Background Heterogeneity is a major obstacle to developing effective treatments for patients with primary Sjögren's syndrome. We aimed to develop a robust method for stratification, exploiting heterogeneity in patient-reported symptoms, and to relate these differences to pathobiology and therapeutic response. Methods We did hierarchical cluster analysis using five common symptoms associated with primary Sjögren's syndrome (pain, fatigue, dryness, anxiety, and depression), followed by multinomial logistic regression to identify subgroups in the UK Primary Sjögren's Syndrome Registry (UKPSSR). We assessed clinical and biological differences between these subgroups, including transcriptional differences in peripheral blood. Patients from two independent validation cohorts in Norway and France were used to confirm patient stratification. Data from two phase 3 clinical trials were similarly stratified to assess the differences between subgroups in treatment response to hydroxychloroquine and rituximab. Findings In the UKPSSR cohort (n=608), we identified four subgroups: Low symptom burden (LSB), high symptom burden (HSB), dryness dominant with fatigue (DDF), and pain dominant with fatigue (PDF). Significant differences in peripheral blood lymphocyte counts, anti-SSA and anti-SSB antibody positivity, as well as serum IgG, κ-free light chain, β2-microglobulin, and CXCL13 concentrations were observed between these subgroups, along with differentially expressed transcriptomic modules in peripheral blood. Similar findings were observed in the independent validation cohorts (n=396). Reanalysis of trial data stratifying patients into these subgroups suggested a treatment effect with hydroxychloroquine in the HSB subgroup and with rituximab in the DDF subgroup compared with placebo. Interpretation Stratification on the basis of patient-reported symptoms of patients with primary Sjögren's syndrome revealed distinct pathobiological endotypes with distinct responses to immunomodulatory treatments. Our data have important implications for clinical management, trial design, and therapeutic development. Similar stratification approaches might be useful for patients with other chronic immune-mediated diseases.     The Western Atlantic Jawfishes of the Opisthognathid Genus Lonchopisthus

    During 1957 two important collections of fishes were obtained from off the coast of Suriname and adjacent regions. The first of these was made by the motor vessel “Coquette”, a commercial shrimp trawler which engaged in exploratory work for the Government of Suriname. Mr. JAMES B. HIGMAN of the United States Fish and Wildlife Service was invited to accompany the “Coquette” during part of this work, and the collection of fishes which resulted was due largely to his efforts. The second collection was obtained by the motor vessel “Oregon”, exploratory vessel of the United States Fish and Wildlife Service. During November, 1957, the “Oregon” occupied over a hundred trawl stations along the northern coast of South America between Venezuela and the Equator. Most of these collections are now in the U. S. National Museum and the Chicago Natural History Museum. Both the “Oregon” and the “Coquette” collections contain representatives of a distinctive new species of Lonchopisthus. The definition of this species has required a review of the western Atlantic species of the genus. I take pleasure in naming this new species of Lonchopisthus from Suriname in honour of its collector
