    Ammonium pertechnetate reacts in mixtures of trifluoromethanesulfonic anhydride and trifluoromethanesulfonic acid under final formation of ammonium pentakis(trifluoromethanesulfonato)oxidotechnetate(V), (NH4)2[TcO(OTf)5]. The reaction proceeds only at exact concentrations and under the exclusion of air and moisture via pertechnetyl trifluoromethanesulfonate, [TcO3(OTf)], and intermediate TcVI species. 99Tc nuclear magnetic resonance (NMR) has been used to study the TcVII compound and electron paramagnetic resonance (EPR), 99Tc NMR and X-ray absorption near-edge structure (XANES) experiments indicate the presence of the reduced technetium species.     Renal Ischemia/Reperfusion Injury in Soluble Epoxide Hydrolase-Deficient Mice

    Aim 20-hydroxyeicosatetraenoic acid (20-HETE) and epoxyeicosatrienoic acids (EETs) are cytochrome P450 (CYP)-dependent eicosanoids that play opposite roles in the regulation of vascular tone, inflammation, and apoptosis. 20-HETE aggravates, whereas EETs ameliorate ischemia/reperfusion (I/R)-induced organ damage. EETs are rapidly metabolized to dihydroxyeicosatrienoic acids (DHETs) by the soluble epoxide hydrolase (sEH). We hypothesized that sEH gene (EPHX2) deletion would increase endogenous EET levels and thereby protect against I/R-induced acute kidney injury (AKI). Methods Kidney damage was evaluated in male wildtype (WT) and sEH-knockout (KO)-mice that underwent 22-min renal ischemia followed by two days of reperfusion. CYP-eicosanoids were analyzed by liquid chromatography tandem mass spectrometry. Results Contrary to our initial hypothesis, renal function declined more severely in sEH-KO mice as indicated by higher serum creatinine and urea levels. The sEH-KO-mice also featured stronger tubular lesion scores, tubular apoptosis, and inflammatory cell infiltration. Plasma and renal EET/DHET-ratios were higher in sEH-KO than WT mice, thus confirming the expected metabolic consequences of sEH deficiency. However, CYP-eicosanoid profiling also revealed that renal, but not plasma and hepatic, 20-HETE levels were significantly increased in sEH-KO compared to WT mice. In line with this finding, renal expression of Cyp4a12a, the murine 20-HETE-generating CYP-enzyme, was up-regulated both at the mRNA and protein level, and Cyp4a12a immunostaining was more intense in the renal arterioles of sEH-KO compared with WT mice. Conclusion These results indicate that the potential beneficial effects of reducing EET degradation were obliterated by a thus far unknown mechanism leading to kidney-specific up- regulation of 20-HETE formation in sEH-KO-mice

    Source apportionment of methane emissions from the Upper Silesian Coal Basin using isotopic signatures

    During the CoMet mission in the Upper Silesian Coal Basin (USCB) ground-based and airborne air samples were taken, and analyzed for the isotopic composition of CH4 to derive the mean signature of the USCB and the source signatures of individual coal mines. Using ή2H signatures, the biogenic emissions from the USCB account for 15–50 % of total emissions, which is underestimated in common emission inventories. This demonstrates the importance of ή2H-CH4 observations for methane source attribution

    IL-33-induced metabolic reprogramming controls the differentiation of alternatively activated macrophages and the resolution of inflammation

    Alternatively activated macrophages (AAMs) contribute to the resolution of inflammation and tissue repair. However, molecular pathways that govern their differentiation have remained incompletely understood. Here, we show that uncoupling protein-2-mediated mitochondrial reprogramming and the transcription factor GATA3 specifically controlled the differentiation of pro-resolving AAMs in response to the alarmin IL-33. In macrophages, IL-33 sequentially triggered early expression of pro-inflammatory genes and subsequent differentiation into AAMs. Global analysis of underlying signaling events revealed that IL-33 induced a rapid metabolic rewiring of macrophages that involved uncoupling of the respiratory chain and increased production of the metabolite itaconate, which subsequently triggered a GATA3-mediated AAM polarization. Conditional deletion of GATA3 in mononuclear phagocytes accordingly abrogated IL-33-induced differentiation of AAMs and tissue repair upon muscle injury. Our data thus identify an IL-4-independent and GATA3-dependent pathway in mononuclear phagocytes that results from mitochondrial rewiring and controls macrophage plasticity and the resolution of inflammation

    Small Ti clusters for catalysis of hydrogen exchange

    Abstract Colloidal Ti has been synthesized following a method described in the literature. Extended x-ray absorption fine structure measurements indicate the presence of small colloidal entities, consisting of only a few core Ti atoms which are coordinated by O atoms from tetrahydrofuran (THF). The results can be explained by the proposed structure of Ti 13 ·6THF, in which the Ti cluster has the shape of a distorted icosahedron. The Ti colloid was used to prepare a functional nanocomposite by ball-milling the clusters with NaAlH 4 . The nanocomposite showed superior hydrogen exchange kinetics when compared to the state of the art in the literature

    Assembly of Compliant Structures with Autonomous Industrial Mobile Manipulators (AIMM) Using an End Effector with Active Deformation Compensation for the Assembly of Flaps

    Composite structures in aeroplanes are often thin-walled and lightweight, resulting in significant compliance, which presents a handling and assembly challenge due to the associated part deformations. In order to counteract these deformations, the parts are held in their specified geometry using stiff and correspondingly heavy fixtures or jigs. Mobile industrial robots are very versatile and widely used in industrial volume production, but they are limited in their payload capacity. High-rate production of large aerospace modules requires highly automated flexible assembly processes. The approach presented in this paper is to combine mobile units with lightweight assembly jigs that have the capability of deformation compensation. The subject of the study is a high-rate assembly process for flap modules using an Autonomous Industrial Mobile Manipulator (AIMM) and a lightweight end effector. The end effector has a shape compensation function, implemented by an integrated Stewart platform, which enables the compensation of manufacturing tolerances as well as gravity effects. The compensation function is used in a closed loop and counteracts shape deviations by appropriate fixture shape adjustments. The paper reports on the conceptual design of the assembly scenario, the design of the end effector, its realization and the successful experimental demonstration at 1:1 scale

    Reducing antipsychotic drugs in stable patients with chronic schizophrenia or schizoaffective disorder: a randomized controlled pilot trial

    As the course of schizophrenic disorders is often chronic, treatment guidelines recommend continuous maintenance treatment to prevent relapses, but antipsychotic drugs can cause many side effects. It, therefore, seems reasonable to try to reduce doses in stable phases of the illness or even try to stop medication. We conducted a 26 weeks, randomized, rater blind, feasibility study to examine individualized antipsychotic dose reduction versus continuous maintenance treatment (Register Number: NCT02307396). We included chronic, adult patients with schizophrenia or schizoaffective disorder, who were treated with any antipsychotic drug except clozapine, who had not been hospitalized in the last 3 years and who were in symptomatic remission at baseline. The primary outcome was relapse of positive symptoms. Symptom severity, social functioning and side effects were also examined as secondary outcomes. 20 patients were randomized. Relapse rates in the two groups were not significantly different. No patient had to be hospitalized. One patient in the control group dropped out. The mean reduction of antipsychotic dose in the individualized dose-reduction group was 42%, however only one patient discontinued drug completely. There were no significant differences in efficacy or safety outcomes. This randomized trial provides evidence, that reduction of antipsychotic medication in chronic stable schizophrenic patients may be feasible. The results need to be confirmed in a larger trial with a longer follow-up period
