    Viral surface glycoproteins, gp120 and gp41, as potential drug targets against HIV-1: Brief overview one quarter of a century past the approval of zidovudine, the first anti-retroviral drug

    The first anti-HIV drug, zidovudine (AZT), was approved by the FDA a quarter of a century ago, in 1985. Currently, anti-HIV drug-combination therapies only target HIV-1 protease and reverse transcriptase. Unfortunately, most of these molecules present numerous shortcoming; such as viral resistances and adverse effects. In addition, these drugs are involved in later stages of infection. Thus, it is necessary to develop new drugs that are able to block the first steps of viral life cycle. Entry of HIV-1 is mediated by its two envelope glycoproteins: gp120 and gp41. Upon gp120 binding to cellular receptors, gp41 undergoes a series of conformational changes from a non-fusogenic to a fusogenic conformation. The fusogenic core of gp41 is a trimer-of-hairpins structure in which three C-terminal helices pack against a central trimeric-coiled coil formed by three N-terminal helices. The formation of this fusogenic structure brings the viral and cellular membranes close together, a necessary condition for membrane fusion to occur. As gp120 and gp41 are attractive targets, the development of entry inhibitors represents an important avenue of anti-HIV drug therapy. The present review will focus on some general considerations about HIV, the main characteristics of gp120, gp41 and their inhibitors, with special emphasis on the advances of computational approaches employed in the development of bioactive compounds against HIV-1 entry process

    A Comparison of Accuracy of Image- versus Hardware-based Tracking Technologies in 3D Fusion in Aortic Endografting

    OBJECTIVES: Fusion of three-dimensional (3D) computed tomography and intraoperative two-dimensional imaging in endovascular surgery relies on manual rigid co-registration of bony landmarks and tracking of hardware to provide a 3D overlay (hardware-based tracking, HWT). An alternative technique (image-based tracking, IMT) uses image recognition to register and place the fusion mask. We present preliminary experience with an agnostic fusion technology that uses IMT, with the aim of comparing the accuracy of overlay for this technology with HWT. METHOD: Data were collected prospectively for 12 patients. All devices were deployed using both IMT and HWT fusion assistance concurrently. Postoperative analysis of both systems was performed by three blinded expert observers, from selected time-points during the procedures, using the displacement of fusion rings, the overlay of vascular markings and the true ostia of renal arteries. The Mean overlay error and the deviation from mean error was derived using image analysis software. Comparison of the mean overlay error was made between IMT and HWT. The validity of the point-picking technique was assessed. RESULTS: IMT was successful in all of the first 12 cases, whereas technical learning curve challenges thwarted HWT in four cases. When independent operators assessed the degree of accuracy of the overlay, the median error for IMT was 3.9 mm (IQR 2.89-6.24, max 9.5) versus 8.64 mm (IQR 6.1-16.8, max 24.5) for HWT (p = .001). Variance per observer was 0.69 mm(2) and 95% limit of agreement ±1.63. CONCLUSION: In this preliminary study, the error of magnitude of displacement from the "true anatomy" during image overlay in IMT was less than for HWT. This confirms that ongoing manual re-registration, as recommended by the manufacturer, should be performed for HWT systems to maintain accuracy. The error in position of the fusion markers for IMT was consistent, thus may be considered predictable

    Essential oils: pharmaceutical applications and encapsulation strategies into lipid-based delivery systems

    Essential oils are being studied for more than 60 years, but a growing interest has emerged in the recent decades due to a desire for a rediscovery of natural remedies. Essential oils are known for millennia and, already in prehistoric times, they were used for medicinal and ritual purposes due to their therapeutic properties. Using a variety of methods refined over the centuries, essential oils are extracted from plant raw materials: the choice of the extraction method is decisive, since it determines the type, quantity, and stereochemical structure of the essential oil molecules. To these components belong all properties that make essential oils so interesting for pharmaceutical uses; the most investigated ones are antioxidant, anti-inflammatory, antimicrobial, wound-healing, and anxiolytic activities. However, the main limitations to their use are their hydrophobicity, instability, high volatility, and risk of toxicity. A successful strategy to overcome these limitations is the encapsulation within delivery systems, which enable the increase of essential oils bioavailability and improve their chemical stability, while reducing their volatility and toxicity.     Ultralow transmission loss in inhibited-coupling guiding hollow fibers

    Attenuation in photonic bandgap guiding hollow-core photonic crystal fiber (HC-PCF) has not beaten the fundamental silica Rayleigh scattering limit (SRSL) of conventional step-index fibers due to strong core-cladding optical overlap, surface roughness at the silica cladding struts, and the presence of interface modes. Hope has been revived recently by the introduction of hypocycloid core contour (i.e., negative curvature) in inhibited-coupling guiding HCPCF. We report on several fibers with a hypocycloid core contour and a cladding structure made of a single ring from a tubular amorphous lattice, including one with a record transmission loss of 7.7 dB/km at ~750 nm (only a factor ~2 above the SRSL) and a second with an ultrabroad fundamental band with loss in the range of 10-20 dB/km, spanning from 600 to 1200 nm. The reduction in confinement loss makes these fibers serious contenders for light transmission below the SRSL in the UV-VIS-NIR spectral range and could find application in high-energy pulse laser beam delivery or gas-based coherent and nonlinear optics

    Nectin-like proteins mediate axon–Schwann cell interactions along the internode and are essential for myelination

    Axon–glial interactions are critical for the induction of myelination and the domain organization of myelinated fibers. Although molecular complexes that mediate these interactions in the nodal region are known, their counterparts along the internode are poorly defined. We report that neurons and Schwann cells express distinct sets of nectin-like (Necl) proteins: axons highly express Necl-1 and -2, whereas Schwann cells express Necl-4 and lower amounts of Necl-2. These proteins are strikingly localized to the internode, where Necl-1 and -2 on the axon are directly apposed by Necl-4 on the Schwann cell; all three proteins are also enriched at Schmidt-Lanterman incisures. Binding experiments demonstrate that the Necl proteins preferentially mediate heterophilic rather than homophilic interactions. In particular, Necl-1 on axons binds specifically to Necl-4 on Schwann cells. Knockdown of Necl-4 by short hairpin RNA inhibits Schwann cell differentiation and subsequent myelination in cocultures. These results demonstrate a key role for Necl-4 in initiating peripheral nervous system myelination and implicate the Necl proteins as mediators of axo–glial interactions along the internode

    International audienceCette communication vise Ă  prĂ©senter les outils mĂ©thodologiques d'analyse/Ă©valuation de la gouvernance territoriale Ă©laborĂ©s dans le cadre du projet de recherche PSDR Gouv.Innov sur les innovations organisationnelles relatives Ă  la gouvernance territoriale. Il s'agit d'Ă©tudier les transformations introduites par les politiques de dĂ©veloppement durable au niveau des dispositifs de gouvernance territoriale visant Ă  favoriser une gestion intĂ©grĂ©e des espaces ruraux. Dans un contexte de recomposition de l'action publique oĂč les procĂ©dures d'amĂ©nagement sont plutĂŽt normĂ©es par des reprĂ©sentations urbaines, l'accent est mis sur la question des modalitĂ©s de reprĂ©sentation des activitĂ©s rurales, pour lesquelles nous faisons l'hypothĂšse qu'elles sont sous reprĂ©sentĂ©es. Les premiers rĂ©sultats mĂ©thodologiques permettent, dans une premiĂšre partie, de proposer une dĂ©finition gĂ©nĂ©rique et pragmatique de la gouvernance territoriale et de prĂ©ciser la notion de dispositifs de gouvernance comme objet d'observation. A partir de cette dĂ©finition une grille d'analyse permettant d'apprĂ©hender l'ensemble des dimensions en jeu dans les processus de gouvernance territoriale est Ă©laborĂ©e. Dans la deuxiĂšme partie nous explorons l'intĂ©rĂȘt de la notion de dispositif pour observer les processus de gouvernance et proposons une grille de collecte et de structuration des informations pour constituer des chroniques des dispositifs Ă©tudiĂ©s. / This paper seeks to present the methodological tools used in the analysis/evaluation of territorial governance developed during research undertaken within the framework of the PSDR Gouv-Innov project on organisational innovations with respect to territorial governance. It involves the study of the changes resulting from the emergence of sustainable development policies in territorial governance systems aiming to implement the integrated management of rural areas. In a context of changes in the way in which public action is implemented (for instance, public-private partnerships), where development procedures are usually standardised by urban representations, the emphasis is placed on the issue of the representation of rural activities, the hypothesis being that they are under-represented. The initial methodological results lead, in the first instance, to a proposition for a generic and pragmatic definition of territorial governance and a clarification of the notion of governance systems as objects of observation. This definition can be used as the basis of a framework to address all the analytical dimensions of the territorial governance process. In the second part, the authors explore the idea of system in order to observe governance processes and suggest a matrix for the collection and structuring of information in order to report on the studied systems

    International audienceDans cet article nous présentons une grille d'analyse de la gouvernance territoriale que nous avons construite dans le cadre d'un projet de recherche pluridisciplinaire à partir d'une définition commune de la gouvernance territoriale. La gouvernance territoriale est devenue une question centrale dans le développement territorial et un enjeu pour l'élaboration des politiques publiques. Elaborer une grille d'analyse sur la gouvernance territoriale ne va pas de soi ; nous illustrerons son opérationnalisation à partir des travaux de terrain réalisés dans trois situations dans la région de Languedoc Roussillon. Les dispositifs et outils sont contextuels et les modes de gouvernances pluriels selon les objectifs et les dynamiques antérieures. Enfin nous en tirerons des enseignements pour l'étude de la gouvernance tant sur la production des connaissances que sur l'opérationnalité de cette grille. / In this paper we present an analytical framework of territorial governance that we built as part of a multidisciplinary research project based on a common definition of territorial governance. Territorial governance has become a central issue in regional development and a challenge for public policy. Develop an analytical framework on territorial governance does not just happen, and we illustrate its operationalization based on fieldwork conducted in three situations in Languedoc-Roussillon. Devices and tools are contextual and plural forms of governance based on the objectives and dynamics earlier. Finally we will draw lessons for the study of governance both in the production of knowledge about the operational capability of the grid
