    Regulation of Op18 during Spindle Assembly in Xenopus Egg Extracts

    Oncoprotein 18 (Op18) is a microtubule-destabilizing protein that is negatively regulated by phosphorylation. To evaluate the role of the three Op18 phosphorylation sites in Xenopus (Ser 16, 25, and 39), we added wild-type Op18, a nonphosphorylatable triple Ser to Ala mutant (Op18-AAA), and to mimic phosphorylation, a triple Ser to Glu mutant (Op18-EEE) to egg extracts and monitored spindle assembly. Op18-AAA dramatically decreased microtubule length and density, while Op18-EEE did not significantly affect spindle microtubules. Affinity chromatography with these proteins revealed that the microtubule-destabilizing activity correlated with the ability of Op18 to bind tubulin. Since hyperphosphorylation of Op18 is observed upon addition of mitotic chromatin to extracts, we reasoned that chromatin-associated proteins might play a role in Op18 regulation. We have performed a preliminary characterization of the chromatin proteins recruited to DNA beads, and identified the Xenopus polo-like kinase Plx1 as a chromatin-associated kinase that regulates Op18 phosphorylation. Depletion of Plx1 inhibits chromatin-induced Op18 hyperphosphorylation and spindle assembly in extracts. Therefore, Plx1 may promote microtubule stabilization and spindle assembly by inhibiting Op18

    Increased Stathmin1 Expression in the Dentate Gyrus of Mice Causes Abnormal Axonal Arborizations

    Pituitary adenylate cyclase-activating polypeptide (PACAP) is involved in multiple brain functions. To clarify the cause of abnormal behavior in PACAP deficient-mice, we attempted the identification of genes whose expression was altered in the dentate gyrus of PACAP-deficient mice using the differential display method. Expression of stathmin1 was up-regulated in the dentate gyrus at both the mRNA and protein levels. PACAP stimulation inhibited stathmin1 expression in PC12 cells, while increased stathmin1expression in neurons of the subgranular zone and in primary cultured hippocampal neurons induced abnormal arborization of axons. We also investigated the pathways involved in PACAP deficiency. Ascl1 binds to E10 box of the stathmin1 promoter and increases stathmin1 expression. Inhibitory bHLH proteins (Hes1 and Id3) were rapidly up-regulated by PACAP stimulation, and Hes1 could suppress Ascl1 expression and Id3 could inhibit Ascl1 signaling. We also detected an increase of stathmin1 expression in the brains of schizophrenic patients. These results suggest that up-regulation of stathmin1 in the dentate gyrus, secondary to PACAP deficiency, may create abnormal neuronal circuits that cause abnormal behavior

    Avec l’émergence des ONG dans nos économies globalisées, les grandes entreprises développent divers types de partenariats ONG-entreprise (POE). Les chercheurs s’interrogent sur leurs enjeux stratégiques. Dans notre thèse, nous étudions la contribution spécifique des POE à l’évolution du business model (BM) de la grande entreprise.Une étude de cas longitudinale processuelle chez Suez-Environnement met en lumière trois grands apports des POE : les POE, selon leur type, influencent la stratégie de l’entreprise par leur concours à la segmentation des activités ; ils contribuent à la conception et au déploiement des BM issus de la nouvelle segmentation ; et ils jouent un rôle moteur dans l’évolution du portefeuille de BM de l’entreprise.À partir de ces résultats, nous proposons : une conception élargie de la finalité du BM intégrant la performance sociétale ; une représentation enrichie du concept de BM, baptisée modèle RCOV-EPs, articulant quatre dimensions en interactions ; et le concept de métabusiness model que nous définissons comme : une métalogique de création de valeur socioéconomique émergeant de relations inter-BM.With the emergence of the NGOs in our globalized economies, large companies developdiverse types of corporate-NGO partnerships (CNPs). Some researchers raised interrogations on their strategic issues. In our thesis, we study the CNPs specific contribution to the evolution of large companies’ business model (BM).Drawing on a longitudinal processual case study at Suez-Environnement, we highlight three main contributions of CNPs: first, some types of CNPs influence company’s strategy by taking part in the activities’ segmentation; second, they contribute to design and to implement the BMs resulting from the new segmentation; third, they play a driving role in the company’s BMs portfolio evolution.From these results, we suggest: an extended approach of BM’s purpose including societalperformance; an enhanced representation of the BM concept, called RCOV-PsE model,structuring four interacting components; and the meta business-model concept that we define as: a socio-economic value creation logic appearing from inter-BM relations
