JUNE-Germany: An Agent-Based Epidemiology Simulation including Multiple Virus Strains, Vaccinations and Testing Campaigns
The June software package is an open-source framework for the detailed
simulation of epidemics based on social interactions in a virtual population
reflecting age, gender, ethnicity, and socio-economic indicators in England. In
this paper, we present a new version of the framework specifically adapted for
Germany, which allows the simulation of the entire German population using
publicly available information on households, schools, universities,
workplaces, and mobility data for Germany. Moreover, JuneGermany incorporates
testing and vaccination strategies within the population as well as the
simultaneous handling of several different virus strains. First validation
tests of the framework have been performed for the state of Rhineland
Palatinate based on data collected between October 2020 and December 2020 and
then extrapolated to March 2021, i.e. the end of the second wave.Comment: 10 pages, 11 figure
Data-Driven Regionalization of Decarbonized Energy Systems for Reflecting Their Changing Topologies in Planning and Optimization
The decarbonization of energy systems has led to a fundamental change in their topologysince generation is shifted to locations with favorable renewable conditions. In planning, this changeis reflected by applying optimization models to regions within a country to optimize the distributionof generation units and to evaluate the resulting impact on the grid topology. This paper proposesa globally applicable framework to find a suitable regionalization for energy system models witha data-driven approach. Based on a global, spatially resolved database of demand, generation, and renewable profiles, hierarchical clustering with fine-tuning is performed. This regionalizationapproach is applied by modeling the resulting regions in an optimization model including asynthesized grid. In an exemplary case study, South Africaâs energy system is examined. The resultsshow that the data-driven regionalization is beneficial compared to the common approach of usingpolitical regions. Furthermore, the results of a modeled 80% decarbonization until 2045 demonstratethat the integration of renewable energy sources fundamentally changes the role of regions withinSouth Africaâs energy system. Thereby, the electricity exchange between regions is also impacted, leading to a different grid topology. Using clustered regions improves the understanding and analysisof regional transformations in the decarbonization process
Towards an integrative, eco-evolutionary understanding of ecological novelty: studying and communicating interlinked effects of global change
Global change has complex eco-evolutionary consequences for organisms and ecosystems, but related concepts (e.g., novel ecosystems) do not cover their full range. Here we propose an umbrella concept of âecological noveltyâ comprising (1) a site-specific and (2) an organism-centered, eco-evolutionary perspective. Under this umbrella, complementary options for studying and communicating effects of global change on organisms, ecosystems, and landscapes can be included in a toolbox. This allows researchers to address ecological novelty from different perspectives, e.g., by defining it based on (a) categorical or continuous measures, (b) reference conditions related to sites or organisms, and (c) types of human activities. We suggest striving for a descriptive, non-normative usage of the term âecological noveltyâ in science. Normative evaluations and decisions about conservation policies or management are important, but require additional societal processes and engagement with multiple stakeholders

This regionalization approach is applied by modeling the resulting regions in an optimization model including a synthesized grid. In an exemplary case study, South Africaâs energy system is examined. The results show that the data-driven regionalization is beneficial compared to the common approach of using political regions. Furthermore, the results of a modeled 80% decarbonization until 2045 demonstrate that the integration of renewable energy sources fundamentally changes the role of regions within South Africaâs energy system. Static and dynamic 68Ga-FAPI PET/CT for the detection of malignant transformation of intraductal papillary mucinous neoplasia of the pancreas.
Purpose: Pancreatic ductal adenocarcinoma (PDAC) may arise from intraductal papillary-mucinous neoplasms (IPMN) with malignant transformation, but a significant portion of IPMN remains to show benign behavior. Therefore, it is important to differentiate between benign IPMN and IPMN lesions undergoing malignant transformation. However, non-operative differentiation by ultrasound, CT, MRI and carbohydrate antigen 19-9 (CA19-9) is still unsatisfactory. Here, we assessed the clinical feasibility of additional assessment of malignancy by positron emission tomography using 68Gallium-labeled Fibroblast Activation Protein Inhibitors (68Ga-FAPI-PET) in 25 patients with magnetic resonance imaging (MRI) - or computed tomography (CT) - proven cystic pancreatic lesions. Methods: 25 patients with cystic pancreatic lesions who were followed up in the European Pancreas Center of Heidelberg University hospital and who were led to surgical resection or fine needle aspiration (FNA) due to suspicious clinical, laboratory chemistry or radiological findings were examined by static (all patients) and dynamic (20 patients) 68Ga-FAPI-PET. Cystic pancreatic lesions were delineated and maximum and mean standardized uptake values (SUVmax / SUVmean) were determined. Time activity curves and dynamic parameters (time to peak, K1, k2, K3, k4) were extracted from dynamic PET data. Receiver operating curves (ROC) of static and dynamic PET parameters were calculated. Results: 11 of the patients suffered from menacing IPMN (high grade IPMN with (6 cases) or without (5 cases) progression into PDAC) and 11 from low grade IPMN, 3 patients from other benign entities. Menacing IMPN showed significantly elevated 68Ga-FAPI uptake compared to low grade IPMN and other benign cystic lesions. In dynamic imaging, menacing IPMN showed increasing time activity curves (TAC) followed by slow decrease afterwards, TAC of low grade IPMN showed an immediate peak followed by rapid decrease for about 10 minutes and slower decrease for the rest of the time. ROC curves showed high sensitivity and specificity (area under the curve (AUC) greater than 80%) of static and dynamic PET parameters for the differentiation of IPMN subtypes. Conclusion: 68Ga-FAPI-PET is a helpful new tool for the differentiation of menacing and low grade IPMN and shows the potential to avoid unnecessary surgery for non-malignant pancreatic IPMN

XCR1 expression distinguishes human conventional dendritic cell type 1 with full effector functions from their immediate precursors
Dendritic cells (DCs) are major regulators of innate and adaptive immune responses. DCs can be classified into plasmacytoid DCs and conventional DCs (cDCs) type 1 and 2. Murine and human cDC1 share the mRNA expression of XCR1. Murine studies indicated a specific role of the XCR1-XCL1 axis in the induction of immune responses. Here, we describe that human cDC1 can be distinguished into XCR1 and XCR1 cDC1 in lymphoid as well as nonlymphoid tissues. Steady-state XCR1 cDC1 display a preactivated phenotype compared to XCR1 cDC1. Upon stimulation, XCR1 cDC1, but not XCR1 cDC1, secreted high levels of inflammatory cytokines as well as chemokines. This was associated with enhanced activation of NK cells mediated by XCR1 cDC1. Moreover, XCR1 cDC1 excelled in inhibiting replication of Influenza A virus. Further, under DC differentiation conditions, XCR1 cDC1 developed into XCR1 cDC1. After acquisition of XCR1 expression, XCR1 cDC1 secreted comparable level of inflammatory cytokines. Thus, XCR1 is a marker of terminally differentiated cDC1 that licenses the antiviral effector functions of human cDC1, while XCR1 cDC1 seem to represent a late immediate precursor of cDC1
