    The Influence of Short-term Quercetin Supplementation on Peak Oxygen Uptake during Simulated Altitude Exposure in Trained Cyclists

    Endurance performance and peak oxygen uptake (VO2peak) decline as altitude increases. Some data exist suggesting that quercetin supplementation improves aerobic capacity in trained and untrained individuals at sea-level (normobaric normoxic conditions). Few studies have examined the effects of quercetin on endurance performance during simulated altitude exposure (normobaric hypoxic conditions). The purpose of this study was to determine the effects of consuming 1000 mg·day-1 of quercetin for two weeks on cycling VO2peak in healthy trained male cyclists performing under normobaric normoxic and hypoxic conditions (NP and HP, respectively). Fourteen age-matched healthy male subjects were randomized to either a placebo or quercetin group. Baseline and post supplementation VO2peak values were quantified during incremental cycling under normobaric normoxic (FIO2 = 20.9%) and normobaric hypoxic (FIO2 = 13.6 + 0.2%) conditions. Subjects consumed capsules twice daily with either 500 mg quercetin or placebo (Tang) for two weeks and were re-assessed. Test order was randomized and assessments were separated by 48-72 hours. At baseline, there were no significant differences between groups for VO2peak normobaric normoxic trials (NPbaseline Placebo vs. NPbaseline Quercetin = 58.7+8.8 and 61.5+7.9 ml . kg-1 . min-1, respectively, p = 0.541) and normobaric hypoxic trials (HPbaseline Placebo vs. HPbaseline Quercetin = 48.5+8.3 and 50.8+4.8 ml . kg-1 . min-1, respectively, p = 0.534). No significant differences were found after treatment (Placebo: NPbaseline vs. NPpost = 58.7+ 8.8 and 56.7+7.4 ml . kg-1 . min-1, respectively, p = 0.10; HPbaseline vs .HPpost = 48.5+8.3 and 47.1+8.3, respectively, p = 0.50; Quercetin: NPbaseline vs. NPpost = 61.5+7.9 and 62.4+7.3 ml . kg-1 . min-1, respectively, p = 0.558; HPbaseline vs. HPpost = 50.8+4.8 and 51.2+3.8 ml . kg-1 . min-1, respectively, p= 0.656. These data suggest that short-term quercetin supplementation at 1000 mg . day-1 does not affect VO2peak elicited via incremental maximal cycle testing under normobaric normoxic and hypoxic conditions in trained male cyclists. Furthermore, quercetin supplementation did not attenuate the decline in VO2peak that was evident in the normobaric hypoxic condition

    Smashing New Results on Aspectual Framing: How People Talk About Car Accidents

    How do people describe events they have witnessed? What role does linguistic aspect play in this process? To provide answers to these questions, we conducted an experiment on aspectual framing. In our task, people were asked to view videotaped vehicular accidents and to describe what happened (perfective framing) or what was happening (imperfective framing). Our analyses of speech and gesture in retellings show that the form of aspect used in the question differentially influenced the way people conceptualized and described actions. Questions framed with imperfective aspect resulted in more motion verbs (e.g., driving), more reckless language (e.g., speeding), and more iconic gestures (e.g., path gesture away from the body to show travel direction) than did questions framed with perfective aspect. Our research contributes novel insights on aspect and the construal of events, and on the semantic potency of aspect in leading questions. The findings are consistent with core assumptions in cognitive linguistics, including the proposal that linguistic meaning, including grammatical meaning, is dynamic and grounded in perceptual and cognitive experience

    Reconciling Epidemiology and Social Justice in the Public Health Discourse Around the Sexual Networks of Black Men Who Have Sex With Men

    Several studies have implicated the sexual networks of Black men who have sex with men (MSM) as facilitating disproportionally high rates of new HIV infections within this community. Although structural disparities place these networks at heightened risk for infection, HIV prevention science continues to describe networks as the cause for HIV disparities, rather than an effect of structures that pattern infection. We explore the historical relationship between public health and Black MSM, arguing that the current articulation of Black MSM networks is too often incomplete and counterproductive. Public health can offer a counternarrative that reconciles epidemiology with the social justice that informs our discipline, and that is required for an effective response to the epidemic among Black MSM

    Developing network adequacy standards for VA Community Care

    OBJECTIVES: To inform how the VA should develop and implement network adequacy standards, we convened an expert panel to discuss Community Care Network (CCN) adequacy and how VA might implement network adequacy standards for community care. DATA SOURCES/STUDY SETTING: Data were generated from expert panel ratings and from an audio-recorded expert panel meeting conducted in Arlington, Virginia, in October 2017. STUDY DESIGN: We used a modified Delphi panel process involving one round of expert panel ratings provided by nine experts in network adequacy standards. Expert panel members received a list of network adequacy standard measures used in commercial and government market and were provided a rating form listing a total of 11 measures and characteristics to rate. DATA COLLECTION METHODS: Items on the rating form were individually discussed during an expert panel meeting between the nine expert panel members and VA Office of Community Care leaders. Attendees addressed discordant views and generated revised or new standards accordingly. Recorded audio data were transcribed to facilitate thematic analysis regarding opportunities and challenges with implementing network adequacy standards in VA Community Care. PRINCIPAL FINDINGS: The five highest ranked standards were network directories for Veterans, regular reporting of network adequacy data to VA, maximum wait time/distance standards, minimum ratio of providers to enrolled population, and qualitative assessments of network adequacy. During the expert panel discussion with VA Community Care leaders, opportunities and challenges implementing network adequacy standards were highlighted. CONCLUSIONS: Our expert panel shed light on priorities for network adequacy to be implemented under CCN contracts, such as developing comprehensive provider directories for Veterans to use when selecting community providers. Remaining questions focus on whether the VA could reasonably develop and implement network adequacy standards given current Congressional restraints on VA reimbursement to community providers

    Evaluation of adenovirus capsid labeling versus transgene expression

    Adenoviral vectors have been utilized for a variety of gene therapy applications. Our group has incorporated bioluminescent, fluorographic reporters, and/or suicide genes within the adenovirus genome for analytical and/or therapeutic purposes. These molecules have also been incorporated as capsid components. Recognizing that incorporations at either locale yield potential advantages and disadvantages, our report evaluates the benefits of transgene incorporation versus capsid incorporation. To this end, we have genetically incorporated firefly luciferase within the early region 3 or at minor capsid protein IX and compared vector functionality by means of reporter readout

    textabstractBackground: There is a need to build the evidence base of early interventions promoting children's health and development in the UK. Malachi Specialist Family Support Services ('Malachi') is a voluntary sector organisation based in the UK that delivers a therapeutic parenting group programme called Inspiring Futures to parents of children identified as having behavioural and emotional difficulties. The programme comprises two parts, delivered sequentially: (1) a group-based programme for all parents for 10-12 weeks, and (2) one-to-one sessions with selected parents from the group-based element for up to 12 weeks. Methods/design: A randomised controlled trial will be conducted to evaluate Malachi's Inspiring Futures parenting programme. Participants will be allocated to one of two possible arms, with follow-up measures at 16 weeks (post-parent group programme) and at 32 weeks (post-one-to-one sessions with selected parents). The sample size is 248 participants with a randomisation allocation ratio of 1:1. The intervention arm will be offered the Inspiring Futures programme. The control group will receive services as usual. The aim is to determine the effectiveness of the Inspiring Futures programme on the primary outcome of behavioural and emotional difficulties of primary school children identified as having behavioural or emotional difficulties. Discussion: This study will further enhance the evidence for early intervention parenting programmes for child behavioural and emotional problems in the UK

    Direct Spectrum of the Benchmark T Dwarf HD 19467 B

    HD 19467 B is presently the only directly imaged T dwarf companion known to induce a measurable Doppler acceleration around a solar-type star. We present spectroscopy measurements of this important benchmark object taken with the Project 1640 integral field unit at Palomar Observatory. Our high-contrast R ≈ 30 observations obtained simultaneously across the JH bands confirm the cold nature of the companion as reported from the discovery article and determine its spectral type for the first time. Fitting the measured spectral energy distribution to SpeX/IRTF T dwarf standards and synthetic spectra from BT-Settl atmospheric models, we find that HD 19467 B is a T5.5 ± 1 dwarf with effective temperature T_eff=978^(+20)_(-43) K. Our observations reveal significant methane absorption affirming its substellar nature. HD 19467 B shows promise to become the first T dwarf that simultaneously reveals its mass, age, and metallicity independent from the spectrum of light that it emits
