    Skin colour changes during experimentally-induced sickness

AH is supported by a studentship from the Biotechnology and Biological Sciences Research Council.Skin colour may be an important cue to detect sickness in humans but how skin colour changes with acute sickness is currently unknown. To determine possible colour changes, 22 healthy Caucasian participants were injected twice, once with lipopolysaccharide (LPS, at a dose of 2 ng/kg body weight) and once with placebo (saline), in a randomised cross-over design study. Skin colour across 3 arm and 3 face locations was recorded spectrophotometrically over a period of 8 hours in terms of lightness (L∗), redness (a∗) and yellowness (b∗) in a manner that is consistent with human colour perception. In addition, carotenoid status was assessed as we predicted that a decrease it skin yellowness would reflect a drop in skin carotenoids. We found an early change in skin colouration 1-3 hours post LPS injection with facial skin becoming lighter and less red whilst arm skin become darker but also less red and less yellow. The LPS injection also caused a drop in plasma carotenoids from 3 hours onwards. However, the timing of the carotenoid changes was not consistent with the skin colour changes suggesting that other mechanisms, such as a reduction of blood perfusion, oxygenation or composition. This is the first experimental study characterising skin colour associated with acute illness, and shows that changes occur early in the development of the sickness response. Colour changes may serve as a cue to health, prompting actions from others in terms of care-giving or disease avoidance. Specific mechanisms underlying these colour changes require further investigation.PostprintPeer reviewe

    Detection of Inflammation via Volatile Cues in Human Urine

    Contagious disease is a major threat to survival, and the cost of relying on the immune system to defeat pathogens is high; therefore, behavioral avoidance of contagious individuals is arguably an adaptive strategy. Animal findings demonstrate the ability to detect and avoid sick individuals by the aid of olfactory cues, and a recent study indicated that human axillary odor also becomes more aversive as a function of immune activation. By injecting healthy human participants with lipopolysaccharide (0.6 ng/kg body weight) to experimentally induce inflammation, this study demonstrates that natural daily rhythms of urine odor—its perceived dimensions and volatile profile—are altered within hours of inflammation onset. Whereas healthy human urine decreases in averseness over the course of a single day, inflammation interrupts this process and results in an increased urine odor averseness and an altered volatile composition. These results support the notion that subtle and early cues of sickness may be detected and avoided, thereby complementing the immune system in its role of keeping us alive and healthy

    Olfactory cues of naturally occurring systemic inflammation: A pilot study of seasonal allergy

    Introduction: In an attempt to avoid contact with infectious individuals, humans likely respond to generalised rather than specific markers of disease. Humans may thus perceive a non-infectious individual as socially less attractive if they look (e.g., have facial discoloration), move (e.g., have a slower walking pace), or sound (e.g., sneeze) sick. This pilot study tested whether humans are averse to the body odour of non-infectious individuals with a low-grade systemic inflammation. Methods: We collected the axillary body odour of individuals with severe seasonal allergy (N = 14) and healthy controls (N = 10) during and outside the allergy season and measured serum levels of two inflammatory cytokines (tumor necrosis factor-α, interleukin-5). Independent participants (N = 67) then sampled and rated these odours on intensity and pleasantness. Results: While individuals with seasonal allergy had nominally more unpleasant and intense body odours during the allergy season - relative to outside of the allergy season and to healthy controls - these effects were not significant. When examining immune markers, the change in perceived pleasantness of an individual’s body odour (from out- to inside pollen season), was significantly related to the change in their interleukin-5 levels but not to tumor necrosis factor-α. Discussion: Our findings tentatively suggest that the human olfactory system could be sensitive to inflammation as present in a non-communicable condition. Larger replications are required to determine the role of olfaction in the perception of infectious and non-infectious (e.g., chronic diseases) conditions.publishedVersio

    UV-Deprived Coloration Reduces Success in Mate Acquisition in Male Sand Lizards (Lacerta agilis)

    Background: Recent work on animal signals has revealed a wide occurrence of UV signals in tetrapods, in particular birds, but also in lizards (and perhaps other Squamate reptiles). Our previous work on the Swedish sand lizard (Lacerta agilis) has verified, both in correlative selection analyses in the wild and with laboratory and field experiments, the importance of the green ‘badge ’ on the body sides of adult males for securing mating opportunities, probably mostly through deterring rival males rather than attracting females. The role of UV in communication has, however, never been examined. Methodology/Principal Findings: Here we show that when measured immediately after spring skin shedding, there is also signaling in the UV. By UV-depriving the signal (reflectance) with sun block chemicals fixated with permeable, harmless spray dressing, we show that males in the control group (spray dressing only) had significantly higher success in mate acquisition than UV-deprived males. Conclusions/Significance: These results suggest that at least two colour traits in sand lizards, badge area and UV, contribute to rival deterrence and/or female choice on UV characters, which elevates success in mate acquisition in UV intact male sand lizards

    Vortex glass transition in a random pinning model

    We study the vortex glass transition in disordered high temperature superconductors using Monte Carlo simulations. We use a random pinning model with strong point-correlated quenched disorder, a net applied magnetic field, longrange vortex interactions, and periodic boundary conditions. From a finite size scaling study of the helicity modulus, the RMS current, and the resistivity, we obtain critical exponents at the phase transition. The new exponents differ substantially from those of the gauge glass model, but are consistent with those of the pure three-dimensional XY model.Comment: 7 pages RevTeX, 4 eps figure

    Monte Carlo calculation of the current-voltage characteristics of a two dimensional lattice Coulomb gas

    We have studied the nonlinear current-voltage characteristic of a two dimensional lattice Coulomb gas by Monte Carlo simulation. We present three different determinations of the power-law exponent a(T)a(T) of the nonlinear current-voltage characteristic, V∼Ia(T)+1V \sim I^{a(T)+1}. The determinations rely on both equilibrium and non-equilibrium simulations. We find good agreement between the different determinations, and our results also agree closely with experimental results for Hg-Xe thin film superconductors and for certain single crystal thin-film high temperature superconductors.Comment: late

    The Transcription Factors SOX9 and SOX10 Are Vitiligo Autoantigens in Autoimmune Polyendocrine Syndrome Type I

    Vitiligo is common in the hereditary disorder autoimmune polyendocrine syndrome type I (APS I). Patients with APS I are known to have high titer autoantibodies directed against various tissue-specific antigens. Using sera from APS I patients for immunoscreening of a cDNA library from human scalp, we identified the transcription factors SOX9 and SOX10 as novel autoantigens related to this syndrome. Immunoreactivity against SOX9 was found in 14 (15%) and against SOX10 in 20 (22%) of the 91 APS I sera studied. All patients reacting with SOX9 displayed reactivity against SOX10, suggesting shared epitopes. Among the 19 patients with vitiligo, 12 (63%) were positive for SOX10 (p0.0001). Furthermore, three of 93 sera from patients with vitiligo unrelated to APS I showed strong reactivity against SOX10, which may indicate a more general role of SOX10 as an autoantigen in vitiligo

    Stable trapping of multiple proteins at physiological conditions using nanoscale chambers with macromolecular gates

    The possibility to detect and analyze single or few biological molecules is very important for understanding interactions and reaction mechanisms. Ideally, the molecules should be confined to a nanoscale volume so that the observation time by optical methods can be extended. However, it has proven difficult to develop reliable, non-invasive trapping techniques for biomolecules under physiological conditions. Here we present a platform for long-term tether-free (solution phase) trapping of proteins without exposing them to any field gradient forces. We show that a responsive polymer brush can make solid state nanopores switch between a fully open and a fully closed state with respect to proteins, while always allowing the passage of solvent, ions and small molecules. This makes it possible to trap a very high number of proteins (500-1000) inside nanoscale chambers as small as one attoliter, reaching concentrations up to 60 gL−1. Our method is fully compatible with parallelization by imaging arrays of nanochambers. Additionally, we show that enzymatic cascade reactions can be performed with multiple native enzymes under full nanoscale confinement and steady supply of reactants. This platform will greatly extend the possibilities to optically analyze interactions involving multiple proteins, such as the dynamics of oligomerization events

    Monte Carlo simulation of a two-dimensional continuum Coulomb gas

    We study the classical two-dimensional Coulomb gas model for thermal vortex fluctuations in thin superconducting/superfluid films by Monte Carlo simulation of a grand canonical vortex ensemble defined on a continuum. The Kosterlitz-Thouless transition is well understood at low vortex density, but at high vortex density the nature of the phase diagram and of the vortex phase transition is less clear. From our Monte Carlo data we construct phase diagrams for the 2D Coulomb gas without any restrictions on the vortex density. For negative vortex chemical potential (positive vortex core energy) we always find a Kosterlitz-Thouless transition. Only if the Coulomb interaction is supplemented with a short-distance repulsion, a first order transition line is found, above some positive value of the vortex chemical potential.Comment: 10 pages RevTeX, 7 postscript figures included using eps
