    The Genetic Profile from HLA and Non-HLA Loci Allows Identification of Atypical Type 2 Diabetes Patients

    The complex diagnosis and treatment of diabetes highlight the need for markers to define how to monitor patients correctly during the course of their disease. Different studies demonstrate the existence of patients who cannot be clearly classified. We have previously shown that it is possible to differentiate “atypical diabetic patients” based on genotyping the HLA. In this work we show that the analysis of non-HLA related to type 1 diabetes in the INS-VNTR, SNP rs689, and rs3842753 improves the identification of these patients. We genotyped 913 individuals comprising controls from the general population and “classic” and “atypical” diabetic patients. We compared the distribution of these loci and analyzed linkage disequilibrium. The haplotype was in LD for all the SNPs that were evaluated. Regarding their association with the disease, the haplotype IAC was associated with type 1 (odds 2.60, 1.82–3.72, CI 95%) and “atypical diabetes” (odds 1.50, 1.01–2.23, CI 95%), whereas we did not observe an association with type 2 diabetes. Therefore, our results confirm that atypical diabetes is a different entity of the disease where the patient presents with a genetic background of T1D and a T2D phenotype, findings that are likely to be relevant for patient diagnosis and management in the clinic

    O objetivo deste trabalho foi avaliar a digestibilidade e o tempo de trânsito gastrintestinal (TTGI) de dietas contendo níveis crescentes de fibra bruta (FB) para pacus. Para avaliar a digestibilidade foram utilizados 288 pacus, em delineamento inteiramente casualizado, alimentados em aquários e transferidos para coletores do tipo GuelfModificado, utilizando-se o método de coleta parcial de fezes. As rações foram marcadas com 1% de óxido de crômio para a determinação da digestibilidade das dietas. No ensaio de TTGI, 288 pacus foram distribuídos em 24 aquários em delineamento inteiramente casualizado e alimentados com rações contendo 1% óxido de titânio ou crômio, queapresentam cores diferentes, verde ou branca, respectivamente. Por meio de massagem abdominal foi averiguada periodicamente a cor das fezes, e o TTGI foi estabelecido quando as fezes de todos os peixes apresentaram cor verde. Os coeficientes de digestibilidade aparente das dietas e o TTGI foram reduzidos com aumento do nível de FB nas dietas. Conclui-se que em dietas para pacus pode-se empregar até 9% de FB sem efeito negativo na digestibilidade da energia, proteína, matéria seca e estrato etéreo e sem alterar o TTGI.This work aimed to evaluate the effect of increasing dietary fiber levels on the digestibility and gastrointestinal transit time (GTT) for pacu. The digestibility trial used 288 pacu in a completely randomized design, fed in aquaria and transferred to a modified Guelf-type feces collector, using the partial sampling method. The inert marker on the diets was chromic oxide (1%) to establish the diet digestibility. In the GTT assay, 288 pacu were distributed in 24 aquaria in a completely randomized design. Fish were fed with two colored diets – green and white – and feces color was determinedafter abdominal pressure. The white and green diets used 1% of titanium and chromic oxides, respectively. Total GTT was determined when all fish showed green feces. The apparent digestibility coefficients and GTT of diets decreased as dietary fiber levelsincreased. These results indicated that 9% dietary fiber can be used in pacu diets without negative effects on energy, protein, crude fiber and ether extract digestibility and GTT

    This study evaluated the apparent protein and energy digestibility of common feed ingredients (soybean meal, fish meal, wheat meal and corn) by juvenile oscars using two different sampling intervals (30 min. and 12h). The 160 juvenile oscar fish tested (22.37 ± 3.06 g BW) were divided into four cylindrical plastic net cages, each one placed in a 1000 L feeding tank. The experiment was completely randomized in a 2 x 4 factorial design (2 feces collection intervals and 4 feed ingredients) with four replications. The statistical tests did not detect an interaction effect of sampling interval and type of ingredient on digestibility coefficients. Sampling interval did not affect protein and energy digestibility. The physical characteristics of juvenile oscar feces likely make them less susceptible to nutrient loss by leaching and can therefore be collected at longer intervals. Protein digestibility of the different ingredients was similar, showing that apparent digestibility of both animal and plant ingredients by juvenile oscars was efficient. Energy digestibility coefficients of fish meal and soybean meal were higher than those of wheat meal and corn. Carbohydrate-rich ingredients (wheat meal and corn) had the worst energy digestibility coefficients and are therefore not used efficiently by juvenile oscars
