    Approximate Entropy of Spiking Series Reveals Different Dynamical States in Cortical Assemblies

    Self-organized criticality theory proved that information transmission and computational performances of neural networks are optimal in critical state. By using recordings of the spontaneous activity originated by dissociated neuronal assemblies coupled to Micro-Electrode Arrays (MEAs), we tested this hypothesis using Approximate Entropy (ApEn) as a measure of complexity and information transfer. We analysed 60 min of electrophysiological activity of three neuronal cultures exhibiting either sub-critical, critical or super-critical behaviour. The firing patterns on each electrode was studied in terms of the inter-spike interval (ISI), whose complexity was quantified using ApEn. We assessed that in critical state the local complexity (measured in terms of ApEn) is larger than in sub-and super-critical conditions (mean \ub1 std, ApEn about 0.93 \ub1 0.09, 0.66 \ub1 0.18, 0.49 \ub1 0.27, for the cultures in critical, sub-critical and super-critical state, respectively\u2014differences statistically significant). Our estimations were stable when considering epochs as short as 5 min (pairwise cross-correlation of spatial distribution of mean ApEn of 94 \ub1 5%). These preliminary results indicate that ApEn has the potential of being a reliable and stable index to monitor local information transmission in a neuronal network during maturation. Thus, ApEn applied on ISI time series appears to be potentially useful to reflect the overall complex behaviour of the neural network, even monitoring a single specific location

    Improved modelling of liquid GeSe2_2: the impact of the exchange-correlation functional

    The structural properties of liquid GeSe2_2 are studied by using first-principles molecular dynamics in conjuncton with the Becke, Lee, Yang and Parr (BLYP) generalized gradient approximation for the exchange and correlation energy. The results on partial pair correlation functions, coordination numbers, bond angle distributions and partial structure factors are compared with available experimental data and with previous first-principle molecular dynamics results obtained within the Perdew and Wang (PW) generalized gradient approximation for the exchange and correlation energy. We found that the BLYP approach substantially improves upon the PW one in the case of the short-range properties. In particular, the Ge−-Ge pair correlation function takes a more structured profile that includes a marked first peak due to homopolar bonds, a first maximum exhibiting a clear shoulder and a deep minimum, all these features being absent in the previous PW results. Overall, the amount of tetrahedral order is significantly increased, in spite of a larger number of Ge−-Ge homopolar connections. Due to the smaller number of miscoordinations, diffusion coefficients obtained by the present BLYP calculation are smaller by at least one order of magnitude than in the PW case.Comment: 6 figure

    Selective modulation of chemical and electrical synapses of Helix neuronal networks during in vitro development

    BACKGROUND: A large number of invertebrate models, including the snail Helix, emerged as particularly suitable tools for investigating the formation of synapses and the specificity of neuronal connectivity. Helix neurons can be individually identified and isolated in cell culture, showing well-conserved size, position, biophysical properties, synaptic connections, and physiological functions. Although we previously showed the potential usefulness of Helix polysynaptic circuits, a full characterization of synaptic connectivity and its dynamics during network development has not been performed. RESULTS: In this paper, we systematically investigated the in vitro formation of polysynaptic circuits, among Helix B2 and the serotonergic C1 neurons, from a morphological and functional point of view. Since these cells are generally silent in culture, networks were chemically stimulated with either high extracellular potassium concentrations or, alternatively, serotonin. Potassium induced a transient depolarization of all neurons. On the other hand, we found prolonged firing activity, selectively maintained following the first serotonin application. Statistical analysis revealed no significant changes in neuronal dynamics during network development. Moreover, we demonstrated that the cell-selective effect of serotonin was also responsible for short-lasting alterations in C1 excitability, without long-term rebounds. Estimation of the functional connections by means of cross-correlation analysis revealed that networks under elevated KCl concentrations exhibited strongly correlated signals with short latencies (about 5 ms), typical of electrically coupled cells. Conversely, neurons treated with serotonin were weakly connected with longer latencies (exceeding 20 ms) between the interacting neurons. Finally, we clearly demonstrated that these two types of correlations (in terms of strength/latency) were effectively related to the presence of electrical or chemical connections, by comparing Micro-Electrode Array (MEA) signal traces with intracellularly recorded cell pairs. CONCLUSIONS: Networks treated with either potassium or serotonin were predominantly interconnected through electrical or chemical connections, respectively. Furthermore, B2 response and short-term increase in C1 excitability induced by serotonin is sufficient to trigger spontaneous activity with chemical connections, an important requisite for long-term maintenance of firing activity

    Photoelectron spectra of aluminum cluster anions: Temperature effects and ab initio simulations

    Photoelectron (PES) spectra from aluminum cluster anions (from 12 to 15 atoms) at various temperature regimes, were studied using ab-initio molecular dynamics simulations and experimentally. The calculated PES spectra, obtained via shifting of the simulated electronic densities of states by the self-consistently determined values of the asymptotic exchange-correlation potential, agree well with the measured ones, allowing reliable structural assignments and theoretical estimation of the clusters' temperatures.Comment: RevTex, 3 gif figures.     Thalamocortical bistable switch as a theoretical model of fibromyalgia pathogenesis inferred from a literature survey

    Fibromyalgia (FM) is an unsolved central pain processing disturbance. We aim to provide a unifying model for FM pathogenesis based on a loop network involving thalamocortical regions, i.e., the ventroposterior lateral thalamus (VPL), the somatosensory cortex (SC), and the thalamic reticular nucleus (TRN). The dynamics of the loop have been described by three differential equations having neuron mean firing rates as variables and containing Hill functions to model mutual interactions among the loop elements. A computational analysis conducted with MATLAB has shown a transition from monostability to bistability of the loop behavior for a weakening of GABAergic transmission between TRN and VPL. This involves the appearance of a high-firing-rate steady state, which becomes dominant and is assumed to represent pathogenic pain processing giving rise to chronic pain. Our model is consistent with a bulk of literature evidence, such as neuroimaging and pharmacological data collected on FM patients, and with correlations between FM and immunoendocrine conditions, such as stress, perimenopause, chronic inflammation, obesity, and chronic dizziness. The model suggests that critical targets for FM treatment are to be found among immunoendocrine pathways leading to GABA/glutamate imbalance having an impact on the thalamocortical system

    Human kallikrein gene 5 (KLK5) expression is an indicator of poor prognosis in ovarian cancer

    Kallikrein gene 5 (KLK5, also known as KLK-L2), located on chromosome 19q13.4, is one of the newly identified members of the kallikrein gene family, which is a subgroup of the serine protease enzyme family. In normal human tissues, KLK5 is highly expressed in skin, mammary gland and testis. Preliminary RT-PCR analysis has indicated that KLK5 is expressed in a subset of ovarian tumours. We have thus hypothesized that KLK5 may be a new prognostic indicator in ovarian cancer. We have examined the mRNA expression of KLK5 in 142 malignant ovarian tissues. Tumours were pulverized, total RNA was extracted, and cDNA was prepared by reverse transcription. KLK5 was amplified by PCR using gene specific primers, and the identity of the PCR product was verified by sequencing. Ovarian tissues were then classified as KLK5 positive or negative, based on ethidium bromide staining of the PCR product on agarose gels. KLK5 was found to be highly expressed in 58/142 (41%) of ovarian cancer samples while its level of expression was very low in normal ovarian tissues. We found a strong positive relation between KLK5 expression and tumour grade (P = 0.006) and disease stage (P = 0.027). Univariate survival analysis revealed that patients with ovarian tumours positive for KLK5 expression had an increased risk for relapse and death (P = 0.018 and 0.022, respectively). In multivariate analysis, KLK5 expression showed independent prognostic value only in the subset of tumours with lower grade disease (grades I and II). We conclude that KLK5 expression is associated with more aggressive forms of epithelial ovarian carcinoma and has indepdent prognostic value in low grade tumours. © 2001 Cancer Research Campaign http://www.bjcancer.co

    Análisis fractal de la precipitación anual en el este de la Provincia de La Pampa, Argentina

    El este de la Provincia de La Pampa (Argentina) se ubica en la franja de transición entre el área sudoccidental de la Región Pampeana, para la cual el documento oficial del Panel Intergubernamental de Cambio Climático (IPCC) proyecta un descenso en las precipitaciones de entre 10 a un 20%, y el área nordeste, para la cual proyecta un ascenso de entre un 10 a un 20%. Con la finalidad de determinar en cuál de las dos tendencias se encuentra comprendido el este de la Provincia de La Pampa (Argentina) se estudió la dimensión fractal de 10 series de precipitación anual de la zona aplicando el coeficiente de Hurst (H), a fin de determinar si las mismas muestran persistencia, y se evaluó, por medio de la prueba de Mann Kendall cuáles fueron la significación y signo de las tendencias observadas. Los resultados confirman que la precipitación anual en el este de la Provincia de La Pampa, Argentina puede ser caracterizada a través de su dimensión fractal, alcanzando valores de dimensión fractal D cercanos a 1, que corroboran un alto nivel de persistencia. Las tendencias positivas estadísticamente significativas a muy significativas exhibidas por todas las localidades utilizadas indican que el régimen de precipitaciones de la zona observó un incremento temporal de sus valores dentro del período analizado, por lo que puede concluirse que el este de la provincia de La Pampa se encuentra incluido dentro del área oriental prevista por el IPCC.Eastern La Pampa Province (Argentina) is located in the transition band between the south-western area of the Pampa Region, where the Intergovernmental Panel Climatic Change (IPCC) official document projects a decrease in the rainfalls between 10 to 20%, and the north-eastern area, where an increase between 10 to 20% is projected. With the purpose of determining in which of the two trends the Eastern La Pampa Province (Argentina) is included, the fractal dimension of 10 series of annual rainfall of the zone was studied by applying the Hurst coefficient (H), in order to determine their degree of persistence, and the significance and sign of the observed trends were evaluated by means of the Mann Kendall test. The results confirm that the annual rainfall in the Eastern La Pampa Province (Argentina) can be characterized by means of his fractal dimension, reaching values of fractal dimension D near to 1, which corroborate a high level of persistence. The positive statistically significant to very significant trends exhibited by all localities indicate that the regime of rainfalls of the zone observed an increase within the analyzed period, allowing the conclusion that Eastern La Pampa Province is included in the north-eastern area foreseen by the IPCC.Fil: Perez, Silvia P..     Supercell technique for total-energy calculations of finite charged and polar systems

    We study the behavior of total-energy supercell calculations for dipolar molecules and charged clusters. Using a cutoff Coulomb interaction within the framework of a plane-wave basis set formalism, with all other aspects of the method (pseudopotentials, basis set, exchange-correlation functional) unchanged, we are able to assess directly the interaction effects present in the supercell technique. We find that the supercell method gives structures and energies in almost total agreement with the results of calculations for finite systems, even for molecules with large dipole moments. We also show that the performance of finite-grid calculations can be improved by allowing a degree of aliasing in the Hartree energy, and by using a reciprocal space definition of the cutoff Coulomb interaction
