    Sull'origine delle terre rosse in Sardegna: gli elementi in traccia nella caratterizzazione di suoli e rocce madri

    The trace elements in the characterization or red soils and parent rocks. The genesis of red soils (Terre Rosse) formed on limestone or dolomite rock bed is yet an unsolved question. A theory suggests that these soils are the final step of an intense decarbonation process of the parent rock followed by the change of the materials in the insoluble residue into iron oxides and clay minerals. A number of trace elements, most1y transition metals, was determined in Sardinian Terre Rosse and parent rocks by instrumental neutron activation analysis performing different irradiations in the TRIGA Mark II nuclear reactor of the University of Pavia. Induced radioactivity measurement was carried out by gamma-ray spectrometry using a High Purity germanium detector coupled to an analyzer-computer system. The same elements were also determined in some standard reference rocks, released by United States Geological Survey, in order to evaluate the accuracy of the employed analytical method. Average values of the trace element content in the Terre Rosse and in the parent rocks are presented and discussed, together with the evaluation of precision and accuracy. Trace element profiles at different horizons are reported as well. A comparison of trace element distribution among soils belonging to the same geological era is also presented

    Seasonal response of benthic foraminifera to anthropogenic pressure in two stations of the Gulf of Trieste (northern Adriatic Sea, Italy): the marine protected area of Miramare versus the Servola water sewage outfall

    A seasonal survey of living benthic foraminifera was performed in 2013 in the Gulf of Trieste (N Adriatic Sea) to compare two marine coastal sites with different degrees of anthropogenic influence. An assessment of ecological quality statuses showed that the station located near the end of an urban pipeline (Ser station), has worse ecological conditions than the site located in a protected marine area (Res station) all year around. Stressed conditions at Ser station were mainly related to high contents of total organic carbon (TOC) and Zn in the bioavailable fraction, which were a limiting factor for the studied foraminiferal communities. Ammonia tepida, Bolivina spp., and Bulimina spp., which characterised this station, were the most tolerant taxa of the studied assemblage. Conversely, Elphidium spp., H. depressula, N. iridea, Quiqueloculina spp., R. nana and Textularia spp., could be considered less tolerant species as they benefitted from the less stressful conditions recorded at Res station, despite slightly higher concentrations of some potentially toxic elements (PTEs), especially Pb, being recorded in this station in comparison to Ser station. Furthermore, foraminiferal assemblages were found to be quite resilient over an annual cycle, being able to recover from a seasonal unbalanced state to a mature one. The beginning of spring and latest summer would be the best period to assess the ecological quality status to avoid any under- or overestimation of the health of the environment

    Responsiveness to 6-n-Propylthiouracil (PROP) Is Associated with Salivary Levels of Two Specific Basic Proline-Rich Proteins in Humans

    Thiourea tasting can be predictive of individual differences in bitter taste responses, general food preferences and eating behavior, and could be correlated with saliva chemical composition. We investigated the possible relationship between PROP bitter taste responsiveness and the salivary proteome in subjects genotyped for TAS2R38 and gustin gene polymorphisms. Taste perception intensity evoked by PROP and NaCl solutions was measured in sixty-three volunteers (21 males, 42 females, age 25±3 y) to establish their PROP taster status, and 24 PROP super-tasters and 21 nontasters were selected to participate in the study. TAS2R38 and gustin gene molecular analysis were performed using PCR techniques. Qualitative and quantitative determination of salivary proteins was performed by HPLC-ESI-MS before and after PROP taste stimulation. PROP super-tastings was strongly associated with the ‘taster’ variant (PAV haplotype) of TAS2R38 and the A allele of rs2274333 polymorphism in the gustin gene and nontasting was associated with the minor alleles at both loci. ANOVA revealed that basal levels of II-2 and Ps-1 proteins, belonging to the basic proline-rich protein (bPRPs) family, were significantly higher in PROP super-taster than in nontaster un-stimulated saliva, and that PROP stimulation elicited a rapid increase in the levels of these same proteins only in PROP super-taster saliva. These data show for the first time that responsiveness to PROP is associated with salivary levels of II-2 peptide and Ps-1 protein, which are products of the PRB1 gene. These findings suggest that PRB1, in addition to TAS2R38 and gustin, could contribute to individual differences in thiourea sensitivity, and the expression of the PROP phenotype as a complex genetic trait

    PROP responsiveness is associated with TAS2R38 haplotypes and fungiform papilla density. Recently, we showed that a polymorphism in the gene coding for the salivary trophic factor, gustin (CA6), affects PROP sensitivity by acting on cell growth and fungiform papillae maintenance, in a genetically homogeneous cohort. Since population homogeneity can lead to over estimation of gene effects, the primary aim of the present work was to confirm gustin’s role in PROP bitterness intensity and fungiform papillae density in a genetically diverse population. Eighty subjects were genotyped for both genes by PCR techniques. PROP responsiveness was assessed by filter paper method and fungiform papilla density was determined in each subject. As expected, PROP bitterness ratings were lower in individuals with the AVI/AVI diplotype of TAS2R38 than in individuals with PAV/PAV and PAV/AVI diplotype. However, no differences in PROP bitterness among genotypes of the gustin gene, and no differences in the density of fungiform papillae related to TAS2R38 diplotype were found. In contrast, the density of fungiform papillae decreased as the number of minor (G) alleles at the gustin locus increased. In addition, the distribution of TAS2R38 genotypes within each gustin genotype group showed that the occurrence of recessive alleles at both loci was infrequent in the present sample compared to other populations. These findings confirm that papillae density is associated with gustin gene polymorphism, rs2274333 (A/G), in an ancestrally heterogeneous population, and suggest that variations in the frequency of allele combinations for these two genes could provide a salient explanation for discrepant findings for gustin gene effects across population

    Dose-Dependent Effects of L-Arginine on PROP Bitterness Intensity and Latency and Characteristics of the Chemical Interaction between PROP and L-Arginine

    Genetic variation in the ability to taste the bitterness of 6-n-propylthiouracil (PROP) is a complex trait that has been used to predict food preferences and eating habits. PROP tasting is primarily controlled by polymorphisms in the TAS2R38 gene. However, a variety of factors are known to modify the phenotype. Principle among them is the salivary protein Ps-1 belonging to the basic proline-rich protein family (bPRP). Recently, we showed that oral supplementation with Ps-1 as well as its related free amino acids (L-Arg and L-Lys) enhances PROP bitterness perception, especially for PROP non-tasters who have low salivary levels of Ps-1. Here, we show that salivary L-Arg levels are higher in PROP super-tasters compared to medium tasters and non-tasters, and that oral supplementation with free L-Arg enhances PROP bitterness intensity as well as reduces bitterness latency in a dose-dependent manner, particularly in individuals with low salivary levels of both free L-Arg and Ps-1 protein. Supplementation with L-Arg also enhanced the bitterness of caffeine. We also used 1H-NMR spectroscopy and quantum-mechanical calculations carried out by Density Functional Theory (DFT) to characterize the chemical interaction between free L-Arg and the PROP molecule. Results showed that the -NH2 terminal group of the L-ArgH+ side chain interacts with the carbonyl or thiocarbonyl groups of PROP by forming two hydrogen bonds with the resulting charged adduct. The formation of this PROP\u2022ArgH+ hydrogen-bonded adduct could enhance bitterness intensity by increasing the solubility of PROP in saliva and its availability to receptor sites. Our data suggest that L-Arg could act as a 'carrier' of various bitter molecules in saliva

    BEACON: A Cloud Network Federation Framework

    This paper presents the BEACON Framework, which will enable the provision and management of cross-site virtual networks for federated cloud infrastructures in order to support the automated deployment of applications and services across different clouds and datacenters. The proposed framework will support different federation architectures, going from tightly coupled (datacenter federation) to loosely coupled (cloud federation and multi-cloud orchestration) architectures, and will enable the creation of Layer 2 and Layer 3 overlay networks to interconnect remote resources located at different cloud sites. A high level description of the main components of the BEACON framework is also introduced
