    Protocol for a realist review of workplace learning in postgraduate medical education and training

    Postgraduate medical education and training (PGMET) is a complex social process which happens predominantly during the delivery of patient care. The clinical learning environment (CLE), the context for PGMET, shapes the development of the doctors who learn and work within it, ultimately impacting the quality and safety of patient care. Clinical workplaces are complex, dynamic systems in which learning emerges from non-linear interactions within a network of related factors and activities. Those tasked with the design and delivery of postgraduate medical education and training need to understand the relationship between the processes of medical workplace learning and these contextual elements in order to optimise conditions for learning. We propose to conduct a realist synthesis of the literature to address the overarching questions; how, why and in what circumstances do doctors learn in clinical environments? This review is part of a funded projected with the overall aim of producing guidelines and recommendations for the design of high quality clinical learning environments for postgraduate medical education and training

    The University of Nebraska–Lincoln Libraries offers several avenues for preserving and providing access to digital and physical research materials. This document outlines the four main avenues for depositing materials with UNL Libraries. Although there are separate repositories with specific missions—Archives & Special Collections, DigitalCommons@University of Nebraska–Lincoln, UNL Data Repository, and UNL Image & Multimedia Collections—all work together toward the goal of preserving the intellectual and creative output of the university and to make our contributions discoverable to state, national, and international communities. This document describes the operations of each repository. The Libraries policy is to publish, or provide online access to, materials (1) when the Libraries holds copyright, (2) when the copyright holder has granted the Libraries permission for online publication, or (3) when the Libraries do not hold copyright but may manage access behind a firewall. The University Libraries is committed to preserving and providing access to the full range of in-tellectual contributions of the faculty and staff at UNL for the benefit of current and future gen-erations. All members of the University of Nebraska–Lincoln are encouraged to deposit content with UNL Libraries. Materials deposited in our institutional repositories are historical and not all historical events confirm to current standards of civility.     Design and methods of the Longitudinal Eating Disorders Assessment Project research consortium for veterans

    Introduction Military service members must maintain a certain body mass index and body fat percentage. Due to weight-loss pressures, some service members may resort to unhealthy behaviors that place them at risk for the development of an eating disorder (ED). Objectives To understand the scope and impact of EDs in military service members and veterans, we formed the Longitudinal Eating Disorders Assessment Project (LEAP) Consortium. LEAP aims to develop novel screening, assessment, classification, and treatment tools for veterans and military members with a focus on EDs and internalizing psychopathology. Methods We recruited two independent nationally representative samples of post-9/11 veterans who were separated from service within the past year. Study 1 was a four-wave longitudinal survey and Study 2 was a mixed-methods study that included surveys, structured-clinical interviews, and qualitative interviews. Results Recruitment samples were representative of the full population of recently separated veterans. Sample weights were created to adjust for sources of non-response bias to the baseline survey. Attrition was low relative to past studies of this population, with only (younger) age predicting attrition at 1-week follow-up. Conclusions We expect that the LEAP Consortium data will contribute to improved information about EDs in veterans, a serious and understudied problem

    Temporospatial shifts in Sandhill Crane staging in the Central Platte River Valley in response to climatic variation and habitat change

    Over 80% of the Mid-Continent Sandhill Crane (Antigone canadensis) Population (MCP), estimated at over 660,000 individuals, stops in the Central Platte River Valley (CPRV) during spring migration from mid-February through mid-April. Research suggests that the MCP may be shifting its distribution spatially and temporally within the CPRV. From 2002 to 2017, we conducted weekly aerial surveys of Sandhill Cranes staging in the CPRV to examine temporal and spatial trends in their abundance and distribution. Then, we used winter temperature and drought severity measures from key wintering and early migratory stopover locations to assess the impacts of weather patterns on annual migration chronology in the CPRV. We also evaluated channel width and land cover characteristics using aerial imagery from 1938, 1998, and 2016 to assess the relationship between habitat change and the spatial distribution of the MCP in the CPRV. We used generalized linear models, cumulative link models, and Akaike’s information criterion corrected for small sample sizes (AICc) to compare temporal and spatial models. Temperatures and drought conditions at wintering and migration locations that are heavily used by Greater Sandhill Cranes (A. c. tabida) best predicted migration chronology of the MCP to the CPRV. The spatial distribution of roosting Sandhill Cranes from 2015 to 2017 was best predicted by the proportion of width reduction in the main channel since 1938 (rather than its width in 2016) and the proportion of land cover as prairie-meadow habitat within 800 m of the Platte River. Our data suggest that Sandhill Cranes advanced their migration by an average of just over 1 day per year from 2002 to 2017, and that they continued to shift eastward, concentrating at eastern reaches of the CPRV. Climate change, land use change, and habitat loss have all likely contributed to Sandhill Cranes coming earlier and staying longer in fewer reaches of the CPRV, increasing their site use intensity. These historically unprecedented densities may present a disease risk to Sandhill Cranes and other waterbirds, including Whooping Cranes (Grus americana). Our models suggest that conservation actions may be maintaining Sandhill Crane densities in areas that would otherwise be declining in use. We suggest that management actions intended to mitigate trends in the distribution of Sandhill Cranes, including wet meadow restoration, may similarly benefit prairie- and braided river–endemic species of concern. Más del 80% de la población de grullas canadienses (Antigone canadensis), de la zona central del continente (MCP por sus siglas en inglés), estimada en más de 660,000, descansa en el valle central del Río Platte (CPRV por sus siglas en inglés) durante su migración de primavera, desde mediados de febrero hasta mediados de abril. Diversos estudios indican que su distribución espacial y temporal podría estar cambiando dentro del CPRV. Desde el año 2002 hasta el 2017 realizamos sondeos aéreos semanales de grullas canadienses en el CPRV para estudiar las tendencias temporales y espaciales relacionadas a su abundancia y distribución. Usamos mediciones de temperatura durante el invierno y de la severidad de la sequía de lugares claves de invernada y de sitios de descanso durante su migración temprana para evaluar el impacto de los patrones climáticos en la cronología migratoria anual del CPRV. También analizamos la amplitud del canal y las características de la cubierta terrestre usando imágenes aéreas de 1938, 1998 y 2016 con el fin de evaluar la relación entre el cambio de hábitat y la distribución espacial de la MCP en el CPRV. Utilizamos modelos lineales generalizados, modelos de enlace acumulativo y el criterio de información de Akaike adecuados a muestras pequeñas (AICc), para comparar modelos temporales y espaciales. Las condiciones climáticas y de sequía en los sitios de invernada y migración más usados por la grulla canadiense mayor (A. c. tabida) predijeron mejor la cronología migratoria de la MCP en el CPRV. La reducción de la amplitud del canal principal desde 1938, junto con el porcentaje de cubierta terrestre como hábitat de pradera dentro de los 800 m del río Platte, fue el mejor predictor de la distribución espacial de la grulla canadiense desde el año 2015 hasta el 2017. Nuestros estudios indican que las grullas canadienses adelantaron su migración en un promedio poco más de un día por año entre el 2002 y el 2017 y que continuaron desplazándose hacia el este, concentrándose en los extremos orientales del CPRV. El cambio climático, el cambio de uso del suelo y la pérdida del hábitat probablemente contribuyeron a la migración temprana de esta especie y a su permanencia más prolongada en algunos sectores del CPRV, aumentando la intensidad del uso del sitio. Estas densidades sin precedentes podrían presentar un riesgo de enfermedad para la grulla canadiense y otras aves acuáticas, incluidas las grullas trompeteras (Grus americana).     Regulation of Neuronal APL-1 Expression by Cholesterol Starvation

    Background: Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by the deposition of b-amyloid plaques composed primarily of the amyloid-b peptide, a cleavage product of amyloid precursor protein (APP). While mutations in APP lead to the development of Familial Alzheimer’s Disease (FAD), sporadic AD has only one clear genetic modifier: the e4 allele of the apolipoprotein E (ApoE) gene. Cholesterol starvation in Caenorhabditis elegans leads to molting and arrest phenotypes similar to loss-of-function mutants of the APP ortholog, apl-1 (amyloid precursor-like protein 1), and lrp-1 (lipoprotein receptor-related protein 1), suggesting a potential interaction between apl-1 and cholesterol metabolism. Methodology/Principal Findings: Previously, we found that RNAi knock-down of apl-1 leads to aldicarb hypersensitivity, indicating a defect in synaptic function. Here we find the same defect is recapitulated during lrp-1 knock-down and by cholesterol starvation. A cholesterol-free diet or loss of lrp-1 directly affects APL-1 levels as both lead to loss of APL-1::GFP fluorescence in neurons. However, loss of cholesterol does not affect global transcription or protein levels as seen by qPCR and Western blot. Conclusions: Our results show that cholesterol and lrp-1 are involved in the regulation of synaptic transmission, similar to apl-1. Both are able to modulate APL-1 protein levels in neurons, however cholesterol changes do not affect global apl-1 transcription or APL-1 protein indicating the changes are specific to neurons.     Intracellular Trafficking and Synaptic Function of APL-1 in Caenorhabditis elegans

    Background: Alzheimer’s disease (AD) is a neurodegenerative disorder primarily characterized by the deposition of b-amyloid plaques in the brain. Plaques are composed of the amyloid-b peptide derived from cleavage of the amyloid precursor protein (APP). Mutations in APP lead to the development of Familial Alzheimer’s Disease (FAD), however, the normal function of this protein has proven elusive. The organism Caenorhabditis elegans is an attractive model as the amyloid precursor-like protein (APL-1) is the single ortholog of APP, and loss of apl-1 leads to a severe molting defect and early larval lethality. Methodology/Principal Findings: We report here that lethality and molting can be rescued by full length APL-1, C-terminal mutations as well as a C-terminal truncation, suggesting that the extracellular region of the protein is essential for viability. RNAi knock-down of apl-1 followed by drug testing on the acetylcholinesterase inhibitor aldicarb showed that loss of apl-1 leads to aldicarb hypersensitivity, indicating a defect in synaptic function. The aldicarb hypersensitivity can be rescued by full length APL-1 in a dose dependent fashion. At the cellular level, kinesins UNC-104/KIF-1A and UNC-116/kinesin-1 are positive regulators of APL-1 expression in the neurons. Knock-down of the small GTPase rab-5 also leads to a dramatic decrease in the amount of apl-1 expression in neurons, suggesting that trafficking from the plasma membrane to the early endosome is important for apl-1 function.     Mitotic Spindle Proteomics in Chinese Hamster Ovary Cells

    Mitosis is a fundamental process in the development of all organisms. The mitotic spindle guides the cell through mitosis as it mediates the segregation of chromosomes, the orientation of the cleavage furrow, and the progression of cell division. Birth defects and tissue-specific cancers often result from abnormalities in mitotic events. Here, we report a proteomic study of the mitotic spindle from Chinese Hamster Ovary (CHO) cells. Four different isolations of metaphase spindles were subjected to Multi-dimensional Protein Identification Technology (MudPIT) analysis and tandem mass spectrometry. We identified 1155 proteins and used Gene Ontology (GO) analysis to categorize proteins into cellular component groups. We then compared our data to the previously published CHO midbody proteome and identified proteins that are unique to the CHO spindle. Our data represent the first mitotic spindle proteome in CHO cells, which augments the list of mitotic spindle components from mammalian cells
