    Sex-partitioning of the <i>Plasmodium falciparum</i> stage V gametocyte proteome provides insight into <i>falciparum</i>-specific cell biology

    One of the critical gaps in malaria transmission biology and surveillance is our lack of knowledge about Plasmodium falciparum gametocyte biology, especially sexual dimorphic development and how sex ratios that may influence transmission from the human to the mosquito. Dissecting this process has been hampered by the lack of sex-specific protein markers for the circulating, mature stage V gametocytes. The current evidence suggests a high degree of conservation in gametocyte gene complement across Plasmodium, and therefore presumably for sex-specific genes as well. To better our understanding of gametocyte development and subsequent infectiousness to mosquitoes, we undertook a Systematic Subtractive Bioinformatic analysis (filtering) approach to identify sex-specific P. falciparum NF54 protein markers based on a comparison with the Dd2 strain, which is defective in producing males, and with syntenic male and female proteins from the reanalyzed and updated P. berghei (related rodent malaria parasite) gametocyte proteomes. This produced a short list of 174 male- and 258 female-enriched P. falciparum stage V proteins, some of which appear to be under strong diversifying selection, suggesting ongoing adaptation to mosquito vector species. We generated antibodies against three putative female-specific gametocyte stage V proteins in P. falciparum and confirmed either conserved sex-specificity or the lack of cross-species sex-partitioning. Finally, our study provides not only an additional resource for mass spectrometry-derived evidence for gametocyte proteins but also lays down the foundation for rational screening and development of novel sex-partitioned protein biomarkers and transmission-blocking vaccine candidates

    Systematic review of economic analyses in patient safety: a protocol designed to measure development in the scope and quality of evidence.

    Recent avoidable failures in patient care highlight the ongoing need for evidence to support improvements in patient safety. According to the most recent reviews, there is a dearth of economic evidence related to patient safety. These reviews characterise an evidence gap in terms of the scope and quality of evidence available to support resource allocation decisions. This protocol is designed to update and improve on the reviews previously conducted to determine the extent of methodological progress in economic analyses in patient safety. A broad search strategy with two core themes for original research (excluding opinion pieces and systematic reviews) in 'patient safety' and 'economic analyses' has been developed. Medline, Econlit and National Health Service Economic Evaluation Database bibliographic databases will be searched from January 2007 using a combination of medical subject headings terms and research-derived search terms (see table 1). The method is informed by previous reviews on this topic, published in 2012. Screening, risk of bias assessment (using the Cochrane collaboration tool) and economic evaluation quality assessment (using the Drummond checklist) will be conducted by two independent reviewers, with arbitration by a third reviewer as needed. Studies with a low risk of bias will be assessed using the Drummond checklist. High-quality economic evaluations are those that score &gt;20/35. A qualitative synthesis of evidence will be performed using a data collection tool to capture the study design(s) employed, population(s), setting(s), disease area(s), intervention(s) and outcome(s) studied. Methodological quality scores will be compared with previous reviews where possible. Effect size(s) and estimate uncertainty will be captured and used in a quantitative synthesis of high-quality evidence, where possible. Formal ethical approval is not required as primary data will not be collected. The results will be disseminated through a peer-reviewed publication, presentations and social media. CRD42017057853

    The Lotic Intersite Nitrogen Experiments: an example of successful ecological research collaboration

    Collaboration is an essential skill for modern ecologists because it brings together diverse expertise, viewpoints, and study systems. The Lotic Intersite Nitrogen eXperiments (LINX I and II), a 17-y research endeavor involving scores of early- to late-career stream ecologists, is an example of the benefits, challenges, and approaches of successful collaborative research in ecology. The scientific success of LINX reflected tangible attributes including clear scientific goals (hypothesis-driven research), coordinated research methods, a team of cooperative scientists, excellent leadership, extensive communication, and a philosophy of respect for input from all collaborators. Intangible aspects of the collaboration included camaraderie and strong team chemistry. LINX further benefited from being part of a discipline in which collaboration is a tradition, clear data-sharing and authorship guidelines, an approach that melded field experiments and modeling, and a shared collaborative goal in the form of a universal commitment to see the project and resulting data products through to completion

    Simultaneous Observations of GRS 1758-258 in 1997 by VLA, IRAM, SEST, RXTE and OSSE: Spectroscopy and Timing

    We report the results of our multi-wavelength observations of GRS 1758-258 made in August 1997. The energy bands include radio, millimeter, X-ray, and gamma-ray. The observations enable us to obtain a complete spectrum of the source over an energy range of 2 - 500 keV. The spectrum shows that GRS 1758-258 was in its hard state. It is well fitted by the Sunyaev-Titarchuk (ST) Compton scattering model. The spectrum is also fit by a power law with an exponential cutoff (PLE) plus a soft black-body component. The temperature of the soft component is about 1.2 keV, and the energy flux is less than 1.5% of the total X- and gamma-ray flux. The deduced hydrogen column density is in the range of (0.93 - 2.0) 10^{22} cm^{-2}. No significant iron lines are detected. The radio emission has a flat energy spectrum. The daily radio, X-ray and gamma-ray light curves show that GRS 1758-258 was stable during the observation period, but was highly variable on smaller time scales in X- and gamma-rays. The power density spectra are typical for the low-state, but we find the photon flux for the 5 to 10 keV band to be more variable than that in the other two energy bands (2 - 5 keV and 10 - 40 keV). Harmonically spaced quasi-periodic oscillations (QPOs) are observed in the power spectra. The phase lags between the hard photons and the soft photons have a flat distribution over a wide range of frequencies. A high coherence of about 1.0 (0.01 - 1 Hz) between the hard photons and the soft photons is also obtained in our observations. We compare these results with two variation models. Our millimeter observations did not reveal any conclusive signatures of an interaction between the jet from GRS 1758-258 and the molecular cloud that lies in the direction of GRS 1758-258.Comment: 32 pages, 13 figures, to appear in ApJ, 2000, V.533, no. 1, Apr. 10.     A Search for Very Active Stars in the Galaxy

    We report the first results of a systematic search near the plane of the Galaxy for the so called very active stars (VAS), which are characterized by a hard X-ray spectrum and activity in the radio domain. Candidates with hard X-ray binary-like spectra have been selected from the Bright ROSAT Source Catalogue in the Zone of Avoidance (b<20o| b | < 20{^o}) and were tentatively identified in GB6/PMM/NVSS radio surveys. Most of them were observed with the ATCA and VLA. Precise radio coordinates have led to unambiguous optical identification for 60 candidates, and a sub-sample of five of themhas been observed with the VLT. Also some discovery and confirmatory spectra were obtained with the AAT (4-m) and BTA (6-m). Spectroscopy with moderate dispersion, made with the FORS1 spectrograph of the VLT has revealed two stellar objects (one of them, VASC J1628-41, is definitivelya binary VAS), one new AGN and two featureless spectrum sources. One of these objects, VASC J1353-66, shows a marginal evidence of proper motion, which, if confirmed, would imply the discovery of a new type of galactic source.Comment: to appear in A&A, 7 figure

    An X-ray study of the SNR G344.7-0.1 and the central object CXOU J170357.8-414302

    Aims. We report results of an X-ray study of the supernova remnant (SNR) G344.7-0.1 and the point-like X-ray source located at the geometrical center of the SNR radio structure. Methods. The morphology and spectral properties of the remnant and the central X-ray point-like source were studied using data from the XMM-Newton and Chandra satellites. Archival radio data and infrared Spitzer observations at 8 and 24 μ\mum were used to compare and study its multi-band properties at different wavelengths. Results. The XMM-Newton and Chandra observations reveal that the overall X-ray emission of G344.7-0.1 is extended and correlates very well with regions of bright radio and infrared emission. The X-ray spectrum is dominated by prominent atomic emission lines. These characteristics suggest that the X-ray emission originated in a thin thermal plasma, whose radiation is represented well by a plane-parallel shock plasma model (PSHOCK). Our study favors the scenario in which G344.7-0.1 is a 6 x 10^3 year old SNR expanding in a medium with a high density gradient and is most likely encountering a molecular cloud on the western side. In addition, we report the discovery of a soft point-like X-ray source located at the geometrical center of the radio SNR structure. The object presents some characteristics of the so-called compact central objects (CCO). However, its neutral hydrogen absorption column (N_{H}) is inconsistent with that of the SNR. Coincident with the position of the source, we found infrared and optical objects with typical early-K star characteristics. The X-ray source may be a foreground star or the CCO associated with the SNR. If this latter possibility were confirmed, the point-like source would be the farthest CCO detected so far and the eighth member of the new population of isolated and weakly magnetized neutron stars.Comment: 9 pages, 8 figures, accepted for publication in Astronomy and Astrophysics.     A review on automatic mammographic density and parenchymal segmentation

    Breast cancer is the most frequently diagnosed cancer in women. However, the exact cause(s) of breast cancer still remains unknown. Early detection, precise identification of women at risk, and application of appropriate disease prevention measures are by far the most effective way to tackle breast cancer. There are more than 70 common genetic susceptibility factors included in the current non-image-based risk prediction models (e.g., the Gail and the Tyrer-Cuzick models). Image-based risk factors, such as mammographic densities and parenchymal patterns, have been established as biomarkers but have not been fully incorporated in the risk prediction models used for risk stratification in screening and/or measuring responsiveness to preventive approaches. Within computer aided mammography, automatic mammographic tissue segmentation methods have been developed for estimation of breast tissue composition to facilitate mammographic risk assessment. This paper presents a comprehensive review of automatic mammographic tissue segmentation methodologies developed over the past two decades and the evidence for risk assessment/density classification using segmentation. The aim of this review is to analyse how engineering advances have progressed and the impact automatic mammographic tissue segmentation has in a clinical environment, as well as to understand the current research gaps with respect to the incorporation of image-based risk factors in non-image-based risk prediction models

    A Comparison of the Morphology and Stability of Relativistic and Nonrelativistic Jets

    We compare results from a relativistic and a nonrelativistic set of 2D axisymmetric jet simulations. For a set of five relativistic simulations that either increase the Lorentz factor or decrease the adiabatic index we compute nonrelativistic simulations with equal useful power or thrust. We examine these simulations for morphological and dynamical differences, focusing on the velocity field, the width of the cocoon, the age of the jets, and the internal structure of the jet itself. The primary result of these comparisons is that the velocity field of nonrelativistic jet simulations cannot be scaled up to give the spatial distribution of Lorentz factors seen in relativistic simulations. Since the local Lorentz factor plays a major role in determining the total intensity for parsec scale extragalactic jets, this suggests that a nonrelativistic simulation cannot yield the proper intensity distribution for a relativistic jet. Another general result is that each relativistic jet and its nonrelativistic equivalents have similar ages (in dynamical time units, = R/a_a, where R is the initial radius of a cylindrical jet and a_a is the sound speed in the ambient medium). In addition to these comparisons, we have completed four new relativistic simulations to investigate the effect of varying thermal pressure on relativistic jets. The simulations generally confirm that faster (larger Lorentz factor) and colder jets are more stable, with smaller amplitude and longer wavelength internal variations. The apparent stability of these jets does not follow from linear normal mode analysis, which suggests that there are available growing Kelvin-Helmholtz modes.     Possible scale invariant linear magnetoresistance in pyrochlore iridates Bi2Ir2O7

    We report the observation of a linear magnetoresistance in single crystals and epitaxial thin films of the pyrochlore iridate Bi2Ir2O7. The linear magnetoresistance is positive and isotropic at low temperatures, without any sign of saturation up to 35 T. As temperature increases, the linear field dependence gradually evolves to a quadratic field dependence. The temperature and field dependence of magnetoresistance of Bi2Ir2O7 bears strikingly resemblance to the scale invariant magnetoresistance observed in the strange metal phase in high Tc cuprates. However, the residual resistivity of Bi2Ir2O7 is more than two orders of magnitude higher than the curpates. Our results suggest that the correlation between linear magnetoresistance and quantum fluctuations may exist beyond high temperature superconductors
