Quinolone Resistance Reversion by Targeting the SOS Response

Suppression of the SOS response has been postulated as a therapeutic strategy for potentiating antimicrobial agents. We aimed to evaluate the impact of its suppression on reversing resistance using a model of isogenic strains of Escherichia coli representing multiple levels of quinolone resistance. E. coli mutants exhibiting a spectrum of SOS activity were constructed from isogenic strains carrying quinolone resistance mechanisms with susceptible and resistant phenotypes. Changes in susceptibility were evaluated by static (MICs) and dynamic (killing curves or flow cytometry) methodologies. A peritoneal sepsis murine model was used to evaluate in vivo impact. Suppression of the SOS response was capable of resensitizing mutant strains with genes encoding three or four different resistance mechanisms (up to 15-fold reductions in MICs). Killing curve assays showed a clear disadvantage for survival (Δlog10 CFU per milliliter [CFU/ml] of 8 log units after 24 h), and the in vivo efficacy of ciprofloxacin was significantly enhanced (Δlog10 CFU/g of 1.76 log units) in resistant strains with a suppressed SOS response. This effect was evident even after short periods (60 min) of exposure. Suppression of the SOS response reverses antimicrobial resistance across a range of E. coli phenotypes from reduced susceptibility to highly resistant, playing a significant role in increasing the in vivo efficacy

Regulon-Specific Control of Transcription Elongation across the Yeast Genome

Transcription elongation by RNA polymerase II was often considered an invariant non-regulated process. However, genome-wide studies have shown that transcriptional pausing during elongation is a frequent phenomenon in tightly-regulated metazoan genes. Using a combination of ChIP-on-chip and genomic run-on approaches, we found that the proportion of transcriptionally active RNA polymerase II (active versus total) present throughout the yeast genome is characteristic of some functional gene classes, like those related to ribosomes and mitochondria. This proportion also responds to regulatory stimuli mediated by protein kinase A and, in relation to cytosolic ribosomal-protein genes, it is mediated by the silencing domain of Rap1. We found that this inactive form of RNA polymerase II, which accumulates along the full length of ribosomal protein genes, is phosphorylated in the Ser5 residue of the CTD, but is hypophosphorylated in Ser2. Using the same experimental approach, we show that the in vivo–depletion of FACT, a chromatin-related elongation factor, also produces a regulon-specific effect on the expression of the yeast genome. This work demonstrates that the regulation of transcription elongation is a widespread, gene class–dependent phenomenon that also affects housekeeping genes

In our current world, the existance of obstacles to acknowledge women’s equal abiliAes is real. Legally there are no barriers. The Sports Act of the government of Andalusia (Law 5/2016, July 19) determines the right to equality between genders. This is what has been called the glass ceiling. Gender stereotypes about behavior profile recreate constraints that occur in everyday pracAces, power relaAons and insAtuAonal arrangements of the general social order. Thee stereotypes are rooted in sports. In this sense. The objecAve of this study was to "interprete different gender stereotypes that exist regarding men’s and women’s profile of conduct in athleAcs.

On the use of many-core machines for the acceleration of a mesh truncation technique for FEM

Finite element method (FEM) has been used for years for radiation problems in the field of electromagnetism. To tackle problems of this kind, mesh truncation techniques are required, which may lead to the use of high computational resources. In fact, electrically large radiation problems can only be tackled using massively parallel computational resources. Different types of multi-core machines are commonly employed in diverse fields of science for accelerating a number of applications. However, properly managing their computational resources becomes a very challenging task. On the one hand, we present a hybrid message passing interface + OpenMP-based acceleration of a mesh truncation technique included in a FEM code for electromagnetism in a high-performance computing cluster equipped with 140 compute nodes. Results show that we obtain about 85% of the theoretical maximum speedup of the machine. On the other hand, a graphics processing unit has been used to accelerate one of the parts that presents high fine-grain parallelism. This work has been fnancially supported by TEC2016-80386-P, TIN2017-82972-R, CAM S2013/ICE-3004 projects and “Ayudas para contratos predoctorales de Formación del Profesorado Universitario FPU”

[EN] In this work is presented the design of a Fuzzy PD+I control system applied to the voltage control of a multi-phase DC-DC power electronic converter. The fuzzy controller has two inputs. The first input, named Error, is the difference between the desired voltage value in the output of the converter and the measured voltage at this particular point; the second input is defined by the changes in the measured voltage of the converter. The control system is embedded in the NI myRIO-1900 development kit, using LabVIEW as programming software employing the embedded system for experimental tests with a prototype of the converter. This control system allows the stabilization of the converter in the buck and boost operation modes, showing an appropriate behavior at the startup and under resistive load changes, presenting acceptable times for DC microgrids future applications while connecting the DC bus with supercapacitors or a battery bank.[ES] En este trabajo se presenta el diseño de un sistema de control difuso PD+I embebido aplicado en el control de voltaje de un convertidor electrónico de potencia bidireccional multi-fase CD-CD. Martínez Nolasco, JJ.; Rodríguez, E.; Rodríguez, H.; Morfin, J.; Padilla, A. Optimal location of exit doors for efficient evacuation of crowds at gathering places

    Get PDF
This work deals with the optimal design for the location of the exit doors at meeting places (such as sports centers, public squares, street markets, transport stations, etc.) to guarantee a safer emergency evacuation in events of a sporting, social, entertainment or religious type. This problem is stated as an optimal control problem of nonlinear partial differential equations, where the state system is a reformulation of the Hughes model (coupling the eikonal equation for a density-weighted walking velocity of pedestrians and the continuity equation for conservation of the pedestrian density), the control is the location of the exit doors at the domain boundary (subject to several geometric constraints), and the cost function is related to the evacuation rate. We provide a full numerical algorithm for solving the problem (a finite element technique for the discretization and a gradient-free procedure for the optimization), and show several numerical results for a realistic case.Ministerio de Ciencia e Innovación | Ref. Towards a more efficient evacuation of crowds by means of an optimal location of exit doors

    Get PDF
In this work we present a new strategy, employing optimal control techniques of partial differential equations, to automate the optimization of locations for a given number of exit doors at gathering places, so that the evacuation of crowds takes place in a safer and faster way. Once given a detailed mathematical formulation of the problem, in order to solve the constrained optimal control problem numerically, we propose its full discretization, with a space semi-discretization via the finite element method over a family of triangular meshes of the domain under study, and a time semi-discretization via the Euler algorithm. Finally, for the resulting discretized minimization problem, we try its optimization by means of a derivative-free algorithm. Numerical examples, corresponding to different scenarios for a real-world study case posed on “Plaza de la Liberacion” (Guadalajara, Mexico), are presented and discussed to assess the effectiveness of our approach
