    The role of hydroxo-bridged dinuclear species and the influence of “innocent” buffers in the reactivity of cis-[CoIII(cyclen)(H2O)2]3+ and [CoIII(tren)(H2O)2]3+ complexes with biologically relevant ligands at physiological pH

    In view of the relevance of the reactivity of inert tetraamine CoIII complexes having two substitutionally active cis positions capable of interact with biologically relevant ligands, the study of the reaction of cis-[Co(cyclen)(H2O)2]3+ and [Co(tren)(H2O)2]3+ with chlorides, inorganic phosphate and 5’-CMP (5’-cytidinemonophosphate) has been pursued at physiological pH. The results indicate that, in addition to the actuation of the expected labilising conjugate-base mechanism, the formation of mono and inert bis hydroxo-bridged species is relevant for understanding their speciation and reactivity. The reactivity pattern observed also indicates the key role played by the “innocent” buffers frequently used in most in vitro studies, which can make the results unreliable in many cases. The differences between the reactivity of inorganic and biologically relevant phosphates has also been found to be remarkable, with outer-sphere hydrogen bonding interactions being a dominant factor for the process. While for the inorganic phosphate substitution process the formation of μ–η2-OPO2O represents the termination of the reactivity monitored, for 5’-CMP only the formation of η1-OPO3 species is observed, which evolve with time to the final dead-end bis hydroxo-bridged complexes. The promoted hydrolysis of the 5’-CMP phosphate has not been observed in any of the processes studied

    Kinetico-mechanistic studies on substitution reactions on cross-bridged cyclen CoIII complexes with nucleosides and nucleotides

    Kinetico-mechanistic studies on the substitution reactivity of the [Co{(μ-ET)cyclen}(H2O)2] 3+ complex cation at pH values within the 6.0-7.0 range with biologically significant ligands have been carried out. The substitution processes have been found to occur exclusively on the mono-hydroxobridged [(Co{(μ-ET)cyclen}(H2O))2(μ-OH)]5+ species formed after equilibration of the cobalt complex in the relevant medium. The studies conducted on the substitution of the aqua/hydroxo ligands of this dinuclear species are indicative of a dominant role of outer-sphere complexation, involving hydrogen-bonding interactions. The values of the outer-sphere complex formation equilibrium constant are in line with the intervention of both the exiting aqua ligands and the NH groups at the encapsulating {(μ-ET)cyclen} ligand. These complexes result in the preferential formation of O- or N-bonded nucleotides depending on the structure of the base moiety of the ligand. Even the entry of the different donor bonded nucleotides is hampered by the hydrogen-bonding interaction with the dangling moiety of an already coordinated ligand. In general the overall substitution processes occur at a faster rate than those published for the fully alkylated encapsulating {(Me)2(μ-ET)cyclen} ligand derivative, as expected for the still available base-catalysing NH groups in the {(μ-ET)cyclen} ligand

    El objetivo de este estudio fue realizar un modelo epidemiológico para analizar los patrones de difusión del virus de la peste porcina clásica en Segovia y evaluar las zonas y tipos de granjas con mayor riesgo epidemiológico. El modelo utilizado fue un modelo de transición de estados estocástico y con componente espacial. Como resultado se obtuvieron una media (95% IP) de 28 (1-98) granjas infectadas, 15 (0-70) sacrificadas y 71 (2-279) inmovilizadas en la zona de protección. El tiempo medio de la duración de la epidemia fueron de 63 (1-168) días y el tiempo medio hasta la detección fue de 14 (6-39) días. Las áreas de mayor riesgo de difusión de Peste Porcina Clásica en Segovia se identificaron en la zona centro-oeste de la región. Este estudio puede ser de utilidad como apoyo en la toma de decisiones respecto a la prevención y el control de futuras epidemias de PPC.The aim of this study was to develop an epidemiological model to analyze the spread patterns of the classical swine fever virus in Segovia and to assess the areas and farm types with more epidemiological risk. The model used was a state-transition stochastic and spatial model. As a result it was obtained a mean (95% IP) of 28 (1-98) infected farms, 15 (0-70) depopulated and 71 (2-279) quarantined farms in the protection zone. The average duration of the epidemic was 63 (1-168) days and the average time to detection was 14 (6-39) days. The areas at higher risk of spread of CSF in Segovia were identified in the central-western region. This study could be useful for decision making regarding the prevention and control of potential epidemics of Classical Swine Fever

    Intervención psicoterapéutica grupal intensiva realizada en un entorno natural para el tratamiento del burnout en un equipo de enfermería oncológica

    This pilot study aims to test the influence of a group psychotherapeutic intervention on stress and burnout in an oncology nursing team. The psychological intervention consisted of implementing a package for stress inoculation, mainly using cognitive-behavioral techniques. The intervention is carried out intensively, through the coexistence of the group for three days in an accommodation located in a natural mountain environment. We used a quasi-experimental design of two groups (experimental and control) with pre and post-treatment measures. The experimental group was composed of nine members of the nursing team that received the psychological treatment and the control group was composed of eleven professionals from other teams of oncology nursing. The dependent variables considered were, on one hand anxiety and depression (evaluated by the Hospital Anxiety and Depression Scale) and, on the other hand, emotional exhaustion, depersonalization, and personal fulfilment (evaluated by the Maslach Burnout Inventory). The experimental group improved significantly in anxiety (Z = -2.44, P = 0.015, d = 0.71), personal fulfilment (Z =-2.54, P= 0.011, d=-1.29) and emotional exhaustion (Z=-2.08; p=0.037; d=0.94) one month post -treatment. In the pre-treatment, The experimental group showed burnout levels higher than the control group, particularly with regard to the lack of personal personal fulfilment (U=14, P = 0.006, d = -1.61), mainly regarding the lack of personal fulfillment (U=14; p=0.006; d=-1.61). in fact the experimental group had requested psychological intervention. One year after the intervention, the treated group is still having a significant positive effect on the personal fulfilment (Z=-2.24; p=0.025; d=-1.15) and on the depersonalization (Z=-2.23; p=0.026; d=0.52). We conclude that an intensive program of group cognitive behavioral therapy in a natural setting outside the hospital can be an effective strategy of burnout treatment in oncology nurses.Este estudio piloto tiene como objetivo valorar la influencia que tiene la intervención psicoterapéutica grupal sobre el estrés y el burnout en un equipo de enfermería oncológica. La intervención psicológica consistió en la aplicación de un paquete para la inoculación al estrés, utilizando principalmente técnicas cognitivo-conductuales, llevándose a cabo de forma intensiva, a través de la convivencia del grupo durante tres días en un alojamiento situado en un entorno natural de montaña. Utilizamos un diseño cuasi-experimental de dos grupos (experimental y control) con medidas pre y post-tratamiento. El grupo experimental estaba compuesto por los nueve miembros del equipo de enfermería que recibió el tratamiento psicológico y el grupo control estuvo compuesto por once profesionales de otros equipos de enfermería oncológica. Las variables dependientes consideradas fueron, de un lado, ansiedad y depresión (evaluadas con la Escala Hospitalaria de Ansiedad y Depresión) y, de otro lado, cansancio emocional, despersonalización y realización personal (evaluadas con el Maslach Burnout Inventory). Los resultados muestran que transcurrido un mes de la intervención psicoterapéutica grupal, en el grupo de tratamiento se había producido una mejoría significativa de la ansiedad (Z=-2,44; p=0,015; d=0,71) y de la realización personal (Z=-2,54; p=0,011; d=-1,29), igualmente se produjo en este período un gran efecto positivo sobre el cansancio emocional (Z=-2,08; p=0,037; d=0,94). En el pre-tratamiento el grupo experimental manifestaba mayores signos de desgaste profesional que el grupo control, principalmente en lo referido a la falta de realización personal (U=14; p= 0,006; d=-1,61), de hecho había demandado la intervención psicológica. Tras la experiencia terapéutica se produjo una equiparación en los valores de las variables dependientes consideradas entre los grupos, respecto a la situación pre-tratamiento. Al año de la intervención, en el grupo tratado sigue habiendo un efecto positivo importante sobre la realización personal (Z=-2,24; p=0,025; d=-1,15) y sobre la despersonalización (Z=-2,23; p=0,026; d=0,52). Concluimos que el tratamiento cognitivo-conductual grupal, intensivo realizado en un medio natural, ajeno al hospital, puede tener notables ventajas para el tratamiento del desgaste profesional en personal de enfermería oncológica

    Probiotics reduce anxiety-related behavior in zebrafish

    [EN] There is increasing evidence that gut microbiome could have effects on neurological processes and on behavior. In this study we used the novel tank test (NTT) to analyze zebrafish exploring behavior after four months’ supplementation with probiotics with probed antioxidant and anti-inflammatory properties. Results showed that prolonged ingestion of Lactobacillus rhamnosus CECT8361 and Bifidobacterium longum CECT7347 significantly alters the swimming pattern and mean swimming speed in the zebrafish model. After treatment, zebrafish strongly reduced their bottom-dwelling geotactic behavior when placed in a new tank, which could be correlated to a lower state of anxiety.S

    Effect of low sperm quality on progeny: a study on zebrafish as model species

    [EN] Nowadays a decrease tendency in human sperm quality has been reported mainly in developed countries. Reproductive technologies have been very valuable in achieving successful pregnancies with low quality sperm samples. However, considering that spermatozoa molecular contribution is increasingly important in recent studies, it is crucial to study whether fertilization with low sperm quality could leave a molecular mark on progeny. This study explores the consequences that fertilization with low sperm quality may have on progeny, using zebrafish as a model. Good and bad breeders were established attending to sperm quality analyses and were individually tracked. Significant differences in fertilization and malformation rates were obtained in progenies between high and low quality sperm samples. Moreover an altered miR profile was found in the progenies of bad zebrafish breeders (upregulation of miR-141 and miR -122 in 24 hpf embryos) and as a consequence, some of their targets involved in male sex development such as dmrt1, suffered downregulation.     Diet Supplemented with Antioxidant and Anti-Inflammatory Probiotics Improves Sperm Quality after Only One Spermatogenic Cycle in Zebrafish Model

    [EN] Infertility is a medical concern worldwide and could also have economic consequences in farmed animals. Developing an efficient diet supplement with immediate effects on sperm quality is a promising tool for human reproduction and for domesticated animal species. This study aims at elucidating the effect of a short-time probiotic supplementation consisting of a mixture of two probiotic bacteria with proven antioxidant and anti-inflammatory activities on zebrafish sperm quality and male behavior. For this purpose, three homogeneous groups of males in terms of motility (<60%) were established. The control group was fed with a normal standard diet. The other received supplements: One group (vehicle control) was fed with maltodextrin and the other received a probiotic preparation based on a mixture (1:1) of Lactobacillus rhamnosus CECT8361 and Bifidobacterium longum CECT7347. The feeding regime was 21 days corresponding with a single spermatogenesis in zebrafish. The preparation did not modify animal weight, positively affected the number of fluent males, increased sperm concentration, total motility, progressive motility, and fast spermatozoa subpopulations. Moreover, the animals fed with the supplement showed different behavior patterns compared to control groups. Our results suggest a diet-related modulation on the exploration activity indicating a lower stress-like conduct. Acciona Infraestructuras, es una de las empresas que ha sabido mantenerse y buscar nuevas alternativas para seguir adelante. Este proyecto se basa en la realización de un estudio económico financiero de Acciona Infraestructuras durante el periodo 2009-2014 para ver cómo ha influido la crisis en ella. Para ello, estudiaremos el entorno a través de la realización del DAFO y de las cinco fuerzas de Porter para después, observar la composición y evolución de su situación económica y financiera. Además, para que el análisis sea más completo, realizaremos una doble comparación: una con su principal competidor (ACS) y otra con la media del sector (a través de la selección de empresas realizada). Por último, la Q de Tobin nos enseñará cómo valora el mercado a la empresa y si estos resultados son los mismos que la evolución que ha llevado a cabo la Bolsa durante este mismo periodo

    Inflammation-related molecules in tears of patients with chronic ocular pain and dry eye disease

    Producción CientíficaThe purpose of this study was to analyze inflammation- and pain-related molecules in tears of patients suffering from chronic ocular pain associated with dry eye (DE) and/or a previous corneal refractive surgery (RS). Based on history, symptomatology, and clinical signs, the subjects (n = 180, 51.0 ± 14.7 years, 118 females, 62 males) in this cross-sectional study were assigned to one of five groups: DE and chronic ocular pain after RS (P/DE-RS, n = 52); asymptomatic subjects, i.e., without DE and chronic ocular pain, after RS (A-RS, n = 30); DE and chronic ocular pain without previous RS (P/DE-nonRS, n = 31); DE, no pain, and no previous RS (DE-nonRS, n = 35); and asymptomatic subjects with no previous RS (controls, n = 32). The tear concentrations of 20 cytokines and substance P (SP) were analyzed by immunobead-based assay and enzyme-linked immunosorbent assay, respectively. We found that tear levels of interleukin (IL)-10 and SP were increased in the RS groups. There were significant differences in IL-8/CXCL8 among the five groups. Nerve growth factor (NGF) tear levels were significantly higher in P/DE-RS than in DE-nonRS and controls. IL-9 had the highest percentage of detection in the P/DE-RS and P/DE-nonRS groups, while macrophage inflammatory protein (MIP)-1α, IL-2, and interferon (IFN)-γ were higher in the P/DE-RS, A-RS, and P/DE-nonRS groups. IL-17A was detected only in the A-RS group. Moderate correlations were observed in the A-RS, P/DE-nonRS, DE-nonRS and controls groups. A positive correlation was obtained between growth related oncogene concentration and tear break-up time (rho = 0.550; p = 0.012), while negative correlation was found between monocyte chemoattractant protein-3/CCL7 and conjunctival staining (rho = −0.560; p = 0.001), both in the A-RS group. IL-10 correlated positively with ocular pain intensity (rho = 0.513; p = 0.003) in the P/DE-nonRS group. Regulated on Activation Normal T Cell Expressed and Secreted/CCL5 correlated negatively with conjunctival staining (rho = −0.545; p = 0.001) in the DE-nonRS group. SP correlated negatively with corneal staining (rho = −0.559; p = 0.001) in the controls. In conclusion, chronic ocular pain was associated with higher IL-9 tear levels. IL-10, SP, MIP-1α/CCL3, IL-2, and IFN-γ were associated with previous RS. Higher levels of IL-8/CXCL8, MIP-1α/CCL3, IL-2, and IFN-γ were associated with DE-related inflammation, while NGF levels were related to chronic ocular pain and DE in RS patients. These findings suggest that improved knowledge of tear cytokines and neuromodulators will lead to a more nuanced understanding of how these molecules can serve as biomarkers of chronic ocular pain, leading to better therapeutic and disease management decisions.Ministerio de Ciencia, Innovación y Universidades (grants SAF-2016-77080-P, FPU17/02715 and FPU15/01443
