    Polymerase chain reaction detection of rabbit DNA in food and animal feed

    [EN] A polymerase chain reaction (PCR) based on oligonucleotide primers targeting the mitochondrial 12S rRNA gene was developed for the specific identification of rabbit DNA (Oryctolagus cuniculus) in food and feedstuffs. The specificity of the primers was verified by PCR analysis of DNA from 32 non-target species including mammals, birds, fish, and plant species. Analysis of experimental mixtures demonstrated the presence of rabbit-derived materials in the range of 0.1-100%. Prolonged heat treatment (up to 133ºC for 20 min at 300 kPa) applied to rabbit muscle/oats binary mixtures did not affect the performance of the method, which could therefore be said to be very useful for the accurate identification of rabbit materials in products submitted to denaturing technologies when other methods are not suitable.This study was supported by Grant Nº.     On the probable composition of ‘Jamaican stone’ aphrodisiac

    A dangerous aphrodisiac, commonly known as ‘Jamaican stone’, banned by the U.S. Food and Drug Administration, has been studied by vibrational spectroscopy in order to solve the controversy on its composition. The results of the ATR-FTIR analysis revealed the presence of the a-pyrone ring, which is characteristic of bufadienolides from toad venom and bulbs of squill (Drimia maritima (L.) Stearn). This conclusion was reached after a comparative study with the spectra for phytochemicals derived from gambir and cat''s claw, two Uncaria species also preconized as aphrodisiacs and deemed as possible constituents of the ‘stone’. Owing to their physiologic similarities to digoxin, bufadienolides have been shown to produce a toxic profile similar to that of digoxin, although the lack one of the side chains found on digoxin should allow the use of hemodialysis to treat ‘Jamaican stone’ overdose

    Pre-main sequence stars in the Lagoon Nebula (M8)

    We report the discovery of new pre-main sequence (PMS) stars in the Lagoon Nebula (M8) at a distance of 1.25 kpc, based on intermediate resolution spectra obtained with the Boller & Chivens spectrograph at the 6.5-m Magellan I telescope (Las Campanas Observatory, Chile). According to the spectral types, the presence of emission lines and the lithium 6708A absorption line, we are able to identify 27 classical T Tauri stars, 7 weak-lined T Tauri stars and 3 PMS emission objects with spectral type G, which we include in a separated stellar class denominated "PMS Fe/Ge class". Using near-infrared photometry either from 2MASS or from our own previous work we derive effective temperatures and luminosities for these stars and locate them in the Hertzsprung-Russell diagram, in order to estimate their masses and ages. We find that almost all of our sample stars are younger than 3 10^6 years and span over a range of masses between 0.8 and 2.5 Msun.     Antifungal ativity against Botryosphaeriaceae fungi of the hydro-methanolic extract of Silybum marianum capitula conjugated with stevioside

    Silybum marianum (L.) Gaertn, viz. milk thistle, has been the focus of research efforts in the past few years, albeit almost exclusively restricted to the medicinal properties of its fruits (achenes). Given that other milk thistle plant organs and tissues have been scarcely investigated for the presence of bioactive compounds, in this study, we present a phytochemical analysis of the extracts of S. marianum capitula during the flowering phenological stage (stage 67). Gas chromatography–mass spectroscopy results evidenced the presence of high contents of coniferyl alcohol (47.4%), and secondarily of ferulic acid ester, opening a new valorization strategy of this plant based on the former high-added-value component. Moreover, the application of the hydro-methanolic extracts as an antifungal agent has been also explored. Specifically, their activity against three fungal species responsible for the so-called Botryosphaeria dieback of grapevine (Neofusicoccum parvum, Dothiorella viticola and Diplodia seriata) has been assayed both in vitro and in vivo. From the mycelial growth inhibition assays, the best results (EC90 values of 303, 366, and 355 μg·mL−1 for N. parvum, D. viticola, and D. seriata, respectively) were not obtained for the hydroalcoholic extract alone, but after its conjugation with stevioside, which resulted in a strong synergistic behavior. Greenhouse experiments confirmed the efficacy of the conjugated complexes, pointing to the potential of the combination of milk thistle extracts with stevioside as a promising plant protection product in organic Viticulture

    The stellar content, metallicity and ionization structure of HII regions

    Observations of infrared fine-structure lines provide direct information on the metallicity and ionization structure of HII regions and indirectly on the hardness of the radiation field ionizing these nebulae. We have analyzed a sample of Galactic and Magellanic Cloud HII regions observed by the Infrared Space Observatory (ISO) to examine the interplay between stellar content, metallicity and the ionization structure of HII regions. The observed [SIV]10.5/[SIII]18.7 mum and [NeIII]15.5/[NeII]12.8 mum line ratios are shown to be highly correlated over more than two orders of magnitude. We have compared the observed line ratios to the results of photoionization models using different stellar energy distributions. The derived characteristics of the ionizing star depend critically on the adopted stellar model as well as the (stellar) metallicity. We have compared the stellar effective temperatures derived from these model studies for a few well-studied HII regions with published direct spectroscopic determinations of the spectral type of the ionizing stars. This comparison supports our interpretation that stellar and nebular metallicity influences the observed infrared ionic line ratios. We can explain the observed increase in degree of ionization, as traced by the [SIV]\[SIII] and [NeIII]\[NeII] line ratios, by the hardening of the radiation field due to the decrease of metallicity.     A photoionization model of the compact HII region G29.96-0.02

    We present a detailed photoionization model of G29.96-0.02 (hereafter G29.96), one of the brightest Galactic Ultra Compact HII (UCHII) regions in the Galaxy. This source has been observed extensively at radio and infrared wavelengths. The most recent data include a complete ISO (SWS and LWS) spectrum, which displays a remarkable richness in atomic fine-structure lines. The number of observables is twice as much as the number available in previous studies. In addition, most atomic species are now observed in two ionization stages. The radio and infrared data on G29.96 are best reproduced using a nebular model with two density components: a diffuse (n_e~680cm-3) extended (~1 pc) component surrounding a compact (~0.1 pc) dense (n_e~57000cm-3) core. The properties of the ionizing star were derived using state-of-the-art stellar atmosphere models. CoStar models yield an effective temperature of \~30^{+2}_{-1} kK whereas more recent non-LTE line blanketed atmospheres with stellar winds indicate somewhat higher values, Teff~32--38 kK. This range in Teff is compatible with all observational constraints, including near-infrared photometry and bolometric luminosity. The range 33-36 kK is also compatible with the spectral type O5-O8 determined by Watson and Hanson (97) when recent downward revisions of the effective temperature scale of O stars are taken into account. The age of the ionizing star of G29.96 is found to be a few 10^6 yr, much older than the expected lifetime of UCHII regions. Accurate gas phase abundances are derived with the most robust results being Ne/S=7.5 and N/O=0.43 (1.3 and 3.5 times the solar values, respectively).Comment: Accepted in Astronomy and Astrophysic

    OptEEmAL: Decision-Support Tool for the Design of Energy Retrofitting Projects at District Level

    Designing energy retrofitting actions poses an elevated number of problems, as the definition of the baseline, selection of indicators to measure performance, modelling, setting objectives, etc. This is time-consuming and it can result in a number of inaccuracies, leading to inadequate decisions. While these problems are present at building level, they are multiplied at district level, where there are complex interactions to analyse, simulate and improve. OptEEmAL proposes a solution as a decision-support tool for the design of energy retrofitting projects at district level. Based on specific input data (IFC(s), CityGML, etc.), the platform will automatically simulate the baseline scenario and launch an optimisation process where a series of Energy Conservation Measures (ECMs) will be applied to this scenario. Its performance will be evaluated through a holistic set of indicators to obtain the best combination of ECMs that complies with user's objectives. A great reduction in time and higher accuracy in the models are experienced, since they are automatically created and checked. A subjective problem is transformed into a mathematical problem; it simplifies it and ensures a more robust decision-making.
