    Epigenetic silencing of SOCS3 expression contributes to fibrosis in Crohn’s disease

    Identified risk polymorphisms affecting the Jak-STAT3 pathway in patients with Crohn’s disease could affect TGF-β1 and collagen I expression and in the pathway’s negative regulator, SOCS3. Genetic factors, however, account for only ~25% of disease. Epigenetic events also shape gene expression. Recent experiments showed that autocrine IL-6 production in mesenchymal cells, subepithelial myofibroblasts (SEMF) and muscle cells, of patients with fibrostenotic Crohn’s disease causes sustained Jak-STAT3 activity, excess TGF-β1 and Collagen I production and fibrosis. SOCS3 paradoxically decreased in these cells. We now identify epigenetic mechanisms that silence SOCS3 expression in SEMF of patients with fibrostenotic Crohn’s disease. In a previous experiment, using isolated SEMF of normal ileum and affected ileum from patients with each Crohn’s phenotype, inflammatory (Montreal B1), fibrostenotic (B2) and penetrating (B3), we confirmed decreased SOCS3 protein levels were unique to B2 patients. Expression of miR-19b increased in SEMF of affected ileum. SOCS3 transcriptional activity decreased after transfection of miR-19b mimic and increased when antagomiR-19b was expressed. Epigenetic silencing of SOCS3 in ileal SEMF of patients with fibrostenotic Crohn’s disease occurs by increased miR-19b mediated inhibition of SOCS3

    Neonatal lamb mortality is often high in Western Australia and losses as high as 33 per cent, have been recorded on private properties in the Esperance area. Surveys by Western Australian Department of Agriculture officers have indicated that more than threequarters of such lamb deaths occur after birth. About half these deaths result from the combined effects of weather and mismothering. Work in Victoria has indicated that lamb mortality may be reduced by intensive lambing systems such as shedding and this trial was established to see if shedding could reduce lamb mortality in the Esperance environment

    One-man electrochemical air revitalization system evaluation

    A program to evaluate the performance of a one man capacity, self contained electrochemical air revitalization system was successfully completed. The technology readiness of this concept was demonstrated by characterizing the performance of this one man system over wide ranges in cabin atmospheric conditions. The electrochemical air revitalization system consists of a water vapor electrolysis module to generate oxygen from water vapor in the cabin air, and an electrochemical depolarized carbon dioxide concentrator module to remove carbon dioxide from the cabin air. A control/monitor instrumentation package that uses the electrochemical depolarized concentrator module power generated to partially offset the water vapor electrolysis module power requirements and various structural fluid routing components are also part of the system. The system was designed to meet the one man metabolic oxygen generation and carbon dioxide removal requirements, thereby controlling cabin partial pressure of oxygen at 22 kN/sq m and cabin pressure of carbon dioxide at 400 N/sq m over a wide range in cabin air relative humidity conditions

    Isolation of microsatellite loci in the Capricorn silvereye, Zosterops lateralis chlorocephalus (Aves : Zosteropidae)

    The Capricorn silvereye (Zosterops lateralis chlorocephalus ) is ideally suited to investigating the genetic basis of body size evolution. We have isolated and characterized a set of microsatellite markers for this species. Seven out of 11 loci were polymorphic. The number of alleles detected ranged from two to five and observed heterozygosities between 0.12 and 0.67. One locus, ZL49, was found to be sex-linked. This moderate level of diversity is consistent with that expected in an isolated, island population

    Discovery of a Second Millisecond Accreting Pulsar: XTE J1751-305

    We report the discovery by the RXTE PCA of a second transient accreting millisecond pulsar, XTE J1751-305, during regular monitoring observations of the galactic bulge region. The pulsar has a spin frequency of 435 Hz, making it one of the fastest pulsars. The pulsations contain the signature of orbital Doppler modulation, which implies an orbital period of 42 minutes, the shortest orbital period of any known radio or X-ray millisecond pulsar. The mass function, f_x = (1.278 +/- 0.003) x 10^{-6} M_sun, yields a minimum mass for the companion of between 0.013 and 0.017 M_sun, depending on the mass of the neutron star. No eclipses were detected. A previous X-ray outburst in June, 1998, was discovered in archival All-Sky Monitor data. Assuming mass transfer in this binary system is driven by gravitational radiation, we constrain the orbital inclination to be in the range 30-85 deg, and the companion mass to be 0.013-0.035 M_sun. The companion is most likely a heated helium dwarf.     Andreev Probe of Persistent Current States in Superconducting Quantum Circuits

    Using the extraordinary sensitivity of Andreev interferometers to the superconducting phase difference associated with currents, we measure the persistent current quantum states in superconducting loops interrupted by Josephson junctions. Straightforward electrical resistance measurements of the interferometers give continuous read-out of the states, allowing us to construct the energy spectrum of the quantum circuit. The probe is estimated to be more precise and faster than previous methods, and can measure the local phase difference in a wide range of superconducting circuits.Comment: Changes made in light of referees comments; to appear in PR

    Expression of alphav integrins and vitronectin receptor identity in breast cancer cells.

    In the present study we have used fluorocytometry and immunoprecipitation to characterize the expression of alphav-containing integrins in a panel of eight human breast cancer cell lines and one normal human mammary epithelial line. We show that the classical vitronectin receptor alphavbeta3 is expressed in only one cell line (MDA-MB-231), whereas alphavbeta5 is expressed on all breast cancer cell lines and alphavbeta1 is expressed on the majority. Using adherence assays to purified ligands in the presence and absence of function-blocking monoclonal antibodies, we have demonstrated that alphavbeta5 mediates adhesion to vitronectin in the majority of these cells. In one cell line, ZR75-1, alphavbeta1 contributes significantly to adhesion to immobilized vitronectin. The formation of focal adhesions containing the alphav and beta1 subunits on vitronectin is also demonstrated by indirect immunofluorescence

    Short gamma-ray bursts within 200 Mpc

    We present a systematic search for short-duration gamma-ray bursts (GRBs) in the local Universe based on 14 yr of observations with the Neil Gehrels Swift Observatory. We cross-correlate the GRB positions with the GLADE catalogue of nearby galaxies, and find no event at a distance ≲100 Mpc and four plausible candidates in the range 100 Mpc ≲ D ≲ 200 Mpc. Although affected by low statistics, this number is higher than the one expected for chance alignments to random galaxies, and possibly suggests a physical association between these bursts and nearby galaxies. By assuming a local origin, we use these events to constrain the range of properties for X-ray counterparts of neutron star mergers. Optical upper limits place tight constraints on the onset of a blue kilonova, and imply either low masses (⁠≲10−3M⊙⁠) of lanthanide-poor ejecta or unfavorable orientations (θ_(obs) ≳ 30 deg). Finally, we derive that the all-sky rate of detectable short GRBs within 200 Mpc is 1.3^(+1.7)_(−0.8) yr⁻¹ (68 per cent confidence interval), and discuss the implications for the GRB outflow structure. If these candidates are instead of cosmological origin, we set a upper limit of ≲2.0 yr⁻¹ (90 per cent confidence interval) to the rate of nearby events detectable with operating gamma-ray observatories, such as Swift and Fermi
