Free Running Single Photon Detection based on a negative feedback InGaAs APD
InGaAs/InP-based semiconductor avalanche photodiode are usually employed for
single-photon counting at telecom wavelength. However they are affected by
afterpulsing which limits the diode performance. Recently, Princeton Lightwave
has commercialised a diode integrating monolithically a feedback resistor. This
solution effectively quenches the avalanche and drastically reduces
afterpulsing. Here, we report the development and characterization of a
detector module based on this diode, implementing an active hold-off circuit
which further reduces the afterpulsing and notably improves the detector
performances. We demonstrate free-running operation with 600 Hz dark count rate
at 10% detection efficiency. We also improved the standard double-window
technique for the afterpulsing characterization. Our algorithm implemented by a
FPGA allows to put the APD in a well-defined initial condition and to measure

Gas-phase metallicity gradients of TNG50 star-forming galaxies
We present the radial gas-phase, mass-weighted metallicity profiles and
gradients of the TNG50 star-forming galaxy population measured at redshifts
0--3. We investigate the redshift evolution of gradients and examine
relations between gradient steepness and galaxy properties. We find that TNG50
gradients are predominantly negative at all redshifts, although we observe
significant diversity among these negative gradients. We determine that the
gradient steepness of all galaxies increases approximately monotonically with
redshift at a roughly constant rate. This rate does not vary significantly with
galaxy mass. We observe a weak negative correlation between gradient steepness
and galaxy stellar mass at redshifts . However, when we normalize
gradients by a characteristic radius defined by the galactic star formation
distribution, we find that these normalized gradients remain invariant with
both stellar mass and redshift. We place our results in the context of previous
simulations and show that TNG50 high-redshift gradients are steeper than those
of models featuring burstier feedback, which may further highlight
high-redshift gradients as important discriminators of galaxy formation models.
We also find that redshift and TNG50 gradients are consistent
with the gradients observed in galaxies at these redshifts, although the
preference for flat gradients observed in redshift galaxies is not
present in TNG50. If future JWST and ELT observations validate these flat
gradients, it may indicate a need for simulation models to implement more
powerful radial gas mixing within the ISM, possibly via turbulence and/or

Pre-Training LiDAR-Based 3D Object Detectors Through Colorization
Accurate 3D object detection and understanding for self-driving cars heavily
relies on LiDAR point clouds, necessitating large amounts of labeled data to
train. In this work, we introduce an innovative pre-training approach, Grounded
Point Colorization (GPC), to bridge the gap between data and labels by teaching
the model to colorize LiDAR point clouds, equipping it with valuable semantic
cues. To tackle challenges arising from color variations and selection bias, we
incorporate color as "context" by providing ground-truth colors as hints during
colorization. Experimental results on the KITTI and Waymo datasets demonstrate
GPC's remarkable effectiveness. Even with limited labeled data, GPC
significantly improves fine-tuning performance; notably, on just 20% of the
KITTI dataset, GPC outperforms training from scratch with the entire dataset.
In sum, we introduce a fresh perspective on pre-training for 3D object
detection, aligning the objective with the model's intended role and ultimately
advancing the accuracy and efficiency of 3D object detection for autonomous
vehicles
Wolbachia and DNA barcoding insects: patterns, potential and problems
Wolbachia is a genus of bacterial endosymbionts that impacts the breeding systems of their hosts. Wolbachia can confuse the patterns of mitochondrial variation, including DNA barcodes, because it influences the pathways through which mitochondria are inherited. We examined the extent to which these endosymbionts are detected in routine DNA barcoding, assessed their impact upon the insect sequence divergence and identification accuracy, and considered the variation present in Wolbachia COI. Using both standard PCR assays (Wolbachia surface coding protein – wsp), and bacterial COI fragments we found evidence of Wolbachia in insect total genomic extracts created for DNA barcoding library construction. When >2 million insect COI trace files were examined on the Barcode of Life Datasystem (BOLD) Wolbachia COI was present in 0.16% of the cases. It is possible to generate Wolbachia COI using standard insect primers; however, that amplicon was never confused with the COI of the host. Wolbachia alleles recovered were predominantly Supergroup A and were broadly distributed geographically and phylogenetically. We conclude that the presence of the Wolbachia DNA in total genomic extracts made from insects is unlikely to compromise the accuracy of the DNA barcode library; in fact, the ability to query this DNA library (the database and the extracts) for endosymbionts is one of the ancillary benefits of such a large scale endeavor – for which we provide several examples. It is our conclusion that regular assays for Wolbachia presence and type can, and should, be adopted by large scale insect barcoding initiatives. While COI is one of the five multi-locus sequence typing (MLST) genes used for categorizing Wolbachia, there is limited overlap with the eukaryotic DNA barcode region
Cardiolipin synthesis in brown and beige fat mitochondria is essential for systemic energy homeostasis
Activation of energy expenditure in thermogenic fat is a promising strategy to improve metabolic health, yet the dynamic processes that evoke this response are poorly understood. Here we show that synthesis of the mitochondrial phospholipid cardiolipin is indispensable for stimulating and sustaining thermogenic fat function. Cardiolipin biosynthesis is robustly induced in brown and beige adipose upon cold exposure. Mimicking this response through overexpression of cardiolipin synthase (Crls1) enhances energy consumption in mouse and human adipocytes. Crls1 deficiency in thermogenic adipocytes diminishes inducible mitochondrial uncoupling and elicits a nuclear transcriptional response through endoplasmic reticulum stress-mediated retrograde communication. Cardiolipin depletion in brown and beige fat abolishes adipose thermogenesis and glucose uptake, which renders animals insulin resistant. We further identify a rare human CRLS1 variant associated with insulin resistance and show that adipose CRLS1 levels positively correlate with insulin sensitivity. Thus, adipose cardiolipin has a powerful impact on organismal energy homeostasis through thermogenic fat bioenergetics

Rare germline variants in DNA repair genes and the angiogenesis pathway predispose prostate cancer patients to develop metastatic disease
Background
Prostate cancer (PrCa) demonstrates a heterogeneous clinical presentation ranging from largely indolent to lethal. We sought to identify a signature of rare inherited variants that distinguishes between these two extreme phenotypes.
Methods
We sequenced germline whole exomes from 139 aggressive (metastatic, age of diagnosis < 60) and 141 non-aggressive (low clinical grade, age of diagnosis ≥60) PrCa cases. We conducted rare variant association analyses at gene and gene set levels using SKAT and Bayesian risk index techniques. GO term enrichment analysis was performed for genes with the highest differential burden of rare disruptive variants.
Results
Protein truncating variants (PTVs) in specific DNA repair genes were significantly overrepresented among patients with the aggressive phenotype, with BRCA2, ATM and NBN the most frequently mutated genes. Differential burden of rare variants was identified between metastatic and non-aggressive cases for several genes implicated in angiogenesis, conferring both deleterious and protective effects.
Conclusions
Inherited PTVs in several DNA repair genes distinguish aggressive from non-aggressive PrCa cases. Furthermore, inherited variants in genes with roles in angiogenesis may be potential predictors for risk of metastases. If validated in a larger dataset, these findings have potential for future clinical application

Alcohol-related hypoglycemia in rural Uganda: socioeconomic and physiologic contrasts
Hypoglycemia is a rare but important complication seen in patients who present with alcohol intoxication. In a study by Marks and Teale, less than one percent of people with alcohol intoxication who presented to an American emergency department were hypoglycemic [1]. It is even more rare to see an intoxicated patient, who had been eating appropriately prior to or during the intoxication, present in a hypoglycemic coma. However, our analysis of the first 500 patients seen in a newly opened five-bed Emergency Department (ED) at Nyakibale Karoli Lwanga Hospital in rural southwestern Uganda, revealed multiple intoxicated patients who presented in hypoglycemic coma within hours of eating a full meal.
