Myalgic encephalomyelitis; Blood analytics; DiagnosisEncefalomielitis miálgica; Análisis de sangre; DiagnósticoEncefalomielitis miàlgica; Anàlisi de sang; DiagnòsticBackground: Routine blood analytics are systematically used in the clinic to diagnose disease or confirm individuals’ healthy status. For myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS), a disease relying exclusively on clinical symptoms for its diagnosis, blood analytics only serve to rule out underlying conditions leading to exerting fatigue. However, studies evaluating complete and large blood datasets by combinatorial approaches to evidence ME/CFS condition or detect/identify case subgroups are still scarce. Methods: This study used unbiased hierarchical cluster analysis of a large cohort of 250 carefully phenotyped female ME/CFS cases toward exploring this possibility. Results: The results show three symptom-based clusters, classified as severe, moderate, and mild, presenting significant differences (p < 0.05) in five blood parameters. Unexpectedly the study also revealed high levels of circulating complement factor C1q in 107/250 (43%) of the participants, placing C1q as a key molecule to identify an ME/CFS subtype/subgroup with more apparent pain symptoms. Conclusions: The results obtained have important implications for the research of ME/CFS etiology and, most likely, for the implementation of future diagnosis methods and treatments of ME/CFS in the clinic. This research was partially funded by the Association of Patients with ME/CFS and Fibromyalgia in Catalonia, Spain (ACAF; www.fibromyalgia.cat (accessed on 23 March 2019) to J.C.-M. and J.A.-M., and by a UCV 2020-270-001 grant to E.O

Loss of Jak2 Selectively Suppresses DC-Mediated Innate Immune Response and Protects Mice from Lethal Dose of LPS-Induced Septic Shock

Given the importance of Jak2 in cell signaling, a critical role for Jak2 in immune cells especially dendritic cells (DCs) has long been proposed. The exact function for Jak2 in DCs, however, remained poorly understood as Jak2 deficiency leads to embryonic lethality. Here we established Jak2 deficiency in adult Cre+/+Jak2fl/fl mice by tamoxifen induction. Loss of Jak2 significantly impaired DC development as manifested by reduced BMDC yield, smaller spleen size and reduced percentage of DCs in total splenocytes. Jak2 was also crucial for the capacity of DCs to mediate innate immune response. Jak2−/− DCs were less potent in response to inflammatory stimuli and showed reduced capacity to secrete proinflammatory cytokines such as TNFα and IL-12. As a result, Jak2−/− mice were defective for the early clearance of Listeria after infection. However, their potency to mediate adaptive immune response was not affected. Unlike DCs, Jak2−/− macrophages showed similar capacity secretion of proinflammatory cytokines, suggesting that Jak2 selectively modulates innate immune response in a DC-dependent manner. Consistent with these results, Jak2−/− mice were remarkably resistant to lethal dose of LPS-induced septic shock, a deadly sepsis characterized by the excessive innate immune response, and adoptive transfer of normal DCs restored their susceptibility to LPS-induced septic shock. Mechanistic studies revealed that Jak2/SATA5 signaling is pivotal for DC development and maturation, while the capacity for DCs secretion of proinflammatory cytokines is regulated by both Jak2/STAT5 and Jak2/STAT6 signaling

Estimation of the age of majority through radiographic evaluation of the third molar maturation degree

Although the third molar is the tooth with the greatest anatomical and developmental variability, some authors consider it important to estimate whether a subject is of legal age or not. The Demirjian?s technique is the most widely used tool to estimate dental age and was therefore used in our study to assess possible correlation between the age of majority and the maturational degree of the lower third molars, evaluated through radiographic images. This observational transversal study was conducted on 180 panoramic radiographs from consecutive patients. The degree of maturation of the lower third molar was independently classified by two observers, according to the Demirjian´s maturational stage method. A total of 180 patients ? 65 men (36.12%) and 115 women (63.88%) ? were included (mean age 21.6 years; standard deviation 5.2). The age range of our subjects corresponded to maturational stages D to H in the Demirjian?s classification. A logistic regression analysis showed that subjects classified into the highest maturational stage H had a significantly higher probability of being considered of legal age by both observers, as compared with subjects in the lowest stage D. Inter-observer agreement was very high. Gender predictive capacity was not observed. Our results showed that subjects with the lower third molars in the highest maturational stage could be estimated to be older than 18 years and therefore considered of legal age, a finding also reported by other authorsThe Demirjian´s tooth maturational stage method, applied to the lower third molars, can be reliably used to estimate whether an individual is of legal age (18 years or older). High concordance between different observers using this method can be expected

On 19 September 2021, a volcanic eruption began on the island of La Palma (Canary Islands, Spain). The eruption has allowed the assessment of an unprecedented multidisciplinary study on the effects of the volcanic plume. This work presents the estimation of the spectral direct radiative forcing (∆F) and efficiency (∆F E f f) from solar radiation measurements at the Izaña Observatory (IZO) located on the island of Tenerife (∼140 km from the volcano). During the eruption, the IZO was affected by different types of aerosols: volcanic, Saharan mineral dust, and a mixture of volcanic and dust aerosols. Three case studies were identified using ground based (lidar) data, satellite-based (Sentinel5P Tropospheric Monitoring Instrument, TROPOMI) data, reanalysis data (Modern-Era Retrospective Analysis for Research and Applications, version 2, MERRA-2), and backward trajectories (Flexible Trajectories, FLEXTRA), and subsequently characterised in terms of optical and micro-physical properties using ground based sun-photometry measurements. Despite the ∆F of the volcanic aerosols being greater than that of the dust events (associated with the larger aerosol load present), the ∆F E f f was found to be lower. The spectral ∆F E f f values at 440 nm ranged between −1.9 and −2.6 Wm−2nm−1AOD−1 for the mineral dust and mixed volcanic and dust particles, and between −1.6 and −3.3 Wm−2nm−1AOD−1 for the volcanic aerosols, considering solar zenith angles between 30◦ and 70◦, respectively. The authors also acknowledge the support of ACTRIS, Ministerio de Ciencia e Innovación of Spain, through the projects SYNERA: PID2020-118793GA-I00 and RT2018- 097864-B-I00, and Junta de Castilla y León grant N◦. A set of samplings were carried out at the rocky intertidal shore of the north of Tenerife island (Canary Islands, Spain) with the objective of compile abundance and size data from the species Epinephelus marginatus (Lowe, 1834) belonging to Serranidae family, known as dusky groupers in colloquial language. This species is an important fishing resource with a wide distribution along the rocky shores, observing juvenile populations inhabiting intertidal pools. In the present research a set of intertidal pools were sampled randomly in function of their size: smalls (80 m3) with the purpose of determine if there is an effect of this factor over the abundance and size values observed. Study area was located in Punta del Hidalgo and Finca El Apio, recording data from 22 tidalpools with a total number of 8 juveniles of E. marginatus. The statistical analysis showed a significant effect of the size of the pools, finding the biggest individuals at the biggest pools
