Parenting Latino Toddlers and Preschoolers: Clinical and Nonclinical Samples
Parenting practices contribute significantly to the social-emotional development of young children. There is limited literature that addresses the role of culture in parenting, particularly among Latino families who have very young children with significant behavior problems. The current study compared the parenting practices of 30 low-income Latino mothers whose young children had been referred for mental health services for their behavior problems with a similar group of 30 mothers of children without behavior problems. Results showed that mothers in the clinical sample nurtured their children less often and used more frequent verbal and corporal punishment as discipline than the nonclinical sample. The clinical sample also had a significantly higher incidence of mental health problems in their families. Results also showed the significant toll that raising young children with challenging behaviors takes on their mothers. The implications of these findings for the early identification of these children are discussed
Olanzapine Attenuates Cue-elicited Craving for Tobacco
Rationale: Recent biological conceptualizations of craving and addiction have implicated mesolimbic dopamine activity as a central feature of the process of addiction. Imaging, and pharmacological studies have supported a role for dopaminergic structures in cue-elicited craving for tobacco.
Objective: If mesolimbic dopamine activity is associated with cue-elicited craving for tobacco, a dopamine antagonist should attenuate cueelicited craving for tobacco. Thus, the aim of the present study was to determine whether an atypical antipsychotic (olanzapine, 5 mg) decreased cue-elicited craving for tobacco.
Method: Participants were randomly assigned to 5 days of pretreatment with olanzapine (5 mg; n=31) or were randomly assigned to 5 days of a matching placebo (n=28). Approximately 8 h after the last dose, participants were exposed to a control cue (pencil) followed by exposure to smoking cues. Participants subsequently smoked either nicotine cigarettes or de-nicotinized cigarettes.
Results: Olanzapine attenuated cue-elicited craving for tobacco but did not moderate the subjective effects of smoking.
Discussion: This study represents one of the first investigations of the effect of atypical antipsychotics on cue-elicited craving for tobacco. The results suggest that medications with similar profiles may reduce cue-elicited craving, which in turn, may partially explain recent observations that atypical antipsychotics may reduce substance use
The Effect of Olanzapine on Craving and Alcohol Consumption
Previous studies have indicated that olanzapine decreases craving after a priming dose of alcohol, that craving after a priming dose of alcohol is greater among individuals with the seven-repeat allele of the DRD4 variable number of tandem repeats (VNTR) polymorphism, and that the effect of olanzapine (a D2/D4 antagonist) is more pronounced among individuals with this allele. The present study tested the hypothesis that olanzapine may be differentially effective at reducing cue-elicited craving and differentially effective as a treatment for alcohol dependence over the course of a 12-week, randomized, placebo-controlled trial among individuals with and without the seven-repeat allele. Participants who met DSM IV criteria for alcohol dependence were randomly assigned to receive olanzapine (5 mg) or a placebo over the course of the trial. After 2 weeks of treatment, participants completed a cue reactivity assessment. The results suggested that participants who were homozygous or heterozygous for the seven (or longer)-repeat allele of the DRD4 VNTR responded to olanzapine with reductions in cue-elicited craving as well as reductions in alcohol consumption over the course of the 12-week trial, whereas individuals with the shorter alleles did not respond favorably to olanzapine
ÎČ-cell dysfunctional ERAD/ubiquitin/proteasome system in type 2 diabetes mediated by islet amyloid polypeptide-induced UCH-L1 deficiency.
ObjectiveThe islet in type 2 diabetes is characterized by ÎČ-cell apoptosis, ÎČ-cell endoplasmic reticulum stress, and islet amyloid deposits derived from islet amyloid polypeptide (IAPP). Toxic oligomers of IAPP form intracellularly in ÎČ-cells in humans with type 2 diabetes, suggesting impaired clearance of misfolded proteins. In this study, we investigated whether human-IAPP (h-IAPP) disrupts the endoplasmic reticulum-associated degradation/ubiquitin/proteasome system.Research design and methodsWe used pancreatic tissue from humans with and without type 2 diabetes, isolated islets from h-IAPP transgenic rats, isolated human islets, and INS 832/13 cells transduced with adenoviruses expressing either h-IAPP or a comparable expression of rodent-IAPP. Immunofluorescence and Western blotting were used to detect polyubiquitinated proteins and ubiquitin carboxyl-terminal hydrolase L1 (UCH-L1) protein levels. Proteasome activity was measured in isolated rat and human islets. UCH-L1 was knocked down by small-interfering RNA in INS 832/13 cells and apoptosis was evaluated.ResultsWe report accumulation of polyubiquinated proteins and UCH-L1 deficiency in ÎČ-cells of humans with type 2 diabetes. These findings were reproduced by expression of oligomeric h-IAPP but not soluble rat-IAPP. Downregulation of UCH-L1 expression and activity to reproduce that caused by h-IAPP in ÎČ-cells induced endoplasmic reticulum stress leading to apoptosis.ConclusionsOur results indicate that defective protein degradation in ÎČ-cells in type 2 diabetes can, at least in part, be attributed to misfolded h-IAPP leading to UCH-L1 deficiency, which in turn further compromises ÎČ-cell viability

Exciton states in monolayer MoSe2 and MoTe2 probed by upconversion spectroscopy
Transitions metal dichalcogenides (TMDs) are direct semiconductors in the
atomic monolayer (ML) limit with fascinating optical and spin-valley
properties. The strong optical absorption of up to 20 % for a single ML is
governed by excitons, electron-hole pairs bound by Coulomb attraction. Excited
exciton states in MoSe and MoTe monolayers have so far been elusive due
to their low oscillator strength and strong inhomogeneous broadening. Here we
show that encapsulation in hexagonal boron nitride results in emission line
width of the A:1 exciton below 1.5 meV and 3 meV in our MoSe and
MoTe monolayer samples, respectively. This allows us to investigate the
excited exciton states by photoluminescence upconversion spectroscopy for both
monolayer materials. The excitation laser is tuned into resonance with the
A:1 transition and we observe emission of excited exciton states up to 200
meV above the laser energy. We demonstrate bias control of the efficiency of
this non-linear optical process. At the origin of upconversion our model
calculations suggest an exciton-exciton (Auger) scattering mechanism specific
to TMD MLs involving an excited conduction band thus generating high energy
excitons with small wave-vectors. The optical transitions are further
investigated by white light reflectivity, photoluminescence excitation and
resonant Raman scattering confirming their origin as excited excitonic states

Spectrally narrow exciton luminescence from monolayer MoS2 exfoliated onto epitaxially grown hexagonal BN
The strong light-matter interaction in transition Metal dichalcogenides
(TMDs) monolayers (MLs) is governed by robust excitons. Important progress has
been made to control the dielectric environment surrounding the MLs, especially
through hexagonal boron nitride (hBN) encapsulation, which drastically reduces
the inhomogeneous contribution to the exciton linewidth. Most studies use
exfoliated hBN from high quality flakes grown under high pressure. In this
work, we show that hBN grown by molecular beam epitaxy (MBE) over a large
surface area substrate has a similarly positive impact on the optical emission
from TMD MLs. We deposit MoS and MoSe MLs on ultrathin hBN films (few
MLs thick) grown on Ni/MgO(111) by MBE. Then we cover them with exfoliated hBN
to finally obtain an encapsulated sample : exfoliated hBN/TMD ML/MBE hBN. We
observe an improved optical quality of our samples compared to TMD MLs
exfoliated directly on SiO substrates. Our results suggest that hBN grown
by MBE could be used as a flat and charge free substrate for fabricating
TMD-based heterostructures on a larger scale.Comment: 5 pages, 3 figure
Revealing exciton masses and dielectric properties of monolayer semiconductors with high magnetic fields
In semiconductor physics, many essential optoelectronic material parameters
can be experimentally revealed via optical spectroscopy in sufficiently large
magnetic fields. For monolayer transition-metal dichalcogenide semiconductors,
this field scale is substantial --tens of teslas or more-- due to heavy carrier
masses and huge exciton binding energies. Here we report absorption
spectroscopy of monolayer MoS, MoSe, MoTe, and WS in very high
magnetic fields to 91~T. We follow the diamagnetic shifts and valley Zeeman
splittings of not only the exciton's ground state but also its excited
, , ..., Rydberg states. This provides a direct experimental
measure of the effective (reduced) exciton masses and dielectric properties.
Exciton binding energies, exciton radii, and free-particle bandgaps are also
determined. The measured exciton masses are heavier than theoretically
predicted, especially for Mo-based monolayers. These results provide essential
and quantitative parameters for the rational design of opto-electronic van der
