Tinjauan Psiko-Teologi Terhadap Fenomena Kekerasan Dalam Pacaran Pada Remaja

    Get PDF
The phenomenon of dating violence does not only occur in closed or private spaces but is very easy to find in public spaces such as school yards, computer rental places, parks, sidewalks, public transportation, on two-wheeled vehicle passengers in the middle of traffic. And when the event took place and was witnessed by the general public, the perpetrators and victims did not feel disturbed, uncomfortable, embarrassed, or stopped. Whereas violence in courtship among adolescents is one access to domestic violence, if this is not handled properly before it is sustained with victims who have an impact on the physical, psychological, social, moral, economic and future generations. The method used is descriptive research by examining the phenomenon of dating violence from the perspective of the Bible and psychology, where psychologically sexual violence that occurs during adolescence has a negative impact on perpetrators and victims of sexual violence. The violence during dating can be caused by adolescents experiencing jumps due to hormone fluctuations and rapid information technology. Theologically sex before marriage is an act of destroying the lives of the perpetrators and violence during dating is an action that is contrary to the concept of Imago dei.

Chromatin Profiles of Chromosomally Integrated Human Herpesvirus-6A

    Get PDF
Human herpesvirus-6A (HHV-6A) and 6B (HHV-6B) are two closely related betaherpesviruses that are associated with various diseases including seizures and encephalitis. The HHV-6A/B genomes have been shown to be present in an integrated state in the telomeres of latently infected cells. In addition, integration of HHV-6A/B in germ cells has resulted in individuals harboring this inherited chromosomally integrated HHV-6A/B (iciHHV-6) in every cell of their body. Until now, the viral transcriptome and the epigenetic modifications that contribute to the silencing of the integrated virus genome remain elusive. In the current study, we used a patient-derived iciHHV-6A cell line to assess the global viral gene expression profile by RNA-seq, and the chromatin profiles by MNase-seq and ChIP-seq analyses. In addition, we investigated an in vitro generated cell line (293-HHV-6A) that expresses GFP upon the addition of agents commonly used to induce herpesvirus reactivation such as TPA. No viral gene expression including miRNAs was detected from the HHV-6A genomes, indicating that the integrated virus is transcriptionally silent. Intriguingly, upon stimulation of the 293-HHV-6A cell line with TPA, only foreign promoters in the virus genome were activated, while all HHV-6A promoters remained completely silenced. The transcriptional silencing of latent HHV-6A was further supported by MNase-seq results, which demonstrate that the latent viral genome resides in a highly condensed nucleosome-associated state. We further explored the enrichment profiles of histone modifications via ChIP-seq analysis. Our results indicated that the HHV-6 genome is modestly enriched with the repressive histone marks H3K9me3/H3K27me3 and does not possess the active histone modifications H3K27ac/H3K4me3. Overall, these results indicate that HHV-6 genomes reside in a condensed chromatin state, providing insight into the epigenetic mechanisms associated with the silencing of the integrated HHV-6A genome

Development and stability evaluation of olive oil nanoemulsion using sucrose ester

    Get PDF
Nanoemulsion is a type of emulsion that consist of fine oil-in-water dispersions, with the droplets covering the size range of 20-200nm. It can be achieved through self-emulsification process. One of the processes is through low energy emulsification method. Olive oil was chosen in this study due to its efficiency in treating skin problem. Pre-emulsion was prepared by mixing oil, sucrose ester and glycerol under heat at 80 °C. After the pre-emulsion was formed, it will be self-emulsified with water under gentle agitation. Olive oil nanoemulsion was prepared using sucrose ester as a surfactant to produce droplets size below 200 nm with low size distribution. The pre-emulsion was very stable when stored at 4 “C. However, it was unstable when stored at 25 °C and 40 °C. Therefore, 4 °C is the ideal storage condition for this pre-emulsion

Higher-Order Chromatin Structures of Chromosomally Integrated HHV-6A Predict Integration Sites

Human herpesvirus -6A and 6B (HHV-6A/B) can integrate their genomes into the telomeres of human chromosomes. Viral integration can occur in several cell types, including germinal cells, resulting in individuals that harbor the viral genome in every cell of their body. The integrated genome is efficiently silenced but can sporadically reactivate resulting in various clinical symptoms. To date, the integration mechanism and the subsequent silencing of HHV-6A/B genes remains poorly understood. Here we investigate the genome-wide chromatin contacts of the integrated HHV-6A in latently-infected cells. We show that HHV-6A becomes transcriptionally silent upon infection of these cells over the course of seven days. In addition, we established an HHV-6-specific 4C-seq approach, revealing that the HHV-6A 3D interactome is associated with quiescent chromatin states in cells harboring integrated virus. Furthermore, we observed that the majority of virus chromatin interactions occur toward the distal ends of specific human chromosomes. Exploiting this finding, we established a 4C-seq method that accurately detects the chromosomal integration sites. We further implement long-read minION sequencing in the 4C-seq assay and developed a method to identify HHV-6A/B integration sites in clinical samples

    Get PDF
Background: The adverse effects of traditional biomass cookstove (TBC) can be reduced by switching to cleaner cookstoves, improving cooking environment or changing cooking behaviour. Improved knowledge and behaviour regarding adverse effects of traditional biomass cookstove play a significant role in the success of any of the initiatives. This study evaluates the knowledge and behaviour regarding the adverse effects of TBC among rural households in selected desert frontline states of Nigeria.Materials and Methods: A cross-sectional study was conducted among rural households in selected communities in desert frontline states of Northern Nigeria between October, 2016 and January, 2017.The study administered pretested structured questionnaire to 392 households via house-to-house survey. The households were selected based on multistage sampling technique.Result: The findings revealed that majority of the households possess good knowledge regarding health and environmental effects (76.0%), hazards and injuries (94.1%) and drudgery and financial burden (91.8%) arising from TBC. Majority of them practice good behaviour to minimise these adverse effects (92.9%). It includes enclosing flames, use of dry fuel, and keeping children away while cooking. Taking precautions such as storing fuelwood far from the main building, ensuring there are ventilations for indoor kitchens and locating kitchens separately from main buildings are also considered in reducing adverse effects of TBC. Findings on households’ knowledge showed a significant association with behaviour towards adverse effects of TBC.Conclusion: It indicates that households have knowledge and approaches to reduce adverse effects of TBC. The study concluded that the households’ knowledge and behaviour signalling their efforts to reducing adverse effects of TBC. Thus, both are considered as the first step in facilitating cookstove improvement programmes. This information is an integral when designing and implementing sustainable intervention strategies for cookstove improvement, particularly in rural households of developing countries

The susceptibility of Varroa destructor against oxalic acid: A study case

    Get PDF
Varroa destructor Anderson et Trueman is an ectoparasitic mite of the honey bee Apis mellifera L. and it must be controlled in managed bee colonies to maintain colony health. Taking into account that these mites are now resistant to synthetic acaricides worldwide, oxalic acid was suggested as an alternative for Varroa control. Oxalic acid is one of the most common natural miticides used against varroosis by spraying and sublimation administration techniques. It is a natural constituent of honey, very active against the Varroa mite, safe to use for beekeepers, and has no residue problems. Nevertheless, some authors have predicted that the risk of developing resistance to oxalic acid in mites is high. The objective of this research was to assess the susceptibility to oxalic acid of a V. destructor population belonging to a commercial apiary where 64 consecutive control treatments with this acid were performed. Bioassays to assess the oxalic acid susceptibility were performed on two mite populations: (1) a „focal‟ population consisting of mites previously exposed to oxalic acid treatments, and (2) a „naïve‟ population that was never exposed to this acid, which allows setting a reference in the absence of historical data on our „focal‟ mites. The results reported here suggest that the Varroa population exposed during 8 successive years to oxalic acid treatments remains susceptible to this acid.Fil: Maggi, Matías Daniel.
