GNAS1 mutation analysis in gastrointestinal tumors
GNAS1 codes for a part of the a-stimulatory subunit (Gsa) of the G protein. Mutation of GNAS1 has been frequently found in myxoid soft tissues, however, in gastrointestinal tumors, little is known about the mutation status of GNAS1. The aim of the study was to analyze the occurrence of GNAS1 mutations indifferent gastrointestinal, gastroenteropancreatic neuroendocrine tumors (GEP-NETs) and colorectal tumors. Mutation status of GNAS1 exon 8 was analyzed in one hundred thirty-five formalin-fixed, paraffin-embedded (FFPE) gastrointestinal tumor samples including 45 tubular-villous adenomas, 11 tubular adenomas, 6 villousadenomas, 10 hyperplastic gastric polyps, 31 GEP-NETs and 32 colorectal adenocarcinomas by using polymerase chain reaction (PCR) and direct sequencing. Five GNAS1 mutations were found in 2 tubular-villous adenomas, 2 villous adenomas and 1 colorectal adenocarcinoma. No mutations were detected in the tubular adenomas, the hyperplastic gastric polyps or GEP-NETs. GNAS1 mutation is not a frequent molecular event in GEP-NETs or hyperplastic gastric polyps. Resultados preliminares de un estudio paleomagnĂ©tico en secuencias sedimentarias del Cenozoico tardĂo expuestas en Entre RĂos, Argentina
Se presenta un estudio paleomagnĂ©tico preliminar realizado en las formaciones La Juanita y Punta Gorda, del Grupo Punta Gorda y la FormaciĂłn Salto Ander Egg. Estas unidades sedimentarias están expuestas en la Provincia de Entre RĂos. Las direcciones de remanencia magnĂ©ticas encontradas permitieron asignar edades magnĂ©ticas que llevaron a las siguientes interpretaciones: 1) considerar una edad Matuyama para el Grupo Punta Gorda, con la Fm. La Juanita ubicada en el techo del Sanandresense mientras la Fm. Punta Gorda se habrĂa depositado durante el evento normal Olduvai. 2) La Fm. La Juanita tendrĂa un edad más joven (≤ 1.77 Ma) dentro del cron Matuyama y la Fm. Punta Gorda podrĂa corresponder con el evento normal Jaramillo o alternativamente a la base del cron Bruhnes. La primera opciĂłn sugiere que el Grupo Punta Gorda se depositĂł en un ciclo de sedimentaciĂłn continua durante el Pleistoceno temprano (2.5-1.77 Ma). Las otras opciones sugieren una secuencia sedimentaria muy extendida temporalmente, desde 2.5 Ma hasta la base del Pleistoceno medio (≤ 0.78 Ma), con unidades separadas por discontinuidades representativas de lapsos significativos. La polaridad normal y la edad Pleistoceno tardĂo previamente asignada sugieren una edad magnĂ©tica Bruhnes < 0.78 Ma para la Fm. Salto Ander Egg.We present a preliminary paleomagnetic study performed on three late Cenozoic continental sedimentary units, exposed at Entre RĂos province: La Juanita and Punta Gorda formations, belonging to the Punta Gorda Group, and the Salto Ander Egg Formation. The magnetic remanence directions allowed to assign magnetic ages that lead to the following interpretations: 1) Consider a Matuyama age for the Punta Gorda group, then La Juanita Fm. would correspond at the top of Sanandresian while the Punta Gorda Fm. may have been deposited during the Olduvai normal event. 2) La Juanita Fm. was deposited during the Matuyama chron but with a younger age (≤ 1.77 Ma). The Punta Gorda Fm. may correspond to the Jaramillo normal event or alternatively to the bottom of Bruhnes chron. The first option suggests that Punta Gorda Group was deposited in a cycle of continuous sedimentation during the Early Pleistocene, (2.5-1.77 Ma). The other options suggest a time-extended sedimentary sequence from 2.5 Ma to the bottom of the Middle Pleistocene (≤ 0.78 Ma), with units separated by unconformities that represent significant periods of time. A normal polarity and the previously assigned age of upper Pleistocene suggest a Bruhnes magnetic age < 0.78 Ma for Salto Ander Egg Fm.Fil: Walther, Ana Maria. Operationsqualität und Zufriedenheit der Patienten der ambulanten Kataraktchirurgie in Mecklenburg-Vorpommern : Retrospektive Langzeituntersuchung
Die nachfolgend dargestellte Untersuchung verfolgt zweierlei. Einerseits die Darstellung
der Qualität der ambulanten Kataraktchirurgie der Klinik für Augenheilkunde des Dietrich-
Bonhoeffer-Klinikums Neubrandenburg unter Alltagsbedingungen auf Grundlage einer im
April 2005 geschlossenen Vereinbarung zur ambulant qualitätsgesicherten Durchführung
der Kataraktchirurgie nach § 140 a SGB V mit der AOK Mecklenburg-Vorpommern. Zur
Messung der Qualität gehört die Erfassung der klinisch objektivierbaren Parameter sowie
die Untersuchung der Patientenangaben zu ihrer Zufriedenheit mit der Kataraktoperation.
Zweitens erfolgte die Reevaluierung vorhandener und die Etablierung neuer,
ortsspezifischer Korrekturfaktoren der von uns dabei implantierten Intraokularlinsen. Ziel
unserer Untersuchung war es, durch die Ermittlung der Qualität der ambulanten
Kataraktchirurgie und der Zufriedenheit der Patienten einen Beitrag zur Versorgungsforschung
im Allgemeinen und im Sinne einer Optimierung der Patientenversorgung
unseres Zentrums im Speziellen zu leisten. In der Kataraktchirurgie sind etablierte und
mehrheitlich genutzte Qualitätsindikatoren die visuelle Rehabilitation und die refraktive
Treffsicherheit. Wir erweiterten sie um den aus Patientensicht wesentlichen Punkt der
Auswertung der vorliegenden postoperativen Zufriedenheitsangaben unserer Patienten
(Patient-reported Outcome Measure, PROM), die wir mit prä-, intra- und postoperativ
erfassten epidemiologischen und ophthalmologischen Daten in Zusammenhang brachten.
Als Patient-reported Outcome Measure positives Ergebnis (PROM+) wurde von uns die
positive Antwort auf die Frage nach der Allgemeinen Zufriedenheit mit der
Kataraktoperation gewertet. Als Clinical Outcome Measure positiv (COM+) definierten wir
einen Visusanstieg ≥ 0,1.
Die Datenerfassung dauerte von Februar 2005 bis Januar 2009. Nach Plausibilitätskontrolle,
AusschlĂĽssen und Modifikation der uns ursprĂĽnglich zur VerfĂĽgung stehenden 3058
Patientendatensätzen (Augen) konnten wir die Datensätze von 2952 Daten nutzen. Unsere Patienten waren durchschnittlich 74,5 Jahre alt, hatten mehrheitlich eine Cataracta
senilis (96,51 %) und 82,96 % von ihnen wiesen zudem operationsrelevante internistische
Erkrankungen auf. Weiter wurden bei 492 (16,63 %) unserer Patienten ophthalmologische
Risiken registriert, wobei das Glaukom dabei fĂĽhrend war. Visusrelevante Befunde kamen
mit 1135 Fällen (38,45 %) noch häufiger vor und die altersbedingte Makuladegeneration,
kurz als AMD bezeichnet, zeigte sich dabei mit 514 betroffenen Patienten am häufigsten.
89 (3,01 %) unserer Operierten gaben an, in der Vergangenheit schon einmal am Auge
operiert worden zu sein.
Unser Kollektiv repräsentiert die Ergebnisse der im Alltag unserer Klinik zu versorgenden
unselektierten Kataraktpatienten.
Operiert wurde durch fünf unterschiedliche Operateure in retrobulbärer Anästhesie (98,34
%) in Ultraschallphakoemulsifikationstechnik, wobei der Zugang ĂĽberwiegend corneal
erfolgte (98,17 %) und bei Verzicht auf eine anschlieĂźende Wundnaht (99,83 %).
Der durchschnittliche Visus unserer Patienten konnte von präoperativ im Mittel 0,36
(LogMar 0,44) auf postoperativ durchschnittlich 0,75 (LogMar 0,13) verbessert werden.
Dabei erreichten 89,02 % einen Visus von ≥ 0,5 (LogMar 0,30), 60,23 % der Fälle einen
von ≥ 0,8 (LogMar 0,10).
Die Abweichung der erreichten postoperativen von der angestrebten Zielrefraktion betrug
unter unseren Operierten im Mittel + 0,22 dpt. Von unseren Patienten reihen sich 78,86 %
(n=2328) in die Gruppe derer ein, die um ≤ 1,0 Dioptrie von der angestrebten Zielrefraktion
abweichen; 49,86 % (n=1472) gehören zu jenen, die ≤ 0,5 Dioptrien abweichen.
Allgemeine Zufriedenheit mit der Operation wurde von 88,48 % der Patienten angegeben,
6,94 % waren noch bedingt und 4,57 % unzufrieden. Wir fanden heraus, dass diese
Zufriedenheit in einem statistisch signifikanten Zusammenhang mit dem postoperativen
Visus, dem tatsächlichen Visusanstieg und auch mit dem Bestehen einer Visusverbesserung
≥ 0,1 steht. Allerdings hängt die allgemeine Patientenzufriedenheit nicht
ausschlieĂźlich von der Entwicklung des Visus ab. Ebenso wurde eine schwache
Korrelationen zwischen ihr und dem Ausgangsvisus in dem Sinne gefunden, dass ein
höherer Ausgangsvisus mit einer postoperativ größeren Zufriedenheit zusammenhängt.
Weiter stellten wir fest, dass sich ein statistisch signifikanter Zusammenhang zwischen der
empfundenen allgemeinen Zufriedenheit und den ophthalmologischen Risiken, den
visusrelevanten Befunden, insbesondere der AMD, sowie dem Auftreten intraoperativer
Komplikationen (3,52 %) und postoperativer pathologischer Befunde nachweisen lässt.
Keine Korrelation fand sich zwischen der allgemeinen Zufriedenheit mit dem
Operationsergebnis und dem Bestehen internistischer Vorerkrankungen. Allerdings wiesen die unzufriedenen Patienten eine fast doppelt so hohe Prävalenz neurologischpsychiatrischer
Erkrankungen auf. Die empfundene Erleichterung bei Alltagsverrichtungen,
der Zugewinn im Bereich der Verkehrssicherheit und auch die Verbesserung des
Lesevermögens korrelieren jeweils mit den Angaben zur allgemeinen Zufriedenheit mit
dem Operationsergebnis und ebenso mit einem Visusanstieg.
Geringe Zufriedenheit lässt sich zu einem gewissen Anteil durch Komorbiditäten oder
Komplikationen (ophthalmologische Risiken, okuläre Voroperationen, visusrelevante
Befunde, intraoperative Besonderheiten sowie Komplikationen, die Notwendigkeit einer
Re-Operation und das Bestehen postoperativer pathologischer Befunde) und einen
fehlenden Visusanstieg ≥ 0,1 erklären. Patient-reported Outcome negativ, kurz PROM- in
Kombination mit einem Clinical Outcome Measure positiven Ergebnis, kurz COM+ , also
PROM-/COM+ waren 1,7 % unserer Patienten. Sie wiesen ohne Komorbidität, ohne
Komplikationen und trotz eines Visusanstieges ≥ 0,1 dennoch Unzufriedenheit auf. Der
größte Anteil der PROM-/COM+ Patienten wurde im Bereich der Zufriedenheit mit dem
Zugewinn des Lesevermögens gefunden (3,9 %).
Sodass wir die Bedeutung der allgemeinen Lebensunzufriedenheit der Patienten, die
möglichen falschen Erwartungen an eine Kataraktoperation gerade vor dem Hintergrund
existierender Komorbiditäten, welcher Art auch immer, sowie die Relevanz der prä- und
auch postoperativen ambulanten Betreuung herausstellen wollen.
FĂĽr insgesamt elf Intraokularlinsentypen, bei denen die vorher festgelegte Fallzahl von
ĂĽber 20 Augen erreicht wurde, haben wir die vorhandenen ortsspezifischen
Korrekturfaktoren der A-Konstanten ĂĽberprĂĽft, gegebenenfalls korrigiert und neue
generiert. Zusammenfassend konnten wir als Ergebnis fĂĽnf neue etablieren (Acri.Smart
36A, MA60AC, ClariFlex, 36A100SV, MA60BM), vier schon vorhandene bestätigen (L-200,
LI61AOV, L-303, L-313) und zwei ortsspezifische Korrekturfaktoren korrigieren (KS3Ai,
MI60).Quality of cataract surgery and patient satisfaction with outpatient cataract surgery
in Mecklenburg-Vorpommern
Retrospective long-term investigation
The following study aims two things - on the one hand we present the objectified results of
cataract surgery in the outpatient area of the Klinik fĂĽr Augenheilkunde of the Dietrich-
Bonhoeffer-Klinikum Neubrandenburg under routine conditions based on a contract of April
2005 for outpatient quality assured cataract surgery due to § 140 a SGB V which in
particular included informations to patient satisfaction. On the other hand we reviewed the
existing specific A-constants and tried to establish new for these intraocular lenses without
one. Purpose of the study was the monitoring of quality of cataract surgery and the patient
satisfaction in the outpatient health care system to contribute to the healthcare research in
general and to the optimization of our centers patient care in particular. Established and
common used quality indicators were the visual rehabilitation and the refractive accuracy.
We expanded these by the postoperative patient satisfaction (Patient-reported Outcome
Measure), which we correlated with pre-, intra- and postoperative epidemiological and
opthalmological datas. The positive answer to our question towards the overall satisfaction
with cataract surgeries was a Patient-reportet Outcome Measure positive result (PROM+).
Clinical Outcome Measure positive (COM+) was the increased visual acuity ≥ 0,1.
The data sampling took place from February 2005 to September 2009. After testing the
plausibility of our datas, elimination of some and modification of others we got an
aggregate of all in all 2952 patients, from originally 3058.
The mean patient was 74,5 years old, suffers from Cataracta senilis (96,51 %) and in
addition he had an internal disease which may affects his surgery. Further we had 492
(16,63 %) patients with ophthalmological risks, followed by the glaucoma. Vision-relevant diseases were registered in 1135 cases (38,45 %) and the age-related macula
degeneration was most frequently (514 patients). 89 of our patients (3,01 %) insisted a
former eye surgery in the past.
All these categories were recorded without making exlusion criterias out of them.
Consequently we can sum up, that our collective reflects the average of the typical
cataract patient.
Five different surgeons did the surgery using retrobulbar anesthesia and
phakoemulsification. The access took place by the cornea in 98,17 % and in the most
cases we could do the surgery without sewing (99,83 %).
The mean best-corrected visual acuity was 0,75 (LogMar 0,13) or better, compared with
the average of the preoperative visual acuity which was 0,36 (LogMar 0,44). 89,02 % of
patients of our data set achieved a postoperative vision of 0,5 (LogMar 0,30) and better
and 60,23 % 0,8 (LogMar 0,10) and better.
The mean absolute biometry prediction error, means the difference between the target
refraction and the spherical equivalent of the final refraction, was +0,22 D. 78.86 % of our
patients (n=2328) received a final refraction within ±1,0 D of the target refraction, 49,86 %
(n=1472) within ±0,5 D.
Our patients commented a good overall satisfaction with the surgical outcome in 88,48 %
of the cases, 6,94 % were limited satisfied and 4,57 % of our patients were unsatisfied. We
can point out, that there is a significant correlation between this satisfaction and the
postoperative vision, the effective increase of the vision and the existence of an increase
of visual acuity more than or equal 0,1. Nevertheless the overall satisfaction with the
surgical results is not only depending on the developement of the visual acuity. Further we
find low correlations between the satisfaction and the preoperative vision in such a way,
that a higher vision preoperative interlinks with a bigger postoperative satisfaction.
In addition we discover a statistically significant correlation between the overall satisfaction
with the surgery results and the presence of ophthalmological risks, vision-relevant
diseases especially the age-related macula degeneration, the presence of complications
during surgery (which was recorded in 3,52 % of the cases) and postoperative pathological
findings. There was no correlation between this reported satisfaction and the presence of
internal diseases, however we can point out a twice the amount presence of neurological
and psychiatric diseases in the group of discontent than in the group of content patients.
The reported improvement of everyday skills, of road safety and reading skills is
correlating with the overall satisfaction with the outcome of the surgery and as well as with
the improvement of the visual acuity. Low overall satisfaction could be traced to the fact of existing comorbidities or
complications and to a lack of increase of visual acuity ≥ 0,1. Onepointseven percent of
our patients were dissatisfied despite of an increase of VA ≥ 0,1, no comorbidities and no
complications (Patient-reported Outcome Measure negative/Clinical Outcome Measure
positiv, PROM-/COM+). We found the highest percentage of not satisfied patients without
objective reason (Patient-reported Outcome Measure negative/Clinical Outcome Measure
positiv, PROM-/COM+ 3,9 %) in the sector of improvement of reading abilities.
Thus we have to emphasize the influence of the general temper and feeling of an
individual, the possible false expectations toward the cataract surgery especially with
preexisting comorbidities plus the relevance of pre- and postoperative outpatient
treatment.
11 types of intraocular lenses meet the determinated number of cases of 20, so we could
review the existing specific A-constants and try to establish new for these intraocular
lenses without one. All in all we are able to point out the establishment of five new
(Acri.Smart 36A, MA60AC, ClariFlex, 36A100SV, MA60BM), the validation of four
preexisting (L-200, LI61AOV, L-303, L-313) and the correction of two preexisting corrected
A-constants (KS3Ai, MI60)
3D Lagrangian VPM: simulations of the near-wake of an actuator disc and horizontal axis wind turbine
The application of a 3-dimensional Lagrangian vortex particle method has been assessed for modelling the near-wake of an axisymmetrical actuator disc and 3-bladed horizontal axis wind turbine with prescribed circulation from the MEXICO (Model EXperiments In COntrolled conditions) experiment. The method was developed in the framework of the open- source Parallel Particle-Mesh library for handling the efficient data-parallelism on a CPU (Central Processing Unit) cluster, and utilized a O(N log N)-type fast multipole method for computational acceleration. Simulations with the actuator disc resulted in a wake expansion, velocity deficit profile, and induction factor that showed a close agreement with theoretical, numerical, and experimental results from literature. Also the shear layer expansion was present; the Kelvin-Helmholtz instability in the shear layer was triggered due to the round-off limitations of a numerical method, but this instability was delayed to beyond 1 diameter downstream due to the particle smoothing. Simulations with the 3-bladed turbine demonstrated that a purely 3-dimensional flow representation is challenging to model with particles. The manifestation of local complex flow structures of highly stretched vortices made the simulation unstable, but this was successfully counteracted by the application of a particle strength exchange scheme. The axial and radial velocity profile over the near wake have been compared to that of the original MEXICO experiment, which showed close agreement between results
GNAS-1 Mutationen in gastrointestinalen Tumoren
Hintergrund:
Weltweit tragen maligne Neoplasien, besonders die des gastrointestinalen Traktes (GIT) zu einer hohen Morbidität und Mortalität bei. Viele dieser Entartungen haben ihren Ursprung in Mutationen. Genetische Sequenzierung gibt uns die Möglichkeit, Gemeinsamkeiten in der Tumorigenese zu finden, Diagnosen schneller zu stellen und eine Therapie einzuleiten. Auf dem Chromosom 20 erstreckt sich der Genlocus des GNAS1 Gens über 13 Exons, welches die α-Subunit eines heterotrimeren membranständigen Rezeptors kodiert, dessen Hauptaufgabe die ubiquitäre Signalübertragung ist. Mutationen dieses Genlocus können Neoplasien zur Folge haben, die schon in vorangegangenen Studien besonders in Weichteiltumoren und in anderen Tumoren nachgewiesen worden. In vorliegender Arbeit wurden unterschiedliche Gruppen von Tumorentitäten des Gastrointenstinaltraktes auf eine mögliche GNAS1 Mutation untersucht.
Methoden:
Insgesamt wurden 135 Proben analysiert, darunter 62 Adenome des Kolons (45 tubulär-villös, elf tubulär, sechs rein villös), zehn hyperplastische Magenpolypen, 31 gastroenteropankreatische neuroendokrine Tumore (GEP-NET´s) und 32 kolorektale Adenokarzinome. Die DNA wurde aus formalinfixierten Paraffinblöcken extrahiert, mittels PCR ein 252 bp messender Abschnitt des GNAS1 Gens von Exon 8 amplifiziert, mit Positiv- und Negativ-Kontrollen verifiziert und anschließend erfolgte die PCR-Produktüberprüfung mittels 1,5-prozentiger gelRED-gefärbter Agarose-Gel-Elektrophorese. Die erhaltene und anschließend aufgereinigte DNA wurde durch Kapillarelektrophorese sequenziert und auf Mutationen ausgewertet.
Ergebnisse:
Von allen analysierten Proben wiesen fĂĽnf (3,7%) eine GNAS1 Mutation auf. Vier der 62 Adenome (6,45%) zeigten eine R201H, sowie ein kolorektales Karzinom (3,13%) eine R201C Mutation. In keinem GEP-NET oder hyperplastischen Magenpolypen konnte eine GNAS1 Mutation nachgewiesen werden.
Schlussfolgerung:
Adenome zeigen teilweise GNAS1 Mutationen, vor allem aber dann, wenn sie histologisch villöse Anteile zeigen. Besonders häufig kommen GNAS1 Mutationen im Rektum vor, scheinen aber in der Entstehung von kolorektalen Karzinomen keine große Rolle zu spielen. GEP-NET´s sowie hyperplastische Magenpolypen entwickeln sich höchstwahrscheinlich ohne Beteiligung einer GNAS1 Mutation. Als genetischer Marker spielen GNAS1 Mutation derzeit in der Diagnostik keine Rolle. Es scheint Unterschiede zwischen GNAS1 Mutations-Vorkommen zwischen dem japanischen und kaukasischen Patientenkohorten zu geben. Dies sollte ein Ansatz für zukünftige wissenschaftliche Studien werden, um die Zusammenhänge zwischen GNAS1 Mutationen in der Tumorigenese verschiedener Entitäten und unterschiedlicher Population besser zu verstehen
Derivation and analysis of the analytical velocity and vortex stretching expressions for an O (N log N)-FMM
In the current paper, a method for deriving the analytical expressions for the velocity and vortex stretching terms as a function of the spherical multipole expansion approximation of the vector potential is presented. These terms are essential in the context of 3D Lagrangian vortex particle methods combined with fast summation techniques. The convergence and computational efficiency of this approach is assessed in the framework of an O(N log N)-type Fast Multipole Method (FMM), by using vorticity particles to simulate a system of coaxial vortex rings for which also the exact results are known. It is found that the current implementation converges rapidly to the exact solution with increasing expansion order and acceptance factor. An investigation into the computational efficiency demonstrated that the O(N log N)-type FMM is already viable for a particle size of only several thousands and that this speedup increases significantly with the number of particles. Finally, i t is shown that the implementation of the FMM with the current analytical expressions is at least twice as fast as when opting for using even the simplest implementation of finite differences instead
Rapid eradication of colon carcinoma by Clostridium perfringens Enterotoxin suicidal gene therapy
Background Bacterial toxins have evolved to an effective therapeutic option
for cancer therapy. The Clostridium perfringens enterotoxin (CPE) is a pore-
forming toxin with selective cytotoxicity. The transmembrane tight junction
proteins claudin-3 and -4 are known high affinity CPE receptors. Their
expression is highly upregulated in human cancers, including breast, ovarian
and colon carcinoma. CPE binding to claudins triggers membrane pore complex
formation, which leads to rapid cell death. Previous studies demonstrated the
anti-tumoral effect of treatment with recombinant CPE-protein. Our approach
aimed at evaluation of a selective and targeted cancer gene therapy of
claudin-3- and/or claudin-4- expressing colon carcinoma in vitro and in vivo
by using translation optimized CPE expressing vector. Methods In this study
the recombinant CPE and a translation optimized CPE expressing vector (optCPE)
was used for targeted gene therapy of claudin-3 and/or -4 overexpressing colon
cancer cell lines. All experiments were performed in the human SW480, SW620,
HCT116, CaCo-2 and HT-29 colon cancer and the isogenic Sk-Mel5 and Sk-Mel5
Cldn-3-YFP melanoma cell lines. Claudin expression analysis was done at
protein and mRNA level, which was confirmed by immunohistochemistry. The CPE
induced cytotoxicity was analyzed by the MTT cytotoxicity assay. In addition
patient derived colon carcinoma xenografts (PDX) were characterized and used
for the intratumoral in vivo gene transfer of the optCPE expressing vector in
PDX bearing nude mice. Results Claudin-3 and -4 overexpressing colon carcinoma
lines showed high sensitivity towards both recCPE application and optCPE gene
transfer. The positive correlation between CPE cytotoxicity and level of
claudin expression was demonstrated. Transfection of optCPE led to targeted,
rapid cytotoxic effects such as membrane disruption and necrosis in claudin
overexpressing cells. The intratumoral optCPE in vivo gene transfer led to
tumor growth inhibition in colon carcinoma PDX bearing mice in association
with massive necrosis due to the intratumoral optCPE expression. Conclusions
This novel approach demonstrates that optCPE gene transfer represents a
promising and efficient therapeutic option for a targeted suicide gene therapy
of claudin-3 and/or claudin-4 overexpressing colon carcinomas, leading to
rapid and effective tumor cell killing in vitro and in vivo
Comparison of physicochemical properties of commercial UHT-treated plant-based beverages and cow's milk
A comparison of consumer-relevant physicochemical and technofunctional properties was performed between plant-based beverages (PBBs) and cow’s milk treated at ultra-high temperatures. The PBBs’ viscosities and pH values were similar to or higher than those in cow’s milk. The PBBs were less white, and their mean particle sizes were usually considerably larger than those of cow’s milk. Foam heights were quite different, from 41.5 mm to 173 mm at room temperature (milk foam height: 134.8 mm) and 50.9 mm to 203.6 mm at 60 °C (milk foam height: 179.3 mm), with a median bubble size radius (root mean square) of 14.0–149.5 μm (milk bubble size: 18 μm) and 31.0–175.5 μm (milk bubble size: 82.8 μm). Our correlation revealed that phytic acid (PA) might affect foam height at 60 °C, the temperature of interest for the consumption of hot beverages. This may be of interest, as PA might be reduced in these beverages for nutritional reasons
