    Exploring with [18F]UCB-H the in vivo cariations in SV2A expression through the kainic acid rat model of temporal lobe epilepsy

    Purpose The main purpose of this study was to understand how the positron emission tomography (PET) measure of the synaptic vesicle 2A (SV2A) protein varies in vivo during the development of temporal lobe epilepsy (TLE) in the kainic acid rat model. Procedures Twenty Sprague Dawley male rats were administered with multiple systemic doses of saline (control group, n = 5) or kainic acid (5 mg/kg/injection, epileptic group, n = 15). Both groups were scanned at the four phases of TLE (early, latent, transition, and chronic phase) with the [F-18]UCB-H PET radiotracer and T2-structural magnetic resonance imaging. At the end of the scans (3 months post-status epilepticus), rats were monitored for 7 days with electroencephalography for the detection of spontaneous electrographic seizures. Finally, the immunofluorescence staining for SV2A expression was performed. Results Control rats presented a significant increase in [F-18]UCB-H binding at the last two scans, compared with the first ones (p < 0.001). This increase existed but was lower in epileptic animals, producing significant group differences in all the phases of the disease (p < 0.028). Furthermore, the quantification of the SV2A expression in vivo with the [F-18]UCB-H radiotracer or ex vivo with immunofluorescence led to equivalent results, with a positive correlation between both. Conclusions Even if further studies in humans are required, the ability to detect a progressive decrease in SV2A expression during the development of temporal lobe epilepsy supports the use of [F-18]UCB-H as a useful tool to differentiate, in vivo, between healthy and epileptic animals along with the development of the epileptic disease

    A Perspective on Recent Advances in Phosphorene Functionalization and its Application in Devices

    Phosphorene, the 2D material derived from black phosphorus, has recently attracted a lot of interest for its properties, suitable for applications in material science. In particular, the physical features and the prominent chemical reactivity on its surface render this nanolayered substrate particularly promising for electrical and optoelectronic applications. In addition, being a new potential ligand for metals, it opens the way for a new role of the inorganic chemistry in the 2D world, with special reference to the field of catalysis. The aim of this review is to summarize the state of the art in this subject and to present our most recent results in preparation, functionalization and use of phosphorene and its decorated derivatives. In particular, we discuss several key points, which are currently under investigation: the synthesis, the characterization by theoretical calculations, the high pressure behaviour of black phosphorus, as well as decoration with nanoparticles and encapsulation in polymers. Finally, device fabrication and electrical transport measurements are overviewed on the basis of recent literature and new results collected in our laboratories

    Differences in the clinical management of women and men after detection of a solitary pulmonary nodule in clinical practice

    Objectives To explore differences in the clinical management of men and women in the 5 years after detecting a solitary pulmonary nodule (SPN) by chest radiograph or CT in routine clinical practice. Methods We followed up 545 men and 347 women with an SPN detected by chest radiograph or CT in a retrospective cohort of 25,422 individuals undergoing routine thoracic imaging in 2010–2011.We compared the frequency of each management strategy (no further test, immediate intervention or follow up) according to sex by means of chi-squared.We estimated the relative risk of women versus men of having been followed up instead of an immediate intervention using multivariate logistic regression. We compared by sex the time between detection of the nodule and lung cancer diagnosis, the time between diagnosis and death by means of Mann-Whitney U test and the cumulative effective dose of radiation in each management strategy by means of t test. Results Women were more likely than men to have follow-up rather than immediate intervention (aRR = 1.8, CI 1.3–2.7, p = 0.002), particularly in those who underwent CT (aRR = 4.2, CI 1.9–9.3, p < 0.001). The median time between SPN detection and lung cancer diagnosis was higher in women (4.2 months, interquartile range (IQR) 5.1) than in men (1.5 months, IQR 16.2). The mean cumulative effective dose was 21.3 mSv, 19.4 mSv in men and 23.9mv in women (p = 0.023). Conclusions Our results could reflect decisions based on a greater suspicion of lung cancer in men. The incidental detection of SPNs is increasing, and it is necessary to establish clear strategies aimed to reduce variability in their management according to patient’s sex

    The Dual Role of Microglia in ALS: Mechanisms and Therapeutic Approaches.

    Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by a non-cell autonomous motor neuron loss. While it is generally believed that the disease onset takes place inside motor neurons, different cell types mediating neuroinflammatory processes are considered deeply involved in the progression of the disease. On these grounds, many treatments have been tested on ALS animals with the aim of inhibiting or reducing the pro-inflammatory action of microglia and astrocytes and counteract the progression of the disease. Unfortunately, these anti-inflammatory therapies have been only modestly successful. The non-univocal role played by microglia during stress and injuries might explain this failure. Indeed, it is now well recognized that, during ALS, microglia displays different phenotypes, from surveillant in early stages, to activated states, M1 and M2, characterized by the expression of respectively harmful and protective genes in later phases of the disease. Consistently, the inhibition of microglial function seems to be a valid strategy only if the different stages of microglia polarization are taken into account, interfering with the reactivity of microglia specifically targeting only the harmful pathways and/or potentiating the trophic ones. In this review article, we will analyze the features and timing of microglia activation in the light of M1/M2 phenotypes in the main mice models of ALS. Moreover, we will also revise the results obtained by different anti-inflammatory therapies aimed to unbalance the M1/M2 ratio, shifting it towards a protective outcome

    Novel experimental model of maintained acute lung injury

    Trabajo presentado en el 23rd Annual Congress European Respiratory Society (ERS) celebrado del 7 al 11 de septiembre de 2013 en Barcelona (España)Abstract publicado en "European Respiratory Journal" 42 (Suppl 57): 50s-51s (2013)Rationale: Several animal models have been developed to study acute lung injury (ALI); however the majority of these studies are focused on different mechanisms within the acute phase. These models do not allow studying the mechanisms in the later phases or testing any possible long-term treatment. The aim of this study was to develop an experimental ALI model simulating bronchial aspiration of gastric contents with bacterial superinfection with alveolar epithelial damage persisting over time. Methods: Sprague-Dawley rats (200-250g) were anesthetized with isofl uorane. ALI was induced by intratracheal instillation of HCl (1 μl/g, 0.1 mol/L pH=1.4) followed by instillation of LPS from Escherichia coli O55:B5 (0, 10, 20, 30 or 40μg/g b.w.) two hours later. Control rats were treated with intratracheal instillations of saline. After 72h, the animals were sacrifi ced and bronchoalveolar lavage fl uid (BALF) was sampled for further analysis of total protein concentration by bicinchoninic acid method. Results: At 72 h, rats suffered a signifi cant loss of weight proportional to the administered dose of LPS (5.6% with 10μg/g b.w, 12.6% with 20μg/g b.w, 14.2% with 30μg/g b.w and 17.7% with 40μg/g b.w). Control rats gained in weight at 72h. LPS at 10, 20, 30 and 40μg/g b.w induced a 1.7, 2.5, 2.9 and 3.4 fold increase in total protein concentration in BAL fl uid, respectively, refl ecting a substantial increase proportional to the LPS dose. Conclusion: The degree of weight loss and the increase of total protein concentration in BAL fl uid in the current model may refl ect disease severity and progression.     Sobre la validez de la Biozona Macrochorobates scalabrinii (Huayqueriense temprano, Mioceno tardío): Análisis multi-proxy del enigmático sitio Arenas Blancas (provincia de Buenos Aires, Argentina)

    Arenas Blancas es una localidad fosilífera poco conocida, ubicada en el curso inferior del arroyo Chasicó (provincia de Buenos Aires, Argentina), pero con gran relevancia desde el punto de vista bioestratigráfico. La Biozona Macrochorobates scalabrinii, propuesta como base bioestratigráfica del Piso/Edad Huayqueriense temprano (Mioceno tardío) fue definida en este sitio, aunque su contexto geológico y registro faunístico nunca han sido estudiados en detalle. En este trabajo analizamos el sitio Arenas Blancas desde diferentes perspectivas, como así también un sitio cercano aquí denominado Curva de la Vaca, a fin de aportar nuevas interpretaciones sobre su origen, edad y bioestratigrafía. Las características sedimentológicas, estratigráficas y geomorfológicas de ambos sitios sugieren que las secuencias aflorantes corresponden a depósitos fluviales/aluviales que representan terrazas cuaternarias. El análisis taxonómico de la asociación faunística de Arenas Blancas permitió reconocer 14 taxones de mamíferos, como así también peces, reptiles y aves. Esta asociación muestra una clara correlación con la asociación de la Formación Cerro Azul asignada al Piso/Edad Chasiquense (Mioceno tardío), también registrada en el curso inferior del arroyo Chasicó. Por otra parte, se considera que el uso de los taxones que caracterizan la Biozona Macrochorobates scalabrinii (incluyendo esta especie) no está avalado por su estatus taxonómico y/o su distribución temporal. El estudio tafonómico muestra que la asociación está constituida por especímenes retrabajados; en este contexto, se propone que los eventos fluviales/aluviales ocurridos durante el Pleistoceno tardío-Holoceno erosionaron el sustrato del Mioceno tardío y retrabajaron los restos fósiles. Sobre la base de la evidencia disponible, se rechaza la validez de la Biozona Macrochorobates scalabrinii como unidad bioestratigráfica.Arenas Blancas is a poorly known fossiliferous site located in the lower reach of the Chasicó creek (Buenos Aires Province, Argentina), with great relevance from a biostratigraphic viewpoint. The Macrochorobates scalabrinii Biozone was defined in this site, proposed as the biostratigraphic basis of the early Huayquerian Stage/Age (Late Miocene); however, the geological context and faunal record of this site have never been studied in detail. In this work, we perform a multi-proxy analysis of the Arenas Blancas site, as well as a nearby site here called Curva de la Vaca, and provide new interpretations on their origin, age, and biostratigraphy. Sedimentological, stratigraphic, and geomorphological characteristics of both sites suggest that the sequences include fluvial/alluvial deposits that represent Quaternary terraces. The taxonomic analysis of the Arenas Blancas faunal assemblage evidences the presence of 14 mammal taxa, together with some fishes, reptiles, and birds; the assemblage is correlated with the assemblage from the Cerro Azul Formation assigned to the Chasicoan Stage/Age (Late Miocene), also recovered from the lower reach of the Chasicó creek. The use of the taxa proposed in previous works to characterize the Macrochorobates scalabrinii Biozone (including this species) is not supported due to their taxonomic status and/or temporal distribution. Taphonomic evaluation indicates that the assemblage is constituted by reworked specimens; in this frame, it is proposed that fluvio/alluvial events, occurred during the Late Pleistocene-Holocene, eroded the Upper Miocene substrate and reworked fossil remains. Based on the present evidence, the Macrochorobates scalabrinii Biozone is here rejected as a valid biostratigraphic unit.Fil: Tomassini, Rodrigo Leandro.
