    O presente trabalho busca analisar, sob a concepção Histórico Cultural, os sentidos atribuídos por licenciandos de física à aprendizagem da docência no processo de estágio desenvolvido na disciplina de Metodologia do Ensino de Física. Os sentidos que licenciandos atribuem à docência se relacionam com os motivos que os mobilizam para ações durante sua atividade de estudo, o que permite apreender sua historicidade. Estudar fatores que colaboram para a atribuição de sentidos permite que, ao serem identificados, possam auxiliar na organização das disciplinas, desencadeando ações que mobilizem futuros professores na aprendizagem da docência de maneira mais significativa para cada sujeito

    Lagochilascariasis leading to severe involvement of ocular globes, ears and meninges

    A case report of a 31 year-old woman from Paraíba State (North-Eastern Brazil) that presented severe involvement of ocular globes, ears and meninges. Diagnosis was established after enucleation of her left eye, when adult worms were seen in the midst of a granulomatous inflammatory process. Her response to the initial treatment with levamisole and cambendazole was good, but there was a relapse after the fifth month of treatment even with maintenance doses of both medications. She later received ivermectin and albendazol and responded well.Paciente do sexo feminino, com 31 anos, procedente da Paraíba, apresentava envolvimento severo de ambos os globos oculares, com perda da visão à esquerda, comprometimento da audição e das meninges. Após enucleação do olho esquerdo fragmento de helminto identificado como Lagochilascaris minor foi observado em processo inflamatório granulomatoso. Inicialmente a paciente foi tratada com levamisol e cambendazol, com bom resultado. Verificou-se, todavia, piora do quadro após cinco meses, com eliminação de larvas do ascarídeo em lesão presente na órbita esquerda, embora a medicação fosse mantida com administração periódica. Houve boa resposta terapêutica, com regressão do quadro, após substituição dos anti-helmínticos anteriores pela associação ivermectina e albendazol

    Pré-tratamento de resíduos agro-industriais e novas perspectivas na produção de bioprodutos

    Visando contribuir para a melhoria das condições ambientais pela eliminação dos resíduos agrícolas/agroindustriais, bem como proporcionar a formação de uma linha de pesquisa com propósito de captar e disseminar informações técnicas e tecnológicas agregando maior valor nestes subprodutos ou resíduos é que o presente trabalho foi realizado. Deste modo, o sabugo de milho in natura foi submetido a dois diferentes processos de prétratamento para a extração das hemiceluloses, como etapa inicial de aplicação integral do resíduo, onde posteriormente estudos e aplicação do complexo celulose-lignina resultante seriam realizados. Dentro deste contexto, o objetivo deste trabalho foi determinar a eficiência de extração de frações de hemiceluloses de sabugo de milho por processos alcalinos (0,75 a 1,25 mol/L em 42,5ºC/180 min), e processos de autohidrólise (165 a 185ºC, razão líquida de 10:1p/p por 40 min). Este procedimento indicou o potencial de pré-tratamentos, em especial dos licores hemicelulosicos obtidos, como parte de um processo que conduza à utilização de materiais lignocelulósicos em diferentes processos biotecnológicos

    Cellular, Molecular and Genotoxic Effects of Digested Titanium Dioxide Nanomaterials

However, the possibility of adverse effects in gastrointestinal tract is unclear. Aiming to study the impact of digestion on the NMs’ properties and their cellular/molecular effects, two human intestinal cell lines were used, Caco-2 and HT29-MTX-E12. After exposure to TiO2NMs(NM-102, NM-103, NM-105), undigested or subjected to standardized static in vitro digestion method (mimicking human digestion), the cells were analyzed for toxicity, genotoxicity, reactive oxygen species, NM uptake and intestinal translocation. We showed that in vitro digestion of TiO2NMs may increase their toxicity and DNA-damaging effect, depending on the NM, more relevant for the rutile/anatase NM-105, possibly due to its smaller hydrodynamic size in the cellular medium. Effects on chromosomal integrity were seen in HT29-MTX-E12 cells, for all tested TiO2NMs, especially after digestion. Internalization into early endosomes was confirmed for NM-103 and NM-105, before and after digestion, in monolayers of both cell lines, and at the apical membrane of polarized Caco-2 cells. The internalized NMs accumulated in late endosomes/multivesicular bodies, partially transversing the basolateral membrane of polarized Caco-2 cells without changing transepithelial electrical resistance or epithelial marker abundance. These results suggest that part of the TiO2NMs can be transcytosed through colonic epithelia without disrupting intestinal barrier integrity.     Prognostic stratification of adult primary glioblastoma multiforme patients based on their tumor gene amplification profiles

    Several classification systems have been proposed to address genomic heterogeneity of glioblastoma multiforme, but they either showed limited prognostic value and/or are difficult to implement in routine diagnostics. Here we propose a prognostic stratification model for these primary tumors based on tumor gene amplification profiles, that might be easily implemented in routine diagnostics, and potentially improve the patients management. Gene amplification profiles were prospectively evaluated in 80 primary glioblastoma multiforme tumors using single-nucleotide polymorphism arrays and the results obtained validated in publicly available data from 267/347 cases. Gene amplification was detected in 45% of patients, and chromosome 7p11.2 including the EGFR gene, was the most frequently amplified chromosomal region – either alone (18%) or in combination with amplification of DNA sequences in other chromosomal regions (10% of cases). Other frequently amplified DNA sequences included regions in chromosomes 12q(10%), 4q12(7%) and 1q32.1(4%). Based on their gene amplification profiles, glioblastomas were subdivided into: i) tumors with no gene amplification (55%); ii) tumors with chromosome 7p/EGFR gene amplification (with or without amplification of other chromosomal regions) (38%); and iii) glioblastoma multiforme with a single (11%) or multiple (6%) amplified DNA sequences in chromosomal regions other than chromosome 7p. From the prognostic point of view, these amplification profiles showed a significant impact on overall survival of glioblastoma multiforme patients (p>0.001).     Ação de insetos (Coleoptera e Isoptera) na destruição de igrejas de importância histórica de Recife e Olinda

    The destruction of historical monuments of great artistic, cultural and economic value is among the most serious types of damage caused by insects. This research aimed at investigating the role of insects in the destruction of churches of historical value according to the “Instituto do Patrimônio Histórico e Artístico Nacional”. We aimed at identifying insects mainly responsible for damages and the methods for prevention and control by means of questionnaires, visits, document surveys and literature review. We observed that insects, especially termites and borers, are the main responsible for damages, according to 90% of the interviewees. We also noticed that the most frequently used method for prevention and control are based on homemade poison mixtures.Entre os danos causados por insetos, destaca-se a destruição de monumentos históricos de grande valor artístico, cultural e econômico. Esta pesquisa teve como objetivo investigar o papel dos insetos na destruição de igrejas de Recife e Olinda, tombadas pelo Instituto do Patrimônio Histórico e Artístico Nacional. Através de visitas, questionários e pesquisa documental e bibliográfica, buscou-se identificar os principais causadores de danos e os métodos preventivos e de controle. Observou-se que os insetos, especialmente os cupins e coleobrocas, são os principais agentes causadores de danos, segundo 90% dos entrevistados. Observou-se também que os métodos de prevenção e controle de insetos mais utilizados baseiam-se no uso de venenos caseiros

    Novel Role for the AnxA1-Fpr2/ALX Signaling Axis as a Key Regulator of Platelet Function to Promote Resolution of Inflammation

    Background: Ischemia reperfusion injury (I/RI) is a common complication of cardiovascular diseases. Resolution of detrimental I/RI-generated prothrombotic and proinflammatory responses is essential to restore homeostasis. Platelets play a crucial part in the integration of thrombosis and inflammation. Their role as participants in the resolution of thromboinflammation is underappreciated; therefore we used pharmacological and genetic approaches, coupled with murine and clinical samples, to uncover key concepts underlying this role. Methods: Middle cerebral artery occlusion with reperfusion was performed in wild-type or annexin A1 (AnxA1) knockout (AnxA1-/-) mice. Fluorescence intravital microscopy was used to visualize cellular trafficking and to monitor light/dye-induced thrombosis. The mice were treated with vehicle, AnxA1 (3.3 mg/kg), WRW4 (1.8 mg/kg), or all 3, and the effect of AnxA1 was determined in vivo and in vitro. Results: Intravital microscopy revealed heightened platelet adherence and aggregate formation post I/RI, which were further exacerbated in AnxA1-/- mice. AnxA1 administration regulated platelet function directly (eg, via reducing thromboxane B2 and modulating phosphatidylserine expression) to promote cerebral protection post-I/RI and act as an effective preventative strategy for stroke by reducing platelet activation, aggregate formation, and cerebral thrombosis, a prerequisite for ischemic stroke. To translate these findings into a clinical setting, we show that AnxA1 plasma levels are reduced in human and murine stroke and that AnxA1 is able to act on human platelets, suppressing classic thrombin-induced inside-out signaling events (eg, Akt activation, intracellular calcium release, and Ras-associated protein 1 [Rap1] expression) to decrease IIbβ3 activation without altering its surface expression. AnxA1 also selectively modifies cell surface determinants (eg, phosphatidylserine) to promote platelet phagocytosis by neutrophils, thereby driving active resolution. (n=5-13 mice/group or 7-10 humans/group.) Conclusions: AnxA1 affords protection by altering the platelet phenotype in cerebral I/RI from propathogenic to regulatory and reducing the propensity for platelets to aggregate and cause thrombosis by affecting integrin (IIbβ3) activation, a previously unknown phenomenon. Thus, our data reveal a novel multifaceted role for AnxA1 to act both as a therapeutic and a prophylactic drug via its ability to promote endogenous proresolving, antithromboinflammatory circuits in cerebral I/RI.     Annona coriacea Mart. fractions promote cell cycle arrest and inhibit autophagic flux in human cervical cancer cell lines

    Plant-based compounds are an option to explore and perhaps overcome the limitations of current antitumor treatments. Annona coriacea Mart. is a plant with a broad spectrum of biological activities, but its antitumor activity is still unclear. The purpose of our study was to determine the effects of A. coriacea fractions on a panel of cervical cancer cell lines and a normal keratinocyte cell line. The antitumor effect was investigated in vitro by viability assays, cell cycle, apoptosis, migration, and invasion assays. Intracellular signaling was assessed by Western blot, and major compounds were identified by mass spectrometry. All fractions exhibited a cytotoxic effect on cisplatin-resistant cell lines, SiHa and HeLa. C3 and C5 were significantly more cytotoxic and selective than cisplatin in SiHa and Hela cells. However, in CaSki, a cisplatin-sensitive cell line, the compounds did not demonstrate higher cytotoxicity when compared with cisplatin. Alkaloids and acetogenins were the main compounds identified in the fractions. These fractions also markedly decreased cell proliferation with p21 increase and cell cycle arrest in G2/M. These effects were accompanied by an increase of H2AX phosphorylation levels and DNA damage index. In addition, fractions C3 and C5 promoted p62 accumulation and decrease of LC3II, as well as acid vesicle levels, indicating the inhibition of autophagic flow.
