    Suv4-20h Abrogation Enhances Telomere Elongation during Reprogramming and Confers a Higher Tumorigenic Potential to iPS Cells

    Reprogramming of adult differentiated cells to induced pluripotent stem cells (iPS) cells has been achieved by over-expression of specific transcription factors. Nuclear reprogramming induces a series of profound changes at the telomeres of the parental differentiated cells, including a telomerase-dependent telomere elongation and the remodeling of telomeric chromatin. In particular, iPS cells show a decreased density of H4K20me3 heterochromatic mark at telomeres compared to the parental cells. Suv4-20h1 and Suv4-20h2 histone methytransferases (HMTases) are responsible for the trimethylation of H4K20 at telomeres, as cells deficient for both HMTases show decreased levels of H4K20me3 at telomeric chromatin. Here, we set to address the role of the Suv4-20h enzymes in telomere reprogramming by generating bona-fide iPS cells from mouse embryonic fibroblasts (MEFs) double null for both HMTases (Suv4-20dn MEFs). We found that Suv4-20h deficiency enhances telomere elongation during reprogramming without altering their ability to protect the chromosome ends or the efficiency of reprogramming. Moreover, teratomas generated from Suv4-20dn iPS cells also have elongated telomeres and an increased growth rate when compared to wild-type controls. These results indicate that abrogation of Suv4-20h enzymes and loss of heterochromatic mark H4K20me3 at telomeric heterochromatin facilitates telomere reprogramming and provides an increased tumorigenic potential to the resulting iPS cells

    We studied the effects of collision energy (ET) on the dynamics of the title reaction using the quasiclassical trajectory method on an analytical triatomic potential energy surface we had derived for this system. We compared the dependence of the scalar and two-vector properties of the reaction on ET with experimental data and obtained a quite good agreement. The results can be explained in terms of the coexistence of two microscopic reaction mechanisms: insertion and abstraction. The former mechanism is the most important one, although the contribution of the latter increases with ET

    A plasmid DNA encoding the viral hemorrhagic septicaemia virus (VHSV)-G glycoprotein under the control of 5′ upstream sequences (enhancer/promoter sequence plus both non-coding 1st exon and 1st intron sequences) from carp β-actin gene (pAE6-GVHSV) was compared to the vaccine plasmid usually described the gene expression is regulated by the human cytomegalovirus (CMV) immediate-early promoter (pMCV1.4-GVHSV). We observed that these two plasmids produced a markedly different profile in the level and time of expression of the encoded-antigen, and this may have a direct effect upon the intensity and suitability of the in vivo immune response. Thus, fish genetic immunisation assays were carried out to study the immune response of both plasmids. A significantly enhanced specific-antibody response against the viral glycoprotein was found in the fish immunised with pAE6-GVHSV. However, the protective efficacy against VHSV challenge conferred by both plasmids was similar. Later analysis of the transcription profile of a set of representative immune-related genes in the DNA immunized fish suggested that depending on the plasmid-related regulatory sequences controlling its expression, the plasmid might activate distinct patterns of the immune system. All together, the results from this study mainly point out that the selection of a determinate encoded-antigen/vector combination for genetic immunisation is of extraordinary importance in designing optimised DNA vaccines that, when required for inducing protective immune response, could elicit responses biased to antigen-specific antibodies or cytotoxic T cells generation

    It was possible to capture and transport BFT YOTYs around 1.5kg from the Murcia shore to land based facilities of IEO in Mazarrón. Nevertheless the survival of individuals did not go beyond 72 hours. The accidental friction with the tank walls and the handling with the rubber net when fish were transfer from the boat to the van (Figure 1) and from the van to the land-based tank could be the reason of the final mortality. Thus, it would be necessary to design a system that minimizes this friction. On the other hand, it seems very important the fish size in terms of survival.     Late Cambrian – Tremadocian faunas and events from Angosto del Moreno Section, Eastern Cordillera, Argentina

    The Santa Victoria Group (SVG, late Upper Cambrian – Caradocian) comprises pre–Ashgillian Ordovician deposits of the Argentinean Eastern Cordillera. The most significant section of the SVG in the western flank of the Eastern Cordillera is located in the Angosto del Moreno area (Figure 1a, b). At this locality, the SVG unconformably overlies the Mesón Group (Cambrian s.l.), and unconformably underlies Cretaceous rocks (Yacoraite Formation). Upper Cambrian to lower Lower Ordovician units are separated from upper Lower to Middle Ordovician units (Parcha and Sepulturas formations) by the Tumbaya unconformity (Figure 2). The Angosto del Moreno Section of the SVG is exceptional in terms of the quality of exposures, continuity of deposits, richness of fossils, and accessibility. Diverse aspects concerning the Ordovician geology of this study area have been discussed by Moya et al. (1994, 1998), Moya and Albanesi (2000), Moya and Monteros (2000), Malanca and Brandán (2000), and Gómez Martínez et al. (2002). Previous data and recent paleontological collections enable a preliminary biostratigraphic scheme (Figure 2) for the Upper Cambrian to Tremadocian units of the SVG.
