    Current observations from a looking down vertical V-ADCP: interaction with winds and tide? The case of Giglio Island (Tyrrhenian Sea, Italy)

    Summary In the context of the environmental monitoring of the Concordia wreck removal project, measurements of currents, winds and sea level height were made along the eastern coast of the Giglio Island, Tyrrhenian Sea (Italy), during 2012–2013. The aim of the study was to investigate the effect of atmospheric forcing and periodic sea-level changes on the coastal currents. Normalised Cross-Correlation Function analysis allowed us to correlate these observations. A marked inter-seasonal variability was found in both current and local wind velocity observations but a significant level of correlation between the data was only found during strong wind events. Current and wind directions appeared to be uncorrelated and current measurements showed a predominant NW–SE direction, presumably linked to the shape and orientation of Giglio Island itself. During strong winds from the SSE, current flow was towards the NNW but it suddenly switched from the NNW to the SE at the end of wind events. The results show that, at Giglio Island, currents are principally dominated by the general cyclonic Tyrrhenian circulation, and, secondly, by strong wind events. The sea level had no effects on the current regime

    Establishing surveillance areas for tackling the invasion of Vespa velutina in outbreaks and over the border of its expanding range

    The yellow-legged hornet Vespa velutina is an invasive alien species in many areas of the world. In Europe, it is considered a species of Union concern and national authorities have to establish surveillance plans, early warning and rapid response systems or control plans. These strategies customarily require the assessment of the areas that could be colonised beyond outbreaks or expanding ranges, so as to establish efficient containment protocols. The hornet is spreading through a mix of natural diffusion and human-mediated transportation. Despite the latter dispersion mode is hardly predictable, natural diffusion could be modelled from nest data of consecutive years. The aim of this work is to develop a procedure to predict the spread of the yellow-legged hornet in the short term in order to increase the efficiency of control plans to restrain the diffusion of this species. We used data on the mean distances of colonial nests between years to evaluate the probability of yellow-legged hornet dispersal around the areas where the species is present. The distribution of nests in Italy was mainly explained by elevation (95% of nests located within 521 m a.s.l.) and distance from source sites (previous years’ colonies; 95% within 1.4–6.2 km). The diffusion models developed with these two variables forecast, with good accuracy, the spread of the species in the short term: 98–100% of nests were found within the predicted area of expansion. A similar approach can be applied in areas invaded by the yellow-legged hornet, in particular beyond new outbreaks and over the border of its expanding range, to implement strategies for its containment. The spatial application of the models allows the establishment of buffer areas where monitoring and control efforts can be allocated on the basis of the likelihood of the species spreading at progressively greater distances

    Extracellular vesicles from human plasma for biomarkers discovery: Impact of anticoagulants and isolation techniques

    Extracellular vesicles (EVs) isolated from plasma are increasingly recognized as promising circulating biomarkers for disease discovery and progression, as well as for therapeutic drug delivery. The scientific community underlined the necessity of standard operative procedures for the isolation and storage of the EVs to ensure robust results. The understanding of the impact of the pre-analytical variables is still limited and some considerations about plasma anticoagulants and isolation methods are necessary. Therefore, we performed a comparison study between EVs isolated by ultracentrifugation and by affinity substrate separation from plasma EDTA and sodium citrate. The EVs were characterized by Nano Tracking Analysis, Western Blot, cytofluorimetric analysis of surface markers, and lipidomic analysis. While anticoagulants did not significantly alter any of the analyzed parameters, the isolation methods influenced EVs size, purity, surface markers expression and lipidomic profile. Compared to ultracentrifugation, affinity substrate separation yielded bigger particles highly enriched in tetraspanins (CD9, CD63, CD81), fatty acids and glycerolipids, with a predominant LDL- and vLDL-like contamination.     Real-world evidence of health outcomes and medication use 24 months after bariatric surgery in the public healthcare system in Brazil: a retrospective, single-center study

    OBJECTIVES: The number of bariatric procedures has significantly increased in Brazil, especially in the public Unified Health System. The present study describes health outcomes and medication use in obese patients treated in a major hospital that performs publicly funded surgery in Brazil. METHODS: A retrospective, single center study was conducted to collect real-world evidence of health outcomes and medication use in 247 obese patients (female, 82.2%) who underwent open Roux-en-Y gastric bypass. Changes in weight and body mass index (BMI), presence of apnea, hypertension, and type 2 diabetes (T2D), and medication use (hypertension, diabetes, and dyslipidemia) were assessed preoperatively and up to 24 months postoperatively. The mean cost of medications was calculated for the 12-month preoperative and 24-month postoperative periods. RESULTS: During the surgery, the mean age of patients was 43.42 years (standard deviation [SD], 10.9 years), and mean BMI was 46.7 kg/m2 (SD, 6.7 kg/m2 ). At 24 months, significant declines were noted in weight (mean, -37.6 kg), BMI (mean, -14.3 kg/m2 ); presence of T2D, hypertension, and apnea (-29.6%, -50.6%, and -20.9%, respectively); and number of patients using medications (-66.67% for diabetes, -41.86% for hypertension, and -55.26% for dyslipidemia). The mean cost of medications (total costs for all medications) decreased by 450% in 12–24 postoperative months compared to that in 12 preoperative months. CONCLUSION: Roux-en-Y gastric bypass successfully reduced weight, BMI, and comorbidities and medication use and cost at 24 months in Brazilian patients treated in the public Unified Health System

    AlpArray-Italy: Site description and noise characterization

    Within the framework of the European collaborative research initiative AlpArray (http://www.alparray.ethz. ch), the Istituto Nazionale di Geofisica e Vulcanolgia (INGV) deployed overall 20 broad-band seismic stations in Northern Italy and on two islands in the Tyrrhenian Sea (Capraia and Montecristo) during Fall-Winter 2015. The temporary deployment (16 stations) will run for two to three years and 4 INGV National Seismic Network accelerometric sites are now equipped with additional per- manent broad-band sensors. The 16 temporary stations are equipped with REF TEK 130 digitizers and Nanometrics Trillium Compact 120 s sensors, a couple have Nanometrics Trillium 120P sensors and one a Streckeisen STS2.     Targeting lysine-specific demethylase 1 (KDM1A/LSD1) impairs colorectal cancer tumorigenesis by affecting cancer cells stemness, motility, and differentiation

    : Among all cancers, colorectal cancer (CRC) is the 3rd most common and the 2nd leading cause of death worldwide. New therapeutic strategies are required to target cancer stem cells (CSCs), a subset of tumor cells highly resistant to present-day therapy and responsible for tumor relapse. CSCs display dynamic genetic and epigenetic alterations that allow quick adaptations to perturbations. Lysine-specific histone demethylase 1A (KDM1A also known as LSD1), a FAD-dependent H3K4me1/2 and H3K9me1/2 demethylase, was found to be upregulated in several tumors and associated with a poor prognosis due to its ability to maintain CSCs staminal features. Here, we explored the potential role of KDM1A targeting in CRC by characterizing the effect of KDM1A silencing in differentiated and CRC stem cells (CRC-SCs). In CRC samples, KDM1A overexpression was associated with a worse prognosis, confirming its role as an independent negative prognostic factor of CRC. Consistently, biological assays such as methylcellulose colony formation, invasion, and migration assays demonstrated a significantly decreased self-renewal potential, as well as migration and invasion potential upon KDM1A silencing. Our untargeted multi-omics approach (transcriptomic and proteomic) revealed the association of KDM1A silencing with CRC-SCs cytoskeletal and metabolism remodeling towards a differentiated phenotype, supporting the role of KDM1A in CRC cells stemness maintenance. Also, KDM1A silencing resulted in up-regulation of miR-506-3p, previously reported to play a tumor-suppressive role in CRC. Lastly, loss of KDM1A markedly reduced 53BP1 DNA repair foci, implying the involvement of KDM1A in the DNA damage response. Overall, our results indicate that KDM1A impacts CRC progression in several non-overlapping ways, and therefore it represents a promising epigenetic target to prevent tumor relapse
