Efeitos de diferentes intervalos de recuperação no desempenho muscular isocinético em idosos
OBJETIVO: Comparar os efeitos de diferentes intervalos de recuperação (IR) entre séries de contração isocinética no desempenho muscular em idosos. MÉTODOS: Vinte idosos voluntários (66,9±3,9 anos, 76,1±13,4kg, 169±5,2cm) foram submetidos a três sessões de exercÃcio resistido isocinético unilateral da musculatura extensora do joelho com diferentes IR (1 minuto, 2 minutos, e 3 minutos) na velocidade de 60° s-1. Cada sessão consistia em três séries de 10 repetições durante as quais foram avaliados o pico de torque (PT), trabalho total (TT) e Ãndice de fadiga (IF). ANOVA fatorial para medidas repetidas, com a análise de Post hoc por Bonferroni, foi utilizada para identificar possÃveis diferenças entre os IR. O nÃvel de significância estatÃstico utilizado foi de p0,05). Apesar de ter sido observado menor desempenho muscular na 3ª série em todos os IR, maiores quedas no PT, TT e IF foram observadas quando adotado o IR de 1minuto (p0.05). Although muscle performance was lower during the third sets with all RI, the greatest decreases in PT, TW and FI occurred with the 1 minute RI (p<0.05). CONCLUSIONS: The results showed that the RI variable has an important influence on isokinetic muscle performance in older adults, particularly from the third sets onwards, which suggests that RI should be increased as successive sets are performed within the same exercise session
Cleaning efficiency in packing lines for fresh market tomatoes
On the last years, in Brazil, sorting and classifying fruits and vegetables using packing lines have increased. This work aimed at characterizing the cleaning process for fresh market tomatoes at two packing lines, one imported and one national located at Campinas, São Paulo State. Characterization included data, number, types and brushes velocity, water use, fruit standing time and cleaning efficiency. Standing time was measured correlating to fruit diameter (CEAGESP). For measuring cleaning efficiency an equipment was developed that was mainly composed of a ring involved with white cloth. Samples were taken before and after the cleaning step and evaluated using a colorimeter HUNTER Lab. The results showed a strong difference between the two equipments. The imported equipment showed lower number on brushes and rotation than national one, however a higher water consumption. For imported equipments this relation was not found. Both packing lines showed the same cleaning efficiency. Cleaning efficiency is related to be an interaction among the studies parameters, and it could be necessary a better management than the one used on both equipments.No Brasil, nos últimos anos, tem aumentado o beneficiamento e a classificação de frutas e hortaliças em equipamentos. O objetivo deste trabalho foi caracterizar a etapa de limpeza de dois equipamentos de beneficiamento de tomate de mesa, um nacional e outro importado, instalados na região metropolitana de Campinas - SP. A caracterização foi realizada quanto ao número, tipo e velocidade de rotação das escovas, aplicação de água, tempo de permanência dos frutos e eficiência de limpeza. O tempo de permanência foi mensurado com relação ao tamanho dos frutos, segundo os padrões da CEAGESP. Para mensurar a eficiência, foi desenvolvido um equipamento composto de um anel deslizante envolvido por um pano branco. As amostragens foram realizadas antes e após a etapa de limpeza e avaliadas utilizando colorÃmetro HUNTER Lab. Por meio dos resultados, observaram-se diferenças entre os dois equipamentos, em que o importado apresentou menor número de escovas, rotações mais baixas e maior consumo de água que o nacional. No equipamento nacional, o tempo de permanência dependeu do tamanho dos frutos, não sendo encontrada essa correlação no equipamento importado, e ambos tiveram a mesma eficiência de limpeza. Rarity of monodominance in hyperdiverse Amazonian forests.
Tropical forests are known for their high diversity. Yet, forest patches do occur in the tropics where a single tree species is dominant. Such "monodominant" forests are known from all of the main tropical regions. For Amazonia, we sampled the occurrence of monodominance in a massive, basin-wide database of forest-inventory plots from the Amazon Tree Diversity Network (ATDN). Utilizing a simple defining metric of at least half of the trees ≥ 10 cm diameter belonging to one species, we found only a few occurrences of monodominance in Amazonia, and the phenomenon was not significantly linked to previously hypothesized life history traits such wood density, seed mass, ectomycorrhizal associations, or Rhizobium nodulation. In our analysis, coppicing (the formation of sprouts at the base of the tree or on roots) was the only trait significantly linked to monodominance. While at specific locales coppicing or ectomycorrhizal associations may confer a considerable advantage to a tree species and lead to its monodominance, very few species have these traits. Mining of the ATDN dataset suggests that monodominance is quite rare in Amazonia, and may be linked primarily to edaphic factors
Intragenic antimicrobial peptides (IAPs) from human proteins with potent antimicrobial and anti-inflammatory activity
Following the treads of our previous works on the unveiling of bioactive peptides encrypted
in plant proteins from diverse species, the present manuscript reports the occurrence of four
proof-of-concept intragenic antimicrobial peptides in human proteins, named Hs IAPs.
These IAPs were prospected using the software Kamal, synthesized by solid phase chemistry,
and had their interactions with model phospholipid vesicles investigated by differential
scanning calorimetry and circular dichroism. Their antimicrobial activity against bacteria,
yeasts and filamentous fungi was determined, along with their cytotoxicity towards erythrocytes.
Our data demonstrates that Hs IAPs are capable to bind model membranes while
attaining α-helical structure, and to inhibit the growth of microorganisms at concentrations
as low as 1μM. Hs02, a novel sixteen residue long internal peptide (KWAVRIIRKFIKGFISNH2)
derived from the unconventional myosin 1h protein, was further investigated in its
capacity to inhibit lipopolysaccharide-induced release of TNF-α in murine macrophages.
Hs02 presented potent anti-inflammatory activity, inhibiting the release of TNF-α in LPSprimed
cells at the lowest assayed concentration, 0.1 μM. A three-dimensional solution
structure of Hs02 bound to DPC micelles was determined by Nuclear Magnetic Resonance.
Our work exemplifies how the human genome can be mined for molecules with biotechnological
potential in human health and demonstrates that IAPs are actual alternatives to antimicrobial
peptides as pharmaceutical agents or in their many other putative applications

MIF Participates in Toxoplasma gondii-Induced Pathology Following Oral Infection
BACKGROUND: Macrophage migration inhibitory factor (MIF) is essential for controlling parasite burden and survival in a model of systemic Toxoplasma gondii infection. Peroral T. gondii infection induces small intestine necrosis and death in susceptible hosts, and in many aspects resembles inflammatory bowel disease (IBD). Considering the critical role of MIF in the pathogenesis of IBD, we hypothesized that MIF participates in the inflammatory response induced by oral infection with T. gondii. METHODOLOGY/PRINCIPAL FINDINGS: Mif deficient (Mif(-/-)) and wild-type mice in the C57Bl/6 background were orally infected with T. gondii strain ME49. Mif(-/-) mice had reduced lethality, ileal inflammation and tissue damage despite of an increased intestinal parasite load compared to wt mice. Lack of MIF caused a reduction of TNF-α, IL-12, IFN-γ and IL-23 and an increased expression of IL-22 in ileal mucosa. Moreover, suppressed pro-inflammatory responses at the ileal mucosa observed in Mif(-/-) mice was not due to upregulation of IL-4, IL-10 or TGF-β. MIF also affected the expression of matrix metalloproteinase-9 (MMP-9) but not MMP-2 in the intestine of infected mice. Signs of systemic inflammation including the increased concentrations of inflammatory cytokines in the plasma and liver damage were less pronounced in Mif(-/-) mice compared to wild-type mice. CONCLUSION/SIGNIFICANCE: In conclusion, our data suggested that in susceptible hosts MIF controls T. gondii infection with the cost of increasing local and systemic inflammation, tissue damage and death
Spatial Evaluation and Modeling of Dengue Seroprevalence and Vector Density in Rio de Janeiro, Brazil
Dengue is a major public health problem in many tropical regions of the world, including Brazil, where Aedes aegypti is the main vector. We present a household study that combines data on dengue fever seroprevalence, recent dengue infection, and vector density, in three neighborhoods of Rio de Janeiro, Brazil, during its most devastating dengue epidemic to date. This integrated entomological–serological survey showed evidence of silent transmission even during a severe epidemic. Also, past exposure to dengue virus was highly associated with age and living in areas of high movement of individuals and social/commercial activity. No association was observed between household infestation index and risk of dengue infection in these areas. Our findings are discussed in the light of current theories regarding transmission thresholds and relative role of mosquitoes and humans as vectors of dengue viruses
