    Insecticide resistance in Aedes aegypti populations from Ceará, Brazil

    <p>Abstract</p> <p>Background</p> <p>Organophosphates and pyrethroids are used widely in Brazil to control <it>Aedes aegypti</it>, the main vector of dengue viruses, under the auspices of the National Programme for Dengue Control. Resistance to these insecticides is widespread throughout Brazil. In Ceará the vector is present in 98% of districts and resistance to temephos has been reported previously. Here we measure resistance to temephos and the pyrethroid cypermethrin in three populations from Ceará and use biochemical and molecular assays to characterise resistance mechanisms.</p> <p>Results</p> <p>Resistance to temephos varied widely across the three studied populations, with resistance ratios (RR<sub>95</sub>) of 7.2, 30 and 192.7 in Juazeiro do Norte, Barbalha and Crato respectively. The high levels of resistance detected in Barbalha and Crato (RR<sub>95 </sub>≥ 30) imply a reduction of temephos efficacy, and indeed in simulated field tests reduced effectiveness was observed for the Barbalha population. Two populations (Crato and Barbalha) were also resistant to cypermethrin, whilst Juazeiro do Norte showed only an altered susceptibility. The <it>Ile1011Met kdr </it>mutation was detected in all three populations and <it>Val1016Ile </it>in Crato and Juazeiro do Norte. <it>1011Met </it>was significantly associated with resistance to cypermethrin in the Crato population. Biochemical tests showed that only the activity of esterases and GSTs, among the tested detoxification enzymes, was altered in these populations when compared with the Rockefeller strain.</p> <p>Conclusions</p> <p>Our results demonstrate that two <it>A. aegypti </it>populations from Ceará are under strong selection pressure by temephos, compromising the field effectiveness of this organophosphate. Our results also provide evidence that the process of reducing resistance to this larvicide in the field is difficult and slow and may require more than seven years for reversal. In addition, we show resistance to cypermethrin in two of the three populations studied, and for the first time the presence of the allele <it>1016Ile </it>in mosquito populations from northeastern Brazil. A significant association between <it>1011M</it>et and resistance was observed in one of the populations. Target-site mechanisms seem not to be implicated in temephos resistance, reinforcing the idea that for the studied populations, detoxification enzymes most likely play a major role in the resistance to this insecticide.</p

    The Pemberton Happiness Index: Validation of the Universal Portuguese version in a large Brazilian sample

    The Pemberton Happiness Index (PHI) is a recently developed integrative measure of well-being that includes components of hedonic, eudaimonic, social, and experienced well-being. The PHI has been validated in several languages, but not in Portuguese. Our aim was to cross-culturally adapt the Universal Portuguese version of the PHI and to assess its psychometric properties in a sample of the Brazilian population using online surveys.An expert committee evaluated 2 versions of the PHI previously translated into Portuguese by the original authors using a standardized form for assessment of semantic/idiomatic, cultural, and conceptual equivalence. A pretesting was conducted employing cognitive debriefing methods. In sequence, the expert committee evaluated all the documents and reached a final Universal Portuguese PHI version. For the evaluation of the psychometric properties, the data were collected using online surveys in a cross-sectional study. The study population included healthcare professionals and users of the social network site Facebook from several Brazilian geographic areas. In addition to the PHI, participants completed the Satisfaction with Life Scale (SWLS), Diener and Emmons' Positive and Negative Experience Scale (PNES), Psychological Well-being Scale (PWS), and the Subjective Happiness Scale (SHS). Internal consistency, convergent validity, known-group validity, and test-retest reliability were evaluated. Satisfaction with the previous day was correlated with the 10 items assessing experienced well-being using the Cramer V test. Additionally, a cut-off value of PHI to identify a happy individual was defined using receiver-operating characteristic (ROC) curve methodology.Data from 1035 Brazilian participants were analyzed (health professionals = 180Facebook users = 855). Regarding reliability results, the internal consistency (Cronbach alpha = 0.890 and 0.914) and test-retest (intraclass correlation coefficient = 0.814) were both considered adequate. Most of the validity hypotheses formulated a priori (convergent and know-group) was further confirmed.     Construção e validação de videoaula sobre ressuscitação cardiopulmonar

    Objetivo: Construir e validar um objeto contemporâneo virtual de ensino, videoaula, sobreressuscitação cardiopulmonar no adulto em suporte básico de vida com o uso do desfibriladorexterno automático no ambiente hospitalar.Métodos: Trata-se de uma pesquisa aplicada, de produção tecnológica, com produção devídeoaula de acordo com a trajetória metodológica proposta por Fleming, Reynolds eWallace. Desenvolvido na Universidade do Estado de Minas Gerais e na Escola deEnfermagem de Ribeirão Preto no período de janeiro de 2017 a março de 2018. Participaram16 enfermeiros expertises na área de urgência e emergência. Para concordância interavaliadoresfoi utilizado a estatística AC1 de Gwet.Resultados: A validação do roteiro/script e storyboard foi alcançada a concordância interavaliadores,de acordo com Landis e Kock, classificada em “concordância moderada”, comAC1=0,59 e p<0,0001.Conclusões: A videoaula construída e validada neste estudo, representa importante estratégiacontemporânea adequada para aplicação no processo de ensino-aprendizagem.Palavras-chave: Ensino.     Amino Acid Biosignature in Plasma among Ischemic Stroke Subtypes

    Ischemic stroke is a neurovascular disorder caused by reduced or blockage of blood flow to the brain, which may permanently affect motor and cognitive abilities. The diagnostic of stroke is performed using imaging technologies, clinical evaluation, and neuropsychological protocols, but no blood test is available yet. In this work, we analyzed amino acid concentrations in blood plasma from poststroke patients in order to identify differences that could characterize the stroke etiology. Plasma concentrations of sixteen amino acids from patients with chronic ischemic stroke (n = 73) and the control group (n = 16) were determined using gas chromatography coupled to mass spectrometry (GC-MS). The concentration data was processed by Partial Least Squares-Discriminant Analysis (PLS-DA) to classify patients with stroke and control. The amino acid analysis generated a first model able to discriminate ischemic stroke patients from control group. Proline was the most important amino acid for classification of the stroke samples in PLS-DA, followed by lysine, phenylalanine, leucine, and glycine, and while higher levels of methionine and alanine were mostly related to the control samples. The second model was able to discriminate the stroke subtypes like atherothrombotic etiology from cardioembolic and lacunar etiologies, with lysine, leucine, and cysteine plasmatic concentrations being the most important metabolites. Our results suggest an amino acid biosignature for patients with chronic stroke in plasma samples, which can be helpful in diagnosis, prognosis, and therapeutics of these patients

    Função sistólica do ventrículo esquerdo pela ecocardiografia em crianças e adolescentes com osteossarcoma tratados com doxorrubicina com e sem dexrazoxane

    OBJETIVO: Avaliar o comportamento da função sistólica do ventrículo esquerdo (VE) pela ecocardiografia em pacientes com osteossarcoma tratados com doxorrubicina com e sem dexrazoxane. MÉTODOS: Foram estudados 55 pacientes com osteossarcoma com ou sem metástase submetidos à quimioterapia (QT) com seis ciclos de doxorrubicina, divididos em dois grupos, conforme o uso de dexrazoxane. Grupo I: 37 pacientes, os quais receberam dexrazoxane (28 do sexo masculino, com média de idade de 15,4 anos). Grupo II: 18 pacientes, que não receberam dexrazoxane (15 do sexo masculino, com média de idade de 15,1 anos). Foram realizadas quatro avaliações ecocardiográficas: 1) antes do início da QT (avaliação inicial); 2) até duas semanas após o terceiro ciclo; 3) até duas semanas após o quinto ciclo e 4) até quatro semanas após o sexto ciclo da QT (avaliação final). A função sistólica do VE foi avaliada pela porcentagem de encurtamento (PE) com o ecocardiograma. Alteração da função contrátil ou toxicidade miocárdica foi definida com valores de PE iguais ou inferiores a 29% e/ou diminuição da PE, em valor absoluto, igual ou superior a 10 unidades do valor inicial de cada paciente. RESULTADOS: Não houve diferença significativa entre os grupos quanto à idade, sexo e raça. A dose cumulativa de doxorrubicina foi significantemente maior no grupo II em todas as fases do tratamento: 174 x 203 mg/m²; 292 x 338 mg/m² e 345 x 405 mg/² (p < 0,0001). A ocorrência de disfunção sistólica do VE, de acordo com os critérios previamente definidos, foi de sete indivíduos no grupo I (18,92%) e de dois no grupo II (11,1%), diferença não significativa (p=0,248). A análise de variância com medidas repetidas não mostrou diferença significativa nas médias da PE ao longo do período de estudo (p=0,967). Entretanto foi encontrada diferença significativa (p=0,029) entre as médias da PE dos grupos I e II nas avaliações 2 (35,67 x 37,21%), 3 (34,95 x 38,47%) e 4 (35,26 x 38,22%). CONCLUSÃO: Os dados do presente estudo indicam que, nos pacientes com osteossarcoma ltratados com doxorrubicina com e sem dexrazoxane, a função sistólica do VE, avaliada pela média da porcentagem de encurtamento, apresentou melhor desempenho no grupo que recebeu a administração de dexrazoxane. A ocorrência de disfunção sistólica, porém, foi semelhante entre os dois grupos de pacientes

    A binuclear silver(I) complex with the amino acid L-buthionine sulfoximine (BSO) of composition Ag 2 C 8 H 16 N 2 O 3 S was synthesized and characterized by chemical and spectroscopic measurements, and DFT (density functional theory) studies. Solid-state 13 C nuclear magnetic resonance (SSNMR) and infrared vibrational spectroscopy (IR) analyses indicate the coordination of the nitrogen and carboxylate groups of the amino acid moiety to one of the silver atoms, while coordination to the second silver atom occurs through the nitrogen of the sulfoximine group.
