    Asymptotic results for the Fourier estimator of the integrated quarticity

    In this paper we prove a central limit theorem for the Fourier quarticity estimator. We obtain a new consistency result and we show that the estimator reaches the parametric rate 1/2. The optimal variance is obtained, with a suitable choice of the number of frequencies employed to compute the Fourier coefficients of the volatility, while the limiting distribution has a bias. As a by-product, thanks to the Fourier methodology, we obtain consistent estimators of any even power of the volatility function and an estimator of the spot quarticity. We assess the finite sample performance of the Fourier quarticity estimator in a numerically exercise with different market micro-structure frictions

    Relationship between retinal microvascular impairment and subclinical atherosclerosis in SLE

    objectives: patients with SLE have higher cardiovascular (CV) risk compared with healthy controls (HC) and are characterised by accelerated atherosclerosis; intima media thickness (IMT), marker of subclinical atherosclerosis, is higher in patients with SLE than in HCs. Retinal microvascular impairment detected through optical coherence tomography angiography (OCTA) was investigated as a marker of systemic vascular involvement in SLE.the aim of the study was to evaluate the relationship between retinal vascular impairment and IMT in SLE. methods: cross-sectional study recruiting patients with SLE and HCs. Data of the study population were collected. CV risk was evaluated through the american college of cardiology/american heart association (ACC/AHA) guidelines, framingham and QRESEARCH risk estimator V.3 (QRISK3) scores. Both groups underwent OCTA and carotid ultrasound with IMT assessment.Statistical analysis was accomplished using Pearson/Spearman, t-test/Mann-Whitney or χ2 test. Variables statistically significant at univariate regression analysis were tested in an age-corrected and sex-corrected multivariate regression model. results: 43 patients with SLE and 34 HCs were recruited.patients with SLE showed higher triglycerides (p=0.019), triglycerides-glucose (TyG) Index (p=0.035), ACC/AHA guidelines (p=0.001), Framingham Risk Scores (p=0.008) and a reduced superficial (p<0.001) and deep (p=0.005) whole retinal vessel density (VD) compared with HCs.In SLE univariate analysis, deep whole VD showed a negative correlation with IMT (p=0.027), age (p=0.001), systolic blood pressure (p=0.011), QRISK3 Score (p<0.001), systemic lupus international collaborating clinics damage index (p=0.006) and apolipoprotein B (p=0.021), while a positive correlation was found with female sex (p=0.029). Age-adjusted and sex-adjusted multivariate analysis confirmed QRISK3 score (p=0.049) and IMT (p=0.039) to be independent risk factors for reduced retinal VD. conclusions: patients with SLE showed lower retinal VD and higher CV risk indicators compared with HCs. Among patients with SLE, QRISK3 Score and IMT were found to be independent risk factors for retinal vascular impairment, suggesting a role of OCTA in evaluating preclinical CV involvement in SLE. moreover, TyG index could represent a biomarker of CV risk in patients with SLE compared with HCs

    The NANOG transcription factor induces type 2 deiodinase expression and regulates the intracellular activation of thyroid hormone in keratinocyte carcinomas

    Type 2 deiodinase (D2), the principal activator of thyroid hormone (TH) signaling in target tissues, is expressed in cutaneous squamous cell carcinomas (SCCs) during late tumorigenesis, and its repression attenuates the invasiveness and metastatic spread of SCC. Although D2 plays multiple roles in cancer progression, nothing is known about the mechanisms regulating D2 in cancer. To address this issue, we investigated putative upstream regulators of D2 in keratinocyte carcinomas. We found that the expression of D2 in SCC cells is positively regulated by the NANOG transcription factor, whose expression, besides being causally linked to embryonic stemness, is associated with many human cancers. We also found that NANOG binds to the D2 promoter and enhances D2 transcription. Notably, blockage of D2 activity reduced NANOG-induced cell migration as well as the expression of key genes involved in epithelial–mesenchymal transition in SCC cells. In conclusion, our study reveals a link among endogenous endocrine regulators of cancer, thyroid hormone and its activating enzyme, and the NANOG regulator of cancer biology. These findings could provide the basis for the development of TH inhibitors as context-dependent anti-tumor agents

    Italian intersociety consensus on management of long covid in children

    Background: Two sequelae of pediatric COVID-19 have been identified, the multisystem inflammatory syndrome in children (MIS-C) and the long COVID. Long COVID is much less precisely defined and includes all the persistent or new clinical manifestations evidenced in subjects previously infected by SARS-CoV-2 beyond the period of the acute infection and that cannot be explained by an alternative diagnosis. In this Intersociety Consensus, present knowledge on pediatric long COVID as well as how to identify and manage children with long COVID are discussed. Main findings: Although the true prevalence of long COVID in pediatrics is not exactly determined, it seems appropriate to recommend evaluating the presence of symptoms suggestive of long COVID near the end of the acute phase of the disease, between 4 and 12 weeks from this. Long COVID in children and adolescents should be suspected in presence of persistent headache and fatigue, sleep disturbance, difficulty in concentrating, abdominal pain, myalgia or arthralgia. Persistent chest pain, stomach pain, diarrhea, heart palpitations, and skin lesions should be considered as possible symptoms of long COVID. It is recommended that the primary care pediatrician visits all subjects with a suspected or a proven diagnosis of SARS-CoV-2 infection after 4 weeks to check for the presence of symptoms of previously unknown disease. In any case, a further check-up by the primary care pediatrician should be scheduled 3 months after the diagnosis of SARS-CoV-2 infection to confirm normality or to address emerging problems. The subjects who present symptoms of any organic problem must undergo a thorough evaluation of the same, with a possible request for clinical, laboratory and / or radiological in-depth analysis in case of need. Children and adolescents with clear symptoms of mental stress will need to be followed up by existing local services for problems of this type. Conclusions: Pediatric long COVID is a relevant problem that involve a considerable proportion of children and adolescents. Prognosis of these cases is generally good as in most of them symptoms disappear spontaneously. The few children with significant medical problems should be early identified after the acute phase of the infection and adequately managed to assure complete resolution. A relevant psychological support for all the children during COVID-19 pandemic must be organized by health authorities and government that have to treat this as a public health issue

    Among the most straining trends that farmers have to face there are: on one side, to guarantee welfare and adequate life conditions for animals and to reduce the environmental footprint, on the other side, to develop new strategies to improve farm management reducing costs. The current conditions and the expected developments of the dairy sector highlight a strong need for more efficient and sustainable farming systems. Studying heat stress, herd management and housing and animals\u2019 productive and reproductive performances is fundamental for the economic and environmental sustainability of the dairy chain. New and effective tools to cope with these challenges have been provided by Precision Livestock Farming (PLF), which is nowadays increasingly applied and makes possible to control quali-quantitative parameters related to production, health, behaviour, and real-time locomotion per animal. The research key challenge is to turn these data into knowledge to provide real-time support in farming optimisation. This research focuses specifically on different systems to collect, process and derive useful information from data on animal welfare and productivity.     SBDS-Deficient Cells Have an Altered Homeostatic Equilibrium due to Translational Inefficiency Which Explains their Reduced Fitness and Provides a Logical Framework for Intervention

    Ribosomopathies are a family of inherited disorders caused by mutations in genes necessary for ribosomal function. Shwachman-Diamond Bodian Syndrome (SDS) is an autosomal recessive disease caused, in most patients, by mutations of the SBDS gene. SBDS is a protein required for the maturation of 60S ribosomes. SDS patients present exocrine pancreatic insufficiency, neutropenia, chronic infections, and skeletal abnormalities. Later in life, patients are prone to myelodisplastic syndrome and acute myeloid leukemia (AML). It is unknown why patients develop AML and which cellular alterations are directly due to the loss of the SBDS protein. Here we derived mouse embryonic fibroblast lines from an SbdsR126T/R126T mouse model. After their immortalization, we reconstituted them by adding wild type Sbds. We then performed a comprehensive analysis of cellular functions including colony formation, translational and transcriptional RNA-seq, stress and drug sensitivity. We show that: 1. Mutant Sbds causes a reduction in cellular clonogenic capability and oncogene-induced transformation. 2. Mutant Sbds causes a marked increase in immature 60S subunits, limited impact on mRNA specific initiation of translation, but reduced global protein synthesis capability. 3. Chronic loss of SBDS activity leads to a rewiring of gene expression with reduced ribosomal capability, but increased lysosomal and catabolic activity. 4. Consistently with the gene signature, we found that SBDS loss causes a reduction in ATP and lactate levels, and increased susceptibility to DNA damage. Combining our data, we conclude that a cell-specific fragile phenotype occurs when SBDS protein drops below a threshold level, and propose a new interpretation of the disease

    Oxygen carrier materials based on La2O2SO4 and promoted by small amounts (1% wt.) of transition metals, namely Co, Mn and Cu, have been synthesized and characterized by means of X-ray diffraction (XRD), Brunauer&#8315;Emmett&#8315;Teller (BET), Temperature-programmed reduction/oxidation (TPR/TPO) and thermogravimetry-mass-Fourier transform infrared spectrometry (TG-MS-FTIR) experiments under alternating feeds in order to investigate their potential use for the Chemical Looping Combustion process using either hydrogen or methane as the fuel. The chemical looping reactivity is based on the reversible redox cycle of sulfur from S6+ in La2O2SO4 to S2&#8722; in La2O2S and entails a large oxygen storage capacity, but it generally requires high temperatures to proceed, challenging material stability and durability. Herein we demonstrate a remarkable improvement of lattice oxygen availability and activity during the reduction step obtained by cost-effective metal doping in the order Co &gt; Mn &gt; Cu. Notably, the addition of Co or Mn has shown a significant beneficial effect to prevent the decomposition of the oxysulfate releasing SO2, which is identified as the main cause of progressive deactivation for the unpromoted La2O2SO4
