    The Letter of James: Literary Structure, Form and Unity

    The aim of this thesis is to investigate the literary structure, form and unity of the letter of James, an investigation which only really began about the middle of the 19th century. This period of time marked a beginning where scholars began to suggest that James was more than a chaotic jumble of sayings and exhortations, a characterisation often describing James in earlier studies. This line of inquiry continued on throughout the 20th and 21st centuries. There still, however, remains a great diversity of opinion and disagreement on the matter. .This thesis offers a sustained study of the literary structure and form of James over seven chapters. Chapter I traces the present state of the question in relation to the literary structure of the letter, and how diverse the range of opinions are. Chapter II focuses specifically on the introduction and conclusion of the letter. Chapter TTI outlines the various methodologies used to decipher the letter’s structure and form, with suggestions on the methods this thesis will take. Chapter IV endeavours to delineate the introduction to the letter while Chapter V focuses on the literary structure of the rest of the letter. Chapter VI will examine the literary form of James which is an important issue that arises from the study of the letter’s literary structure. Here, the epistolary status of James will be assessed. Finally, Chapter VII will then seek to establish the unity of the letter, with a focus upon the themes of perfection and double mindedness as unifying themes

    Aircraft wing structure detail design

    The provisions of this project call for the design of the structure of the wing and carry-through structure for the Viper primary trainer, which is to be certified as a utility category trainer under FAR part 23. The specific items to be designed in this statement of work were Front Spar, Rear Spar, Aileron Structure, Wing Skin, and Fuselage Carry-through Structure. In the design of these parts, provisions for the fuel system, electrical system, and control routing were required. Also, the total weight of the entire wing planform could not exceed 216 lbs. Since this aircraft is to be used as a primary trainer, and the SOW requires a useful life of 107 cycles, it was decided that all of the principle stresses in the structural members would be kept below 10 ksi. The only drawback to this approach is a weight penalty

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Population structures and the role of genetic exchange in the zoonotic pathogen Cryptosporidium parvum

    Apicomplexan protozoan parasites include some of the most globally important human and animal pathogens, all of which have obligatory sexual cycles in their definitive hosts. Despite their importance and the relevance of understanding the population genetic structure and role of genetic exchange in generating diversity, population genetic analysis has largely been restricted to Plasmodium spp. and Toxoplasma gondii. These species show a considerable diversity of population structure suggesting different strategies for transmission and survival in mammalian hosts. We have undertaken a population genetic analysis of a further apicomplexan species (Cryptosporidium parvum) to extend our understanding of the diversity of genetic structures and test whether it has a clonal population structure. Nothing is known about the population structure of this parasite. We have analyzed 180 parasite isolates from both humans and cattle derived from a single discrete geographical area, using three minisatellite and four microsatellite markers that define 38 multilocus genotypes. Analysis of linkage disequilibria between pairs of loci combined with measures of genetic distance and similarity provides evidence that the sample comprises four genetically isolated populations. One group of human isolates consists primarily of two closely related multilocus genotypes (clonal), while the major subtypes of a second group, common to both humans and animals, show a panmictic population structure. The data provide an important step in understanding the role of genetic exchange in these parasites, which is an essential prerequisite for determining the value of multilocus genotyping for the analysis of sources of human infection as well as future molecular epidemiological studies

    The Invisible Woman? A comparative study of women's sports coverage in the UK national press before and after the 2012 Olympic Games

    While the coverage of women's sport in UK media rises to comparable levels to men's sports during large sporting events like the Olympics, academics agree that “routine” women's sports coverage is under-represented. According to the Women's Sport and Fitness Foundation, “81% of people think that the female athletes at London 2012 make better role models for young girls than other celebrities.” This article examines the representation of women in sport and compares routine coverage of women's sports in the UK national press across a week in February 2012, six months before the London Olympics, with coverage in a week in February 2013, six months after the Olympics, to see if there has been an Olympic “legacy” that increased coverage. It also examines coverage at the same time of year a decade earlier, to see how far, if at all, women's sports coverage in newspapers has progressed. The results suggest that there has been minimal change in everyday coverage of women's sports after the Olympics, and that female athletes continue to be hugely under-represented in the UK pres
