    Osteocytes Influence on Bone Matrix Integrity Affects Biomechanical Competence at Bone-Implant Interface of Bioactive-Coated Titanium Implants in Rat Tibiae

    Osseointegration is a prerequisite for the long-term success of implants. Titanium implants are preferred for their biocompatibility and mechanical properties. Nonetheless, the need for early and immediate loading requires enhancing these properties by adding bioactive coatings. In this preclinical study, extracellular matrix properties and cellular balance at the implant/bone interface was examined. Polyelectrolyte multilayers of chitosan and gelatin or with chitosan and Hyaluronic acid fabricated on titanium alloy using a layer-by-layer self-assembly process were compared with native titanium alloy. The study aimed to histologically evaluate bone parameters that correlate to the biomechanical anchorage enhancement resulted from bioactive coatings of titanium implants in a rat animal model. Superior collagen fiber arrangements and an increased number of active osteocytes reflected a significant improvement of bone matrix quality at the bone interface of the chitosan/gelatin-coated titan implants over chitosan/hyaluronic acid-coated and native implants. Furthermore, the numbers and localization of osteoblasts and osteoclasts in the reparative and remodeling phases suggested a better cellular balance in the chitosan/Gel-coated group over the other two groups. Investigating the micro-mechanical properties of bone tissue at the interface can elucidate detailed discrepancies between different promising bioactive coatings of titanium alloys to maximize their benefit in future medical applications

    Core-clock genes regulate proliferation and invasion via a reciprocal interplay with MACC1 in colorectal cancer cells

    The circadian clock coordinates the timing of several cellular processes including transcription, the cell cycle, and metabolism. Disruptions in the clock machinery trigger the abnormal regulation of cancer hallmarks, impair cellular homeostasis, and stimulate tumourigenesis. Here we investigated the role of a disrupted clock by knocking out or knocking down the core-clock (CC) genes ARNTL, PER2 or NR1D1 in cancer progression (e.g., cell proliferation and invasion) using colorectal cancer (CRC) cell lines HCT116, SW480 and SW620, from different progression stages with distinct clock phenotypes, and identified mechanistic links from the clock to altered cancer-promoting cellular properties. We identified MACC1 (metastasis-associated in colon cancer 1), a known driver for metastasis and an EMT (epithelial-to-mesenchymal transition)-related gene, to be significantly differentially expressed in CC manipulated cells and analysed the effect of MACC1 manipulation (knockout or overexpression) in terms of circadian clock phenotype as well as cancer progression. Our data points to a bi-directional MACC1-circadian clock interplay in CRC, via CC genes. In particular, knocking out MACC1 reduced the period of oscillations, while its overexpression increased it. Interestingly, we found the MACC1 protein to be circadian expressed in HCT116 WT cells, which was disrupted after the knockout of CC genes, and identified a MACC1-NR1D1 protein-protein interaction. In addition, MACC1 manipulation and CC knockout altered cell invasion properties of HCT116 cells, pointing to a regulation of clock and cancer progression in CRC, possibly via the interaction of MACC1 with core-clock genes

    The Pristine survey - XV. A CFHT ESPaDOnS view on the Milky Way halo and disc populations

    We present a one-dimensional, local thermodynamic equilibrium homogeneous analysis of 132 stars observed at high resolution with ESPaDOnS. This represents the largest sample observed at high resolution (R ∼40 000) from the Pristine survey. This sample is based on the first version of the Pristine catalogue and covers the full range of metallicities from [Fe/H] ∼-3 to ∼+0.25, with nearly half of our sample (58 stars) composed of very metal-poor (VMP) stars ([Fe/H] ≤ -2). This wide range of metallicities provides the opportunity of a new detailed study of the Milky Way stellar population. Because it includes both dwarf and giant stars, it also enables the analysis of any potential bias induced by the Pristine selection process. Based on Gaia EDR3, the orbital analysis of this Pristine-ESPaDOnS sample shows that it is composed of 65 halo stars and 67 disc stars. After a general assessment of the sample chemical properties with the α-elements Mg and Ca, we focus on the abundance of carbon and the neutron capture elements Ba and Sr. While most of our VMP subsample is carbon normal, we also find that 14 stars out of the 38 stars with [Fe/H] ≤ -2 and measured carbon abundances turn out to be carbon-enhanced metal-poor (CEMP) stars. We show that these CEMP stars are nearly exclusively (i.e. 12 stars out of 14) in the regime of low luminosity, unevolved, dwarf stars, which we interpret as the consequence of bias of the Pristine filter against C-rich giants. Among the VMP stars, we identify two CEMP stars with no enhancement in neutron-capture process elements and another one enriched in s-process element. Finally, one VMP star is found with a very low [Sr/Fe] abundance ratio for its metallicity, as expected if it had been accreted from an ultra-faint dwarf galaxy

    Saudi SCD patients’ symptoms and quality of life relative to the number of ED visits

    Background Individuals living with sickle cell disease (SCD) have significantly increased emergency department (ED) use compared to the general population. In Saudi Arabia, health care is free for all individuals and therefore has no bearing on increased ED visits. However, little is known about the relationship between quality of life (QoL) and frequency of acute care utilization in this patient population. Methods A cross-sectional study was conducted on 366 patients with SCD who attended the outpatient department at King Fahad Hospital, Hofuf, Saudi Arabia. Data were collected through self-administered surveys, which included: demographics, SCD-related ED visits, clinical issues, and QoL levels. We assessed the ED use by asking for the number of SCD-related ED visits within a 6-month period. Results The self-report survey of ED visits was completed by 308 SCD patients. The median number of SCD-related ED visits within a 6-month time period (IQR) was four (2-7 visits). According to the unadjusted negative binomial model, the rate of SCD-related ED visits increased by (46, 39.3, 40, and 53.5 %) for patients with fever, skin redness with itching, swelling, and blood transfusion, respectively. Poor QoL tends to increase the rate of SCD-related ED visits. Well education and poor general health positively influenced the rate of SCD-related ED visits. Well education tends to increase the rate of SCD-related ED visits by 50.2 %. The rate of SCD-related ED visits decreased by 1.4 % for every point increase in general health. Conclusion Saudi patients with sickle cell disease reported a wide range of SCD-related ED visits. It was estimated that six of 10 SCD patients had at least three ED visits within a 6-month period. Well education and poor general health resulted in an increase in the rate of SCD-related ED visits

    The purpose of this two-part series is to review the composition, properties, products, and clinical aspects of mineral trioxide aggregate (MTA) materials.search engines to include relevant scientific citations from the peer-reviewed journals published in English. MTA is a refined form of the parent compound, Portland cement (PC). It demonstrates a strong biocompatible nature owing to the high pH and its ability to form hydroxyapatite. MTA materials provide a better seal than traditional endodontic materials as observed in dye leakage, fluid filtration, protein leakage, and bacterial penetration leakage studies, and it has been recognized as a bioactive material. Currently a variety of MTA commercial products are available, including Proroot® Gray MTA and White MTA both from DENTSPLY Tulsa Dental Specialties , and MTA Angelus  Although these materials are indicated for various dental uses. This first of this series highlights and discusses the composition, physical, and/or chemical properties of MTA. A subsequent article will offer an overview of the material aspect (commercial products) and clinical considerations for MTA materials

    Endodontic treatments tend to have very high successful rates in spite of the fact that they are quite an invasive, difficult procedure, especially in complex anatomies. As a consequence, retreatment has become a well-defined field of knowledge and expertise in endodontics, since most complications can be solved with proper surgical or nonsurgical retreatments. Failure to grasp the rationale behind cleaning and shaping concepts can increase the occurrence of needless complications, such as ledges. Extension of the access cavity to provide unobstructed access to the root canals, precurving and not forcing instruments, using NiTi files, using passive step-back and balanced force techniques, and instrumenting the canal to its full length will all help to prevent ledge formation. Initial negotiation and bypassing the ledge can be achieved using a small file with a distinct curve at the tip, whereas a slight rotation motion of the file combined with a picking motion can often help advance the instrument. Blockage, laceration, and foraminal damage are the most common results. Each alters the reliability of the procedure and must be prevented if one is to obtain the best possible prognosis for the patient

It is most commonly seen at the cervical necks of teeth, but can occur in any area, even inter-dentally from vigorous and incorrect use of dental floss. Clinically, at the beginning it is small horizontal groove near the cemento-enamel junction; However, then the walls form a wedge with polished, glassy surfaces and tactile sensitivity. The treatment approach for cervical abrasion must not be based only on restorative procedures since a variety of causative and aggravating factors are related to their formation. The current article discusses a treatment protocol and techniques for the restoration of non-carious Class V lesions and presents a clinical case in which restoration is achieved with the conventional Glass Ionomer Cement
