    Melanotrope Secretory Cycle is Regulated by Physiological Inputs via the Hypothalamus

    Previously, it has been shown that background color conditions regulate the overall activity of the frog intermediate lobe by varying the proportions of the two subtypes of melanotropes existing in the gland, the highly active or secretory melanotropes and hormone-storage melanotropes, depending on melanocyte-stimulating hormone ( -MSH) requirements. However, the factors and mechanisms underlying these background-induced changes are still unknown. In the present study, we investigated whether hypothalamic factors known to regulate melanotrope cell function can induce changes in vitro similar to those caused by background adaptation in vivo. We found that the inhibitors apomorphine (a dopamine receptor agonist) and NPY decreased the number of active melanotropes and increased simultaneously that of storage melanotropes. On the other hand, the stimulator TRH increased the number of active cells and concomitantly reduced that of storage cells. Inasmuch as none of these treatments modified the apoptotic and proliferation rates in melanotrope cells, it appears that these hypothalamic factors caused actual interconversions of cells from a subpopulation to its counterpart. When taken together, these findings suggest that the hypothalamus would control melanotrope activity not only through short-term regulation of hormone synthesis and release, but also through a long-term regulation of the secretory phenotype of these cells whereby the activity of the intermediate lobe would be adjusted to fulfill the hormonal requirements imposed by background conditions

    Bioprospecting and characterization of poly-bhydroxyalkanoate (PHAs) producing bacteria isolated from Colombian sugarcane producing areas

    Bioprospecting for poly-b-hydroxyalkanoate (PHA)-accumulating micro-organisms was carried out in sugarcane growing areas of Colombia. They were isolated in unbalanced culture medium (highcarbon/nitrogen ratio) with sucrose, fructose and glucose used as carbon source. PHAs producing bacteria were identified by staining with Sudan black and solubilising cellular components in sodiumhypochlorite. A arbitrary scale was established (ranging from 1 to 4) for selecting the best strains, acording to growth, staining with Sudan black and solubilising cellular material. 108 isolates rated higher than 2 were obtained by using this scale; 44 of these were selected for evaluating them in a balloon flask for their ability to grow in sucrose. Scheffe’s test grouped the isolates evaluated in the balloon flask for polymer production and productivity. The 6 best isolates were evaluated in a fermenter to determine their kinetic growth profiles, substrate consumption and polymer accumulation. Differential scanning calorimeter (DSC) was used on the recovered polymer for determining fusion temperature and the conclusion was reached that 2 strains accumulated poly--hydroxybutyrate (PHB) and another 4 accumulated hydroxy-butyrate copolymers and other monomer units. These 6 strains were molecularly characterised by partially sequencing the 16s rRNA ribosomal gene, localizing them in 4 clusters on the taxonomic tree

    Resistin Regulates Pituitary Lipid Metabolism and Inflammation In Vivo and In Vitro

    The adipokine resistin is an insulin-antagonizing factor that also plays a regulatory role in inflammation, immunity, food intake, and gonadal function and also regulates growth hormone (GH) secretion in rat adenopituitary cells cultures with the adipokine. Although adipose tissue is the primary source of resistin, it is also expressed in other tissues, including the pituitary. The aim of this study is to investigate the possible action of resistin on the lipid metabolism in the pituitary gland in vivo (rats in two different nutritional status, fed and fast, treated with resistin on acute and a chronic way) and in vitro (adenopituitary cell cultures treated with the adipokine). Here, by a combination of in vivo and in vitro experimental models, we demonstrated that central acute and chronic administration of resistin enhance mRNA levels of the lipid metabolic enzymes which participated on lipolysis and moreover inhibiting mRNA levels of the lipid metabolic enzymes involved in lipogenesis.     The ATLAS inner detector trigger performance in pp collisions at 13TeV during LHC Run 2

In 2016, 2017, and 2018 the ATLAS detector recorded35.6fb−1,46.9fb−1,and60.6fb−1 respectively of proton–proton collision data at a centre-of-mass energy of 13TeV. In order to deal with the very high interaction multiplicities per bunch crossing expected with the 13TeV collisions the inner detector trigger was redesigned during the long shutdown of the Large Hadron Collider from 2013 until 2015. An overview of these developments is provided and the performance of the tracking in the trigger for the muon, electron, tau and b-jet signatures is discussed. The high performance of the inner detector trigger with these extreme interaction multiplicities demonstrate show the inner detector tracking continues to lie at the heart of the trigger performance and is essential in enabling the ATLAS physics programm

    Caracterización de las condiciones de salud respiratoria de los trabajadores expuestos a polvo de carbón en minería subterránea en Boyacá, 2013.

    Introducción: la neumoconiosis de los mineros de carbón es una enfermedad pulmonar ocupacional asociada a factores individuales y condiciones laborales específcas. Se manifesta progresiva e irreversiblemente, afectando la salud de los trabajadores y la productividad laboral. Objetivo: caracterizar las condiciones de salud respiratoria e identifcar los factores de riesgo asociados al desarrollo de neumoconiosis en trabajadores que han laborado expuestos a polvo de carbón en minas de socavón en Boyacá, 2013. Materiales y métodos: Se realizó un estudio descriptivo de corte transversal en trabajadores de minería de socavón expuestos a polvo de carbón en el departamento de Boyacá. Se llevó a cabo un registro completo de historia clínica ocupacional, examen físico completo, toma de radiografía de tórax, prueba de tuberculina y espirometría. Resultados: participaron 170 trabajadores masculinos. 75% de la población tenía antigüedad en el sector minero mayor a 12,5 años. Roncus, sibilancias y estertores predominaron en trabajadores con más de 20 años de antigüedad. Expectoración y tos se encontraron más frecuentemente en quienes realizaban la extracción mecanizada. El 15,9% de las radiografías de tórax fueron compatibles con neumoconiosis, 17,1% de las pruebas de tuberculina se encontraron positivas. 5,3% de las espirometrías mostraron alteraciones de vía aérea periférica y 2,4% mostraron patrón obstructivo. Discusión y conclusión: Los cambios radiológicos sugestivos de neumoconiosis fueron más frecuentes en trabajadores con antigüedad menor de 20 años. Los hallazgos espirométricos anormales predominaron en el grupo con antigüedad mayor a 20 años. Results: 170 male workers participated. 75% of the population had worked in the mining sector for more than12.5 years old. Rhonchi, wheezing and rattling predominated in workers over 20 years old. Expectoration and cough were most frequently found in those who made mechanized extraction. 15.9% of chest X rays were compatible with pneumoconiosis, 17.1% of tuberculin tests were found positive. 5.3% of spirometries showed peripheral airway abnormalities and 2.4% showed obstructive pattern. Discussion and conclusion: Radiological changes suggestive of pneumoconiosis were more frequent in workers under 20 years old. Abnormal spirometric fndings predominated in the group with more than 20 years old.     The E6 Oncoprotein from HPV16 Enhances the Canonical Wnt/?-Catenin Pathway in Skin Epidermis In Vivo

    The contribution of the Wnt signaling pathway to human papilloma virus (HPV)-induced carcinogenesis is poorly understood. In high-grade dysplastic lesions that are caused by high-risk HPVs (HR-HPV), ?-catenin is often located in the cell nucleus, which suggests that Wnt pathway may be involved in the development of HPV-related carcinomas. Most of the oncogenic potential of HR-HPVs resides on the PDZ-binding domain of E6 protein. We hypothesized that the PDZ-binding domain of the HPV16-E6 oncoprotein induces the nuclear accumulation of ?-catenin due to its capacity to degrade PDZ-containing cellular targets. To test this hypothesis, we evaluated the staining pattern of ?-catenin in the skin epidermis of transgenic mice expressing the full-length E6 oncoprotein (K14E6 mice) and measured LacZ gene expression in K14E6 mice that were crossed with a strain expressing LacZ that was knocked into the Axin2 locus (Axin2+/LacZ mice). Here, we show that the E6 oncoprotein enhances the nuclear accumulation of ?-catenin, the accumulation of cellular ?-catenin–responsive genes, and the expression of LacZ. None of these effects were observed when a truncated E6 oncoprotein that lacks the PDZ-binding domain was expressed alone (K14E6?PDZ mice) or in combination with Axin2+/LacZ. Conversely, cotransfection with either E6 or E6?PDZ similarly enhanced canonical Wnt signaling in short-term in vitro assays that used a luciferase Wnt/?-catenin/TCF-dependent promoter. We propose that the activation of canonical Wnt signaling could be induced by the HPV16-E6 oncoprotein; however, the participation of the E6 PDZ-binding domain seems to be important in in vivo models only.
