    Localization of damage in beams using interferometric techniques

    Two interferometric techniques and their applications in structural damage identification are presented in this paper. Out-of-plane displacement fields of modal response are measured with a pulsed electronic speckle pattern interferometric system(ESPI). The modal rotation fields, defined as the spatial derivative of the displacement field, are measured with a pulsed speckle shearography system. The measurements using these two interferometric systems are compared with measurements from experimental modal analysis and results from finite element analysis. This comparison shows that these two interferometric techniques, which allow non-contact, full-field measurements, are well suited to the measurement of modal response. A damaged beam with free-free boundary conditions is analyzed. The damages studied are small cuts perpendicular to the beams longitudinal axis. The bending moments and shear forces, which are related to the second and third order spatial derivatives of the modal displacement field, in the undamaged and damaged states are computed using numerical differentiation techniques. The damage is localized by looking for maximum values and/or perturbations of damage indicators based on bending moments and shear forces along the beam. The pulsed speckle shearography system leads to a significant improvement in the computation of bending moments and shear forces and, therefore, better damage localizations than the ones obtained with the pulsed ESPI system.The authors greatly appreciate the financial support of FCOMP -01-0124-FEDER-010236 through Project Ref.     Damage localisation in beams using the ritz method and speckle shear interferometry

    A novel numerical-experimental technique is developed in order to minimise some of the difficulties exhibited by others damage localisation approaches. The present technique relies on the computation of undamaged rotation fields using the Ritz method and the Timoshenko beam theory, while the measurement of damaged rotation fields is performed by speckle shear interferometry. Two damage localisation indicators are also presented, which, instead of being based on the second derivative of displacement fields, are based on the first spatial derivative of rotation fields. These damage localisation indicators, the modified curvature difference (MCD) and the modified damage index (MDI), were applied successfully in the localisation of damage in two clamped-clamped aluminium beams.The authors greatly appreciate the financial support of FCT through Project FCT PTDC/EME-PME/102095/2008

    In vitro biomodels in stenotic arteries to perform blood analogues flow visualizations and measurements: a review

    Cardiovascular diseases are one of the leading causes of death globally and the most common pathological process is atherosclerosis. Over the years, these cardiovascular complications have been extensively studied by applying in vivo, in vitro and numerical methods (in silico). In vivo studies represent more accurately the physiological conditions and provide the most realistic data. Nevertheless, these approaches are expensive, and it is complex to control several physiological variables. Hence, the continuous effort to find reliable alternative methods has been growing. In the last decades, numerical simulations have been widely used to assess the blood flow behavior in stenotic arteries and, consequently, providing insights into the cardiovascular disease condition, its progression and therapeutic optimization. However, it is necessary to ensure its accuracy and reliability by comparing the numerical simulations with clinical and experimental data. For this reason, with the progress of the in vitro flow measurement techniques and rapid prototyping, experimental investigation of hemodynamics has gained widespread attention.     Diagnosis methods for COVID-19: a systematic review

    At the end of 2019, the coronavirus appeared and spread extremely rapidly, causing millions of infections and deaths worldwide, and becoming a global pandemic. For this reason, it became urgent and essential to find adequate tests for an accurate and fast diagnosis of this disease. In the present study, a systematic review was performed in order to provide an overview of the COVID-19 diagnosis methods and tests already available, as well as their evolution in recent months. For this purpose, the Science Direct, PubMed, and Scopus databases were used to collect the data and three authors independently screened the references, extracted the main information, and assessed the quality of the included studies. After the analysis of the collected data, 34 studies reporting new methods to diagnose COVID-19 were selected. Although RT-PCR is the gold-standard method for COVID-19 diagnosis, it cannot fulfill all the requirements of this pandemic, being limited by the need for highly specialized equipment and personnel to perform the assays, as well as the long time to get the test results. To fulfill the limitations of this method, other alternatives, including biological and imaging analysis methods, also became commonly reported.     GD1a modulates GM-CSF-induced cell proliferation

    AbstractGangliosides have been extensively described to be involved in the proliferation and differentiation of various cell types, such including hematopoietic cells. Our previous studies on murine models of stroma-mediated myelopoiesis have shown that gangliosides are required for optimal capacity of stromal cells to support proliferation of myeloid precursor cells, being shed to the supernatant and selectively incorporated into myeloid cell membranes. Here we describe the effect of gangliosides on the specific granulocyte–macrophage colony-stimulating factor (GM-CSF)-induced proliferation. For that, we used the monocytic FDC-P1 cell line, which is dependent upon GM-CSF for survival and proliferation. Cells were cultured in the presence of GM-CSF and exogenous gangliosides (GM3, GD1a or GM1) or in the absence of endogenous ganglioside synthesis by the use of a ceramide-synthase inhibitor, d-PDMP. We observed that exogenous addition of GD1a enhanced the GM-CSF-induced proliferation of the FDC-P1 cells. Also, we detected an increase in the expression of the α isoform of the GM-CSF receptor (GMRα) as well as of the transcription factor C/EBPα. On the contrary, inhibition of glucosylceramide synthesis was accompanied by a decrease in cell proliferation, which was restored upon the addition of exogenous GD1a. We also show a co-localization of GD1a and GMR by immunocytochemistry. Taken together, our results suggest for the first time that ganglioside GD1a play a role on the modulation of GM-CSF-mediated proliferative response, which might be of great interest not only in hematopoiesis, but also in other immunological processes, Alzheimer disease, alveolar proteinosis and wherever GM-CSF exerts its effects

    Histological and ultrastructural feature and nitrite production of caprine preantral follicles in vitro cultured in the presence or absence of serum

    Avaliou-se o efeito da adição de diferentes tipos e concentrações de soro sobre o desenvolvimento e a sobrevivência de folículos ovarianos pré-antrais (FOPA) caprinos in vitro. Além disso, verificou-se a relação entre as concentrações de nitrito presentes no meio de cultivo e a viabilidade folicular. Cada par ovariano foi dividido em 29 fragmentos, sendo um destinado ao controle. Os fragmentos foram cultivados por um ou sete dias em meio essencial mínimo suplementado (MEM+) ou MEM+ com diferentes concentrações (10 ou 20%) de soro fetal bovino (SFB), soro de cabra em estro (SCE) ou soro de cabra em diestro (SCD). Na análise morfológica após sete dias, apenas o tratamento com 10% de SFB apresentou percentual de FOPA normais similar ao MEM+ (P>0,05). A análise ultra-estrutural dos folículos cultivados por sete dias com MEM+ ou MEM+ com 10% de SFB mostrou danos oocitários, porém células da granulosa normais. A análise do meio de cultivo revelou correlação positiva entre a viabilidade folicular e a produção de nitrito. A suplementação com soro não melhorou a viabilidade de FOPA e a concentração de nitrito no meio de cultivo funcionou como um indicador da viabilidade das células da granulosa de FOPA caprinos cultivados in vitro. ______________________________________________________________________________________________________________ ABSTRACTThe effect of the addition of different types and concentrations of sera on the viability and development of caprine preantal follicles (PAF) in vitro cultured was analyzed. In addition, it was evaluated the correlation between nitrite concentrations in culture medium and folicular viability. Each ovarian pair was divided in 29 fragments and one was used as control. The fragments were cultured for one or seven days in minimal essential medium (MEM+) or MEM+ with different concentrations of (10 or 20%) bovine fetal serum (BFS), estrous goat serum (EGS), or diestrous goat serum (DGS). After seven days, the morphological analysis showed that only the treatment with 10% BFS maintained the percentage of normal PAF similar to MEM+ (P>0.05). The ultrastructural analysis of follicles cultured for seven days in MEM+ or MEM+ with 10% BFS showed some oocyte damage, although the granulosa cells were normal. Analysis of culture medium revealed a positive correlation between follicular viability and nitrite production. Supplementation with serum did not improve the viability of PAF and nitrite levels in culture medium served as an indicator of viability of granulose cells from caprine PAF in vitro cultured

    Graviton production from extra dimensions

    Graviton production due to collapsing extra dimensions is studied. The momenta lying in the extra dimensions are taken into account. A DD-dimensional background is matched to an effectively four-dimensional standard radiation dominated universe. Using observational constraints on the present gravitational wave spectrum, a bound on the maximal temperature at the beginning of the radiation era is derived. This expression depends on the number of extra dimensions, as well as on the DD-dimensional Planck mass.     A global optimization approach applied to structural dynamic updating

    In this paper, the application of stochastic global optimization tech- niques, in particular the GlobalSearch and MultiStart solvers from MatLab®, to improve the updating of a structural dynamic model, are presented. For com- parative purposes, the efficiency of these global methods relatively to the local search method previously used in a Finite Element Model Updating program is evaluated. The obtained solutions showed that the GlobalSearch and MultiStart solvers are able to achieve a better solution than the local solver previously used, in the updating of a structural dynamic model. The results show also that the GlobalSearch solver is more efficient than the MultiStart, since requires less computational effort to obtain the global solution.Fundação para a Ciência e a Tecnologia (FCT

    Activated Random Walkers: Facts, Conjectures and Challenges

    We study a particle system with hopping (random walk) dynamics on the integer lattice Zd\mathbb Z^d. The particles can exist in two states, active or inactive (sleeping); only the former can hop. The dynamics conserves the number of particles; there is no limit on the number of particles at a given site. Isolated active particles fall asleep at rate λ>0\lambda > 0, and then remain asleep until joined by another particle at the same site. The state in which all particles are inactive is absorbing. Whether activity continues at long times depends on the relation between the particle density ζ\zeta and the sleeping rate λ\lambda. We discuss the general case, and then, for the one-dimensional totally asymmetric case, study the phase transition between an active phase (for sufficiently large particle densities and/or small λ\lambda) and an absorbing one. We also present arguments regarding the asymptotic mean hopping velocity in the active phase, the rate of fixation in the absorbing phase, and survival of the infinite system at criticality. Using mean-field theory and Monte Carlo simulation, we locate the phase boundary. The phase transition appears to be continuous in both the symmetric and asymmetric versions of the process, but the critical behavior is very different. The former case is characterized by simple integer or rational values for critical exponents (β=1\beta = 1, for example), and the phase diagram is in accord with the prediction of mean-field theory. We present evidence that the symmetric version belongs to the universality class of conserved stochastic sandpiles, also known as conserved directed percolation.
