    This study aims to analyze the factors that affect the going concern audit covering audit quality, audit tenure, the previous audit opinion and the company's growth. This research was conducted through secondary data searches and using 68 samples from the list of manufacturing companies in Indonesia Stock Exchange during the period 2011, 2012, 2013, 2014. Logistic regression analysis was used as techniques in this study to test the hypothesis. From the processed data can be concluded that only the variable quality of the audit and audit opinion the previous year affecting the going-concern audit and variables that do not affect the going concern audit opinion is audit tenure and the company's growth

    Isolasi dan Identifikasi Aspergillus Sp. pada Kantung Hawa Puyuh (Cortunix Japonica)

    Aspergillosis merupakan penyakit yang disebabkan oleh jamur Aspergillus sp. yang dapat menyerang puyuh muda secara akut dan sebagai penyakit menahun pada puyuh dewasa. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Aspergillus sp. pada kantung hawa puyuh (Coturnix japonica). Sampel yang digunakan dalam penelitian ini adalah 35 kantung hawa puyuh yang tidak produktif lagi (afkir) diambil secara acak. Isolasi dan identifikasi Aspergillus sp. dilakukan sesuai dengan menggunakan metode Thompson. Sampel dicuci dengan aquadest steril berisi antibiotic gentamicin selanjutnya ditanamkan pada media Sabouraud’s Dextrose Agar (SDA) kemudian diinkubasikan pada suhu kamar selama 3-7 hari. Pertumbuhan koloni Aspergillus sp. diamati morfologinya secara makroskopis dan mikroskopis. Data yang diperoleh dianalisis secara deskriptif. Hasil pemeriksaan menunjukkan bahwa Aspergillus sp. dapat diisolasi dan diidentifikasi pada 24 dari 35 sampel kantung hawa. Berdasarkan hasil penelitian dapat disimpulkan bahwa 68,57% sampel yang diperiksa positif terinfeksi Aspergillus sp. dengan persentase masing-masing spesies Aspergillus fumigatus 40%, Aspergillus flavus 25,7%, dan Aspergillus niger 17,1%

    Deteksi Antibodi terhadap Virus Newcastle Disease pada Burung Trucukan (Pycnonotus goiavier)

    Burung trucukan (Pycnonotus goiavier) merupakan salah satu burung liar yang sering dipelihara karena kicauannyayang merdu. Infeksi ND dapat berupa infeksi yang akut atau kronis yang menyerang burung trucukan dan jenis burunglainnya. Burung liar merupakan salah satu sumber virus ND yang dapat menyebarkan virus ke unggas peliharaan.Penelitian ini bertujuan untuk mengetahui titer antibodi terhadap penyakit tetelo pada burung trucukan liar. Sampel yangdigunakan adalah serum darah dari 50 ekor burung trucukan liar. Parameter yang diamati adalah titer antibodi dalamserum berdasarkan metode Hirst menggunakan uji Haemagglutination Inhibition (HI) untuk melihat titer antibodi. Untukmenghitung titer virus maka uji Haemaglutination (HA) dilakukan sebelumnya. Untuk memastikan titer antigen yangdigunakan adalah 4HAU dilkukan juga back titrasi sehingga didapatkan nilai virus optimal yang akan dipakai pada uji HI.Hasil penelitian menunjukkan 45 dari 50 sampel (90%) mempunyai antibodi positif terhadap ND, dengan titer berkisar 21-29. Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa 90% burung trucukan di Kabupaten Aceh Besar sudahpernah terpapar ND, tetapi hanya 80% yang memiliki titer antibodi yang dapat melingdungi terhadap serangan virus ND.Oleh sebab itu dapat disimpulkan bahwa burung trucukan liar dengan titer antibodi yang protektif berarti sudah pernahterpapar virus ND sebelumnya akan berpotensi sebagai pembawa virus yang dapat ditularkan ke unggas peliharaanmasyarakat di sekitarnya

    AbstractBiofilm  is one of parameters for determining pathogenic pathways of a bacterium. Bacterial virulence activities can be attenuated by antibiotics with inhibiting biofilm. The aim of this study was to examine the characteristics and activities of antibiotics against the biofilm of Stapylococcus aureus isolate which taken from Aceh cattle prepuce. The study was exploratory and experimental, using a Completely Randomized Design (RAL) factorial pattern using three types of antibiotics, three concentrations and replications. Antibiotics were tetracycline, oxytetracycline and phosphomycin with concentrations of 10%, 20% and 30% (mg/mL), respectively. Therefore, the aim of this study was to examine the characteristics and activities of antibiotics to the biofilm of Stapylococcus aureus from Aceh cattle prepuce isolate. U-bottom polystyrene 96 wells microplate and Nutrient Broth (NB)  medium were used to biofilm. Bacterial  while biofilm was stained on crystals violet 1%. Optical Density (OD) of biofilm were  λ 595 nm. Data were analyzed by using Analysis Of Variance (ANOVA). The results showed that the activity of Staphylococcus aureus was moderately positive, i.e 2,667cell/ΌL. It showed that antibiotics and concetrations have significant effect (P<0.05) to inhibit biofilm staphylococcus aureus. In additioon, tetracycline 30% (mg / mL) has the best ability to inhibit biofilms from 52,18% to 1,262 cell/ΌL. Based on that fact, it can be concluded that the virulence of Staphylococcus aureus was moderate positive to be false positive. Keywords: biofilm, antibiotics, Staphylococcus aureus, cows aceh.                Abstrak Biofilm merupakan salah satu parameter untuk menentukan jalur patogenisitas suatu bakteri. Antibiotik dapat melemahkan aktivitas virulensi bakteri dengan menghambat pembentukan biofilm. Penelitian ini bertujuan untuk melihat karaksteristik dan aktivitas antibiotik terhadap biofilm Stapylococcus aureus isolat preputium sapi Aceh. Penelitian bersifat eksplorasi dan eksperimental, menggunakan Rancangan Acak Lengkap (RAL) pola faktorial dengan menggunakan tiga jenis antibiotik, tiga konsentrasi dan tiga kali ulangan. Antibiotik yang digunakan adalah tetrasiklin, oksitetrasiklin dan fosfomisin dengan konsentrasi masing-masing 10%, 20% dan 30% (mg/mL). Pengujian biofilm dilakukan pada media Nutrient Broth (NB), menggunakan mikroplat U-bottom polystyrene 96 wells. Pewarnaan biofilm menggunakan kristal violet 1%. Optical Density (OD) biofilm dibaca pada    λ 595 nm. Data hasil penelitian dianalisis dengan menggunakan Analysis Of Variance (ANOVA). Hasil penelitian menunjukkan aktivitas virulensi Staphylococcus aureus isolat preputium sapi Aceh berdasarkan karaksteristik biofilm tergolong positif moderat, yaitu 2,667cell/”L. Uji Biofilm menunjukkan jenis dan konsentrasi antibiotik berpengaruh secara nyata (P<0,05) terhadap daya hambat biofilm Staphylococcus aureus isolat preputium sapi Aceh. Tetrasiklin 30% (mg/mL) mempunyai kemampuan paling baik menghambat pembentukan biofilm Staphylococcus aureus isolat preputium sapi Aceh sebesar 52,18% menjadi 1,262 cell/”L, sehingga aktivitas virulensi Staphylococcus aureus isolat preputium sapi Aceh berdasarkan karaksteristik biofilm dari positif moderat menjadi positif palsu. Kata Kunci: biofilm, antibiotik, Staphylococcus aureus, sapi aceh

    Targeting negative regulation of p53 by MDM2 and WIP1 as a therapeutic strategy in cutaneous melanoma

    BACKGROUND: Cutaneous melanoma is the most serious skin malignancy and new therapeutic strategies are needed for advanced melanoma. TP53 mutations are rare in cutaneous melanoma and hence activation of wild-type p53 is a potential therapeutic strategy in cutaneous melanoma. Here, we investigated the WIP1 inhibitor, GSK2830371, and MDM2–p53 binding antagonists (nutlin-3, RG7388 and HDM201) alone and in combination treatment in cutaneous melanoma cell lines and explored the mechanistic basis of these responses in relation to the genotype and induced gene expression profile of the cells. // METHODS: A panel of three p53WT (A375, WM35 and C8161) and three p53MUT (WM164, WM35-R and CHL-1) melanoma cell lines were used. The effects of MDM2 and WIP1 inhibition were evaluated by growth inhibition and clonogenic assays, immunoblotting, qRT–PCR gene expression profiling and flow cytometry. // RESULTS: GSK2830371, at doses (10 ΌM) that alone had no growth-inhibitory or cytotoxic effects on the cells, nevertheless significantly potentiated the growth-inhibitory and clonogenic cell killing effects of MDM2 inhibitors in p53WT but not p53MUT melanoma cells, indicating the potentiation worked in a p53-dependent manner. The siRNA-mediated knockdown of p53 provided further evidence to support the p53 dependence. GSK2830371 increased p53 stabilisation through Ser15 phosphorylation and consequent Lys382 acetylation, and decreased ubiquitination and proteasome-dependent degradation when it was combined with MDM2 inhibitors. These changes were at least partly ATM mediated, shown by reversal with the ATM inhibitor (KU55933). GSK2830371 enhanced the induction of p53 transcriptional target genes, cell cycle arrest and apoptosis. // CONCLUSIONS: GSK2830371, a WIP1 inhibitor, at doses with no growth-inhibitory activity alone, potentiated the growth-inhibitory and cytotoxic activity of MDM2 inhibitors by increasing phosphorylation, acetylation and stabilisation of p53 in cutaneous melanoma cells in a functional p53-dependent manner

    Co-directional replication-transcription conflicts lead to replication restart

    August 24, 2011Head-on encounters between the replication and transcription machineries on the lagging DNA strand can lead to replication fork arrest and genomic instability1, 2. To avoid head-on encounters, most genes, especially essential and highly transcribed genes, are encoded on the leading strand such that transcription and replication are co-directional. Virtually all bacteria have the highly expressed ribosomal RNA genes co-directional with replication3. In bacteria, co-directional encounters seem inevitable because the rate of replication is about 10–20-fold greater than the rate of transcription. However, these encounters are generally thought to be benign2, 4, 5, 6, 7, 8, 9. Biochemical analyses indicate that head-on encounters10 are more deleterious than co-directional encounters8 and that in both situations, replication resumes without the need for any auxiliary restart proteins, at least in vitro. Here we show that in vivo, co-directional transcription can disrupt replication, leading to the involvement of replication restart proteins. We found that highly transcribed rRNA genes are hotspots for co-directional conflicts between replication and transcription in rapidly growing Bacillus subtilis cells. We observed a transcription-dependent increase in association of the replicative helicase and replication restart proteins where head-on and co-directional conflicts occur. Our results indicate that there are co-directional conflicts between replication and transcription in vivo. Furthermore, in contrast to the findings in vitro, the replication restart machinery is involved in vivo in resolving potentially deleterious encounters due to head-on and co-directional conflicts.     Adaptive access and rate control of CSMA for energy, rate and delay optimization

    In this article, we present a cross-layer adaptive algorithm that dynamically maximizes the average utility function. A per stage utility function is defined for each link of a carrier sense multiple access-based wireless network as a weighted concave function of energy consumption, smoothed rate, and smoothed queue size. Hence, by selecting weights we can control the trade-off among them. Using dynamic programming, the utility function is maximized by dynamically adapting channel access, modulation, and coding according to the queue size and quality of the time-varying channel. We show that the optimal transmission policy has a threshold structure versus the channel state where the optimal decision is to transmit when the wireless channel state is better than a threshold. We also provide a queue management scheme where arrival rate is controlled based on the link state.
