Práce obsahuje informace o jednotlivých typech VLJ, jejich provozní náklady a srovnání s konkurencí, informace o podílovém vlastnictví a možnostech provozování. Je zde provedena analýza trhu pro tyto letouny a rozebrána problematika letecké dopravy aerotaxi.Master’s thesis deals with problematic of operating aspects and competitiveness of VLJ aircraft in Czech and Europe market. The Thesis contain information about different types of VLJ, their operational costs and comparisons with competitors, some information about aircraft fractional ownership and their operation. There is written a market analysis for this aircraft and air taxi service in analyzed.
Plugin for Simplification of Programming Using Visual Studio 2010

Aplikace napovídá ukázky zdrojového kódu v prostředí Visual Studia 2010 dle aktuálně používaného programovacího jazyku. Aplikace je typu klient-server, server zajišťuje vyhledávání ukázek zdrojových kódů.This thesis is focused on tool for simplification development in new programing language for non-experienced programmers. Application suggests sample source codes for current programming language in Visual Studio 2010. The application is a client-server, the server implements searching of the source code examples.

Bakalářská práce se zabývá větracími systémy a požadavky na větrání společenských místností. Autor se nejprve zaměřuje na základní systémy větrání. Ty by měly napomoci vytvořit teoretický náhled na problematiku větrání. Poté jsou podrobně probrány hygienické požadavky na větrání společenských sálů s podrobnějším zaměřením na kina a divadla. Na závěr se autor uvádí vhodné systémy větrání splňující požadavky stanovené hygienickými předpisy.Bachelor’s thesis deals with ventilation systems and requirements for ventilation of cultural rooms. First of all author focuses on basic ventilation systems. This should be useful to create some theoretical view about this theme. In next topic hygienic requirements for cultural rooms with specification on theatres are discussed in details. In final part author presents suitable ventilation systems capable to fit stated hygienic requirements.
Statistic Evaluation of Efficient Characteristic of UL and VLA Aircraft

In total 65 types of aircraft. In the Thesis is described statistic evaluation of performance and geometrical characteristics by mathematics method of correlation.
Evaluation of NAD(+)-dependent DNA ligase of mycobacteria as a potential target for antibiotics
Mycobacteria contain genes for several DNA ligases, including ligA, which encodes a NAD+-dependent enzyme that has been postulated to be a target for novel antibacterial compounds. Using a homologous recombination system, direct evidence is presented that wild-type ligA cannot be deleted from the chromosome of Mycobacterium smegmatis. Deletions of native ligA in M. smegmatis could be obtained only after the integration of an extra copy of M. smegmatis or Mycobacterium tuberculosis ligA into the attB site of the chromosome, with expression controlled by chemically inducible promoters. The four ATP-dependent DNA ligases encoded by the M. smegmatis chromosome were unable to replace the function of LigA. Interestingly, the LigA protein from M. smegmatis could be substituted with the NAD+-dependent DNA ligase of Escherichia coli or the ATP-dependent ligase of bacteriophage T4. The conditional mutant strains allowed the analysis of the effect of LigA depletion on the growth of M. smegmatis. The protein level of the conditional mutants was estimated by Western blot analysis using antibodies raised against LigA of M. tuberculosis. This revealed that a strong overproduction or depletion of LigA did not affect the growth or survival of mycobacteria under standard laboratory conditions. In conclusion, although NAD+-dependent DNA ligase is essential for mycobacterial viability, only low levels of protein are required for growth. These findings suggest that very efficient inhibition of enzyme activity would be required if NAD+-dependent DNA ligase is to be useful as an antibiotic target in mycobacteria. The strains developed here will provide useful tools for the evaluation of the efficacy of any appropriate compounds in mycobacteria
NHEJ protects mycobacteria in stationary phase against the harmful effects of desiccation
The physiological role of the non-homologous end-joining (NHEJ) pathway in the repair of DNA double-strand breaks (DSBs) was examined in Mycobacterium smegmatis using DNA repair mutants (DeltarecA, Deltaku, DeltaligD, Deltaku/ligD, DeltarecA/ku/ligD). Wild-type and mutant strains were exposed to a range of doses of ionizing radiation at specific points in their life-cycle. NHEJ-mutant strains (Deltaku, DeltaligD, Deltaku/ligD) were significantly more sensitive to ionizing radiation (IR) during stationary phase than wild-type M. smegmatis. However, there was little difference in IR sensitivity between NHEJ-mutant and wild-type strains in logarithmic phase. Similarly, NHEJ-mutant strains were more sensitive to prolonged desiccation than wild-type M. smegmatis. A DeltarecA mutant strain was more sensitive to desiccation and IR during both stationary and especially in logarithmic phase, compared to wild-type strain, but it was significantly less sensitive to IR than the DeltarecA/ku/ligD triple mutant during stationary phase. These data suggest that NHEJ and homologous recombination are the preferred DSB repair pathways employed by M. smegmatis during stationary and logarithmic phases, respectively
RNase HI Is Essential for Survival of Mycobacterium smegmatis
RNases H are involved in the removal of RNA from RNA/DNA hybrids. Type I RNases H
are thought to recognize and cleave the RNA/DNA duplex when at least four ribonucleotides
are present. Here we investigated the importance of RNase H type I encoding genes
for model organism Mycobacterium smegmatis. By performing gene replacement through
homologous recombination, we demonstrate that each of the two presumable RNase H
type I encoding genes, rnhA and MSMEG4305, can be removed from M. smegmatis genome
without affecting the growth rate of the mutant. Further, we demonstrate that deletion
of both RNases H type I encoding genes in M. smegmatis leads to synthetic lethality. Finally,
we question the possibility of existence of RNase HI related alternative mode of initiation
of DNA replication in M. smegmatis, the process initially discovered in Escherichia coli. We
suspect that synthetic lethality of double mutant lacking RNases H type I is caused by formation
of R-loops leading to collapse of replication forks. We report Mycobacterium smegmatis
as the first bacterial species, where function of RNase H type I has been found
