    La formazione della più antica società picena tra Bronzo tardo e prima età del Ferro

    Questa ricerca di dottorato ha avuto come obiettivo finale la ricostruzione dei processi formativi della cosidetta “civiltà” o “cultura” picena, in un arco cronologico compreso tra la fine dell’età del Bronzo e la prima età del Ferro. Dopo un’accurata revisione del materiale documentario e dei contesti inediti a disposizione, la ricerca si è però concentrata essenzialmente sulla prima età del Ferro, che forniva una base quantitativamente più coerente e consistente, rispetto ai pochi e scarsamente documentati contesti databili nell’ambito del Bronzo finale. Tuttavia, nel capitolo VI, relativo alle dinamiche insediamentali in area medio-adriatica, nell’analisi sono stati riconsiderati anche i contesti noti databili nell’ambito del Bronzo finale, al fine di evidenziare eventuali cambiamenti e forme di continuità/discontinuità insediativa con la fase successiva. La problematica della formazione della civiltà o cultura picena è stata tradizionalmente affrontata sulla base della tipologia e della distribuzione di alcuni elementi distintivi tra Marche e Abruzzo centro-settentrionale, pertinenti soprattutto all’ambito funerario, mentre si sono tralasciati tutta un’altra serie di aspetti, quali quelli legati ai contesti e ai materiali da abitato. L’obiettivo preliminare della ricerca è stato dunque quello di costruire una solida seriazione cronologica, inizialmente considerando i soli materiali ceramici, da contesti perlopiù abitativi e in minor misura funerari. In questo senso la disponibilità dei materiali dai siti inediti di Montedoro, Montelupone e Teramo ha ampliato notevolmente la base documentaria a disposizione . In un secondo momento si è deciso di considerare anche i materiali non ceramici da contesti funerari, al fine di agganciare la cronotipologia dei materiali ceramici a tipi metallici noti e cronologicamente affidabili. Questa griglia cronotipologica ha costituito la base per la seconda parte del lavoro, incentrato sulla distribuzione di tipi di manufatti, di strutture e di rituali funerari, nonché sull’analisi delle forme di insediamento e dell’organizzazione territoriale. Il lavoro svolto ha permesso di verificare l’esistenza di un’entità culturale picena, piuttosto omogenea al proprio interno, già a partire dalla prima età del Ferro, di ridefinirne i confini e talvolta di sfumarli, così come ha permesso di intravedere nella scarsa coesione e omogeneità delle manifestazioni culturali di parte dell’Abruzzo interno e meridionale, le origini della successiva frammentazione dell’area in diverse entità culturali

    Indeterminate Thyroid Nodules: From Cytology to Molecular Testing

    Thyroid cancer is the most common malignancy of the endocrine system. Fine-needle aspiration (FNA) biopsy of thyroid nodules has become the gold standard procedure, in terms of cost and efficacy, for guiding clinicians towards appropriate patients’ management. One challenge for cytopathologists is to accurately classify cytological specimens as benign or malignant based on cytomorphological features. In fact, with a frequency ranging from 10% to 30%, nodules are diagnosed as indeterminate. In recent years, the mutational landscape of thyroid tumors has been extensively described, and two molecular profiles have been identified: RAS-like (NRAS, HRAS, and KRAS mutations; EIF1AX mutations; BRAF K601E mutation; and PPARG and THADA fusions) and BRAFV600E-like (including BRAFV600E mutation and RET and BRAF fusions).     Plio-quaternary geology of L’Aquila-Scoppito basin (Central Italy)

    We present a geological map at 1: 25,000 scale of the Plio-Quaternary L'Aquila-Scoppito intermontane basin (central Italy), which corresponds to the epicentral area of the 6th April 2009 L'Aquila earthquake (Mw: 6.29). The map is derived from geological field surveys at 1: 5000 scale and takes into account previously published maps. It is supported by a fine-scale LiDAR digital elevation model, paleontological analyses, C-14 dating, well log analyses of deep boreholes, and geophysical data interpretation. By taking into account that the remarkable historical and present-day seismicity of the area is the result of its PlioQuaternary geological evolution, the aim of this study is to produce a fine-scale geological map of the study area through a comprehensive analysis of the occurring Plio-Quaternary synthems and the tectonic processes that are taking place within it

    MicroRNA expression profiling of RAS-mutant thyroid tumors with follicular architecture: microRNA signatures to discriminate benign from malignant lesions

    Purpose: RAS mutations represent common driver alterations in thyroid cancer. They can be found in benign, low-risk and malignant thyroid tumors with follicular architecture, which are often diagnosed as indeterminate nodules on preoperative cytology. Therefore, the detection of RAS mutations in preoperative setting has a suboptimal predictive value for malignancy. In this study, we investigated differentially expressed microRNA (miRNA) in benign and malignant thyroid tumors with follicular architecture carrying mutations in RAS genes. Methods: Total RNA was purified from 60 RAS-mutant follicular-patterned thyroid tumors, including follicular adenoma (FA), noninvasive follicular thyroid neoplasm with papillary-like nuclear features (NIFTP), papillary and follicular thyroid carcinoma cases (PTC, FTC); 22 RAS-negative FAs were used as controls. The expression analysis of 798 miRNAs was performed by digital counting (nCounter nanoString platform). Results: Comparing RAS-mutant and RAS-negative FAs, 12 miRNAs showed significant deregulation, which was likely related to the oncogenic effects of RAS mutations. Twenty-two miRNAs were differentially expressed in RAS-mutant benign versus malignant tumors. Considering the tumor type, 24 miRNAs were deregulated in PTC, 19 in NIFTP, and seven in FTC and compared to FA group; among these, miR-146b-5p, miR-144-3p, and miR-451a showed consistent deregulation in all the comparisons with the highest fold change. Conclusions: The miRNA expression analysis of follicular-patterned thyroid tumors demonstrated that RAS mutations influences miRNA profile in benign tumors. In addition, several miRNAs showed a histotype-specific deregulation and could discriminate between RAS-mutant benign and RAS-mutant malignant thyroid lesions, thus deserving further investigation as potential diagnostic markers

    New insights into earthquake precursors from InSAR

    We measured ground displacements before and after the 2009 L’Aquila earthquake using multitemporal InSAR techniques to identify seismic precursor signals. We estimated the ground deformation and its temporal evolution by exploiting a large dataset of SAR imagery that spans seventy-two months before and sixteen months after the mainshock. These satellite data show that up to 15 mm of subsidence occurred beginning three years before the mainshock. This deformation occurred within two Quaternary basins that are located close to the epicentral area and are filled with sediments hosting multi-layer aquifers. After the earthquake, the same basins experienced up to 12 mm of uplift over approximately nine months. Before the earthquake, the rocks at depth dilated, and fractures opened. Consequently, fluids migrated into the dilated volume, thereby lowering the groundwater table in the carbonate hydrostructures and in the hydrologically connected multi-layer aquifers within the basins. This process caused the elastic consolidation of the fine-grained sediments within the basins, resulting in the detected subsidence. After the earthquake, the fractures closed, and the deep fluids were squeezed out. The pre-seismic ground displacements were then recovered because the groundwater table rose and natural recharge of the shallow multi-layer aquifers occurred, which caused the observed uplift.Published120356T.     Site response analyses for complex geological and morphological conditions: relevant case-histories from 3rd level seismic microzonation in Central Italy

    The paper presents the results of 5 case studies on complex site e ects selected within the project for the level 3 seismic microzonation of several municipalities of Central Italy dam- aged by the 2016 seismic sequence. The case studies are characterized by di erent geo- logical and morphological con gurations: Monte San Martino is located along a hill slope, Montedinove and Arquata del Tronto villages are located at ridge top whereas Capitignano and Norcia lie in correspondence of sediment- lled valleys. Peculiarities of the sites are constituted by the presence of weathered/jointed rock mass, fault zone, shear wave veloc- ity inversion, complex surface and buried morphologies. These factors make the de ni- tion of the subsoil model and the evaluation of the local response particularly complex and di cult to ascertain. For each site, after the discussion of the subsoil model, the results of site response numerical analyses are presented in terms of ampli cation factors and acceleration response spectra in selected points. The physical phenomena governing the site response have also been investigated at each site by comparing 1D and 2D numerical analyses. Implications are deduced for seismic microzonation studies in similar geological and morphological conditions.Published5741–57775T.     Telomerase reverse transcriptase promoter alterations across cancer types as detected by next-generation sequencing: A clinical and molecular analysis of 423 patients.

    BACKGROUND: Telomerase reverse transcriptase (TERT) promoter mutations that may affect telomerase activity have recently been described in human malignancies. The purpose of this study was to investigate the clinical correlates of TERT promoter abnormalities in a large cohort of patients with diverse cancers. METHODS: This study analyzed TERT promoter alterations and clinical characteristics of 423 consecutive patients for whom molecular testing by next-generation sequencing was performed between August 2014 and July 2015. RESULTS: Of the 423 patients, 61 (14.4%) had TERT promoter mutations, and this placed TERT promoter alterations among the most prevalent aberrations after tumor protein 53 (TP53; 39%) and KRAS and cyclin-dependent kinase inhibitor 2A/B (CDKN2A/B) alterations (15% each) in this population. TERT promoter alterations were more frequent in men (P = .031) and were associated with brain cancers (P = .001), skin cancers/melanoma (P = .001), and a higher number of aberrations (P = .0001). A co-alteration analysis found that TERT promoter alterations were significantly correlated with CDKN2A/B (P = .001) and BRAF abnormalities (P = .0003). Patients harboring TERT promoter alterations or TP53 or CDKN2A/B alterations and those with 4 or more alterations demonstrated shorter survival (hazard ratio for normal TERT promoters vs aberrant ones, 0.44; P = .017). However, only a higher number of alterations remained significant in the multivariate analysis. CONCLUSIONS: Overall, TERT promoter alterations were among the most prevalent aberrations in this population, with very high rates in brain cancers (48% of patients) and melanomas (56% of patients). These aberrations frequently coexist with a high number of other aberrations, with the latter feature also significantly associated with poorer overall survival. Therapeutic options for targeting tumors with TERT promoter mutations are currently limited, although a variety of novel approaches are under development.     Web Based Postgraduate Thesis/Dessertation System - A Prototype

    With the advancement of information communication technology in Malaysia, education field should take advantage to upgrade their learning and management techniques. Students should be allowed to learn anytime, anywhere and at their own place. However administration and lecture should be able to manage their work more effective and flexible. The web-based system is effective way to learning and managing education works. This report outlines the development of a web-based postgraduate thesis/dissertation management system (WPTS), which aimed to assist thesis/dissertation administration, supervisor and students in the better integration during students doing the thesis/dissertation works. This prototype system base on case study with a group of MSC(IT), administration, lecturer and students who participate in thesis/dissertation management activities. This report also presenting the tests conducted with users, it also contributed some perspective regarding benefits that gain by administration, supervisor and students, and recommends future application of the approach

    Understanding TERT promoter mutations: a common path to immortality

    Telomerase (TERT) activation is a fundamental step in tumorigenesis. By maintaining telomere length, telomerase relieves a main barrier on cellular lifespan, enabling limitless proliferation driven by oncogenes. The recently discovered, highly recurrent mutations in the promoter of TERT are found in over 50 cancer types, and are the most common mutation in many cancers. Transcriptional activation of TERT, via promoter mutation or other mechanisms, is the rate-limiting step in production of active telomerase. Although TERT is expressed in stem cells, it is naturally silenced upon differentiation. Thus, the presence of TERT promoter mutations may shed light on whether a particular tumor arose from a stem cell or more differentiated cell type. It is becoming clear that TERT mutations occur early during cellular transformation, and activate the TERT promoter by recruiting transcription factors that do not normally regulate TERT gene expression. This review highlights the fundamental and widespread role of TERT promoter mutations in tumorigenesis, including recent progress on their mechanism of transcriptional activation. These somatic promoter mutations, along with germline variation in the TERT locus also appear to have significant value as biomarkers of patient outcome.
