    Electrochemical studies on small electron transfer proteins using membrane electrodes

    Journal of Electroanalytical Chemistry 541 (2003) 153-162Membrane electrodes (ME) were constructed using gold, glassy carbon and pyrolytic graphite supports and a dialysis membrane, and used to study the electrochemical behavior of small size electron transfer proteins: monohemic cytochrome c522 from Pseudomonas nautica and cytochrome c533 as well as rubredoxin from Desulfovibrio vulgaris . Different electrochemical techniques were used including cyclic voltammetry (CV), square wave voltammetry (SW) and differential pulse voltammetry (DP). A direct electrochemical response was obtained in all cases except with rubredoxin where a facilitator was added to the protein solution entrapped between the membrane and the electrode surface. Formal potentials and heterogeneous charge transfer rate constants were determined from the voltammetric data. The influence of the ionic strength and the pH of the medium on the electrochemical response at the ME were analyzed. The benefits from the use of the ME in protein electrochemistry and its role in modulating the redox behavior are analyzed. A critical comparison is presented with data obtained at non-MEs. Finally, the interactions that must be established between the proteins and the electrode surfaces are discussed, thereby modeling molecular interactions that occur in biological systems

    Num momento em que a gestão sustentável dos recursos hídricos se assume como uma das maiores preocupações à escala mundial, as regiões de montanha têm vindo a ser reconhecidas como Reservatórios de Água (PROGRAMA UNESCO IHP-VI). O facto de muitas destas áreas se localizarem a altitudes elevadas e serem recortadas por rodovias obriga a frequentes operações de limpeza de neve, especialmente no inverno. A aplicação de substâncias químicas como cloreto de sódio e cloreto de cálcio na promoção da fusão do gelo e na limpeza da neve das estradas constitui um problema ambiental de elevado interesse científico, social e económico. Devido às suas características geológicas, geomorfológicas e climáticas, a Serra da Estrela é considerada estratégica para a gestão da água em Portugal. No presente trabalho apresentam-se alguns resultados hidrogeoquímicos que indicam que a utilização dos referidos sais na limpeza do gelo e da neve das rodovias contribui para a contaminação dos recursos hídricos

    Undamning the Douro river catchment: a stepwise approach for prioritizing dam removal

    Dams provide water supply, flood protection, and hydropower generation benefits, but also harm native species by altering the natural flow regime, and degrading the aquatic and riparian habitats. In the present study, which comprised the Douro River basin located in the North of Portugal, the cost-benefit assessment of dams was based upon a balance between the touristic benefits of a dammed Douro, and the ecological benefits of less fragmented Douro sub-catchments. Focused on four sub-catchments (Sabor, Tâmega, Côa and Corgo), a probabilistic stream connectivity model was developed and implemented to recommend priorities for dam removal, where this action could significantly improve the movement of potadromous fish species along the local streams. The proposed model accounts for fish movement across the dam or weir (permeability), which is a novel issue in connectivity models. However, before any final recommendation on the fate of a dam or weir, the connectivity results will be balanced with other important socio-economic interests. While implementing the connectivity model, an inventory of barriers (dams and weirs) was accomplished through an observation of satellite images. Besides identification and location of any obstacles, the inventory comprised the compilation of data on surrounding land use, reservoir water use, characteristics of the riparian gallery, and permeability conditions for fish, among others. All this information was stored in a geospatial dataset that also included geographical information on the sub-catchment drainage network. The linear (drainage network) and point (barriers) source data were processed in a computer program that provided or returned numbers for inter-barrier stream lengths (habitat), and the barrier permeability. These numbers were finally used in the same computer program to calculate a habitat connector index, and a link improvement index, used to prioritize dam removal based upon structural connectivity criteria. The results showed that habitat patch connectivity in the Sabor, Tâmega and Côa sub-catchments is not dramatically affected by the installed obstacles, because most link improvement values were generally low. For the opposite reason, in the Corgo sub-catchment, obstacles may constitute a relatively higher limitation to connectivity, and in this case the removal of eight obstacles could significantly improve this connectivity. Using the probabilistic model of structural connectivity, it was possible to elaborate a preliminary selection of dams/weirs that critically limit stream connectivity, and that will be the focus of field hydraulic characterization to precisely determine fish movement along the associated river stretches.     Four-dimensional anti-de Sitter black holes from a three-dimensional perspective: Full complexity

    The dimensional reduction of black hole solutions in four-dimensional (4D) general relativity is performed and new 3D black hole solutions are obtained. Considering a 4D spacetime with one spacelike Killing vector, it is possible to split the Einstein-Hilbert-Maxwell action with a cosmological term in terms of 3D quantities. Definitions of quasilocal mass and charges in 3D spacetimes are reviewed. The analysis is then particularized to the toroidal charged rotating anti-de Sitter black hole.     Complex geomorphologic assemblage of terrains in association with the banded terrain in Hellas basin, Mars

    Hellas basin acts as a major sink for the southern highlands of Mars and is likely to have recorded several episodes of sedimentation and erosion. The north-western part of the basin displays a potentially unique Amazonian landscape domain in the deepest part of Hellas, called “banded terrain”, which is a deposit characterized by an alternation of narrow band shapes and inter-bands displaying a sinuous and relatively smooth surface texture suggesting a viscous flow origin. Here we use high-resolution (HiRISE and CTX) images to assess the geomorphological interaction of the banded terrain with the surrounding geomorphologic domains in the NW interior of Hellas to gain a better understanding of the geological evolution of the region as a whole. Our analysis reveals that the banded terrain is associated with six geomorphologic domains: a central plateau named Alpheus Colles, plain deposits (P1 and P2), reticulate (RT1 and RT2) and honeycomb terrains. Based on the analysis of the geomorphology of these domains and their cross-cutting relationships, we show that no widespread deposition post-dates the formation of the banded terrain, which implies that this domain is the youngest and latest deposit of the interior of Hellas. Therefore, the level of geologic activity in the NW Hellas during the Amazonian appears to have been relatively low and restricted to modification of the landscape through mechanical weathering, aeolian and periglacial processes. Thermophysical data and cross-cutting relationships support hypotheses of modification of the honeycomb terrain via vertical rise of diapirs such as ice diapirism, and the formation of the plain deposits through deposition and remobilization of an ice-rich mantle deposit. Finally, the observed gradual transition between honeycomb and banded terrain suggests that the banded terrain may have covered a larger area of the NW interior of Hellas in the past than previously thought.     Flow cytometry immunophenotyping for diagnostic orientation and classification of pediatric cancer based on the EuroFlow Solid Tumor Orientation Tube (STOT)

    Early diagnosis of pediatric cancer is key for adequate patient management and improved outcome. Although multiparameter flow cytometry (MFC) has proven of great utility in the diagnosis and classification of hematologic malignancies, its application to non-hematopoietic pediatric tumors remains limited. Here we designed and prospectively validated a new single eight-color antibody combination-solid tumor orientation tube, STOT-for diagnostic screening of pediatric cancer by MFC. A total of 476 samples (139 tumor mass, 138 bone marrow, 86 lymph node, 58 peripheral blood, and 55 other body fluid samples) from 296 patients with diagnostic suspicion of pediatric cancer were analyzed by MFC vs. conventional diagnostic procedures. STOT was designed after several design-test-evaluate-redesign cycles based on a large panel of monoclonal antibody combinations tested on 301 samples. In its final version, STOT consists of a single 8-color/12-marker antibody combination (CD99-CD8/(nu)myogenin/CD4-EpCAM/CD56/GD2/(sm)CD3-CD19/(cy)CD3-CD271/CD45). Prospective validation of STOT in 149 samples showed concordant results with the patient WHO/ICCC-3 diagnosis in 138/149 cases (92.6%). These included: 63/63 (100%) reactive/disease-free samples, 43/44 (98%) malignant and 4/4 (100%) benign non-hematopoietic tumors together with 28/38 (74%) leukemia/lymphoma cases; the only exception was Hodgkin lymphoma that required additional markers to be stained.& nbsp;In addition, STOT allowed accurate discrimination among the four most common subtypes of malignant CD45(-) CD56(++) non-hematopoietic solid tumors: 13/13 (GD2(++) (nu)myogenin(-) CD271(-/+) (nu)MyoD1(-) CD99(-) EpCAM(-)) neuroblastoma samples, 5/5 (GD2(-) (nu)myogenin(++) CD271(++) (nu)MyoD1(++) CD99(-/+) EpCAM(-)) rhabdomyosarcomas, 2/2 (GD2(-/+) (nu)myogenin(-) CD271(+) (nu)MyoD1(-) CD99(+) EpCAM(-)) Ewing sarcoma family of tumors, and 7/7 (GD2(-) (nu)myogenin(-) CD271(+) (nu)MyoD1(-) CD99(-) EpCAM(+)) Wilms tumors.
