    Synthesis of N'-Substituted-2-(5-(4-Chlorophenyl)-1,3,4- oxadiazol-2-ylthio)acetohydrazide Derivatives as Suitable Antibacterial Agents

    Purpose: To evaluate antibacterial activity of a series of molecules bearing 1,3,4-oxadiazole and azomethine moieties.Methods: The 4-chlorobenzoic acid (1) was precursor to N'-substituted-2-(5-(4-chlorophenyl)-1,3,4- oxadiazol-2-ylthio)acetohydrazide, 8a-p, through a multistep synthesis of corresponding ester, 2, hydrazide, 3 and 1,3,4-oxadiazole, 4. The molecule, 4, was subjected to electrophilic substitution by ethyl-2-bromoacetate to yield 5 which was stepped to 2-(5-(4-chlorophenyl)-1,3,4-oxadiazol-2- ylthio)acetohydrazide (6). The target molecules, 8a-p, were synthesized by nucleophilic addition of 6 to arylaldehydes, 7a-p. The proposed structures of all the synthesized molecules were elucidated by Infra Red (IR), Proton Nuclear Magnetic Resonance (1H-NMR) and Electron Impact Mass Spectrometry (EIMS) spectral data. Antibacterial activity was evaluated by the principle that microbial growth is in a log phase of growth and so results in increased absorbance of broth medium which is observed.Results: The molecule, 8b, was active against S. aureus and 8c against S. typhi only. The molecule, 8p, was the most active against S. typhi with minimum inhibitory concentration (MIC) value of 10.04 ± 1.25 μM while 8e was active against E. coli with MIC of 9.45 ± 1.00 μM, both relative to the reference standard, ciprofloxacin, which displayed MIC of 9.13 ± 2.00 and 8.90 ± 1.65 μM, respectively.Conclusion: Most of the synthesized molecules exhibit 50 % antibacterial activity relative to the reference.     Synthesis and Evaluation of some New 5-Substituted-1,3,4- oxadiazol-2yl-4-(morpholin-4yl Sulfonyl)benzyl Sulfides as Antibacterial Agent

    Purpose: To synthesise a new series of 5-substituted-1,3,4-Oxadiazol-2yl-4-(morpholin-4yl sulfonyl)benzyl sulfide and evaluate their antibacterial activity.Methods: Different organic acids were converted consecutively into corresponding esters, hydrazides and 5-substituted-1,3,4-Oxadiazol-2-thiols (4a-e). The targets, 6a-e were synthesized by stirring 4a-e with 4-(4-(bromomethyl)phenylsulfonyl) morpholine (5) in the presence of N,N-dimethylformamide (DMF) and sodium hydride (NaH). All the structures were elucidated by modern spectroscopic techniques and screened against bacteria using standard procedure and ciprofloxacin drug as positive control.Results: The yield of the synthesized compounds (4a-e and 6a-e) were moderate (65 - 90 %). Compounds 6a-e had antibacterial activity against Pseudomonas aeruginosa, Bacillis subtilis and Staphylococcus aureus while some had activity against the other bacteria used. One of the compounds, 6b, exhibited significant activity against all the bacterial strains, i.e., S. typhi (-), E. coli (-), K. pneumoniae (-), P. aeruginosa (-), B. subtilis (+) and S. aureus (+) with  MIC (μM) values of 11.01 ± 0.31, 15.37 ± 3.33, 16.11 ± 1.14, 9.70 ± 1.96, 10.01 ± 2.70 and 9.15 ± 0.29, respectively. However, none of the compounds had any inhibitory activity against any bacteria as high as that of ciprofloxacin.Conclusion: Five new compounds with antibacterial activities have been synthesized.     A comparative study of pulsed laser and pulsed TIG welding of Ti-5Al-2.5Sn titanium alloy sheet

    Pulsed Nd:YAG laser beam welding (P-LBW) and pulsed tungsten inert gas (P-TIG) welding were used to prepare full penetration bead-on-plate weldments of 1.6 mm thick Ti-5Al-2.5Sn alpha titanium alloy sheet. The influence of welding phenomenon on the microstructure, micro-hardness, tensile properties, surface and sub-surface residual stress distribution and deformation and distortion of both the weldments were studied. Higher cooling rate in P-LBW resulted in complete α’ martensitic transformation in fusion zone whereas in P-TIG weldment α’ and acicular α was formed within equiaxed β matrix due to lower cooling rate. Hardness in fusion zone of P-LBW was higher than that of the fusion zone of P-TIG weldment due to faster cooling rate in P-LBW. The welded zone in both the weldments showed higher hardness and strength than that of the parent metal since a ductile fracture occurred in the un-welded section during tensile testing. Residual stresses in both P-LBW and P-TIG weldments showed similar trend but the distribution was much narrower in P-LBW due to less width of heat affected zone. P-LBW resulted in more nonuniformity in through thickness stress profile because of greater top to bottom width ratio. Less residual stresses, deformation and distortion and superior mechanical properties in P-LBW made the process more feasible than P-TIG for the welding of Ti-5Al-2.5Sn alloy sheet

    TAT-peptide conjugated repurposing drug against SARS-CoV-2 main protease (3CLpro): potential therapeutic intervention to combat COVID-19

    The Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) that originated in Chinese city of Wuhan has caused around 906,092 deaths and 28,040,853 confirmed cases worldwide (WHO, 11 September, 2020). In a life-threatening situation, where there is no specific and licensed anti-COVID-19 vaccine or medicine available; the repurposed drug might act as a silver bullet. Currently, more than 211 vaccines, 80 antibodies, 31 antiviral drugs, 35 cell-based, 6 RNA-based and 131 other drugs are in clinical trials. It is therefore utter need of the hour to develop an effective drug that can be used for the treatment of COVID-19 before a vaccine can be developed. One of the best-characterized and attractive drug targets among coronaviruses is the main protease (3CL^{pro}). Therefore, the current study focuses on the molecular docking analysis of TAT-peptide^{47–57} (GRKKRRQRRRP)-conjugated repurposed drugs (i.e., lopinavir, ritonavir, favipiravir, and hydroxychloroquine) with SARS-CoV-2 main protease (3CL^{pro} to discover potential efficacy of TAT-peptide (TP) - conjugated repurposing drugs against SARS-CoV-2. The molecular docking results validated that TP-conjugated ritonavir, lopinavir, favipiravir, and hydroxychloroquine have superior and significantly enhanced interactions with the target SARS-CoV-2 main protease. In-silico approach employed in this study suggests that the combination of the drug with TP is an excelling alternative to develop a novel drug for the treatment of SARS-CoV-2 infected patients. The development of TP based delivery of repurposing drugs might be an excellent approach to enhance the efficacy of the existing drugs for the treatment of COVID-19. The predictions from the results obtained provide invaluable information that can be utilized for the choice of candidate drugs for in vitro, in vivo and clinical trials. The outcome from this work prove crucial for exploring and developing novel cost-effective and biocompatible TP conjugated anti-SARS-CoV-2 therapeutic agents in immediate future

    Identifying patients with PTSD utilizing resting-state fMRI data and neural network approach

    Purpose: The primary aim of the study is to identify the existence of the post-traumatic stress disorder (PTSD) in an individual and to detect the dominance level of each affected brain region in PTSD using rs-fMRI data. This will assist the psychiatrists and neurologists to distinguish impartially between PTSD individuals and healthy controls for the brain-based treatment of PTSD. Methods: Twenty-eight individuals (14 with PTSD, 14 healthy controls) were assessed to obtain rs-fMRI data of their six brain regions-of-interest. The rs-fMRI data analyzed by the Artificial Neural Network (ANN), adopting the training-validation-testing approach to classify PTSD and to identify the most affected brain region due to PTSD. The classification accuracy is justified by a variety of different methods and metrics. Results: Three ANN models were established to attain the study’s purpose using the susceptible regions in the right, left, and both hemispheres, and the classification accuracy of ANN models achieved 79%, 93.5%, and 94.5%, respectively. The prediction accuracy even increased in the independent holdout sample using trained models. The developed models are reliable, intellectually attractive and generalize. Additionally, the most dominant region in the PTSD individuals was the left hippocampus and the least was the right hippocampus. Conclusion: The present investigation achieved high classification accuracy and identified the brain regions those contributed most to differentiating PTSD individuals from healthy controls. The results indicated that the left hippocampus is the most affected brain region in PTSD individuals. Therefore, our findings are helpful for practitioners for diagnostic, medication, and therapy of the affected brain regions by knowing the strength of infected regions

    Species and tissue-specificity of prokinetic, laxative and spasmodic effects of Fumaria parviflora

    <p>Abstract</p> <p>Background</p> <p><it>Fumaria parviflora </it>Linn. (<it>Fumariaceae</it>), is a small branched annual herb found in many parts of the world including Saudi Arabia and Pakistan. This study was designed to provide pharmacological basis for the medicinal use of <it>Fumaria parviflora </it>in gut motility disorders.</p> <p>Methods</p> <p>The <it>in-vivo </it>prokinetic and laxative assays were conducted in mice. Isolated intestinal preparations (ileum and jejunum) from different animal species (mouse, guinea-pig and rabbit) were separately suspended in tissue baths containing Tyrode's solution bubbled with carbogen and maintained at 37°C. The spasmogenic responses were recorded using isotonic transducers coupled with PowerLab data acquisition system.</p> <p>Results</p> <p>The aqueous-methanol extract of <it>Fumaria parviflora </it>(Fp.Cr), which tested positive for the presence of alkaloids, saponins, tannins and anthraquinones showed partially atropine-sensitive prokinetic and laxative activities in the <it>in-vivo </it>in mice at 30 and 100 mg/kg. In the <it>in-vitro </it>studies, Fp.Cr (0.01-1 mg/ml) caused a concentration-dependent atropine-sensitive stimulatory effect both in mouse tissues (jejunum and ileum), and rabbit jejunum but had no effect in rabbit ileum. In guinea-pig tissues (ileum and jejunum), the crude extract showed a concentration-dependent stimulatory effect with higher efficacy in ileum and the effect was partially blocked by atropine, indicating the involvement of more than one types of gut-stimulant components (atropine-sensitive and insensitive). This could be a plausible reason for the greater efficacy of Fp.Cr in gut preparations of guinea-pig than in rabbit or mouse.</p> <p>Conclusions</p> <p>This study shows the prokinetic, laxative and spasmodic effects of the plant extract partially mediated through cholinergic pathways with species and tissue-selectivity, and provides a sound rationale for the medicinal use of <it>Fumaria parviflora </it>in gut motility disorders such as, indigestion and constipation. This study also suggests using different species to know better picture of pharmacological profile of the test material.</p

    Antispasmodic and vasodilator activities of Morinda citrifolia root extract are mediated through blockade of voltage dependent calcium channels

    <p>Abstract</p> <p>Background</p> <p><it>Morinda citrifolia </it>(Noni) is an edible plant with wide range of medicinal uses. It occurs exclusively in tropical climate zone from India through Southeast Asia and Australia to Eastern Polynesia and Hawaii. The objective of this study was to explore the possible mode(s) of action for its antispasmodic, vasodilator and cardio-suppressant effects to rationalize its medicinal use in gut and cardiovascular disorders.</p> <p>Methods</p> <p>Isolated tissue preparations such as, rabbit jejunum, rat and rabbit aorta and guinea pig atria were used to test the antispasmodic and cardiovascular relaxant effects and the possible mode of action(s) of the 70% aqueous-ethanolic extract of <it>Morinda citrifolia </it>roots (Mc.Cr).</p> <p>Results</p> <p>The Mc.Cr produced a concentration-dependent relaxation of spontaneous and high K<sup>+ </sup>induced contractions in isolated rabbit jejunum preparations. It also caused right ward shift in the concentration response curves of Ca<sup>++</sup>, similar to that of verapamil. In guinea-pig right atria, Mc.Cr caused inhibition of both atrial force and rate of spontaneous contractions. In rabbit thoracic aortic preparations, Mc.Cr also suppressed contractions induced by phenylephrine (1.0 μM) in normal- Ca<sup>++ </sup>and Ca<sup>++</sup>-free Kerb's solutions and by high K<sup>+</sup>, similar to that of verapamil. In rat thoracic aortic preparations, Mc.Cr also relaxed the phenylephrine (1.0 μM)-induced contractions. The vasodilatory responses were not altered in the presence of L-NAME (0.1 mM) or atropine (1.0 μM) and removal of endothelium.</p> <p>Conclusions</p> <p>These results suggest that the spasmolytic and vasodilator effects of Mc.Cr root extract are mediated possibly through blockade of voltage-dependent calcium channels and release of intracellular calcium, which may explain the medicinal use of <it>Morinda citrifolia </it>in diarrhea and hypertension. However, more detailed studies are required to assess the safety and efficacy of this plant.</p

    Assessment of post-competition peak blood lactate in male and female master swimmers aged 40–79 years and its relationship with swimming performance

    The main purpose of this study was to measure the postcompetition blood lactate concentration ([La]b) in master swimmers of both sexes aged between 40 and 79 years in order to relate it to age and swimming performance. One hundred and eight swimmers participating in the World Master Championships were assessed for [La]b and the average rate of lactate accumulation (La’;mmol l-1 s-1) was calculated. In addition, 77 of them were also tested for anthropometric measures. When the subjects were divided into 10-year age groups, males exhibited higher [La]b than women (factorial ANOVA, P < 0.01) and a steeper decline with ageing than female subjects. Overall, mean values (SD) of [La]b were 10.8 (2.8), 10.3 (2.0), 10.3 (1.9), 8.9 (3.2) mmol l-1 in women, and 14.2 (2.5), 12.4 (2.5), 11.0 (1.6), 8.2 (2.0) mmol l-1 in men for, respectively, 40–49, 50–59, 60–69, 70–79 years’ age groups. When, however, [La]b values were normalised for a ‘‘speed index’’, which takes into account swimming speed as a percentage of world record, these sex-related differences, although still present, were considerably attenuated. Furthermore, the differences in La’ between males and females were larger in the 40–49 age group (0.34 vs 0.20 mmol l-1 s-1 for 50-m distance) than in the 70–79 age group (0.12 vs 0.14 mmol l-1 s-1 for 50-m distance).     Comprehensive assessment of sequence variation within the copy number variable defensin cluster on 8p23 by target enriched in-depth 454 sequencing

    <p>Abstract</p> <p>Background</p> <p>In highly copy number variable (CNV) regions such as the human defensin gene locus, comprehensive assessment of sequence variations is challenging. PCR approaches are practically restricted to tiny fractions, and next-generation sequencing (NGS) approaches of whole individual genomes e.g. by the 1000 Genomes Project is confined by an affordable sequence depth. Combining target enrichment with NGS may represent a feasible approach.</p> <p>Results</p> <p>As a proof of principle, we enriched a ~850 kb section comprising the CNV defensin gene cluster DEFB, the invariable DEFA part and 11 control regions from two genomes by sequence capture and sequenced it by 454 technology. 6,651 differences to the human reference genome were found. Comparison to HapMap genotypes revealed sensitivities and specificities in the range of 94% to 99% for the identification of variations.</p> <p>Using error probabilities for rigorous filtering revealed 2,886 unique single nucleotide variations (SNVs) including 358 putative novel ones. DEFB CN determinations by haplotype ratios were in agreement with alternative methods.</p> <p>Conclusion</p> <p>Although currently labor extensive and having high costs, target enriched NGS provides a powerful tool for the comprehensive assessment of SNVs in highly polymorphic CNV regions of individual genomes. Furthermore, it reveals considerable amounts of putative novel variations and simultaneously allows CN estimation.</p
