    Recruitment and growth of Megabalanus azoricus (Pilsbry, 1916) on artificial substrates: first steps towards commercial culture in the Azores

    This paper reports the results of preliminary research evaluating the potential of farming Megabalanus azoricus. From March 2006 to September 2006, artificial substrates were immersed bi-monthly in a shallow water bank located in the channel separating the islands of Faial and Pico, Azores. They were monitored monthly for a period of 12 months by scuba-diving. M. azoricus rapidly colonised the immersed substrates, establishing itself as the dominant organism. Even though recruitment was observed year round, there was a peak in recruitment between June and October. The substrate immersed for the longest period (15 months) presented final densities of 1109 ind. m-2, from which 20.2% were of edible size. Growth was monitored for 18 isolated barnacles for 8 months. Growth rate proved to be high (base diameter; 0.08 ± 0.02 mm day-1; mean ± SD), with individuals reaching diameters of 21.35 ± 6.2 mm after 8 months. Considering this growth rate, it was estimated that after a further 9 months, over 80% of the total densities (on the oldest substrate ) would be harvestable, thus suggesting a production cycle of 17 to 24 months. Although much further research is needed, the species presents all characteristics of a potential candidate to initiate the aquaculture industry in the Azores

    Melatonin MT1 receptors as a target for the psychopharmacology of bipolar disorder: a translational study

    The treatment of bipolar disorder (BD) still remains a challenge. Melatonin (MLT), acting through its two receptors MT1 and MT2, plays a key role in regulating circadian rhythms which are dysfunctional in BD. Using a translational approach, we examined the implication and potential of MT1 receptors in the pathophysiology and psychopharmacology of BD. We employed a murine model of the manic phase of BD (Clock mutant (ClockΔ19) mice) to study the activation of MT1 receptors by UCM871, a selective partial agonist, in behavioral pharmacology tests and in-vivo electrophysiology. We then performed a high-resolution Nuclear Magnetic Resonance study on isolated membranes to characterize the molecular mechanism of interaction of UCM871. Finally, in a cohort of BD patients, we investigated the link between clinical measures of BD and genetic variants located in the MT1 receptor and CLOCK genes. We demonstrated that: 1) UCM871 can revert behavioral and electrophysiological abnormalities of ClockΔ19 mice; 2) UCM871 promotes the activation state of MT1 receptors; 3) there is a significant association between the number of severe manic episodes and MLT levels, depending on the genetic configuration of the MT1 rs2165666 variant.     Peri-operative red blood cell transfusion in neonates and infants: NEonate and Children audiT of Anaesthesia pRactice IN Europe: A prospective European multicentre observational study

    BACKGROUND: Little is known about current clinical practice concerning peri-operative red blood cell transfusion in neonates and small infants. Guidelines suggest transfusions based on haemoglobin thresholds ranging from 8.5 to 12 g dl-1, distinguishing between children from birth to day 7 (week 1), from day 8 to day 14 (week 2) or from day 15 (≄week 3) onwards. OBJECTIVE: To observe peri-operative red blood cell transfusion practice according to guidelines in relation to patient outcome. DESIGN: A multicentre observational study. SETTING: The NEonate-Children sTudy of Anaesthesia pRactice IN Europe (NECTARINE) trial recruited patients up to 60 weeks' postmenstrual age undergoing anaesthesia for surgical or diagnostic procedures from 165 centres in 31 European countries between March 2016 and January 2017. PATIENTS: The data included 5609 patients undergoing 6542 procedures. Inclusion criteria was a peri-operative red blood cell transfusion. MAIN OUTCOME MEASURES: The primary endpoint was the haemoglobin level triggering a transfusion for neonates in week 1, week 2 and week 3. Secondary endpoints were transfusion volumes, 'delta haemoglobin' (preprocedure - transfusion-triggering) and 30-day and 90-day morbidity and mortality. RESULTS: Peri-operative red blood cell transfusions were recorded during 447 procedures (6.9%). The median haemoglobin levels triggering a transfusion were 9.6 [IQR 8.7 to 10.9] g dl-1 for neonates in week 1, 9.6 [7.7 to 10.4] g dl-1 in week 2 and 8.0 [7.3 to 9.0] g dl-1 in week 3. The median transfusion volume was 17.1 [11.1 to 26.4] ml kg-1 with a median delta haemoglobin of 1.8 [0.0 to 3.6] g dl-1. Thirty-day morbidity was 47.8% with an overall mortality of 11.3%. CONCLUSIONS: Results indicate lower transfusion-triggering haemoglobin thresholds in clinical practice than suggested by current guidelines. The high morbidity and mortality of this NECTARINE sub-cohort calls for investigative action and evidence-based guidelines addressing peri-operative red blood cell transfusions strategies.     Genetic diversity and structure of Megabalanus azoricus in the Azores: Implications for aquaculture management

    Megabalanus azoricus giant barnacles are the most traditional seafood of the Azores archipelago (NE Atlantic). This valuable commercial species has been highly exploited in the past and it is considered one of the key species for the development of aquaculture in the region. Despite the importance for conservation and aquaculture there is still a lack of basic information about M. azoricus genetic diversity and population structure. Here we used seven microsatellites markers to analyse 300 samples collected at six out of nine islands of the Azores archipelago, including also different locations from a single island, to provide information on the scale of genetic diversity and population structure of this species. Parameters like heterozygosity, allelic richness and effective number of alleles indicated a high genetic diversity and variability among islands. Pairwise comparisons and PCoA analysis on FST and Jost's DEST showed significant and evident differentiation among sampling locations. Additionally, AMOVA allocates a small (6.02%) but statistically significant portion of the variance to the among Island level revealing also a weak resolution (1.87%) at finer scale. Additionally Monte Carlo resampling methods indicated the most likely sources of the recruits were the local or adjacent populations. Genetic risks associated with the giant barnacle potential production scheme should be taken into account in a future management plan delimiting, as precautionary measure, this culture at a single island or at groups of islands here identified. Moreover a monitoring strategy should be implemented with the aim to evaluate possible changes in genetic parameters of native populations

    Developing a Sustainable Aquaculture Industry in the Azores : Proceedings of the International Workshop

    International Workshop, Horta 2-5-June 2008.The first workshop on developing a sustainable aquaculture industry in the Azores was organised as a joint venture between the Institute of Marine Research at the Department of Oceanography and Fisheries of the University of the Azores (IMAR/DOP) and the private enterprise seaExpert. The aim of the meeting was to determine the necessary steps to establish a sustainable Aquaculture Industry in the Azores and to identify priorities for further aquaculture research in the Region. Experts were invited from diverse areas, bringing specialized knowledge and understanding of a broad variety of factors involved in setting up an aquaculture industry. Speakers from across the world shared experience of aquaculture projects from other island ecosystems including: Madeira, the Canaries, Malta, Cyprus and Hawaii. Representatives of official institutions such as the Food and Agricultural Organisation of the United Nations (FAO), the National Oceanic and Atmosphere Administration (NOAA) of the U.S.A., and Centro TecnolĂłgico del Mar of Spain (FundaciĂłn Cetmar) offered advice regarding planning, management and policy issues. A specialist from an independent company, Akvaplan-niva, with vast experience in Aquaculture development in Europe was also invited to offer suggestions and advice to local government representatives and stakeholders. The first part of the meeting was composed of a series of lectures: issuing the status of Aquaculture Worldwide and on the Portuguese scale; regulatory measures for aquaculture in the EU; trade, marketing, quality and safety of aquaculture products; oceanic and land-based production systems; and partitioned systems. Attention was also given to specific species that could be potentially produced in the Azores Region. A series of discussion forums were launched during the second part of the meeting. These were designed to determine the specific requirements and steps needed in order to launch an aquaculture industry in the Azores.     Long-term outcomes of extracorporeal shock wave therapy for acute Peyronie's disease: a 10-year retrospective analysis

    : The aim of this paper was to describe the long-term outcomes of extracorporeal shock wave therapy (ESWT) in patients with acute Peyronie'disease (PD). An observational retrospective study was conducted in men with acute PD who underwent ESWT between 2009-2013 at a single institution. ESWT protocol consisted of 1 session (3000 shock waves, 0.10-0.25 mJ/mm^2, 4-6 Hz) per week for 4 weeks. Penile pain was chosen as the primary outcome. Penile curvature angle, erectile function, and satisfaction with ESWT were selected as secondary long-term outcomes. A total of 194 patients were included. The mean follow-up duration after ESWT was 125.6 months. Mean penile curvature worsened significantly at 3 months (18.3 vs. 21.5 degrees; p = 0.023) and 12 months (21.5 vs. 28.6 degrees; p = 0.001) and stabilized over the long-term (28.6 vs. 28.8 degrees; p = 0.335). Mean penile pain improved significantly at 3 months (6.5 vs. 3.1 points; p < 0.001) and 12 months (3.1 vs. 1.0 points; p = 0.001), remaining stable over time (1.0 vs. 1.0 points; p = 0.074). The mean five-item version of the International Index of Erectile Function (IIEF-5) increased significantly at 3 months (14.5 vs. 17.9 points; p = 0.001), remaining stable at 12 months (17.9 vs. 18.5 points; p = 0.082), and deteriorating in the long-term (18.5 vs. 15.8 points; p = 0.003). A high satisfaction rate with ESWT was recorded at 3 months (92.3%), remaining similar at 12 months (91.2%) and over the long-term (90.2%). No new acute phase and low rate of PD surgery (4.1%) were recorded in the long-term analysis. In patients with acute PD, ESWT seems to be associated with early and persistent relief of penile pain, transient improvement in erectile function, no significant effect on penile curvature, and a high rate of patient satisfaction constant over time
