    Polymerized Laminin-332 Matrix Supports Rapid and Tight Adhesion of Keratinocytes, Suppressing Cell Migration

    Laminin-332 (α3ß3γ2) (Lm332) supports the stable anchoring of basal keratinocytes to the epidermal basement membrane, while it functions as a motility factor for wound healing and cancer invasion. To understand these contrasting activities of Lm332, we investigated Lm332 matrices deposited by normal human keratinocytes and other Lm332-expressing cell lines. All types of the cells efficiently deposited Lm332 on the culture plates in specific patterns. On the contrary, laminins containing laminin ß1 and/or γ1 chains, such as Lm511 and Lm311, were not deposited on the culture plates even if secreted into culture medium. The Lm332 deposition was not inhibited by function-blocking antibodies to the α3 and α6 integrins but was inhibited by sodium selenate, suggesting that sulfated glycosaminoglycans on cell surface, e.g. heparan sulfate proteoglycans, might be involved in the process. HEK293 cells overexpressing exogenous Lm332 (Lm332-HEK) almost exclusively deposited Lm332 on the plates. The deposited Lm332 matrix showed a mesh-like network structure as analyzed by electron microscopy, suggesting that Lm332 was highly polymerized. When biological activity was analyzed, the Lm332 matrix rather suppressed the migration of keratinocytes as compared with purified Lm332, which highly promoted the cell migration. The Lm332 matrix supported adhesion of keratinocytes much more strongly and stably than purified Lm332. Integrin α3ß1 bound to the Lm332 matrix at a three times higher level than purified Lm332. Normal keratinocytes prominently showed integrin α6ß4-containing, hemidesmosome-like structures on the Lm332 matrix but not on the purified one. These results indicate that the polymerized Lm332 matrix supports stable cell adhesion by interacting with both integrin α6ß4 and α3ß1, whereas unassembled soluble Lm332 supports cell migration

    Application of ozone ultrafine bubble water for cleaning and sterilizing the dialysis circuit

    The dialysis circuit was washed by the ultrafine bubble (UFB) water and ozone UFB water and sodium hypochlorite aqueous solution. The effect of washing time, ozone concentration of UFB water, influence of UFB water on artificial dialysis equipment, etc. were investigated. The cleaning and sterilizing effect of ozone UFB water passed through the filter for artificial dialysis was verified. It was confirmed that there were no colony bacteria in the circuit which was washed by the ozone UFB water for 1 hour. Ozone UFB water has the same bactericidal effect as sodium hypochlorite. As an alternative of sodium hypochlorite, we are expecting its application to the cleaning and sterilization of ozone UFB water for dialysis circuit

    This is a study conducted by our research group about the psychological background of local people in Manado,Indonesia. Interviews were made to a group of locals concerning their approach to death and consequent state of mind focused on their religious beliefs on Obon festival,conversation with spirits,reincarnation and the influence of holy men. We recognized the fantasy they believed that spirit comes back from afterworld in the ceremony. In addition,we understood the event that spirit returns from afterworld once a year and“offering” is common sense. However,these custom had difference depended on regions and their social rank. As a result,these local people were difficult to share the common sense about the event. Reporting to spirit and conversation to spirit were common events in whole regions in Indonesia,but the quality might be different.     Effects of long-term cigarette smoke exposure on bone metabolism, structure, and quality in a mouse model of emphysema

    Smoking is a common risk factor for both chronic obstructive pulmonary disease (COPD) and osteoporosis. In patients with COPD, severe emphysema is a risk factor for vertebral fracture; however, the effects of smoking or emphysema on bone health remain largely unknown. We report bone deterioration in a mouse model of emphysema induced by nose-only cigarette smoke (CS) exposure. Unexpectedly, short-term exposure for 4-weeks decreased bone turnover and increased bone volume in mice. However, prolonged exposure for 20- and 40-weeks reversed the effects from suppression to promotion of bone resorption. This long-term CS exposure increased osteoclast number and impaired bone growth, while it increased bone volume. Strikingly, long-term CS exposure deteriorated bone quality of the lumbar vertebrae as illustrated by disorientation of collagen fibers and the biological apatite c-axis. This animal model may provide a better understanding of the mechanisms underlying the deterioration of bone quality in pulmonary emphysema caused by smoking.Effects of long-term cigarette smoke exposure on bone metabolism, structure, and quality in a mouse model of emphysema.     A novel locally operated master-slave robot system for single-incision laparoscopic surgery

    Purpose: Single-incision laparoscopic surgery (SILS) provides more cosmetic benefits than conventional laparoscopic surgery but presents operational difficulties. To overcome this technical problem, we have developed a locally operated master-slave robot system that provides operability and a visual field similar to conventional laparoscopic surgery. Material and methods: A surgeon grasps the master device with the left hand, which is placed above the abdominal wall, and holds a normal instrument with the right hand. A laparoscope, a slave robot, and the right-sided instrument are inserted through one incision. The slave robot is bent in the body cavity and its length, pose, and tip angle are changed by manipulating the master device; thus the surgeon has almost the same operability as with normal laparoscopic surgery. To evaluate our proposed system, we conducted a basic task and an ex vivo experiment. Results: In basic task experiments, the average object-passing task time was 9.50 sec (SILS cross), 22.25 sec (SILS parallel), and 7.23 sec (proposed SILS). The average number of instrument collisions was 3.67 (SILS cross), 14 (SILS parallel), and 0.33 (proposed SILS). In the ex vivo experiment, we confirmed the applicability of our system for single-port laparoscopic cholecystectomy.     Enhanced stability of hippocampal place representation caused by reduced magnesium block of NMDA receptors in the dentate gyrus

    BACKGROUND: Voltage-dependent block of the NMDA receptor by Mg(2+) is thought to be central to the unique involvement of this receptor in higher brain functions. However, the in vivo role of the Mg(2+) block in the mammalian brain has not yet been investigated, because brain-wide loss of the Mg(2+) block causes perinatal lethality. In this study, we used a brain-region specific knock-in mouse expressing an NMDA receptor that is defective for the Mg(2+) block in order to test its role in neural information processing. RESULTS: We devised a method to induce a single amino acid substitution (N595Q) in the GluN2A subunit of the NMDA receptor, specifically in the hippocampal dentate gyrus in mice. This mutation reduced the Mg(2+) block at the medial perforant path–granule cell synapse and facilitated synaptic potentiation induced by high-frequency stimulation. The mutants had more stable hippocampal place fields in the CA1 than the controls did, and place representation showed lower sensitivity to visual differences. In addition, behavioral tests revealed that the mutants had a spatial working memory deficit. CONCLUSIONS: These results suggest that the Mg(2+) block in the dentate gyrus regulates hippocampal spatial information processing by attenuating activity-dependent synaptic potentiation in the dentate gyrus

No use, distribution or reproduction is permitted which does not comply with these terms.Chimeric rodent malaria parasites with the endogenous circumsporozoite protein (csp) gene replaced with csp from the human parasites Plasmodium falciparum (Pf) and P. vivax (Pv) are used in preclinical evaluation of CSP vaccines. Chimeric rodent parasites expressing PfCSP have also been assessed as whole sporozoite (WSP) vaccines. Comparable chimeric P. falciparum parasites expressing CSP of P. vivax could be used both for clinical evaluation of vaccines targeting PvCSP in controlled human P. falciparum infections and in WSP vaccines targeting P. vivax and P. falciparum. We generated chimeric P. falciparum parasites expressing both PfCSP and PvCSP. These Pf-PvCSP parasites produced sporozoite comparable to wild type P. falciparum parasites and expressed PfCSP and PvCSP on the sporozoite surface. Pf-PvCSP sporozoites infected human hepatocytes and induced antibodies to the repeats of both PfCSP and PvCSP after immunization of mice.     プールの健康教室に参加することによる心身への影響―低身体活動高齢女性の実態調査―

    目的:群馬県内のAプールが実施している健康教室に参加することが心身に及ぼす影響を明らかにすることを目的とした. 方法:研究参加に同意した65歳以上の低身体活動の女性11名を対象者とした.教室参加前に年齢,健康状態,気分を,教室当日に運動前,中,後の血圧・脈拍,主観的運動強度を,教室終了後に気分を調査した.任意で体力測定を行った. 結果:体力測定の結果は,握力,上体起こし,長座体前屈,開眼片足立ち,10m障害物歩行の全てで全国平均よりも低かった.気分は,教室参加後に緊張・不安が有意に低下した.収縮期血圧は運動後では運動前よりも有意に上昇し,脈拍は運動前よりも有意に低下した.主観的運動強度は運動前よりも有意に上昇した. 結論:対象者は,肥満,筋力低下傾向にあるが,集団で実施する水中運動の強度は適度であった.健康教室により,緊張・不安が改善し,適度な運動強度においても血圧や脈拍の有意な変動が認められた.Objectives: The purpose of this study was to investigate the effects of a health class offered at a swimming pool. Methods: The subjects were 11 women aged 65 years or older who had low active mass. We investigated age, health condition, and mood before the health class. On the day of the health class, we measured blood pressure, pulse, and perceived exertion before, during, and after aquatic exercise. We then investigated mood after the health class. We also offered an optional physical fitness test to the subjects. Results: Scores for grip strength, sit-ups, toe touching, one-leg standing with eyes open, and 10-m obstacle walking test results were below healthy levels. The mood test showed that tension and anxiety were improved. Systolic blood pressure was significantly higher, pulse was significantly lower, and the rate of perceived exertion was significantly higher after exercise than before. Conclusions: The subjects were obese and had muscle weakness, but the exercise was of moderate intensity. Tension and anxiety improved after the health class. Significant changes in blood pressure and pulse were found after exercise.原
