Na Universidade Federal do Paraná, UFPR, foi desenvolvido o projeto SAGU (Sistema de Apoio ao Gerenciamento Universitário), que é um sistema de suporte à decisão, que tem por objetivo prover os administradores daquela universidade com informações que auxiliarão na direção de seus objetivos. O principal componente deste sistema é um data warehouse. Neste artigo nós apresentamos a especificação formal (utilizando Semântica de Ações) do componente Integrador do data warehouse no contexto do projeto SAGU. A especificação obtida foi utilizada para auxiliar no processo de construção do data warehouse e pode ser considerada parte da documentação do projeto de um data warehouse real

    For some, the transnationalization of political action and communicative space in the European Union heralds an emergent cosmopolitan order. Need that be so? There are supranational institutions in the EU as well as transnational political and cultural spaces and cross-border communicative flows. However, the Union's member states remain key controllers of citizenship rights and purveyors of collective identities. And for many purposes they still maintain strongly bounded national public spheres. Because the EU's overall character as a polity remains unresolved, this has consequences for the organization of communicative spaces. The EU is a field of tensions and contradictions that is inescapably rooted in institutional realities. Wishful thinking about cosmopolitanism can get in the way of clear analysis

    X-ray diffraction reveals the intrinsic difference in the physical properties of membrane and soluble proteins.

    Membrane proteins are distinguished from soluble proteins by their insertion into biological membranes. This insertion is achieved via a noticeable arrangement of hydrophobic amino acids that are exposed at the surface of the protein, and renders the interaction with the aliphatic tails of lipids more energetically favorable. This important difference between these two categories of proteins is the source of the need for a specific handling of membrane proteins, which transpired in the creation of new tools for their recombinant expression, purification and even crystallization. Following this line, we show here that crystals of membrane proteins display systematically higher diffraction anisotropy than those of soluble proteins. This phenomenon dramatically hampers structure solution and refinement, and has a strong impact on the quality of electron-density maps. A farther search for origins of this phenomenon showed that the type of crystallization, and thus the crystal packing, has no impact on anisotropy, nor does the nature or function of the membrane protein. Membrane proteins fully embedded within the membrane display equal anisotropy compared to the ones with extra membranous domains or fusions with soluble proteins. Overall, these results overturn common beliefs and call for a specific handling of their diffraction data

    Distribution of knock-down resistance mutations in Anopheles gambiae molecular forms in west and west-central Africa

    <p>Abstract</p> <p>Background</p> <p><it>Knock-down </it>resistance (<it>kdr</it>) to DDT and pyrethroids in the major Afrotropical vector species, <it>Anopheles gambiae </it>sensu stricto, is associated with two alternative point mutations at amino acid position 1014 of the voltage-gated sodium channel gene, resulting in either a leucine-phenylalanine (L1014F), or a leucine-serine (L1014S) substitution. In <it>An. gambiae </it>S-form populations, the former mutation appears to be widespread in west Africa and has been recently reported from Uganda, while the latter, originally recorded in Kenya, has been recently found in Gabon, Cameroon and Equatorial Guinea. In M-form populations surveyed to date, only the L1014F mutation has been found, although less widespread and at lower frequencies than in sympatric S-form populations.</p> <p>Methods</p> <p><it>Anopheles gambiae </it>M- and S-form specimens from 19 sites from 11 west and west-central African countries were identified to molecular form and genotyped at the <it>kdr </it>locus either by Hot Oligonucleotide Ligation Assay (HOLA) or allele-specific PCR (AS-PCR).</p> <p>Results</p> <p>The <it>kdr </it>genotype was determined for about 1,000 <it>An. gambiae </it>specimens. The L1014F allele was found at frequencies ranging from 6% to 100% in all S-form samples (N = 628), with the exception of two samples from Angola, where it was absent, and coexisted with the L1014S allele in samples from Cameroon, Gabon and north-western Angola. The L1014F allele was present in M-form samples (N = 354) from Benin, Nigeria, and Cameroon, where both M- and S-forms were sympatric.</p> <p>Conclusion</p> <p>The results represent the most comprehensive effort to analyse the overall distribution of the L1014F and L1014S mutations in <it>An. gambiae </it>molecular forms, and will serve as baseline data for resistance monitoring. The overall picture shows that the emergence and spread of <it>kdr </it>alleles in <it>An. gambiae </it>is a dynamic process and that there is marked intra- and inter-form heterogeneity in resistance allele frequencies. Further studies are needed to determine: i) the importance of selection pressure exerted by both agricultural and public health use of pyrethroid insecticides, ii) the phenotypic effects, particularly when the two mutations co-occur; and iii) the epidemiological importance of <it>kdr </it>for both pyrethroid- and DDT-based malaria control operations, particularly if/when the two insecticides are to be used in concert.</p

    Development of MKIDs in the Optical and Near-infrared Bands for SPIAKID

    SpectroPhotometric Imaging in Astronomy with Kinetic Inductance Detectors (SPIAKID) aims at designing, building, and deploying on the sky a spectrophotometric imager based on microwave kinetic inductance detectors (MKIDs) in the optical and near-infrared bands. MKIDs show a fast response and the ability to resolve photon energy compared to the conventional Charge-coupled Devices (CCDs). In this paper, we present the design and simulation of the MKID arrays for SPIAKID. The detectors consist of four arrays with each array of 20,000 lumped-element pixels, and each array will be read with 10 readout lines. %The array is designed to have resonances between 4-8GHz with a frequency spacing of 2 MHz and a coupling quality factor (Qc) of about 50000. The meander material of the resonators is trilayer TiN/Ti/TiN to have better uniformity of the critical temperature across the array. We also present the measurement result for a test array with 30×3030\times30 pixels which is a subset of the designed 2000-pixel array to verify the design and fabrication. The current measured best energy resolving power R=E/ΔER = E/\Delta E is 2.4 at λ=405 \lambda = 405~nm and the current medium R is around 1.7.     Role of Anopheles (Cellia) rufipes (Gough, 1910) and other local anophelines in human malaria transmission in the northern savannah of Cameroon: a crosssectional survey

    Background As part of a study to determine the impact of insecticide resistance on the effectiveness of longlasting insecticide treated nets (LLINs) in the north of Cameroon, the unexpectedly high density and anthropophilic behaviour of Anopheles rufipes lead us to investigate this species bionomics and role in human malaria parasite transmission. Methods For four consecutive years (2011–2014), annual cross-sectional sampling of adult mosquitoes was conducted during the peak malaria season (September-October) in three health districts in northern Cameroon. Mosquitoes sampled by human landing catch and pyrethrum spray catch methods were morphologically identified, their ovaries dissected for parity determination and Anopheles gambiae siblings were identified by molecular assay. Infection with P. falciparum and blood meal source in residual fauna of indoor resting anopheline mosquitoes were determined by enzyme-linked-immunosorbent assays. Results Anopheles gambiae (sensu lato) (s.l.) comprised 18.4% of mosquitoes collected with An. arabiensis representing 66.27% of the sibling species. The proportion of An. rufipes (2.7%) collected was high with a humanbiting rate ranging between 0.441 and 11.083 bites/person/night (b/p/n) and an anthropophagic rate of 15.36%. Although overall the members of An. gambiae complex were responsible for most of the transmission with entomological inoculation rates (EIR) reaching 1.221 infective bites/person/night (ib/p/n), An. arabiensis and An.coluzzii were the most implicated. The roles of An. funestus, An. pharoensis and An. paludis were minor. Plasmodium falciparum circumsporozoite protein rate in Anopheles rufipes varied from 0.6 to 5.7% with EIR values between 0.010 and 0.481 ib/p/n Conclusions The study highlights the epidemiological role of An. rufipes alongside the members of the An.gambiae complex, and several other sympatric species in human malaria transmission during the wet season in northern Cameroon. For the first time in Cameroon, An. rufipes has been shown to be an important local malaria vector, emphasising the need to review the malaria entomological profile across the country as pre-requisite to effective vector management strategies

    The Rat Grimace Scale: A partially automated method for quantifying pain in the laboratory rat via facial expressions

    We recently demonstrated the utility of quantifying spontaneous pain in mice via the blinded coding of facial expressions. As the majority of preclinical pain research is in fact performed in the laboratory rat, we attempted to modify the scale for use in this species. We present herein the Rat Grimace Scale, and show its reliability, accuracy, and ability to quantify the time course of spontaneous pain in the intraplantar complete Freund's adjuvant, intraarticular kaolin-carrageenan, and laparotomy (post-operative pain) assays. The scale's ability to demonstrate the dose-dependent analgesic efficacy of morphine is also shown. In addition, we have developed software, Rodent Face Finder®, which successfully automates the most labor-intensive step in the process. Given the known mechanistic dissociations between spontaneous and evoked pain, and the primacy of the former as a clinical problem, we believe that widespread adoption of spontaneous pain measures such as the Rat Grimace Scale might lead to more successful translation of basic science findings into clinical application

    Radiological markers of neurological manifestations of post-acute sequelae of SARS-CoV-2 infection: a mini-review

    The neurological impact of COVID-19 is a rising concern among medical professionals, as patients continue to experience symptoms long after their recovery. This condition, known as neurological post-acute sequelae of COVID-19 (Neuro-PASC), can last for more than 12 weeks and includes symptoms such as attention disorders, brain fog, fatigue, and memory loss. However, researchers and health professionals face significant challenges in understanding how COVID-19 affects the brain, limiting the development of effective prevention and treatment strategies. In this mini-review, we provide readers with up-to-date information on the imaging techniques currently available for measuring the neurological impact of post-SARS-CoV-2 infection. Our search of PubMed and Google Scholar databases yielded 38 articles on various brain imaging techniques, including structural MRI (magnetic resonance imaging), functional MRI, diffusion MRI, susceptibility-weighted imaging, SPECT (single-photon emission computed tomography) imaging, and PET (positron emission tomography) imaging. We also discuss the optimal usage, limitations, and potential benefits of these techniques. Our findings show that various cerebral imaging techniques have been evaluated to identify a reliable marker for Neuro-PASC. For instance, 18F-FDG-PET/CT and functional MRI have demonstrated hypometabolism in cerebral regions that are directly linked to patient symptoms. Structural MRI studies have revealed different findings, such as infarcts, white matter atrophy, and changes in gray matter volumes. One SPECT imaging study noted frontal lobe hypometabolism, while diffusion MRI showed increased diffusivity in the limbic and olfactory cortical systems. The sequence SWI showed abnormalities primarily in white matter near the gray-white matter junction. A study on 18F-amyloid PET/CT found amyloid lesions in frontal and anterior cingulate cortex areas, and a study on arterial spin labeling (ASL) found hypoperfusion primarily in the frontal lobe. While accessibility and cost limit the widespread use of 18F-FDG-PET/CT scans and functional MRI, they seem to be the most promising techniques. SPECT, SWI sequence, and 18F-amyloid PET/CT require further investigation. Nevertheless, imaging remains a reliable tool for diagnosing Neuro-PASC and monitoring recovery
