    Net gain: Low-cost, trawl-associated eDNA samplers upscale ecological assessment of marine demersal communities

    Marine biodiversity stewardship requires costly and time-consuming capture-based monitoring techniques, which limit our understanding of the distribution and status of marine populations. Here, we reconstruct catch and demersal community compo- sition in a set of 24 fishing sites in the central Tyrrhenian Sea by gathering environ- mental DNA (eDNA) aboard commercial bottom-trawl fishing vessels. We collected genetic material from two sources: the water draining from the net after the end of hauling operations (ā€œslushā€), and custom-made rolls of gauze tied to a hollow perfo- rated sphere placed inside the fishing net (ā€œmetaprobeā€). Species inventories were generated using a combination of fish-specific (Tele02 12S) and universal metazoan (COI) molecular markers. DNA metabarcoding data recovered over 90% of the caught taxa and accurately reconstructed the overall structure of the assemblages of the examined sites, reflecting expected differences linked to major drivers of community structure in Mediterranean demersal ecosystems, such as depth, distance from the coast, and fishing effort. eDNA also returned a ā€œbiodiversity bonusā€ mostly consisting of pelagic species not catchable by bottom trawl but present in the surrounding en- vironment. Overall, the ā€œmetaprobeā€ gauzes showed a greater biodiversity detection power as compared to ā€œslushā€ water, both qualitatively and quantitatively, strengthen- ing the idea that these low-cost sampling devices can play a major role in upscaling the gathering of data on both catch composition and the broader ecological charac- teristics of marine communities sustaining trawling activities. This approach has the potential to drastically expand the reach of ecological monitoring, whereby fishing vessels operating across the oceans may serve as opportunistic scientific platforms to increase the strength and granularity of marine biodiversity data

    Neoadjuvant eribulin mesylate following anthracycline and taxane in triple negative breast cancer: Results from the HOPE study

    Background Eribulin mesylate (E) is indicated for metastatic breast cancer patients previously treated with anthracycline and taxane. We argued that E could also benefit patients eligible for neoadjuvant chemotherapy. Methods Patients with primary triple negative breast cancer 2 cm received doxorubicin 60 mg/m2 and paclitaxel 200 mg/m2 x 4 cycles (AT) followed by E 1.4 mg/m2 x 4 cycles. Primary endpoint was pathological complete response (pCR) rate; secondary and explorative endpoints included clinical/metabolic response rates and safety, and biomarker analysis, respectively. Using a two-stage Simon design, 43 patients were to be included provided that 4 of 13 patients had achieved pCR in the first stage of the study. Results In stage I of the study 13 women were enrolled, median age 43 years, tumor size 2ā€“5 cm in 9/13 (69%), positive nodal status in 8/13 (61%). Main grade 3 adverse event was neutropenia (related to AT and E in 4 and 2 cases, respectively). AT followed by E induced clinical complete + partial responses in 11/13 patients (85%), pCR in 3/13 (23%). Median measurements of maximum standardized uptake value (SUVmax) resulted 13, 3, and 1.9 at baseline, after AT and E, respectively. Complete metabolic response (CMR) occurred after AT and after E in 2 and 3 cases, respectively. Notably, 2 of the 5 (40%) patients with CMR achieved pCR at surgery. Immunostaining of paired pre-/post-treatment tumor specimens showed a reduction of Ī²-catenin, CyclinD1, Zeb-1, and c-myc expression, in the absence of N-cadherin modulation. The study was interrupted at stage I due to the lack of the required patients with pCR. Conclusions Despite the early study closure, preoperative E following AT showed clinical and biological activity in triple negative breast cancer patients. Furthermore, the modulation of Ī²-catenin pathway core proteins, supposedly outside the domain of epithelialā€“mesenchymal transition, claims for further investigation.     A Bioinspired Plasmonic Nanocomposite Actuator Sunlight-Driven by a Photothermal-Hygroscopic Effect for Sustainable Soft Robotics

    Combined photothermal-hygroscopic effects enable novel materials actuation strategies based on renewable and sustainable energy sources such as sunlight. Plasmonic nanoparticles have gained considerable interest as photothermal agents, however, the employment in sunlight-driven photothermal-hygroscopic actuators is still bounded, mainly due to the limited absorbance once integrated into nanocomposite actuators and the restricted plasmonic peaks amplitude (compared to the solar spectrum). Herein, the design and fabrication of an AgNPs-based plasmonic photothermal-hygroscopic actuator integrated with printed cellulose tracks are reported (bioinspired to Geraniaceae seeds structures). The nanocomposite is actuated by sunlight power density (i.e., 1 Sun = 100 mW cmāˆ’2). The plasmonic AgNPs are in situ synthesized on the PDMS surface through a one-step and efficient fluoride-assisted synthesis (surface coverage ā‰ˆ40%). The nanocomposite has a broadband absorbance in the VIS range (>1) and a Photothermal Conversion Efficiency ā‰ˆ40%. The actuator is designed exploiting a mechanical model that predicted the curvature and forces, featuring a ā‰ˆ6.8 Ā± 0.3 s response time, associated with a ā‰ˆ43% change in curvature and a 0.76 Ā± 0.02 mN force under 1 Sun irradiation. The plasmonic nanocomposite actuator can be used for multiple tasks, as hinted through illustrative soft robotics demonstrators, thus fostering a bioinspired approach to developing embodied energy systems driven by sunlight

    The geometrical nature of optical resonances : from a sphere to fused dimer nanoparticles

    We study the electromagnetic response of smooth gold nanoparticles with shapes varying from a single sphere to two ellipsoids joined smoothly at their vertices. We show that the plasmonic resonance visible in the extinction and absorption cross sections shifts to longer wavelengths and eventually disappears as the mid-plane waist of the composite particle becomes narrower. This process corresponds to an increase of the numbers of internal and scattering modes that are mainly confined to the surface and coupled to the incident field. These modes strongly affect the near field, and therefore are of great importance in surface spectroscopy, but are almost undetectable in the far field
