    Seismic resilience timber connection-adoption of shape memory alloy tubes as dowels

    This study investigates a novel timber dowel-type connection system using superelastic shape memory alloy (SMA) bar and tubes as dowels, in order to provide self-centering effect. Double-shear connections with SMA and mild steel dowels were tested under dynamic loadings at different displacement levels. The results showed that SMA dowel-type connections have good self-centering behaviours and can mitigate the residual deformation effectively compared with steel dowel-type connections after excessive deformation; although the steel dowel-type connections present higher strength. These tests reveal that the connection with tube dowels show higher equivalent viscous damping ratio than those use solid bar as tube would allow larger deformation to dissipate energy. To demonstrate application of the benefit of this system, an analytical model of a 3-storey timber framed structure was built for parametric study. The results showed that the structures with conventional dowel-type type connections exhibit large unrecoverable deformation after timber framed structures experience an earthquake. In comparison, those with the connections developed in this project show limited unrecoverable deformation due to the self-centering capacity of the connections

    A practical approach to achieving agility : a theory of constraints perspective

    This article documents an action research (AR) project aimed at identifying the practical steps needed to become an agile manufacturer through a combination of the theory of constraints (TOC) and resource- based view (RBV) approaches in a small to medium enterprise (SME) in the Australian manufacturing sector. To date, lean production has been highlighted as a possible catalyst for creating an agile manufacturer, despite the evidence suggesting that lean manufacturing lacks the responsiveness and adaptability to effectively handle a rapidly changing market place and only works well in a stable environment. A more flexible system of production is required to fully encompass the agile characteristics needed to attain a competitive advantage. This research provides empirical evidence that the TOC perspective can be used as a practical approach for becoming an agile manufacturer. The study provides a workable approach for small firms to achieve ‘Agility’ in practice

    SPATIAL DISTRIBUTION OF ATMOSPHERIC POLLUTION IN DUHOK URBAN AREA BY USING GIS TOOLS

    Air pollution is becoming a serious challenge in thickly populated areas in the world. The air pollution in Kurdistan region is clearly noticed, especially in areas where pollution sources and human population are concentrated. Urbanization and economic growth are proceeding at a rapid pace, accompanied by increasing emissions of a mixture of greenhouse gases especially from transporting sector, that positively contribute to accelerate climate change around the globe, and also have impacts on public health and vegetation. In order to decrease the effects that caused by atmospheric pollution, suitable monitoring systems are urgently needed that can rapidly and reliably detect and quantify polluting sources and concentration for monitoring by local authorities in order to restrain more damage of the current pollution levels. In this study, GIS with portable gas detector (K-60 IV) KELISAIKE safety equipment, China, have been used to assess the status of NOx, VOCs and noise pollution at 54 randomly selected urban locations of Duhok city. The measurements were taken during the week and weekend days started from 9 am until 4 pm. The results showed that there is a variation in the values of the three variables NOx, VOCs and noise, where the highest values recorded during working days in the locations distributed along the highway and the city center with heavy traffic load and dense human population in comparison to lowest values obtained during weekend days in locations distributed outskirt of the city with lower population and traffic loads. Therefore, likely such connections exist between the urban traffic density and  low air pollution quality within urban locations around Duhok city

    PIP5KIβ Selectively Modulates Apical Endocytosis in Polarized Renal Epithelial Cells

    Localized synthesis of phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] at clathrin coated pits (CCPs) is crucial for the recruitment of adaptors and other components of the internalization machinery, as well as for regulating actin dynamics during endocytosis. PtdIns(4,5)P2 is synthesized from phosphatidylinositol 4-phosphate by any of three phosphatidylinositol 5-kinase type I (PIP5KI) isoforms (α, β or γ). PIP5KIβ localizes almost exclusively to the apical surface in polarized mouse cortical collecting duct cells, whereas the other isoforms have a less polarized membrane distribution. We therefore investigated the role of PIP5KI isoforms in endocytosis at the apical and basolateral domains. Endocytosis at the apical surface is known to occur more slowly than at the basolateral surface. Apical endocytosis was selectively stimulated by overexpression of PIP5KIβ whereas the other isoforms had no effect on either apical or basolateral internalization. We found no difference in the affinity for PtdIns(4,5)P2-containing liposomes of the PtdIns(4,5)P2 binding domains of epsin and Dab2, consistent with a generic effect of elevated PtdIns(4,5)P2 on apical endocytosis. Additionally, using apical total internal reflection fluorescence imaging and electron microscopy we found that cells overexpressing PIP5KIβ have fewer apical CCPs but more internalized coated structures than control cells, consistent with enhanced maturation of apical CCPs. Together, our results suggest that synthesis of PtdIns(4,5)P2 mediated by PIP5KIβ is rate limiting for apical but not basolateral endocytosis in polarized kidney cells. PtdIns(4,5)P2 may be required to overcome specific structural constraints that limit the efficiency of apical endocytosis. © 2013 Szalinski et al

    New structural analogues of curcumin exhibit potent growth suppressive activity in human colorectal carcinoma cells

    <p>Abstract</p> <p>Background</p> <p>Colorectal carcinoma is one of the major causes of morbidity and mortality in the Western World. Novel therapeutic approaches are needed for colorectal carcinoma. Curcumin, the active component and yellow pigment of turmeric, has been reported to have several anti-cancer activities including anti-proliferation, anti-invasion, and anti-angiogenesis. Clinical trials have suggested that curcumin may serve as a potential preventive or therapeutic agent for colorectal cancer.</p> <p>Methods</p> <p>We compared the inhibitory effects of curcumin and novel structural analogues, GO-Y030, FLLL-11, and FLLL-12, in three independent human colorectal cancer cell lines, SW480, HT-29, and HCT116. MTT cell viability assay was used to examine the cell viability/proliferation and western blots were used to determine the level of PARP cleavages. Half-Maximal inhibitory concentrations (IC<sub>50</sub>) were calculated using Sigma Plot 9.0 software.</p> <p>Results</p> <p>Curcumin inhibited cell viability in all three of the human colorectal cancer cell lines studied with IC<sub>50 </sub>values ranging between 10.26 μM and 13.31 μM. GO-Y030, FLLL-11, and FLLL-12 were more potent than curcumin in the inhibition of cell viability in these three human colorectal cancer cell lines with IC<sub>50 </sub>values ranging between 0.51 μM and 4.48 μM. In addition, FLLL-11 and FLLL-12 exhibit low toxicity to WI-38 normal human lung fibroblasts with an IC-50 value greater than 1,000 μM. GO-Y030, FLLL-11, and FLLL-12 are also more potent than curcumin in the induction of apoptosis, as evidenced by cleaved PARP and cleaved caspase-3 in all three human colorectal cancer cell lines studied.</p> <p>Conclusion</p> <p>The results indicate that the three curcumin analogues studied exhibit more potent inhibitory activity than curcumin in human colorectal cancer cells. Thus, they may have translational potential as chemopreventive or therapeutic agents for colorectal carcinoma.</p

    Spatial heterogeneity of habitat suitability for Rift Valley fever occurrence in Tanzania: an ecological niche modelling approach

    Despite the long history of Rift Valley fever (RVF) in Tanzania, extent of its suitable habitat in the country remains unclear. In this study we investigated potential effects of temperature, precipitation, elevation, soil type, livestock density, rainfall pattern, proximity to wild animals, protected areas and forest on the habitat suitability for RVF occurrence in Tanzania. Presence-only records of 193 RVF outbreak locations from 1930 to 2007 together with potential predictor variables were used to model and map the suitable habitats for RVF occurrence using ecological niche modelling. Ground-truthing of the model outputs was conducted by comparing the levels of RVF virus specific antibodies in cattle, sheep and goats sampled from locations in Tanzania that presented different predicted habitat suitability values. Habitat suitability values for RVF occurrence were higher in the northern and central-eastern regions of Tanzania than the rest of the regions in the country. Soil type and precipitation of the wettest quarter contributed equally to habitat suitability (32.4% each), followed by livestock density (25.9%) and rainfall pattern (9.3%). Ground-truthing of model outputs revealed that the odds of an animal being seropositive for RVFV when sampled from areas predicted to be most suitable for RVF occurrence were twice the odds of an animal sampled from areas least suitable for RVF occurrence (95% CI: 1.43, 2.76, p < 0.001). The regions in the northern and central-eastern Tanzania were more suitable for RVF occurrence than the rest of the regions in the country. The modelled suitable habitat is characterised by impermeable soils, moderate precipitation in the wettest quarter, high livestock density and a bimodal rainfall pattern. The findings of this study should provide guidance for the design of appropriate RVF surveillance, prevention and control strategies which target areas with these characteristics
