    Model selection and prediction of outcomes in recent onset schizophrenia patients who undergo cognitive training.

    Predicting treatment outcomes in psychiatric populations remains a challenge, but is increasingly important in the pursuit of personalized medicine. Patients with schizophrenia have deficits in cognition, and targeted cognitive training (TCT) of auditory processing and working memory has been shown to improve some of these impairments; but little is known about the baseline patient characteristics predictive of cognitive improvement. Here we use a model selection and regression approach called least absolute shrinkage and selection operator (LASSO) to examine predictors of cognitive improvement in response to TCT for patients with recent onset schizophrenia. Forty-three individuals with recent onset schizophrenia randomized to undergo TCT were assessed at baseline on measures of cognition, symptoms, functioning, illness duration, and demographic variables. We carried out 10-fold cross-validation of LASSO for model selection and regression. We followed up on these results using linear models for statistical inference. No individual variable was found to correlate with improvement in global cognition using a Pearson correlation approach, and a linear model including all variables was also found not to be significant. However, the LASSO model identified baseline global cognition, education, and gender in a model predictive of improvement on global cognition following TCT. These findings offer guidelines for personalized approaches to cognitive training for patients with schizophrenia

    Cell-extracellular matrix interactions regulate neural differentiation of human embryonic stem cells

    <p>Abstract</p> <p>Background</p> <p>Interactions of cells with the extracellular matrix (ECM) are critical for the establishment and maintenance of stem cell self-renewal and differentiation. However, the ECM is a complex mixture of matrix molecules; little is known about the role of ECM components in human embryonic stem cell (hESC) differentiation into neural progenitors and neurons.</p> <p>Results</p> <p>A reproducible protocol was used to generate highly homogenous neural progenitors or a mixed population of neural progenitors and neurons from hESCs. This defined adherent culture system allowed us to examine the effect of ECM molecules on neural differentiation of hESCs. hESC-derived differentiating embryoid bodies were plated on Poly-D-Lysine (PDL), PDL/fibronectin, PDL/laminin, type I collagen and Matrigel, and cultured in neural differentiation medium. We found that the five substrates instructed neural progenitors followed by neuronal differentiation to differing degrees. Glia did not appear until 4 weeks later. Neural progenitor and neuronal generation and neurite outgrowth were significantly greater on laminin and laminin-rich Matrigel substrates than on other 3 substrates. Laminin stimulated hESC-derived neural progenitor expansion and neurite outgrowth in a dose-dependent manner. The laminin-induced neural progenitor expansion was partially blocked by the antibody against integrin α6 or β1 subunit.</p> <p>Conclusion</p> <p>We defined laminin as a key ECM molecule to enhance neural progenitor generation, expansion and differentiation into neurons from hESCs. The cell-laminin interactions involve α6β1 integrin receptors implicating a possible role of laminin/α6β1 integrin signaling in directed neural differentiation of hESCs. Since laminin acts in concert with other ECM molecules <it>in vivo</it>, evaluating cellular responses to the composition of the ECM is essential to clarify further the role of cell-matrix interactions in neural derivation of hESCs.</p

    The Role of Ki-67 Immunoexpression in Diagnosis of Molar Pregnancy and Differentiating its Subtypes

    الهدف:-&nbsp; لدراسة دور التعبير المناعي ل Ki-67 &nbsp;في تشخيص الحمل العنقودي وتمييزها عن الانواع الأخرى لأنواع اجهاض الحمل. العمليات:- ثمان وستون نموذج لنواتج الحمل المثبتة بالفورمالين والمطمورة بالفورمالين ، والتي شملت أجهاض الحمل في الأشهر الأولى (15 نموذج)، حمل عنقودي جزئي ( 24 نموذج)، حمل عنقودي كامل ( 24 نموذج)&nbsp; ومشيمة الحمل الكامل&nbsp; (5 نموذج). جميع النماذج تم جمعها من مختبرات فحص الأنسجة&nbsp; في مستشفى الولادة في اربيل / العراق خلال الفترة من ايلول 2012- ايلول 2013. تم استخدام التصبيغ المناعي الهيستوكيمياوي ل Ki-67 وباستعمال المضاد الأحادي MIB-1 وباستخدام الطريقة المعتادة . المؤشر التعريفي لKi-67 (عدد الأنوية المصبوغة/عدد الأنوية الكلي) لكل من الزغابات المشيمة&nbsp; والجذعة الغذائية الخلوية والجذعة الغذائية المخلاوية والخلايا السدوية تم حسابها بصورة منفصلة. الحسابات الأحصائية تم اجراءها بأستخدام مؤشر Fisher&nbsp; واذا كانت قيمة الp أقل من 0.05 تم اعتباره ذو قيمة أحصائية. النتائج:-&nbsp; أظهرت الدراسة أن مؤشر ال Ki-67 في كل امراض الزغابات المشيمية كان عالي وبصورة خاصة في الزغابات المشيمية للحمل العنقودي الكامل&nbsp; (&gt;50%&nbsp; ) ثم في الحمل العنقودي الجزئي (&gt;20%). كذلك تم ايجاد علاقة ذو قيمة احصائية&nbsp; للتعبير المناعي لل Ki-67 &nbsp;والتي كانت مفيدة في التمييز بين الأجهاض والحمل العنقودي الكامل ( p&lt;0.01) والحمل العنقودي الجزئي ( p&lt;0.05). الأستنتاج:- المؤشر التعريفي ل Ki-67&nbsp; في الزغابات المشيمية كان ذو قيمة في تشخيص وتمييز الحمل العنقودي من الأجهاض في الأشهر الأولى للحمل وكذلك في تمييز الحمل العنقودي الكامل من الحمل العنقودي الجزئي .Objectives: The study is intended to evaluate the role of Ki-67 immunoexpression in the diagnosis of molar pregnancy &amp; differential diagnosis of its subgroups from miscarriage. Methods: Sixty eight formalin-fixed, paraffin- embedded specimens of products of conception , including 1st trimester miscarriage (n=15), partial hydatidiform mole PHM (n=24), complete hydatidiform CHM (n=24) and full term placenta (n=5), all were examined at Histopathology Department of Maternity Teaching Hospital in Erbil, Iraq during the period Sep.2012-Sep.2013. Ki-67 immunohistochemical staining was performed by using the monoclonal antibody MIB-1 and the standard streptavidin-biotin immunoperoxidase method. The labeling index (number of positive nuclei/total number of nuclei) for villous, cytotrophoblasts , syncytiotrophoblasts and stromal cells were evaluated separately. Statistical analysis was carried out by Fisher’s exact test &amp; statistical significance was determined at p &lt; 0.05. Results: The study shows that all villous trophoblastic lesions showed high Ki-67 in all villous components especially cytotrophoblasts, being the highest in CHM mole(&gt;50%) followed by PHM (&gt;20%). Also found a statistically significant differences in immunoexpressins of Ki-67 that was useful in separating miscarriage from CHM, p&lt;0.01 (highly significant), and partial hydatidiform mole p&lt;0.05, (significant). Conclusion: Ki-67 labeling index of villous cells ,especially cytotrophoblasts, is valuable in diagnosis and differentiation of hydatidiform mole from 1st trimester miscarriage as well as between different subgroups of hydatidiformmoles (CHM &amp; PHM)

    Drosophila tan Encodes a Novel Hydrolase Required in Pigmentation and Vision

    Many proteins are used repeatedly in development, but usually the function of the protein is similar in the different contexts. Here we report that the classical Drosophila melanogaster locus tan encodes a novel enzyme required for two very different cellular functions: hydrolysis of N-β-alanyl dopamine (NBAD) to dopamine during cuticular melanization, and hydrolysis of carcinine to histamine in the metabolism of photoreceptor neurotransmitter. We characterized two tan-like P-element insertions that failed to complement classical tan mutations. Both are inserted in the 5′ untranslated region of the previously uncharacterized gene CG12120, a putative homolog of fungal isopenicillin-N N-acyltransferase (EC 2.3.1.164). Both P insertions showed abnormally low transcription of the CG12120 mRNA. Ectopic CG12120 expression rescued tan mutant pigmentation phenotypes and caused the production of striking black melanin patterns. Electroretinogram and head histamine assays indicated that CG12120 is required for hydrolysis of carcinine to histamine, which is required for histaminergic neurotransmission. Recombinant CG12120 protein efficiently hydrolyzed both NBAD to dopamine and carcinine to histamine. We conclude that D. melanogaster CG12120 corresponds to tan. This is, to our knowledge, the first molecular genetic characterization of NBAD hydrolase and carcinine hydrolase activity in any organism and is central to the understanding of pigmentation and photoreceptor function

    Chinese, spoken by approximately 20% of the world’s population, is the most commonly spoken language in the world (Fung, 1990). There is some debate regarding whether varieties of spoken Chinese should be considered dialects or separate language. Here, we regard them as separate languages. The various languages of Chinese are considered united by a common written system, although this characterization is problematic for Cantonese, which has many colloquial expressions with no written form (Bauer & Benedict, 1997). In this chapter, we focus on two of the most common Chinese languages, Cantonese and Mandarin (Putonghua). Cantonese is spoken in Hong Kong, Southern China and many overseas Chinese communities. Mandarin, also known as Standard Chinese, is the official national language of China and Taiwan, and is one of the four official languages of Singapore.caslpub3214pu

    Ultralow phase noise microwave generation with an Er:fiber-based optical frequency divider

    We present an optical frequency divider based on a 200 MHz repetition rate Er:fiber mode-locked laser that, when locked to a stable optical frequency reference, generates microwave signals with absolute phase noise that is equal to or better than cryogenic microwave oscillators. At 1 Hz offset from a 10 GHz carrier, the phase noise is below -100 dBc/Hz, limited by the optical reference. For offset frequencies > 10 kHz, the phase noise is shot noise limited at -145 dBc/Hz. An analysis of the contribution of the residual noise from the Er:fiber optical frequency divider is also presented.Comment: 4 pages, 3 figure

    On faces of the Kunz cone and the numerical semigroups within them

    A numerical semigroup is a cofinite subset of the non-negative integers that is closed under addition and contains 0. Each numerical semigroup SS with fixed smallest positive element mm corresponds to an integer point in a rational polyhedral cone Cm\mathcal C_m, called the Kunz cone. Moreover, numerical semigroups corresponding to points in the same face FCmF \subseteq \mathcal C_m are known to share many properties, such as the number of minimal generators. In this work, we classify which faces of Cm\mathcal C_m contain points corresponding to numerical semigroups. Additionally, we obtain sharp bounds on the number of minimal generators of SS in terms of the dimension of the face of Cm\mathcal C_m containing the point corresponding to SS
