    High level expression of human basic fibroblast growth factor in Escherichia coli: Evaluating the effect of the GC content and rare codons within the first 13 codons

    High-level expression of recombinant human basic fibroblast growth factor in Escherichia coli presents research opportunities such as analysis of hbFGF expression after translation initiation region (TIR) mutagenesis. In our study, hbfgf-cDNA was expressed in three stains of E. coli comprising OrigamiB (DE3), BL21 (DE3) and modified strain carrying copies for rare codon tRNAs (BL21 (DE3)-codonplus- RP). During the course of these experiments, we investigated the role of rare codon replacement and ofGC content reduction in N-terminal, just downstream of the ATG start codon. As standardized procedure, two forward primers were designed for modification of N-terminal of hbfgf-cDNA. Nterminally modified genes were PCR amplified and cloned into the expression vector, pET-22b.Meanwhile, wild-type gene remarkably expressed in all the strains especially in codon plus strain, rare codon substituted hbFGF gene construct surprisingly displayed undetectable levels of protein production; modified gene construct with reduction in GC content of the first 13 codons contributes to 2.5 folds increased expression level. In addition, recombinant hbFGF were purified and the biological activity of the recombinant growth factor was demonstrated by its ability to stimulate proliferation of NIH/3T3 cells. Purified rhbFGF exhibited proliferative activity comparable to commercial rhbFGF

    Coordinated regulation of the ESCRT-III component CHMP4C by the chromosomal passenger complex and centralspindlin during cytokinesis

    The chromosomal passenger complex (CPC)-composed of Aurora B kinase, Borealin, Survivin and INCENP-surveys the fidelity of genome segregation throughout cell division. The CPC has been proposed to prevent polyploidy by controlling the final separation (known as abscission) of the two daughter cells via regulation of the ESCRT-III CHMP4C component. The molecular details are, however, still unclear. Using atomic force microscopy, we show that CHMP4C binds to and remodels membranes in vitro\textit{in vitro}. Borealin prevents the association of CHMP4C with membranes, whereas Aurora B interferes with CHMP4C's membrane remodelling activity. Moreover, we show that CHMP4C phosphorylation is not required for its assembly into spiral filaments at the abscission site and that two distinctly localized pools of phosphorylated CHMP4C exist during cytokinesis. We also characterized the CHMP4C interactome in telophase cells and show that the centralspindlin complex associates preferentially with unphosphorylated CHMP4C in cytokinesis. Our findings indicate that gradual dephosphorylation of CHMP4C triggers a 'relay' mechanism between the CPC and centralspindlin that regulates the timely distribution and activation of CHMP4C for the execution of abscission.This work was supported by Cancer Research UK grant no.     ChromatoShiny: an interactive R/Shiny App for plotting chromatography profiles [version 2; peer review: 2 approved]

    BackgroundUnicorn™ software on Äkta liquid chromatography instruments outputs chromatography profiles of purified biological macromolecules. While the plots generated by the instrument software are very helpful to inspect basic chromatogram properties, they lack a range of useful annotation, customization and export options.MethodsWe use the R Shiny framework to build an interactive app that facilitates the interpretation of chromatograms and the generation of figures for publications.ResultsThe app allows users to fit a baseline, to highlight selected fractions and elution volumes inside or under the plot (e.g. those used for downstream biochemical/biophysical/structural analysis) and to zoom into the plot. The app is freely available at https://ChromatoShiny.bio.ed.ac.uk.ConclusionsIt requires no programming experience, so we anticipate that it will enable chromatography users to create informative, annotated chromatogram plots quickly and simply.FPLC instruments used to purify macromolecules output the UV intensity values over the elution volume. However, the software used with these instruments is not usually used to generate the figures for publication. To facilitate the analysis of chromatograms and generation of publication figures, we developed a web app which is possible to use without programming skills. The app is working on Äkta .txt files and is able to fit a baseline, to highlight fractions on and under the plot and to zoom into the plot. The app is designed for Äkta instruments, but the data from other softwares can be entered in the provided template and plotted accordingly. The app is well suited for plotting many similar plots. The plots can be downloaded in various formats. The app is equipped with instructions and has a user friendly interface. We hope that the app will become a helpful tool for displaying chromatograms from various FPLC softwares

    EFFECTS OF THE PRODUCTS AND SERVICES OF MICROFINANCE INSTITUTIONS ON THE ECONOMIC DEVELOPMENT OF THEIR MEMBERS IN VINTAR, ILOCOS NORTE, PHILIPPINES

    This study determined the effects of the products and services of Microfinance Institutions on the economic development of their members in Vintar, Ilocos Norte, Philippines.           The descriptive correlational research design was adopted using a survey questionnaire to gather data. A total of 603 respondents were involved. They were taken from the members of two Microfinance Institutions (CARD Bank Inc. and ASA Philippines Foundation Inc.,) in Vintar, Ilocos Norte, Philippines who have been a member for at least two years. The study was delimited into the top three barangays with the most members which are composed of Barangays Parut, Alejo Malasig, and Pedro F. Alviar. Frequency, percentage, weighted mean, Chi-square test, and Pearson r of correlation were used to analyze and interpret the data gathered.           The following were the highlights of the findings of the study. The MFIs are giving their members quality products offered on loan applications and loan features as manifested by a very satisfied and satisfied rating of their members, respectively, as it is significantly and positively related to their economic development. The MFI members’ economic development in terms of financial, personal, and business status are significantly and highly related to their level of satisfaction with the customer services offered by the MFIs. The MFI’s services offered have met the satisfaction level of their members as expressed by a rating of very satisfied and satisfied on customer service and non-financial services, respectively. It was recommended that: On the part of Microfinance Institutions, they should consider the financial status of their members in terms of the reasonable service fee, and allow repayment and reasonable penalties for nonpayment/default to lessen their burden on their expenses, microfinance institutions should also conduct community development projects in different municipalities to increase their members' economic development, and microfinance institutions should provide business training to improve or expand their members’ business strategies.  On the part of the members, they should pay their payments on time and actively participate in the activities of the MFIs. On the part of future researchers, they may also conduct studies in other places that offer products and services of Microfinance Institutions, they may also consider other variables, such as their incentives and benefits

    Generational intelligence tests score changes in Spain: are we asking the right question?

    Generational intelligence test score gains have been documented worldwide in the twentieth century. However, recent evidence suggests these increased scores are coming to an end in some world regions. Here we compare two cohorts of university freshmen. The first cohort (n = 311) was assessed in 1991, whereas the second cohort (n = 349) was assessed thirty years later (2022). These cohorts completed the same intelligence battery including eight standardized speeded and power tests tapping reasoning (abstract and quantitative), language (vocabulary, verbal comprehension, and verbal meanings), rote calculation, and visuospatial relations. The results revealed a global gain of 3.5 IQ points but also upward and downward changes at the test level. The 2022 cohort outperformed the 1991 cohort on reasoning (abstract and quantitative), verbal comprehension, and vocabulary, whereas the 1991 cohort outscored the 2022 cohort on rote calculation, visuospatial relations (mental rotation and identical figures), and verbal meanings. These findings are thought to support one key claim made by James Flynn: generational changes on the specific cognitive abilities and skills tapped by standardized tests should be expected without appreciable or substantive changes in the structure of the intelligence construct identified within generations. This main conclusion is discussed with respect to theoretical causal implications putatively derived from current intelligence psychometric model

    Informational literacy and the use of technologies by Secondary Education students in Spain: A descriptive study. The development of Information and Communication Technologies (ICTs), together with their application to research carried out on educational areas, are factors which contribute to the promotion of a new educative model constructed on literacy-based competences and skills, and which sets technologies as essential tools for a life-long learning process (Unesco, 2005). This is the framework where we can insert the research that we are currently carrying out, funded by the research Program I+D+I of the Spanish Ministry of Education, and in the frame of which we are developing a diagnostic assessment of informational literacy competence in students between 14 and 16 years (Secondary Education), based on the fact that one of the main aspects in knowledge generation and acquisition is the capacity to use information extracted from documents and electronic resources, available in informational networks in a correct way. In this paper we present the results obtained from evidences on the contrast existing between the level of use of technologies (videogames, social networks...) and the level of informational literacy shown by students. The data were gathered from a sample of more than 1000 Secondary Education students who are around 15 years old

    Intraday rallies and crashes : spillovers of trading halts

    This paper analyses a set of intraday rally and crash events at the firm level during the single stock circuit breaker (SSCB) program, and documents the cross-sectional spillover effects of such events on non-halted stocks. We test whether such major price jumps, and subsequent trading halts, affect related stocks through the destabilizing arbitrage channel. We find that extreme price movements that trigger the circuit breakers at the firm level are accompanied by a massive surge in volume, spread and short-term volatility, which gradually revert back to normal. Speculative strategies of arbitrageurs such as momentum and pairs trading cause cross-sectional spillovers in volume and volatility during the trading halt

    Psychometric Evaluation of a New Instrument in Spanish to Measure Self-Care Requisites in Patients with Schizophrenia

    PURPOSE: The purpose of this study was to develop and validate a new instrument in Spanish designed to measure self-care requisites in patients with schizophrenia treated in the community. DESIGN AND METHODS: The first phase was conducted to develop the questionnaire through a panel of experts and evaluate for content validity. Psychometric evaluation was then conducted with a consecutive sample of 341 patients. FINDINGS: The scale demonstrated good internal consistency and stability over time. The discriminant and convergent validity was satisfactory. The confirmatory factor analysis showed that the theoretical model fits the self-care requisites proposed by Orem's nursing theory from which it originated. PRACTICE IMPLICATIONS: This scale is a valid and reliable instrument for use in clinical practice, guiding the nurse in developing the most appropriate care plan for each patient

    Wireless magneto-ionics: voltage control of magnetism by bipolar electrochemistry

    Modulation of magnetic properties through voltage-driven ion motion and redox processes, i.e., magneto-ionics, is a unique approach to control magnetism with electric field for low-power memory and spintronic applications. So far, magneto-ionics has been achieved through direct electrical connections to the actuated material. Here we evidence that an alternative way to reach such control exists in a wireless manner. Induced polarization in the conducting material immersed in the electrolyte, without direct wire contact, promotes wireless bipolar electrochemistry, an alternative pathway to achieve voltage-driven control of magnetism based on the same electrochemical processes involved in direct-contact magneto-ionics. A significant tunability of magnetization is accomplished for cobalt nitride thin films, including transitions between paramagnetic and ferromagnetic states. Such effects can be either volatile or non-volatile depending on the electrochemical cell configuration. These results represent a fundamental breakthrough that may inspire future device designs for applications in bioelectronics, catalysis, neuromorphic computing, or wireless communications.Comment: 32 pages, 4 figures, Supplementary Information (9 figures
