    Non-intrusive measurements of wave-induced flow over dikes by means of a combined ultrasound doppler velocimetry and videography

    The performance of non-intrusive instruments, such as acoustic profilers and cameras, to describe the wave-induced flow processes over maritime dike crest was investigated in experiments carried out at the University of Bologna. Direct and derived measurements from the acoustic probes deployed along the structure crest were discussed in relation to the observed backscatter rates. Image processing was implemented by means of clustering algorithm, in order to detect the free surface during overtopping events and characterize wave front propagation over the dike crest. UVP data were processed to indirectly derive flow depths and overtopping rates and compare them with the direct measurements in order to assess the measurement reliability and discuss their limits. Individual overtopping volume distribution as obtained by UVP data were estimated and compared with well-consolidated formulations, showing a good agreement. Finally, suggestions for an appropriate use of non-intrusive instruments to characterize a shallow, transient and aerated flow were provided, such as the control of the artificial seeding density, the use of a bi-static UVP configuration and adjustments to light exposure

    Essential oils in the diet of young bulls: Effect on animal temperament.

    Plant extracts and essential oils can be alternative products to antibiotics, because several plants produce secondary metabolites with antimicrobial properties. Additionally, they may act on the olfactory system, which sends signals to the central nervous system releasing endorphins that may affect the feeling of an animal''s welfare while altering an animal''s temperament. This study was conducted to evaluate the animal temperament of 40 young crossbred bulls (one-half Brown Swiss and one-half Nellore) 10 ± 2.2 mo old with an average BW of 219 ± 11.7 kg. Young bulls were randomly assigned to 1 of 5 diets: control, which had no clove or cinnamon; clove leaf included to supply 3, 500 mg/animal per day; clove leaf included to supply 7, 000 mg/animal per day; essential oil of leaf cinnamon to supply 3, 500 mg/animal per day; or essential oil of leaf cinnamon to supply 7, 000 mg/animal per day. The animal temperament was evaluated by calculating the chute score, exit score, and temperament score for periods (each 28 d during 6 periods) and among diets. The data were submitted to an ANOVA using GLM procedures with SAS version 9.0. The diet and period were considered fixed effects, whereas the animals were considered a random effect. Differences between means were evaluated using a Tukey test of 5% of significance. The addition of clove or cinnamon essential oils did not alter (P > 0.05) animal temperament. The analysis of temperament score (P = 0.55), chute score (P = 0.71), and exit score (P = 0.06) did not show significant differences among the diets. The exit score was similar among the 6 periods (1.57, 1.80, 2.98, 2.02, 2.73, and 2.66, respectively); however, chute score and temperament score were greater (P < 0.001) in the first period compared with other periods. The values for chute score were 1.85, 1.40, 1.35, 1.33, 1.20, 1.17, respectively, and the values for temperament score were 2.31, 1.81, 1.67, 1.69, 1.50, and 1.56, respectively, for periods 1 to 6. The results of this study suggest that clove and cinnamon essential oils can be added as an additive in high-grain finished diets without changing the animal temperament

    Scalar meson dynamics in Chiral Perturbation Theory

    A comparison of the linear sigma model (Lσ\sigmaM) and Chiral Perturbation Theory (ChPT) predictions for pion and kaon dynamics is presented. Lowest and next-to-leading order terms in the ChPT amplitudes are reproduced if one restricts to scalar resonance exchange. Some low energy constants of the order p4p^4 ChPT Lagrangian are fixed in terms of scalar meson masses. Present values of these low energy constants are compatible with the Lσ\sigmaM dynamics. We conclude that more accurate values would be most useful either to falsify the Lσ\sigmaM or to show its capability to shed some light on the controversial scalar physics.Comment: 9 pages, REVTeX 4.0.     No Brasil, pouca atenção se tem dado às formas e caminhos com que os fluxos energéticos se distribuem nos sistemas produtivos. Na agropecuária, a atenção tem sido voltada a novas fontes de energia (biomassa) ou em tecnologia alternativa, visando a racionalização do uso de energia fóssil ou elétrica. Entretanto, a agricultura tem se desenvolvido baseada fortemente na utilização intensiva de máquinas agrícolas, com conseqüente uso de combustíveis fósseis. Um fator de estrangulamento muito forte no consumo energético geral tem sido a utilização massiva de fertilizantes derivados do petróleo nos agroecossistemas. Estudos de Balanços Energéticos visam determinar os pontos de estrangulamento energético fundamentando a busca por tecnologias poupadoras de energia, especialmente aquelas de origem fóssil (combustível, fertilizante, agrotóxicos, energia despendida na fabricação das máquinas e implementos, etc.). No Brasil, a Região Sul, é onde se encontram vários trabalhos buscando uma agricultura mais auto-sustentável, do ponto de vista da utilização da energia. Em vista da possibilidade de eventuais futuras crises energéticas, o presente trabalho procura analisar o estado-da-arte dos estudos em Balanço Energético, no Brasil e no Mundo, como uma ferramenta de indicação da sustentabilidade dos sistemas agropecuários

    Genome-wide association analysis of susceptibility and clinical phenotype in multiple sclerosis

    Multiple sclerosis (MS), a chronic disorder of the central nervous system and common cause of neurological disability in young adults, is characterized by moderate but complex risk heritability. Here we report the results of a genome-wide association study performed in a 1000 prospective case series of well-characterized individuals with MS and group-matched controls using the Sentrix® HumanHap550 BeadChip platform from Illumina. After stringent quality control data filtering, we compared allele frequencies for 551 642 SNPs in 978 cases and 883 controls and assessed genotypic influences on susceptibility, age of onset, disease severity, as well as brain lesion load and normalized brain volume from magnetic resonance imaging exams. A multi-analytical strategy identified 242 susceptibility SNPs exceeding established thresholds of significance, including 65 within the MHC locus in chromosome 6p21.3. Independent replication confirms a role for GPC5, a heparan sulfate proteoglycan, in disease risk.     Inherited cutis laxa, or inelastic, sagging skin is a genetic condition of premature and generalised connective tissue ageing, affecting various elastic components of the extracellular matrix. Several cutis laxa syndromes are inborn errors of metabolism and lead to severe neurological symptoms. In a patient with cutis laxa, a choreoathetoid movement disorder, dysmorphic features and intellectual disability we performed exome sequencing to elucidate the underlying genetic defect. We identified the amino acid substitution R275W in phosphatidylinositol 4-kinase type II alpha, caused by a homozygous missense mutation in thePI4K2Agene. We used lipidomics, complexome profiling and functional studies to measure phosphatidylinositol 4-phosphate synthesis in the patient and evaluated PI4K2A deficient mice to define a novel metabolic disorder. The R275W residue, located on the surface of the protein, is involved in forming electrostatic interactions with the membrane. The catalytic activity of PI4K2A in patient fibroblasts was severely reduced and lipid mass spectrometry showed that particular acyl-chain pools of PI4P and PI(4,5)P(2)were decreased. Phosphoinositide lipids play a major role in intracellular signalling and trafficking and regulate the balance between proliferation and apoptosis. Phosphatidylinositol 4-kinases such as PI4K2A mediate the first step in the main metabolic pathway that generates PI4P, PI(4,5)P(2)and PI(3,4,5)P-3. Although neurologic involvement is common, cutis laxa has not been reported previously in metabolic defects affecting signalling. Here we describe a patient with a complex neurological phenotype, premature ageing and a mutation inPI4K2A, illustrating the importance of this enzyme in the generation of inositol lipids with particular acylation characteristics.Medical Biochemistr

    Early life risk factors and their cumulative effects as predictors of overweight in Spanish children

    Objectives: To explore early life risk factors of overweight/obesity at age 6 years and their cumulative effects on overweight/obesity at ages 2, 4 and 6 years. Methods: Altogether 1031 Spanish children were evaluated at birth and during a 6-year follow-up. Early life risk factors included: parental overweight/obesity, parental origin/ethnicity, maternal smoking during pregnancy, gestational weight gain, gestational age, birth weight, caesarean section, breastfeeding practices and rapid infant weight gain collected via hospital records. Cumulative effects were assessed by adding up those early risk factors that significantly increased the risk of overweight/obesity. We conducted binary logistic regression models. Results: Rapid infant weight gain (OR 2.29, 99% CI 1.54–3.42), maternal overweight/obesity (OR 1.93, 99% CI 1.27–2.92), paternal overweight/obesity (OR 2.17, 99% CI 1.44–3.28), Latin American/Roma origin (OR 3.20, 99% CI 1.60–6.39) and smoking during pregnancy (OR 1.61, 99% CI 1.01–2.59) remained significant after adjusting for confounders. A higher number of early life risk factors accumulated was associated with overweight/obesity at age 6 years but not at age 2 and 4 years. Conclusions: Rapid infant weight gain, parental overweight/obesity, maternal smoking and origin/ethnicity predict childhood overweight/obesity and present cumulative effects. Monitoring children with rapid weight gain and supporting a healthy parental weight are important for childhood obesity prevention

    Prognostic factors of a lower CD4/CD8 ratio in long term viral suppression HIV infected children

    Background Combination antiretroviral therapy (cART) is associated with marked immune reconstitution. Although a long term viral suppression is achievable, not all children however, attain complete immunological recovery due to persistent immune activation. We use CD4/CD8 ratio like a marker of immune reconstitution. Methods Perinatal HIV-infected children who underwent a first-line cART, achieved viral suppression in the first year and maintained it for more than 5 years, with no viral rebound were included. Logistic models were applied to estimate the prognostic factors, clinical characteristics at cART start, of a lower CD4/CD8 ratio at the last visit. Results 146 HIV-infected children were included: 77% Caucasian, 45% male and 28% CDC C. Median age at cART initiation was 2.3 years (IQR: 0.5-6.2). 42 (30%) children received mono-dual therapy previously to cART. Time of undetectable viral load was 9.5 years (IQR: 7.8, 12.5). 33% of the children not achieved CD4/CD8 ratio >1. Univariate analysis showed an association between CD4/CD8 <1 with lower CD4 nadir and baseline CD4; older age at diagnosis and at cART initiation; and a previous exposure to mono-dual therapy. Multivariate analysis also revealed relationship between CD4/CD8 <1 and lower CD4 nadir (OR: 1.002, CI 95% 1.000-1.004) as well as previous exposure to mono-dual therapy (OR: 0.16, CI 95% 0.003-0.720). Conclusions CD4/CD8 > 1 was not achieved in 33% of the children. Lower CD4 nadir and previous exposure to suboptimal therapy, before initiating cART, are factors showing independently association with a worse immune recovery (CD4/CD8 < 1)
