    Molecular and biochemical alterations in tubular epithelial cells of patients with isolated methylmalonic aciduria

    Methylmalonic acidurias (MMAurias) are a group of inherited disorders in the catabolism of branched-chain amino acids, odd-chain fatty acids and cholesterol caused by complete or partial deficiency of methylmalonyl-CoA mutase (mut0 and mut- subtype respectively) and by defects in the metabolism of its cofactor 5′-deoxyadenosylcobalamin (cblA, cblB or cblD variant 2 type). A long-term complication found in patients with mut0 and cblB variant is chronic tubulointerstitial nephritis. The underlying pathomechanism has remained unknown. We established an in vitro model of tubular epithelial cells from patient urine (hTEC; 9 controls, 5 mut0, 1 cblB). In all human tubular epithelial cell (hTEC) lines we found specific tubular markers (AQP1, UMOD, AQP2). Patient cells showed disturbance of energy metabolism in glycolysis, mitochondrial respiratory chain and Krebs cycle in concert with increased reactive oxygen species (ROS) formation. Electron micrographs indicated increased autophagosome production and endoplasmic reticulum stress, which was supported by positive acridine orange staining and elevated levels of LC3 II, P62 and pIRE1. Screening mTOR signaling revealed a release of inhibition of autophagy. Patient hTEC produced and secreted elevated amounts of the pro-inflammatory cytokine IL8, which was highly correlated with the acridine orange staining.     Spike-based coupling between single neurons and populations across rat sensory cortices, perirhinal cortex, and hippocampus

    Cortical computations require coordination of neuronal activity within and across multiple areas. We characterized spiking relationships within and between areas by quantifying coupling of single neurons to population firing patterns. Single-neuron population coupling (SNPC) was investigated using ensemble recordings from hippocampal CA1 region and somatosensory, visual, and perirhinal cortices. Within-area coupling was heterogeneous across structures, with area CA1 showing higher levels than neocortical regions. In contrast to known anatomical connectivity, between-area coupling showed strong firing coherence of sensory neocortices with CA1, but less with perirhinal cortex. Cells in sensory neocortices and CA1 showed positive correlations between within- and between-area coupling; these were weaker for perirhinal cortex. All four areas harbored broadcasting cells, connecting to multiple external areas, which was uncorrelated to within-area coupling strength. When examining correlations between SNPC and spatial coding, we found that, if such correlations were significant, they were negative. This result was consistent with an overall preservation of SNPC across different brain states, suggesting a strong dependence on intrinsic network connectivity. Overall, SNPC offers an important window on cell-to-population synchronization in multi-area networks. Instead of pointing to specific information-coding functions, our results indicate a primary function of SNPC in dynamically organizing communication in systems composed of multiple, interconnected areas

    Constraints on neutrino-photon interactions from rare Z decays

    It is shown that the rare decays ZνnuˉγZ \to \nu \bar{nu} \gamma and ZννˉγγZ \to \nu \bar{\nu} \gamma \gamma are useful to put model-independent bounds on neutrino-one-photon and neutrino-two-photon interactions. The results are then used to constrain the τ\tau neutrino magnetic moment and the double radiative decay νjνiγγ\nu_j \to \nu_i \gamma \gamma. It is found that the decay ZνnuˉγZ \to \nu \bar{nu} \gamma gives a more stringent bound on the τ\tau neutrino magnetic moment than that obtained from ZννˉγγZ \to \nu \bar{\nu} \gamma \gamma; the latter decay in turn gives limits on the neutrino-two-photon interaction that are less stringent than those obtained for a sterile neutrino νs\nu_s from the analysis of νμNνsN\nu_\mu N \to \nu_s N conversion.Comment: 11 pages, 5 figures, elsart forma

    New hadrons as ultra-high energy cosmic rays

    Ultra-high energy cosmic ray (UHECR) protons produced by uniformly distributed astrophysical sources contradict the energy spectrum measured by both the AGASA and HiRes experiments, assuming the small scale clustering of UHECR observed by AGASA is caused by point-like sources. In that case, the small number of sources leads to a sharp exponential cutoff at the energy E<10^{20} eV in the UHECR spectrum. New hadrons with mass 1.5-3 GeV can solve this cutoff problem. For the first time we discuss the production of such hadrons in proton collisions with infrared/optical photons in astrophysical sources. This production mechanism, in contrast to proton-proton collisions, requires the acceleration of protons only to energies E<10^{21} eV. The diffuse gamma-ray and neutrino fluxes in this model obey all existing experimental limits. We predict large UHE neutrino fluxes well above the sensitivity of the next generation of high-energy neutrino experiments. As an example we study hadrons containing a light bottom squark.     Synaptic processes and immune-related pathways implicated in Tourette syndrome.

    Tourette syndrome (TS) is a neuropsychiatric disorder of complex genetic architecture involving multiple interacting genes. Here, we sought to elucidate the pathways that underlie the neurobiology of the disorder through genome-wide analysis. We analyzed genome-wide genotypic data of 3581 individuals with TS and 7682 ancestry-matched controls and investigated associations of TS with sets of genes that are expressed in particular cell types and operate in specific neuronal and glial functions. We employed a self-contained, set-based association method (SBA) as well as a competitive gene set method (MAGMA) using individual-level genotype data to perform a comprehensive investigation of the biological background of TS. Our SBA analysis identified three significant gene sets after Bonferroni correction, implicating ligand-gated ion channel signaling, lymphocytic, and cell adhesion and transsynaptic signaling processes. MAGMA analysis further supported the involvement of the cell adhesion and trans-synaptic signaling gene set. The lymphocytic gene set was driven by variants in FLT3, raising an intriguing hypothesis for the involvement of a neuroinflammatory element in TS pathogenesis. The indications of involvement of ligand-gated ion channel signaling reinforce the role of GABA in TS, while the association of cell adhesion and trans-synaptic signaling gene set provides additional support for the role of adhesion molecules in neuropsychiatric disorders. This study reinforces previous findings but also provides new insights into the neurobiology of TS

    Two-Loop O(alpha_s G_F M_Q^2) Heavy-Quark Corrections to the Interactions between Higgs and Intermediate Bosons

    By means of a low-energy theorem, we analyze at O(alpha_s G_F M_Q^2) the shifts in the Standard-Model W^+W^-H and ZZH couplings induced by virtual high-mass quarks, Q, with M_Q >> M_Z, M_H, which includes the top quark. Invoking the improved Born approximation, we then find the corresponding corrections to various four- and five-point Higgs-boson production and decay processes which involve the W^+W^-H and ZZH vertices with one or both of the gauge bosons being connected to light-fermion currents, respectively.     Motivation and satisfaction of volunteers for community-based urban agriculture programmes

    Urban agriculture means cultivating plants and raising livestock within cities for food and other uses. A Community‐based Urban Agriculture Programme is where people from residential areas get together as volunteers to practise urban agriculture in an empty space within residential areas. However, the programme encounters problems when it is incapable of attracting enough volunteers and retaining them in order to establish a sustainable programme. This study aims to determine the relationship between the dimensions of motivation and satisfaction of volunteers on the Community‐based Urban Agriculture Programme. Data collected from 375 volunteers on the Community‐based Urban Agriculture Programme in Klang Valley, Malaysia were analysed using descriptive analysis, reliability analysis, correlation analysis, and hierarchical multiple regression analysis. It was found that the most significant predictor of Community‐based Urban Agriculture Programme volunteers’ satisfaction was favoured by external factors such as campaigns, support groups, Department of Extension, and community as well as government policy, followed by love of farming, social referents, and values. Therefore, there should be a focus on the above‐mentioned dimensions of motivation in order to enhance the satisfaction of volunteers towards the Community‐based Urban Agriculture Programme
