    STUDY OBJECTIVES: The majority of studies investigating the association between sleep and Alzheimer's disease (AD) biomarkers have been performed in healthy participants. Our objective was to investigate the association between sleep and several biomarkers that reflect distinct aspects of AD physiopathology. METHODS: The cohort included 104 individuals with mild-moderate AD. The participants were submitted to one-night polysomnography, and cerebrospinal fluid was collected in the following morning to measure the selected biomarkers associated with amyloid deposition, tau pathology, neurodegeneration, axonal damage, synaptic integrity, neuroinflammation, and oxidative damage. RESULTS: There was a positive correlation between neurofilament light (NF-L) and the time spent in stage 1 of non-rapid eyes movement (NREM) (N1) sleep and a negative correlation between this marker and the time spent in stage 3 of NREM (N3) sleep. Accordingly, we observed that deep sleep was associated with lower levels of NF-L, whereas light sleep increased the probability of having higher levels of this marker. Furthermore, chitinase-3-like-1 (YKL-40) was negatively correlated with sleep efficiency, the time spent in stage 2 of NREM (N2) sleep, and the time spent in N3 sleep. Conversely, there was a positive correlation between N3 sleep and the oxidative protein damage markers N-ε-(carboxyethyl)lysine and N-ε-(malondialdehyde)lysine. CONCLUSIONS: There were significant correlations between sleep parameters and AD biomarkers related to axonal damage and neuroinflammation, such as NF-L and YKL-40. A lack of deep sleep was associated with higher levels of NF-L. This highlights a potential role for NF-L as a biomarker of sleep disruption in patients with mild-moderate AD in addition to its role in predicting neurodegeneration and cognitive decline

    Gain of DNA methylation is enhanced in the absence of CTCF at the human retinoblastoma gene promoter

    <p>Abstract</p> <p>Background</p> <p>Long-term gene silencing throughout cell division is generally achieved by DNA methylation and other epigenetic processes. Aberrant DNA methylation is now widely recognized to be associated with cancer and other human diseases. Here we addressed the contribution of the multifunctional nuclear factor CTCF to the epigenetic regulation of the human <it>retinoblastoma </it>(<it>Rb</it>) gene promoter in different tumoral cell lines.</p> <p>Methods</p> <p>To assess the DNA methylation status of the <it>Rb </it>promoter, genomic DNA from stably transfected human erythroleukemic K562 cells expressing a <it>GFP </it>reporter transgene was transformed with sodium bisulfite, and then PCR-amplified with modified primers and sequenced. Single- and multi-copy integrants with the CTCF binding site mutated were isolated and characterized by Southern blotting. Silenced transgenes were reactivated using 5-aza-2'-deoxycytidine and Trichostatin-A, and their expression was monitored by fluorescent cytometry. <it>Rb </it>gene expression and protein abundance were assessed by RT-PCR and Western blotting in three different glioma cell lines, and DNA methylation of the promoter region was determined by sodium bisulfite sequencing, together with CTCF dissociation and methyl-CpG-binding protein incorporation by chromatin immunoprecipitation assays.</p> <p>Results</p> <p>We found that the inability of CTCF to bind to the <it>Rb </it>promoter causes a dramatic loss of gene expression and a progressive gain of DNA methylation.</p> <p>Conclusions</p> <p>This study indicates that CTCF plays an important role in maintaining the <it>Rb </it>promoter in an optimal chromatin configuration. The absence of CTCF induces a rapid epigenetic silencing through a progressive gain of DNA methylation. Consequently, CTCF can now be seen as one of the epigenetic components that allows the proper configuration of tumor suppressor gene promoters. Its aberrant dissociation can then predispose key genes in cancer cells to acquire DNA methylation and epigenetic silencing.</p

    Geodetic Observations at the Eolian Islands (Southern Italy)

    In order to investigate the dynamic processes actually in progress in the Aeolian Islands a volcanic area in Southern Italy, high precision geodetic observations are employed. the knowledge of the kinematic field (vertical and horizontal displacements) and of the time-space gravity variations is an important constrain for any model describing the geodynamic processes involving the investigated area. Gravimetric (absolute, relative and gradiometric), photogrammetric, altimetric and GPS surveys are periodically carried out on networks spanning over the whole aeolian archipelagus. Recently a GPS network has been designed and surveyed to obtain a more complete knowledge of the kinematic field taking place in the area. This will improve the understanding of the tectonic processes acting in the area. The result of the interdisplinary approach are presented and discussed.Published33-501.3.     VEZF1 elements mediate protection from DNA methylation

    There is growing consensus that genome organization and long-range gene regulation involves partitioning of the genome into domains of distinct epigenetic chromatin states. Chromatin insulator or barrier elements are key components of these processes as they can establish boundaries between chromatin states. The ability of elements such as the paradigm &#946;-globin HS4 insulator to block the range of enhancers or the spread of repressive histone modifications is well established. Here we have addressed the hypothesis that a barrier element in vertebrates should be capable of defending a gene from silencing by DNA methylation. Using an established stable reporter gene system, we find that HS4 acts specifically to protect a gene promoter from de novo DNA methylation. Notably, protection from methylation can occur in the absence of histone acetylation or transcription. There is a division of labor at HS4; the sequences that mediate protection from methylation are separable from those that mediate CTCF-dependent enhancer blocking and USF-dependent histone modification recruitment. The zinc finger protein VEZF1 was purified as the factor that specifically interacts with the methylation protection elements. VEZF1 is a candidate CpG island protection factor as the G-rich sequences bound by VEZF1 are frequently found at CpG island promoters. Indeed, we show that VEZF1 elements are sufficient to mediate demethylation and protection of the APRT CpG island promoter from DNA methylation. We propose that many barrier elements in vertebrates will prevent DNA methylation in addition to blocking the propagation of repressive histone modifications, as either process is sufficient to direct the establishment of an epigenetically stable silent chromatin stat

En este sentido, IRTA ha realizado un estudio pionero en el marco del Crupo Operativo ‘Rumprint: huella ambiental de la carne y la leche de vacuno’. El objetivo era calcular la huella ambiental de estos productos, aplicando la metodologia recomendada por la Comision Europea de ‘Huella Ambiental de Producto’.     A new chronological framework and site formation history for Cova del Gegant (Barcelona): Implications for Neanderthal and Anatomically Modern Human occupation of NE Iberian Peninsula

    The chronological framework for Neanderthal occupation and demise across Europe continues to be debated. In particular, there is still uncertainty regarding the nature, timing and regional expressions of the Middle to Upper Palaeolithic transition associated with the disappearance of Neanderthals and the broader expansion of modern human populations in Europe around 42e40 thousand years ago (ka). The geographical and chronological distribution of Neanderthal populations also remains difficult to evaluate owing to the practical challenges of directly dating human fossils at many sites, and the fact that a large proportion of Neanderthals sites lie close to, or well-beyond, the limits of radiocarbon dating. Cova del Gegant e one of the few sites in north-eastern Iberian Peninsula to yield Neanderthal fossil remains, associated Mousterian archaeological layers, and occupations related to the Middle and Upper Palaeolithic transition e is a key locality for informing these ongoing debates. Here we provide a comprehensive chronological framework for the Cova del Gegant site using multiple radiometric dating techniques (uranium-thorium (UeTh), radiocarbon and luminescence dating), sedimentological and micromorphological analyses, and Bayesian modelling. This integrated chronostratigraphic approach enables us to reliably reconstruct site formation processes and history, and undertake improved correlations with other sites regionally. The results allow us to sub-divide the Cova del Gegant sequence into three sections spanning ~94 ka to ~32 ka, namely: a Middle Palaeolithic sequence covering ~94e59 ka, a Châtelperronian/Aurignacian section spanning ~43e39 ka, and a Late Aurignacian/Gravettian section spanning ~34e32 ka. The Neanderthal fossil remains accumulated in the cave between the end of Marine Isotope Stage (MIS) 5/MIS 4 and the beginning of MIS 3, during two different events dated to ~72e67 ka and ~60e52 ka. The chronological framework for Cova del Gegant is in accordance with that reported for other Middle and Upper Palaeolithic sites in north-eastern Iberian Peninsula, and reveals a record of successive human occupation coinciding with a period of progressive global cooling and lowering sea levels (end of MIS 5 through to MIS 2). Sedimentological evidence points to the emergence of a coastal platform in front of the cave and indicates that local palaeoenvironmental conditions likely benefited human displacements along the littoral margin, and favoured repeated occupation of the cave during the Late Pleistocene
